Abstract
Die Erfindung betrifft mehrere Herstellungsverfahren neuer, an der Aminofunktion durch einen Heterozyklus substituierter Trifluoraethylamine der Formel I. Ziel der Erfindung ist die Bereicherung des Standes der Technik durch pharmakologisch interessante Verbindungen. Die neuen Verbindungen wirken neurologisch und blutdrucksenkend bei niedriger Toxizitaet und koennen in der Medizin als Heilmittel eingesetzt werden.

Verfahren zur Herstellung substituierter Trifluoräthylamine Anwendungsgebiet der Erfindung
Die Erfindung betrifft Herstellungsverfahren neuer substituierter Trifluoräthylamine, deren Aminofunktion durch einen stickstoffhaltigen Heterozyklus substituiert ist. Die neuen Verbindungen weisen interessante pharmakologische Wirksamkeiten auf, sie sind insbesondere als Neurologika und Antihypertonikum in der Medizin einsetzbar·
Es sind bisher keine vergleichbaren Lösungen bekannt geworden.
Es ist Ziel der Erfindung, den Stand der Technik durch neue pharmakologisch, insbesondere neurologisch und blutdrucksenkend wirkende Mittel zu bereichern.
Der Erfindung liegt die Aufgabe zugrunde, Verfahren zur Herstellung neuer, an der Aminofunktion mit einem Heterozyklus substituierter Trifluoräthylamine bereitzustellen.

zu gewinnen, in der die Definition von R, m und ρ unverändert bleibt, von der, falls erforderlich, durch Zugabe einer Mineralsäure oder organischen Säure ein Salz hergestellt werden kann oder die durch Salzbildung mit Hilfe einer optisch aktiven Säure in ihre optischen Isomere gespalten werden kann.
Das erfindungsgemäße Verfahren kann noch durch folgende, gegenwärtig bevorzugte Ausführungsarten definiert werden:
a) Die Kondensation des Amins der allgemeinen Formel II mit dem Isocyanat oder Isothiocyanat der allgemeinen Formel III findet bei einer Temperatur im Bereich von 0 bis 10 in einem tragen Lösungsmittel wie einem linearen oder zyklischen Äther statt.
b) Die Zyklisierung des LO-Halogenalkylharnstoffes oder -thioharnstoffe der allgemeinen Formel IV findet durch Erhitzen auf eine Temperatur zwischen vorzugsweise 50 und 150 ° in wäßrigem Medium und in Gegenwart oder in Abwesenheit eines Hydrazidakzeptoren wie beispielsweise einem Alkalimetallkarbonat oder einem Trialkylamin statt.
c) Die Kondensation des Amins der allgemeinen Formel II mit einem zyklischen S-Methylisothioharnstoff der allgemeinen Formel V wird in einem polaren Lösungsmittel wie Pyridin, Dimethylformamid, Dimethylacetamid oder Hexamethylphosphortriamid bei einer Temperatur zwischen 80 ° und 120 durchgeführt.
d) Der S-Methylisothioharnstoff wird in versalzter Form vorgezogen, und die Kondensation liefert ein Salz der Verbindung der allgemeinen Formel I"»

θ) Das Alkalimetallthiocyanat ist vorzugsweise das Ammoniumthiocyanat·
f) Das Azylierungsinittel ist vorzugsweise ein Säurechlorid und spezieller das Chlorid einer niederen aliphatischen Säure, das Benzoylchlorid oder das Chlorid einer substituierten Benzoesäure.
g) Die Verseifung des Azylthioharnstoffs der allgemeinen Formel VI wird mit Hilfe eines Alkalimetallhydroxide wie beispielsweise Soda oder Kaliumkarbonat durchgeführt.
h) Das Alkylierungsmittel ist ein niederes Alkylhalogenid, ein niederes Alkylsulfat oder ein niederes Alkylarylsulfonat und vorzugsweise Methyljodid oder Äthylsulfat, um das Methyljodid oder ein Methylsulfat oder Athylthio-Uronium-Sulfat zu gewinnen.
i) Die Kondensation zwischen dem Thio-Uronium-Salz der allgemeinen Formel IX und dem Alkylendiamin der allgemeinen Formel X wird durch Erhitzen in einem Lösungsmittel mit hohem Siedepunkt wie Pyridin, Butanol oder Isopropanol durchgeführt.
Die Spaltung der Verbindung der allgemeinen Formel I kann durch Salzbildung mittels einer chiralen Säure wie der D-Weinsäure, der D-Ditoluyl-Weinsäure, der D-Dibenzoyl-Weinsäure, der D-Kampfersäure, der NN-Diäthyltartramsaure, der D-Glukose-Phosphorsäure oder der D-Dampfersulfonsäure durchgeführt werden.
Die Spaltung kann auch entweder an der Ausgangsverbindung der allgemeinen Formel II oder an einer der Zwischenverbindungen der Formel IV, VI oder VIII durchgeführt werden. Die

Spaltungsreagenzien sind die gleichen wie diejenigen, die für die Spaltung der Verbindungen der Formel I verwendet werden.
Die Verbindungen der allgemeinen Formel II, bei denen R ein niederes Cycloalkyl-Radikal mit 3 bis 7 Kohlenstoffatomen darstellt, können auch ausgehend von einer aromatischen Ver bindung der Formel XI

Diese Variante bei der Gewinnung der Verbindungen der allgemeinen Formel II ermöglicht es, Ausgangsverbindungen mit Substituenten am Kern wie zum Beispiel die niederen Alkyl-Radikale oder niederen Alkoxyradikale herzustellen.
Die Verbindungen der allgemeinen Formel II können allgemein ausgehend von einem Trifluormethylketon der allgemeinen Formel XII

hergestellt werden, in der R und m wie vorher definiert sind, das der Einwirkung des Hydroxylamins ausgesetzt wird, um das entsprechende Oxim zu bilden, das durch Einwirkung eines me-

tallischen Reduktionsmittels, eines gemischten Alkalimetallhydrids oder des Diborans zu einem Amin reduziert wird.
Soweit es um die Erfindung geht, bezeichnete der Begriff "niederes Alkyl" eine Kohlenwasserstoffkette mit 1 bis 6 Atomen in gerader oder verzweigter Kette, die gegebenenfalls durch eine Hydroxy-Acyloxy- oder eine Diäthylamino-Gruppe substituiert ist» Beispiele von niederen Alkoxy-Radikalen können eine Methoxy-, eine Äthoxy-, eine Isopropoxy-, eine sekundäre Butyloxy-, eine Neopentyloxy-, eine tert. Butyloxy-, eine n-Hexyloxy, eine /b-Äthoxy (h-Äthoxy- und eine p-Diäthylaminoäthoxy-Gruppe sein.
Wenn R ein Cycloalkyl-Radikal ist, kann es ein cyklanisches Radikal mit einem oder mehreren niederen Alkyl-Substituenten oder niederen Alkoxy-Substituenten wie Cyclopropyl-, 2,2-Dimethylcyklopropyl-, l-tert-Butylcyklopropyl-, Cyklobutyl-, Cyklopentyl-, Cyklohexyl-, 2,6-Dimethylcyklohexyl-, 3,4,5-Trimethoxycyklohexyl-oder Cykloheptyl sein.
Von den Verbindungen der allgemeinen Formel I können leicht durch eine Mineralsäure oder eine organische Säure wie beispielsweise Salzsäure, Bromwasserstoff3äure, Phosphorsäure, Schwefelsäure oder Salpetersäure; Ameisensäure, Essigsäure, Di-n-propylessigsäure, Weinsäure, Zitronensäure, Maleinsäure, Itakonsäure, Benzoesäure, Thiazol-5-karboxylsäure, Nipekoteäure, Glukose-l~phoshorsäure, Methansulfonsäure, Äthansulfonsäure, Isethionsäure oder Benzolsulfonsäure Salze hergestellt werden.
DieStruktur der Verbindungen der allgemeinen Formel I enthält wenigstens ein asymmetrisches Kohlenstoffatom, und deshalb können die Verbindungen der allgemeinen Formel !.entweder in razemischer oder in gespaltener Form vorkommen. Die

optisch aktiven Isomere wie die razemischen Verbindungen sind Bestandteil der Verbindung.
Wenn außerdem R ein Cycloalkylradikal oder ein substituiertes Alkylradikal ist, erscheint mindestens ein zusätzliches Asymmetrie-Zentrum, und die entsprechenden Verbindungen können in ihre eis- und trans-Diastereoisomere gespalten werden. Oedes Diastereoisomer kann danach in seine linksdrehenden und rechts· drehenden Enantiomere umgewandelt werden.
Die Verbindungen der allgemeinen Formel I und ihre Salze weisen interessante pharmakologische Eigenschaften, insbesondere hypotensive Eigenschaften auf. Die erfindungsgemäßen Verbindungen haben nur abgeschwächte neurologische Auswirkungen und können deshalb zu therapeutischen Zwecken als Antihypertensionsmittel ohne störende Nebenwirkungen, wie zum Beispiel eine bei der Beruhigung möglicherweise auftretende Schläfrigkeit oder ein Betäubungszustand, eingesetzt werden.
Pharmazeutische Mischungen können hergestellt werden, die als Wirkstoff wenigstens eine Verbindung der allgemeinen Formel I oder eines ihrer Salze mit einer Mineralsäure oder organischen Säure in Verbindung mit einem inerten, nicht-toxischen, pharmazeutisch vertretbaren Arzneiträger enthalten.
Pharmazeutische Mischungen, die neben einer Verbindung der allgemeinen Formel I oder einem ihrer Salze einen Wirkstoff von ähnlicher oder synergetischer Wirkung enthält, wie zum Beispiel ein Diuretikum oder einen Antagpnisten des Aldosterons, können ebenfalls hergestellt werden. Als Beispiel für solche Wirkstoffe können ein Thiazid oder ein Sulfamid wie das Furosemid oder das Hydrochlorthiazid, das Canceron

- 11 oder das Kaliumcanrenoat genannt werden.
Die eobenstehend beschriebenen pharmazeutischen Mischungen können parenteral, oral, sublingual, percutan oder rektal in Form von einfachen oder überzogenen Tabletten, von Gelatinekapseln, Kapseln, Dragees, Emulsionen oder Lösungen zum Einnehmen, Tropfen, injizierbaren Lösungen oder Suspensionen, Lösungen in einem polaren Lösungsmittel zur perku· tanen Anwendung, von Sublingualia oder von Zäpfchen verabreicht werden.
Die injizierbaren Lösungen oder Suspensionen können vor allem in Ampullen, mehrdosigen Flaschen oder in Wegwerf-Spritzen abgefüllt sein.
Die zweckmäßige Dosierung bewegt sich in weiten Grenzen je nach Alter und Gewicht des Patienten, der Verabreichung und der therapeutischen Indikation.
Gewöhnlich staffelt sich in der Humanmedizin die Einheitsdosierung zwischen O mgl und 2 mg und die Tagesdosierung zwischen O mgl und 5 mg beim Erwachsenen.
Unter den Verbindungen, die nach dem erfindungsgemäßen Verfahren gewonnen wurden, sind insbesondere zu nennen:

Die folgenden Beispiele dienen zur Veranschaulichung der Erfindung. Sie sind in keiner Weise einschränkend*
DL 2-(<S—Trifluormethyl-äthylamino)-oxazolin und sein Chlorhydrat
Stadium A DU 1-Trifluormethyl-äthylamin
Nachdem 9 gl Lithium- und Aluminiumhydrid in 180 ml Äther aufgelöst wurden, werden in einer halben Stunde ungefähr 15 g c^ÄJoC-Trifluoracetonoxim zugesetzt, die vorher in 100 ml Äther aufgelöst wurden. Das Reaktionsgemisch wird unter Rückfluß eine Stunde lang in inerter Atmosphäre gekocht· Nach Wiedererreichung der Raumtemperatur wird der Hydridüberschuß durch Zugabe von wäßriger Soda zersetzt. Die ätherische Phase wird über Natriumsulfat getrocknet und filtriert,

Danach wird 36%ige Salzsäure bis zu einem pH von etwa 2 zugesetzt· Der auftretende Chlorhydratniederschlag wird durch Filtration getrennt, mit Äther gewaschen und unter Vakuum getrocknet. Es werden somit 10 g DL-1-Trifluormethyl-äthylamin, d. h. eine Ausbeute von 57 % gewonnen.
Das Produkt entspricht den analytischen Normen der Fachliteratur. Das Oxim des Trifluoracetons wird nach dem von R. A. Shepard in D. Org. Chenu 31, (1966), S. 964-5 beschriebenen Verfahren gewonnen.
Stadium S
N-( /3j-Chloräthyl)-N'-(l-trif luormethyl-äthyl)-Harnstof f
Es werden 5 g DL-1-Trifluormethyl-äthylamin-chlorhydrat in 10 ml 2N-Soda aufgelöst, und die wäßrige Stammlösung wird 3 mal mit 10 ml Äther ausgeschüttelt. Den gesamten Ätherlösungen wird eine Lösung von 3 g 1 ß-Chloräthylisocyanat in 5 ml Äther unter Abkühlung des Gemisches auf ungefähr O ° zugesetzt. Es wird danach 12 Stunden lang geröhrt, wobei man die Temperatur des Reaktionsgemische auf etwa 15 ° ansteigen läßt.
Der /^-Chloräthyl-Harnstoff fällt nach und nach aus. Er wird durch Filtration abgetrennt; die Kristalle werden mit Eisäther gewaschen, dann unter Vakuum getrocknet. Es werden somit 4,7 g N- (β-ChloräthylJ-N'-Cl-trzfluormethyläthyl)-Harnstoff, d. h. eine Ausbeute von 77 % gewonnen. Das Produkt wird für die Analyse mit Isopropyläther umkristallisiert. Es schmilzt unter Sublimation bei 108 bis 113 °.

3 g N-(/S-Chloräthyl)-Nt-(l-trifluormethyl-äthyl)-Harnstoff werden in 20 ml Wasser und 3,75 ml Triäthylamin suspendiert, Es wird drei Stunden lang unter Rückfluß dee Lösungsmittels gekocht, dann läßt man das Medium auf die Raumtemperatur abkühlen. Nach Ansäuerung der wäßrigen Phase wird nun ausgeäthert. Die Ätherphasen werden abgetrennt, und die wäßrige Phase wird durch Zugabe von Ammoniak bis zu einem pH 10 alkalisiert. Die wäßrige Phase wird dann dreimal mit Methylenchlorid extrahiert. Die organischen Phasen werden getrennt, mit Wasser ausgewaschen, getrocknet und unter vermindertem Druck zur Trockne eingeengt. Somit werden 1,95 g Kristalle, d. h. eine Ausbeute von 78 % gewonnen«
Das Produkt wird durch Sublimation unter verringertem Druck bei 55 ° gereinigt. Eine reine Probe schmilzt bei 102 bis 105 ° (Sublimation).

Cyclopropyl-trifluormethyl-keton
In 50 ml Äther werden 12,5 g Magnesiumspäne unter Stickstoffatmosphäre suspendiert und einige kleine Oodkristalle zugegeben. Nach und nach werden 60,5 g in 400 ml Äther gelöstes Cyclopropylbromid zugegeben. Das Zugeben dauert ungefähr eine Stunde. Danach wird das Gemisch unter Rückfluß des Lösungsmittels eine Stunde lang gekocht«
Es wird eine Lithiumderivat-Lösung der Tpifluoressigsäure hergestellt indem 4 g Lithiumhydrid unter Stickstoff mit 50,5 g Trifluoressigsäure in 100 ml Äther zur Reaktion gebracht werden, wobei die Temperatur des Gemischs durch Eintauchen in eine Eis-Salz-Kühlmischung auf null gehalten wird.
Dann wird die Lösung des Lithiumsalzes der Tpifluoressigsäure in die magnesiumhaltige Lösung gegossen, die wieder Raumtemperatur erreicht hat. Während der gesamten Dauer des Zugebens wird das Gemisch gekühlt, um einen zu starken Temperaturanstieg zu vermeiden.
Nach dem Zusetzen dieses Reagens wird das Gemisch zwei Stunden lang unter Rückfluß gekocht, danach läßt man es wieder eine Temperatur von ungefähr 20 ° erreichen. Der Magnesiumküberschuß wird durch vorsichtiges Zusetzen von 200 ml 6N Salzsäure beseitigt. Die organische Phase wird dekantiert,

mit Wasser gewaschen, dann über Magnesium-sulfat getrocknet. Der Äther wird durch fraktionierte Destillation abdestilliert. Die eingedickte Restlösung besteht aus einem Gemisch aus Trifluor-methyl-cyclopropyl-keton und Tetrahydrofuran. Sie wird als solche für das nächste Stadium verwendet;
Stadium B
Cyclopropyl-trifluormethyl-ketoxim.
Der Tetrahydrofuran-Lösung des Stadiums A werden '3,49 g Hydroxylaminchlorhydrat und 4 gl wasserfreies Natriumacetat zugesetzt. Es wird 6 Stunden lang unter Rückfluß gekocht, danach wird abgekühlt» Das Gemisch wird eine Nacht lang in den Kühlschrank gestellt, dann werden die Oximkristalle durch Filtrieren, Waschen mit Wasser und Vakuum-Trocknung abgetrennt.
Das Cyclopropyl-trifluormethyl-ketoxim verdunstet bei 55 bis 62 ° unter 35 mmHg.

Die Hydrierung des Gemisches erfolgt 4 Stunden lang bei 110 ° unter 180 bar, dann läßt man es wieder die Raumtemperatur erreichen. Das 2-^l-Trifluormethyl-l-cyclohexyl)-methylamino7-oxazolin ist in verdünnter Salzsäure löslich, wobei das Chlorhydrat entsteht· Das Ausgangsprodukt, das &-Trifluormethyl-benzylarain (rechtsdrehendes Isomer) wird nach dem Verfahren hergestellt, das in dem französischen Patent 2.358.890 der anmeldenden Gesellschaft beschrieben ist.
Das (1-Trifluormethyl-l»cyclohexyl)-methylamin (rechtsdrehendes Isomer) kann auch ausgehend von Cyclohexylbrornid durch Bildung des entsprechenden Cyclohexylmagnesiumsalzes, Kondensation mit Hilfe von TrifIuores3igsäure gewonnen werden, um das Cyclohexyl-trifluormethylketon nach der von F. E. Horkes» in 0. Org. Chem. 32 (1967) 1311 - 1318 beschriebenen Methode zu bilden.
Das Cyclohexyl-trif luormethylketon wird in ein Oxim umgewandelt, das zu razemischem (1-Trifluormethyl-1-cyclohexyl)-methylamin reduziert wird. Dieses letztere wird durch Salzbildung durch die D-Weinsäure oder Dibenzoyl-D-VVeineäure in seine optischen Isomere gespalten.

Es werden 12,3 g Ammoniumthiocyanat in 80 ml Azeton gelöst und 11,25 ml Benzoylchlorid innerhalb von 15 Minuten zugesetzt. Das Gemisch wird unter Rückfluß des Lösungsmittels gekocht« Dieser Lösung wird eine Lösung von 27,8 g (1-Trifluormethyl-l-cyclohexyl)~methylamin (rechtsdrehendes Isomer) in 100 ml Azeton zugesetzt. Es wird erneut zwei Stunden lang unter Rückfluß gekocht. Danach wird zur Trockne eingeengt und der Rückstand in 15 ml Methylenchlorid aufgenommen« Die Methylenphase wird bis zum Verschwinden der Chloride mit Wasser gewaschen, dann wird sie zur Trockne eingeengt. Der Rückstand, der 47,5 g wiegt, weist eine ölige Konsistenz auf. Das somit gewonnene N-Benzoyl-Derivat wird durch 150 ml Soda und 25Ö ml Äthanol 24 Stunden lang bei Zimmertemperatur verseift·»
Danach wird das Äthanol abgedampft, der Rückstand wird in Äther aufgenommen und mit Wasser gewaschen, bis das Waschwasser neutral ist (pH = 7,8 ungefähr)e Die Ätherphase

wird getrocknet, dann zur Trockne eingedampft. Es werden somit 23 g (? Zahl unleserlich, Anm. d. Obers·) Thioharnstoff, d. h, eine Ausbeute von 75 % gewonnen.

N-^£l-Cyclohexyl-l-trifluormethyl)-methyl7-isothioharnstoffjodid (linksdrehendes Isomer)
Es werden 27 g des im Stadium A gewonnenen Thioharnstoffe in 150 ml wasserfreiem Aceton gelöst, und es werden 15 ml Methyljodid zugegeben. Es wird 4 Stunden lang unter Rückfluß gekocht, dann das Lösungsmittel zur Trockne eingedampft. Es werden 41 g Kristalle gewonnen, die bei 160 bis 170 ° (unter Zersetzung) schmelzen. Nach der Umkristallisation aus Acetonitril erhält man 34,3 g reines Produkt, d. h. eine Ausbeute von 78 %, Das reine Produkt schmilzt bei 170 ° (unter Zersetzung),

Es werden 21 g des iiii Stadium B gewonnenen Methylisothioharnstoffjodids mit 3 ml Äthylendiamin in 270 ml Isoamylalkohol 6 Stunden lang erhitzt. Danach wird abgekühlt, dann das Lösungsmittel zur Trockene eingedampft· Der Rückstand wird in einer sehr geringen Menge warmen Äthylazetat aufgenommen, und das Imidazolinjodhydrat kristallisiert durch Abkühlung aus· Nach einer Nacht im Kühlschrank werden die Kristalle durch Filtrieren abgetrennt und zentrifugiert getrocknet. Sie werden in 120 ml Soda N und 100 ml Äthanol aufgenommen. Nach 12 Stunden Kontakt wird die Base mit Methylenchlorid extrahiert· Nach Eindampfen des Lösungsmittels erhält man 63 g 2-/Xl-Trifluormethyl-1-cyclohexyl )-methylamino7-methylamino7.-imidazolin, das bei 148 bis 157 ° (Sublimation) schmilzt. Nach Umkristallisieren aus Heptan beträgt der Schmelzpunkt 154 bis

Wenn nach der Arbeitsweise des Beispiels VI verfahren wird undim Stadium B 2,85 ml Propyldiamin verwendet werden, erhält man das 2-^~(l-Trifluormethyl-l-cyclohexyl)-methylamino7-tetrahydropyrimidin (rechtsdrehende3 Isomer)
l-Trifluormethyl-l-cyclohexylj-methylamino-thiazolin (rechtsdrehendes Isomer)
In 75 ml Dimethylformamid werden 7 gl rechtsdrehendes 1-Trifluormethyl-l-cyclohexylmethylamin und 26 g 2-Methylthiothiazolin-jodhydrat gelöst· Es wird eine Stunde lang unter Ruckfluß gekocht, dann läßt man das Ganze wieder Normaltemperatur erreichen· Das Reaktionsgemisch wird mit Isopropyläther in gleicher Menge verdünnt. Die Kristallisation des Oodhydrats wird durch Kratzen eingeleitet, dann läßt man das Ganze eine Nacht im Kühlschrank.
Durch Filtrieren wird das 2-^I-Trifluormethyl-l-cyclohexylmethylamino7-thiazolin-jodhydrat gewonnen, das geschleudert, mit einigen ml Isopropyläther gespült und dann unter Vakuum getrpcknet wird·
Das Oodhydrat wird dann in Wasser gelöst, danach durch Zugabe von Natriumkarbonat bis zur deutlichen Aktivität ineine Base umgewandelt· Die wäßrige Lösung wird dreimal mit Isopropyläther ausgeäthert, die ätherhaltigen Lösungen zusammengegossen,mit Wasser gewaschen, getrocknet, filtriert, dann zur Trockne eingedampft.
Auf diese Weise wird das 2-^1/ -Trifluormethyl-cyclohexyl« methylamino7~thiazolin (rechtsdrehend) gewonnen, das nach Umkristallisation aus Acetonitril bei 135 bis 138 ° schmilzt·

Pharmakolo.qische Untersuchung der erfindunqsqemäßen Verbindunp en
a) Bestimmung der akuten Toxizität
Die akute Toxizität wurde auf intraperitonealem Weg an Losen von 10 Mäusen (Stamm Swiss) mit einem Gewicht von ungefähr 20 g bestimmt, die zunehmende Dosen der zu testenden erfindungsgemäßen Verbindung aufnahmen«
Die Tiere wurden acht Tage lang beobachtet, und die toten, soweit vorhanden, wurden gezählt. Die mittlere letale Dosis wird graphisch nach der Methode von Tainter und Miller ermittelt.

Die erfindungsgemäßen Verbindungen wurden in Dosen verabreicht, die sich von 20 bis 200 mg/kg staffelten, Die durchschnittliche letale Dosis beträgt im allgemeinen 200 mg/kg. Bei dieser Dosis zeigen sich bei den Tieren Beruhigung und gesträubte Haare.
b) Bestimmung der blutdrucksenkenden Aktivität
Die erfindungsgemäßen Verbindungen wurden auf intravenösem Weg Losen von Hunden, die vocher mit Nembutal betäubt wurden, in Dosen zwischen 0,005 mg/kg bis 0,5 mg/kg verabreicht.
Bei den geringsten Dosen (5 und 10X/kg) ist das Absinken des Blutdrucks vorübergehend, doch die Herzfrequenz wird min lang signifikant verringert·
Größere Dosen führen zu einer leichten Erhöhung, und dann zu einer merklichen Verringerung des Blutdrucks. Die Verringerung der Herzfrequenz kann 50 % des bei den Versuchstieren beobachteten Wertes erreichen.
c) Untersuchung, einer neurologischen Wirkung
Bei der Maus CD führen intraperitonal verabreichte Dosen von 1 bis 2 mg/kg zu einer Reduzierung der Motilität, der Körpertemperatur und der Nahrungs- und Flüssigkeitsaufnahme.
Bei der Ratte LE führen vergleichbare Dosen zu Erregung und Sträuben der Haare, höhere Dosen bewirken eine Verminderung der Motilität und der Muskelkraft.
Bei der Katze rufen die erfindungsgemäßen Verbindungen anfänglich Erregtheit, dann eine leichte Abgeschlagenheit

Die erfindungsgemäßen Verbindungen treten folglich als Stimulanzien der zentralen nor-adrenergischen Rezeptoren auf, die eine Inhibition des Tonus sympathicus. Hypotension und Bradykardie hervorrufen. Sie sind sowohl auf parenteralem als auch oralem Wege wirksam.
