Description
Die vorliegende Erfindung betrifft neuartige Formulierungen des bekannten antimykotischen Bifonazols der Formel die eine Depot-Wirkung und eine höhere Bioverfügbarkeit des Wirkstoffes aufweisen und dadurch eine Kurzzeittherapie ermöglichen.
Für die Behandlung von Mykosen beim Menschen, vor allem der Mykosen der Haut sind bereits Zubereitungen von antimykotischen Derivaten bekannt geworden. Mit diesen Zubereitungen wurden für eine vollständige Sanierung >21 Tage Therapiezeit benötigt.
Um zu einer Verkürzung der Therapiedauer zu kommen, benötigt man, besonders zur Eliminierung der Keime, bzw. um eine mykologische Sanierung zu erzielen, eine gewisse Depot-Wirkung und eine höhere Bioverfügbarkeit des Wirkstoffes. Dafür sind die bekannten Formulierungen nur begrenzt geeignet, weil sich von dem vorhandenen Wirkstoffangebot nur ein kleiner Anteil im Flüssigvolumen am Ort der Infektion löst. Wenn man nun ohne weitere Erhöhung der Wirkstoffkonzentration eine Verkürzung der Therapiedauer, z. B. auf einige Tage bei täglich einmaliger Applikation, erreichen will, muss man für eine optimale Bioverfügbarkeit des Wirkstoffes Sorge tragen.
Es wurde nun gefunden, dass solche Formulierungen des Bifonazols, die 10—60 Gewichtsteile Spreitmittel und 20—80 Gewichtsteile Lösungsmittel sowie gegebenenfalls übliche Formulierungshilfstoffe enthalten, eine optimale Freisetzung des Wirkstoffes und damit eine auf einige Tage verkürzte Therapiedauer durch das Erreichen von hohen Konzentrationen des Wirkstoffes ermöglichen. Dieser Effekt wird dadurch erreicht, dass die Wirkung des in den Formulierungen enthaltenen Bifonazols durch Spreitmittel und Lösungsmittel erhöht wird und dadurch die Wirkstoff-Freiset-zung bis ins Zehnfache gesteigert werden kann. Die erfin-dungsgemässen Zubereitungen stellen somit ein neues Applikationsprinzip von Bifonazol zur dermalen Behandlung von Mykosen dar..
Die erfindungsgemässen Zubereitungen können sowohl Lösungen als auch Sprays sein. Sie werden hergestellt, indem man in an sich bekannter Weise entweder
(a) das Carbinol der Formel

gegebenenfalls in Gegenwart eines Verdünnungsmittels, umsetzt, und das dabei erhaltene Azolyl-(l)-methan gegebenenfalls mit physiologisch verträglichen Säuren umsetzt, das dabei erhaltene Produkt mit 10—60 Gewichtsteilen Spreitmittel und 20—80 Gewichtsteilen Lösungsmittel vermischt, und die so erhaltene Lösung gegebenenfalls mit einem Treibmittel vermischt.
Bifonazol ist in den erfindungsgemässen Mitteln im allgemeinen in Mengen von 0,05—1,5%, vorzugsweise 1 %, vorhanden.
Unter Spreitmitteln werden ölige Flüssigkeiten verstanden, die sich auf der Haut besonders gut verteilen. (R. Key-mer, Pharm. Ind. 32 (1970), S. 77—S. 81.) Für die erfindungsgemässen Mittel eignen sich als Spreitmittel insbesondere folgende Verbindungen:
Silikonöle verschiedener Viskosität.
Fettsäureester, wie Ethylstearat, Di-n-butyl-adipat, Lau-rinsäurehexylester, Dipropylen-glykolpelargonat, Ester einer verzweigten Fettsäure mittlerer Kettenlänge mit gesättigten Fettalkoholen Ci6—C]8, Isopropylmyristat, Isopropylpalmi-tat, Capryl/Caprinsäureester von gesättigten Fettalkoholen der Kettenlänge C!2—Ci8, Isopropylstearat, Ölsäureoleyl-ester, Ölsäuredecylester, Ethyloleat, Milchsäureethylester, wachsartige Fettsäureester wie künstliches Entenbürzeldrü-senfett, Dibutylphthalat, Adipinsäurediisopropylester, letzterem verwandte Estergemische u. a.
Triglyceride, wie Capryl/Caprinsäuretriglycerid, Trigly-ceridgemische mit Pflanzenfettsäuren der Kettenlänge Cg—Cu oder anderen speziell ausgewählten natürlichen Fettsäuren, Partialglyceridgemische gesättigter oder ungesättigter evtl. auch hydroxylgruppenhaltige Fettsäuren, Mo-noglyceride der Cg/Cio-Fettsäuren u. a.
Fettalkohole, wie Isotridecylalkohol, Cetylstearyl-Alko-hol, Oleylalkohol.
Fettsäuren, wie z. B. Ölsäure.
Besonders gut geeignete spreitende Öle sind die folgenden: Isopropylmyristat, Isopropylpalmitat, Isopropylstearat,
Capryl/Caprinsäureester von gesättigten Fettalkoholen der Kettenlänge Ci2 —C|8, wachsartige Fettsäureester wie künstliches Entenbürzeldrüsenfett, Siiikonöle, Isopropylmyristat-Isopropylpalmitat-Isopropylstearat-Gemisch und Kokos-fettsäureisopropylester.
Als Lösungsmittel kommen beispielsweise Alkanole, wie Ethanol und Isopropylalkohol, Methyl-Cellosolve, Cellosol-ve, Ester, Morpholine, Dioxan, Dimethylsulfoxid, Dimethyl-formamid, Tetrahydrofuran, Cyclohexanon, etc. in Betracht.
Bei der Herstellung der erfindungsgemässen Formulierungen können ein oder mehrere Lösungsmittel eingesetzt werden.
Als Formulierungshilfsstoffe kommen beispielsweise in Frage:
a) Tenside (beinhaltet Emulgatoren und Netzmittel),
z.B.
1. anionaktive, wie Na-Laurylsulfat, Fettalkoholethersulfa-te, Mono/Dialkylpolyglykoletherorthophosphorsäu-reester-Monoethanolaminsalz;
2. kationaktive, wie Cetyltrimethylammoniumchlorid;
3. ampholytische, wie Di-Na-N-lauryl-ß-ininodipropionat oder Lecithin;
4. nicht ionogene, z. B. polyoxyethyliertes Rizinusöl, poly-oxyethyliertes Sorbitan-Monooleat, Sorbitan-Monostea-rat, Cetylalkohol, Glycerinmonostearat, Polyoxyethylen-stearat, Alkylphenolpolyglykolether.
b) Stabilisatoren zur Verhinderung des bei einigen Wirkstoffen eintretenden chemischen Abbaues, wie Antioxydan-tien, z. B. Tocopherole, Butylhydroxyanisol.
Wirksamkeits-Testung der erfindungsgemässen Mittel am Trichophyton-infizierten Meerschweinchen.
Als Testmodell zur vergleichenden Wirksamkeitsprüfung der erfindungsgemässen Mittel verwendeten wir Trichophyton infizierte Pirbright-white-Meerschweinchen mit einem durchschnittlichen Gewicht von 600 g. Die Tiere wurden auf dem Rücken mit einer elektrischen Haarschneidemaschine so geschoren, dass ca. 1/10 mm lange Haarstümpfe stehen blieben.
Die Infektion mit Trichophyton mentagrophytes erfolgte durch leichtes Verreiben einer 24 Stunden in Sabouroud-Nährlösung angekeimten Sporensuspension des Erregers auf einer ca. 2 x 2 cm grossen Fläche des geschorenen Rückens der Tiere. Aufgetragen wurden pro Tier 0,5 ml Keimsuspension, die 1 —3 x 105 infektiöse Pilzpartikel enthielten.
Bei diesem Infektionsmodus zeigen sich 2 —3 Tage post infectionem die ersten Symptome der Dermatophytose als Rötung und Schuppung der Haut. Bei unbehandelten Tieren ist ca. 14 Tage p.i. die Dermatophytose maximal ausgeprägt. Flächiger Haarausfall und blutige Integument-Defekte innerhalb einer entzündlich veränderten, schuppigen Randzone.
Die zu prüfenden Formulierungen wurden lmal, am 2. Tag post infektionem, lokal auf die gerötete Infektionsstelle der Tiere appliziert. Es wurden jeweils 0,5 ml der Formulierungen = 5 mg Wirkstoff (1 %ige Formulierung) aufgetragen. Die Bewertung des Infektionsablaufs erfolgte täglich bis zum 20. Tag p.i.
In diesem Test zeigten die erfindungsgemässen Formulierungen eine sehr gute Wirkung.
Verwendet man zum Vergleich solche Formulierungen, die neben Bifonazol weder Spreitmittel noch Lösungsmittel enthalten, so erzielt man nur eine schwache Wirkung.
In den nachstehenden Beispielen sind Rezepturen für er-findungsgemässe Mittel angegeben. Die einzelnen Komponenten werden bei Zimmertemperatur miteinander vermischt und gehen dabei in Lösung.

Herstellung des Wirkstoffs Bifonazol
Verfahrensvariante (a)
13,6 g (0,2 Mol) Imidazol werden in 150 ml Acetonitril gelöst und bei 10° C mit 3,5 ml Thionylchlorid versetzt. Zu der so erhaltenen Lösung von Thionyl-bis-imidazol gibt man 13 g (0,05 Mol) Diphenyl-phenyl-carbinol. Nach 15-stündi-gem Stehen bei Raumtemperatur wird das Lösungsmittel durch Abdestillieren im Vakuum entfernt. Der Rückstand wird in Chloroform aufgenommen und mit Wasser gewaschen. Die organische Phase wird abgetrennt, über Natriumsulfat getrocknet, filtriert und das Lösungsmittel im Vakuum abdestilliert. Der ölige Rückstand wird in Essigester gelöst und durch Filtration von unlöslichen, harzigen Bestandteilen befreit. Das Lösungsmittel wird erneut im Vakuum abdestilliert und der Rückstand durch Umkristallisa-tion aus Acetonitril gereinigt. Man erhält 8,7 g (56% der Theorie) Diphenyl-imidazolyl-(l)-phenyl-methan (Bifonazol) vom Schmelzpunkt 142° C.
Ausgangsprodukt
38,8 g (0,15 Mol) 4-Phenyl-benzophenon werden in 200 ml Ethanol gelöst und mit 3 g (0,075 Mol) Natriumborhydrid versetzt. Nach 15-stündigem Erhitzen unter Rück-fluss wird das erkaltete Reaktionsgemisch mit schwach salz-s saurem Wasser hydrolysiert. Der dabei entstandene Feststoff wird durch Umkristallisation aus Ethanol gereinigt. Man erhält 36 g (89% der Theorie) Diphenyl-phenyl-carbi-nol vom Schmelzpunkt 72—73° C.
io Verfahrensvariante (b/3.)
167 g (0,6 Mol) Diphenyl-phenyl-chlor-methan und 92 g (0,66 Mol) Trimethylsilylimidazol, gelöst in 500 ml Acetonitril, werden 15 Stunden unter Rückfluss erhitzt. Nach Abdestillieren des Lösungsmittels wird der kristalline Rückstand 15 durch Umkristallisation aus Essigester gereinigt. Man erhält 97 g (52% der Theorie) Diphenyl-imidazolyl-(l)-phenyl-methan (Bifonazol) vom Schmelzpunkt 142° C.

Sprays
Die gemäss Beispiel 1 und 2 hergestellten Bifonazollö-sungen können auch zu Sprays verarbeitet werden. Zu die-35 sem Zweck vermischt man z. B. 60—90% Lösung mit 20 —40% der gebräuchlichen Treibmittel, z. B. N2, N20, C02, Propan, Butan, Halogenkohlenwasserstoff usw.

Claims (10)
1. mit Imidazol umsetzt, oder
1. Antimykotische Mittel mit höherer Freisetzung der Wirkstoffe, enthaltend Bifonazol, dadurch gekennzeichnet, dass sie 10—60 Gewichtsteile Spreitmittel und 20 — 80 Gewichtsteile Lösungsmittel enthalten.
2. mit Silber- oder Alkalisalzen des Imidazols umsetzt, oder
2. Antimykotische Mittel nach Anspruch 1, dadurch gekennzeichnet, dass sie Bifonazol in Mengen von 0,05—1,5 Gew.-%, vorzugsweise von 1 Gew.-%, enthalten.

3. Antimykotisches Mittel nach Anspruch 1, dadurch gekennzeichnet, dass es eine Lösung ist.
4. Antimykotisches Mittel nach Anspruch 1, dadurch gekennzeichnet, dass es ein Treibmittel enthält und ein Spray ist.
5. Verfahren zur Herstellung von Zubereitungen des Bi-fonazols, dadurch gekennzeichnet, dass man entweder
(a) das Carbinol der Formel

6. Verfahren nach Anspruch 5, dadurch gekennzeichnet, dass bei der Variante (a) die Umsetzung des genannten Car-binols mit dem Thionyl-bis-imidazol in Gegenwart eines Verdünnungsmittels erfolgt.
7. Verfahren nach Anspruch 5, dadurch gekennzeichnet, dass bei der Variante (b) die Umsetzung des genannten Ha-logen-methans mit dem Imidazol in Gegenwart eines Säurebindemittels und gegebenenfalls in Gegenwart eines Verdünnungsmittels erfolgt.
8. Verfahren nach Anspruch 5, dadurch gekennzeichnet, dass bei der Variante (b) die Umsetzung des genannten Ha-logen-methans mit Silber- oder Alkalisalzen des Imidazols in Gegenwart eines Verdünnungsmittels erfolgt.
9. Verfahren nach Anspruch 5, dadurch gekennzeichnet, dass bei der Variante (b) die Umsetzung des genannten Halogen-methans mit dem Trimethylsilylimidazol in Gegenwart eines Verdünnungsmittels erfolgt.
10. Verfahren nach einem der Ansprüche 5 bis 9, dadurch gekennzeichnet, dass man die so erhaltene Lösung mit einem Treibmittel vermischt.
