Abstract

通常已知泡沫塑料可通过挤出含挥发性发泡剂的熔融物来制备。
还已知,例如EP-A240904通过挤出含活性成分的聚合物熔融物并随后成型来制备固体药物形式。
原则上,快速释放的药物形式可通过采用不溶的、但可溶胀的并导致药物形式快速崩解的崩解剂获得。
快速释放还可利用通过用相对低分子量的水溶性热塑性聚合物作为基质聚合物的熔融物挤出生产的药物形式获得。然而,该种形式的缺点是这类聚合物的使用常会给最终药物形式的贮存稳定性带来问题。
本发明的一个目的是寻找可采用有经济吸引力的熔融挤出方法获得的含活性成分的制剂,该制剂允许活性成分的控制释放。
我们发现该目的通过开头所定义的活性成分形式得以实现。我们还发现了生产这类活性成分形式的方法。
本发明的固体的发泡的含活性成分的制剂可包含所有的在加工条件下均可掺入聚合物熔融物中而不会被分解的物质做为活性成分。
适宜的活性成分实例是:

依据本发明,特别优选的活性成分是布洛芬、乙酰水杨酸、对乙酰氨基酚、氨替比林、氟比洛芬、卡托普利、硝苯吡啶、乙酰半胱氨酸、萘呋胺、戊脉安和呋塞米。
其他适宜的活性成分是谷物保护试剂、其他杀生剂或兽药。
依据本发明,用作聚合物基质的适宜的热塑性可加工聚合物是非晶态的热塑性聚合物。
特别适宜的聚合物是水溶性的、热塑性可加工的N-乙烯基吡咯烷酮的均聚物或共聚物或者这类聚合物的混合物。通常这类聚合物的玻璃态化温度在80-190,优选在90-170℃范围内。适宜的均聚物的实例是Fikentscher K值在10-30范围内的聚合物。适宜的共聚物可包含作为共聚单体的不饱和羧酸,如甲基丙烯酸、巴豆酸、马来酸、衣康酸和它们与具有1-12,优选1-8个碳原子的醇的酯,还有羟乙基或羟丙基的丙烯酸酯和甲基丙烯酸酯、(甲基)丙烯酰胺、马来酸和衣康酸的酸酐和半酯(优选仅在聚合后形成的半酯)或乙烯基单体,如N-乙烯基己内酰胺、乙酸乙烯酯、丁酸乙烯酯和丙酸乙烯酯或所述共聚单体的混合物。适宜的实例是N-乙烯基吡咯烷酮、乙酸乙烯基酯和丙酸乙烯酯的三元共聚物。
优选的共聚单体是丙烯酸,尤其优选乙烯基乙酸酯。共聚单体的含量可为20-70%(重量)。依据本发明,特别优选的是那些由60%(重量)的N-乙烯基吡咯烷酮和40%(重量)的乙酸乙烯基酯获得的共聚物。
适宜的聚合物实例还有氯乙烯、聚乙烯醇、聚苯乙烯、聚酯(poly…rates)、聚羟基丁酸酯的均聚物或共聚物以及乙烯和乙烯基乙酸酯的共聚物。
活性成分制剂还可包含淀粉、降解的淀粉、酪蛋白、果胶、壳多糖、脱乙酰壳多糖、明胶或虫胶作为基质成分,它们可在熔融物中添加常用增塑剂的条件下进行加工。
依据本发明,制剂中还可含有重量可高达约50%的常规药物辅助物质,如填充剂、润滑剂、脱模剂、流动调节剂、增塑剂、颜料和稳定剂。这些和下文所述的含量均是基于制剂的总重量(=100%)。
填充剂的实例可列举镁、铝、硅和钛的氧化物、乳糖、甘露醇、山梨醇、木糖醇、季戊四醇及其衍生物,填充剂的量是约0.02-50,优选0.2-20%(重量)。
流动调节剂的实例可列举长链脂肪酸,如C12 C14、C16和C18脂肪酸的单、二或三甘油酯;蜡,如巴西棕榈蜡和卵磷脂,其量是约0.1-30,优选0.1-5%(重量)。
除低分子量的聚氧化亚烷基,如聚乙二醇、聚丙二醇和聚乙/丙二醇以外,增塑剂的实例还可列举多元醇,如丙二醇、甘油、季戊四醇和山梨醇,以及二乙基硫代琥珀酸钠、甘油的单、二和三乙酸酯以及聚乙二醇的硬脂酸酯。增塑剂的量在约0.5-15%,优选在0.5-5%(重量)。
润滑剂的实例可列举硬脂酸铝或硬脂酸钙、以及滑石和聚硅氧烷,其量为约0.1-5,优选为0.1-3%(重量)。
稳定剂的实例可列举光稳定剂、抗氧剂、自由基清除剂和抗微生物的稳定剂,其量优选为约0.01-0.05%(重量)。
为生产本发明的制剂,可将活性成分预先与聚合物混合,随后挤出或者在挤出过程中将活性成分计量加入到含发泡剂的聚合物熔融物中。
制剂中各种成分的比例可在较宽范围内变化。根据活性成分的有效剂量和释放速率,活性成分的量可为活性成分制剂重量的0.1-90%。聚合物的量可为10-99.9%(重量)。此外,还可含有0-50%(重量)的一种或多种辅助物质。
本发明的完全泡沫化的活性成分制剂优选通过挤出熔融物生产,除一种或多种活性成分外,熔融物中还含有一种或多种热塑性可加工聚合物,并且如果需要还含有常规的辅助物质,熔融物还充有挥发性的生理可接受的发泡剂。
适宜的挥发性生理可接受的发泡剂是气体发泡剂,如二氧化碳、氮气、空气、惰性气体,如氦气或氩气,氯氟烃或二氧化氮(笑气),其中二氧化碳和/或氮气是优选的。
熔融物优选在挤出机,尤其优选在双螺旋挤出机中进行生产。一种或多种活性成分与聚合物和,如果需要,其它添加剂的混合可在聚合物熔融之前或之后按照常规工业方法进行。对于温度敏感的活性成分,特别建议仅在热塑性物质熔融后加入。可在50-200,优选在100-180℃获得熔融物,适宜的温度尤其是取决于聚合物的玻璃态温度。聚合物通常在高于它们的玻璃态温度下熔融。
优选在10-300巴,尤其优选在50-200巴的压力下往熔融物中充入发泡剂。在该条件下,有约1-15%(重量)的发泡剂溶于熔融物。
增塑性发泡剂如CO2的充入可降低熔融物的粘度,这样含发泡剂的熔融物可在比相应的不含发泡剂的熔融物更低的温度下挤出。含发泡剂的聚合物熔融物的这种特性有益于热敏感的活性成分的加入。
优选将含发泡剂的熔融物冷却到0-50℃的高于不含发泡剂的聚合物或聚合物混合物的玻璃态温度的温度。
对于温度特别敏感的活性成分,特别建议在添加发泡剂并降低温度后加到熔融物中。
本发明的方法可在有不同温度区的单挤出机中进行。但由两台挤出机联合在一起的串联挤出装置是优选的,在第一台挤出机中熔融聚合物,填充熔融物的发泡剂,优选在有足够搅拌功能的双螺旋挤出机中进行;第二台挤出机是带有轻微剪切功能和快速冷却功能的单螺旋挤出机。
从塑模口中出来的仍具塑性的挤出物在挤压机之外的大气压下膨胀为泡沫。
本发明的活性成分制剂的泡沫化程度可通过加入的发泡剂量和挤出温度来控制。高程度泡沫化导致活性成分剂型的低密度和快速溶解。如果需要较高的密度,可通过直接在塑模缝口脱气降低有益于制造的高发泡剂含量直至获得的产品仅是轻微泡沫化的。随后将泡沫化的活性成分制剂成型为所需的活性成分剂型,例如采用已知的方法制成小丸、颗粒或片剂。
固体的完全泡沫化的活性成分制剂通常的密度在200-1000g/l,优选在200-800g/l。
本发明方法的另一种实施方案涉及通过共挤出生产含活性成分的部分或完全泡沫化的多层剂型的方法。该剂型包含至少两种组合物,所述至少两种组合物中都至少含有一种所述的热塑性粘合剂,其中至少一种组合物含有活性成分并且至少一种组合物以上述方式填充有气态的生理可接受的发泡剂,将这两种组合物共同挤出并随后成型为所需剂型。

将各挤出机中的熔融或塑性组合物加入公用的共挤出塑模中,成形并挤出。共挤出塑模的形状取决于所需的活性成分形式。例如称为宽缝塑模的,带有平直出口缝的塑模,和具有环形缝隙状出口截面塑模是适宜的。喷嘴的设计取决于使用的聚合物粘合剂和所需的形状。
从共挤出塑模中挤出后,将其成型为所需的活性成分剂型或药物剂型。在该阶段,可根据共挤出塑模和成型方式生产各种形状的产品。例如,可由从宽缝塑模中挤出的并且特别地具有两或三层的挤出物通过如冲压或切割例如用炽热的线切割来生产开放的多层片剂。或者,开放的多层片剂可通过具有环形缝状出口截面的塑模采用热切方法分开,即通过立即切割或切削刚从喷嘴中出来的挤出物,或者优选采用冷切割方法分开,即至少在部分冷却后切割或切削挤出物。
封闭的活性成分形式,即其中含活性成分的层完全被不含活性成分的层所包封的形式尤其可通过具有环形缝状出口截面的塑模处理在适宜的压辊装置(例如在附图1和附图2中所示的)中的挤出物获得,这将在下面的实施例中加以解释。在这种情况下,如果在外层冷却后,在进入辊压装置中时多层片的内层仍能塑性变形是有益的。用这种方法可生产尤其是片剂,优选椭圆形片剂,包衣片剂、锭剂和丸剂。
在另一种变换的方法中,含活性成分的泡沫化剂型可以这样的方式制备:挤出除含有一种或多种活性成分外还含有至少一种热塑性粘合剂的熔融物,随后将仍具有塑性的熔融物成型,然后在压力下往含活性成分的固体中充入上述的气态发泡剂,例如在常规的加压釜中在10-300巴,优选在50-200巴的压力下填充,随后发泡。一旦气压解除,填充的剂型就发泡为部分或完全泡沫化的形式。
泡沫化的程度取决于填充的时间,并是可任意调节的。该实施方案优选适于生产部分泡沫化的剂型,它具有泡沫化外壳和未泡沫化的内核,这样显示了阶段化的释放特性。
泡沫化形式可以具有常规的允许活性成分渗透的包衣,这样可以简单的方式获得漂浮剂型。这类漂浮剂型可用于药学、兽药或农业产品,例如缓慢沉降的鱼食方面。
根据本发明获得的固体的泡沫化的活性成分制剂,它含有均匀分散在聚合物基质中的活性成分,溶解非常快速,因此可使活性成分快速释放。该泡沫化的活性成分制剂可采用本发明的方法以简单和经济的方式获得。通过发泡剂的粘度降低作用,挤出可在明显低于不加发泡剂时的温度下进行是有利的这样活性成分可承受较低的热负荷。
实施例1
将20%(重量)的布洛芬和80%(重量)K值为30的聚合物(由60%(重量)N-乙烯基吡咯烷酮和40%(重量)乙酸乙烯酯制备的)在双螺旋挤出机中于150℃熔融并在100巴的挤出机内部压力下往挤出机中充入二氧化碳,这样有4%(重量)的二氧化碳溶于熔融物中。在单螺旋挤出机中使带有发泡剂的熔融物冷却到80℃,并挤出。挤出物在进入大气压环境的同时冷却到室温,得到固体的发泡的含活性成分的制剂。经显微镜肉眼观察,含活性成分的泡沫在水中的溶解速率比在无发泡剂存在下通过挤压获得的相应压制的活性成分快30倍。
实施例2
将26%(重量)的布洛芬和74%(重量)K值为30的共聚物(由60%(重量)N-乙烯基吡咯烷酮和40%(重量)乙酸乙烯酯制成)在双螺旋挤出机中于150℃熔融并挤出。采用EPA240906描述的方法使仍具热塑性的挤出物形成为重650mg的椭圆形片剂,随后在高压釜中于40℃和100巴的二氧化碳压力下填充4小时,然后解除压力使其冷却到室温。用USP XII浆式装置测定释放速率。30分钟后,有85%的活性成分释放。与此相比,以类似的但无填充和泡沫化步骤的方法制备的椭圆形片剂在30分钟后仅释放了40%的活性成分。
Claims (9)
1.一种固体的部分或完全泡沫化的活性成分制剂,它含有至少一种热塑性聚合物。
2.权利要求1的活性成分制剂,它含有N-乙烯基吡咯烷酮均聚物或共聚物作为热塑性聚合物。
3.权利要求1或2的活性成分制剂,它含有的活性成分选自布洛芬、酮洛芬、氟比洛芬、乙酰水杨酸、戊脉安、对乙酰氨基酚、硝苯吡啶、咖啡因、卡托普利和维生素或两种或多种这些活性成分的混合物。
4.一种生产权利要求1-3任一项的活性成分制剂的方法,其特征在于,挤出含一种或多种活性成分和至少一种热塑性聚合物以及挥发性生理可接受的发泡剂的熔融物,使发泡的挤出物成型为所需的活性成分制剂。
5.一种生产权利要求1-3任一项的活性成分制剂的方法,其特征在于,共挤出至少两种熔融物,所述至少两种熔融物中均含有热塑性聚合物,并且至少一种熔融物中含有一种或多种活性成分,以及至少一种熔融物中含有生理可接受的发泡剂,随后使发泡的挤出物成型为所需的活性成分制剂。
6.一种生产权利要求1-3任一项的活性成分制剂的方法,其特征在于,挤出含一种或多种活性成分和至少一种热塑性粘合剂的熔融物,随后使仍具塑性的熔融物成型,在加压下往固体活性成分制剂中充入挥发性的、生理可接受的发泡剂,然后发泡。
7.采用权利要求4的方法获得的活性成分制剂。
8.采用权利要求5的方法获得的活性成分制剂。
9.采用权利要求6的方法获得的活性成分制剂。
