Verfahren zur Darstellung eines morphinfreien Opiumalkaloidpräparates. Bei Opiumpräparaten, in denen die Opium alkaloide in ihrem natürlichen Mengenver hältnis vorhanden sind, herrscht das Morphin sowohl an Gewicht, wie in pharmakologischer Wirkung vor. Solche Präparate sind nicht in allen Fällen der Opiummedikation in gleicher Weise günstig.
In gewissen Fällen. zum Bei spiel bei Funktionsstörungen, die mit einem Krampfe der glatten Muskulatur des Ver dauungstraktes einhergehen, ist ein relativ morphinarmes oder gar ein morphinfreies Opiumalkaloidpräparat zweckentsprechender als ein morphinreiches.
Gegenstand vorliegender Erfindung ist nun ein Verfahren zur Herstellung eines morphinfreien Präparates, das die übrigen Alkaloide des Opiums in ihrer gesamten Menge enthält. Durch Zusatz verschieden grosser Mengen von reinem Morphin zu die sem Präparat kann man die pharmakologische Wirkung des Präparates weitgehend und zu verlässig regulieren, ohne dass der anerkannte Vorteil verloren geht, der in dem günstigen Zusammenwirken der Gesamtalkaloide des Opiums besteht.
Das neue Opiiimall,:aolidpräparat enthält alle Nebenalkaloide des Opiums, also die Ge samtalkaloide der of fizinellen Droge mit Aus nahme des Morphins in ihrem ursprünglichen Verhältnis und frei von fremden Begleit- stoffen. Als Ausgangsmaterial zu dessen Herstellung kann man sowohl eine Mischung von Opium und natürlicher Zellensubstanz, als auch eine Mischung eines aus der Droge hereiteten geeigneten Extraktes und natür licher Zellstubstanz verwenden, als welche zur Fixation der Alkaloide vorteilhaft Blatt hulver verwendet; wird.
Das Verfahren beruht darauf, dass man dureb Zusatz von sauer wirkenden Reagen- tien zu pflanzlichem Ausgangsinaterial die Alkaloide in natürlicher Zellstubstanz gegen über mit Wasser nicht mischbaren, niedrig siedenden Fettlösungsmitteln fixiert, worauf man dem Material durch erschöpfende Vor- extrAtion mit einem dieser Fettlösungs mittel die löslichen,
nicht basischen Begleit- stoffe der AllLaloide entzieht, dann das so vorgereinigte Extraktiunsgut in neuen 1Vfen- gen Fettlösungsmittel suspendiert und alka- lisch macht, worauf man die in Freiheit ge setzten Alkaloide extraliiert und den so gewonnenen Extrakt bei niedriger Tempe ratur einengt.
Der niorphinhaltige Rückstand wird in der Kälte, vorteilhaft bei (-r'e,yyenwart von etwas @@"asser, durch Behandeln mit einem Fettlösiizt-:inittel, in dem Morphin nahezu unlöslich ist. wie z. B. Benzin, in die darin leicht löslichen Nebenalkaloide und in das bis auf geringe Spuren sich ausscheidende Nebenprodukt llorpliin getrennt, worauf die Lösun-, der Nelicnall@aloide von Morphin ab- filtritert und bei niedriger Temperatur zur Trockne verdampft wird.
Das hinterbleibende Alkaloidgeiniseh kann zwecks Reinigung mit ausreichender verdünnter Säure aufgenommen und die Flüssigkeit von einer kleinen Menge unlöslicher Flocken abfiltriert werden. Die nun reine, nur in grösserer Konzentration gelb lich gefärbte Alkaloidlösung kann als solche verwendet und eingedampft werden.
Der Mannit-fa.ltigkeit der Opiumalkaloide in ihrer Basizitä t und in ihrer Löslichkeit in Fettlösungsmitteln trägt das Verfahren Rechnung: Zur Fixation der starkbasischen. Alkaloide, z. B. der Morphingruppe, wird mit Rücksicht auf ihre Säureempfindlichkeit vor teilhaft eine nur sehwach angesäuerte Zell- substanz, welche die schwachbasischen Al kaloide, z.
B. den, Narkotingruppe, gegenüber Fettlösungsmitteln noch nicht festzuhalten vermal'', angewandt, iv iilirend zu der darauf folgenden Fixation der sclüvachen Basen die Zellsubstanz zweckmässig mit viel Mineral säure, z.
B. Scliv-efelsä.tire, im Überschuss, be handelt wird, so dass aus einer Probe der Zellsulistanz mit einem Gemisch von .1 Teilen Alkohol und f Teilen Benzol freie Mineral säure extrahiert werden kann.
Soll das Verfahren dazu dienen, ausser den niorphinfreien Nelienall@aloiden gleich zeitig auch das l\ebenprodulzt Morphin in sei ner ganzen Men..-e aus Opium oder daraus bereiteten geeigneten Extrakten zu gewinnen, so kann man.

Der kleine Morphinanteil, der die Neben alkaloide im Benzolextrakte benleitet, wird abgetrennt durch Minengen der Lösung auf ein nicht zii kleines Volumen, woraus sich beim Stehen in der Kälte, namentlich bei Zu satz von etwas Wasser,
das Morphin bis auf eine praktisch verschwindend ](leine Menge kristallinisch ausselieidet und von den in Lö sung bleibenden übrigen Alkaloiden durch Filtration abgetrennt werden kann. Man dampft hierauf das Filtrat vollständig ein und trocknet das brä-unliclie Alkaloidgemisch im Vakuumexsikkator. Das Präparat.
enthält neben den Alkaloiden noch Spuren von Farb stoff und geringe Mengen von wachsartigen Stoffen, die beim Aufnehmen der Basen finit ausreichender verdünnter Siiure ungelöst blei ben und den Farbstoff aufnehmen, so class durch Filtration eine nur sehwach gelblich gefärbte Lösung erhalten wird. Sie enthält die Alkaloide frei von Fremdstoffen und kann als solche verwendet oder eingedampft wer den.
