Abstract
Die Erfindung betrifft ein Verfahren zur Herstellung von 9-(1,3-Dihydroxy-2-propoxymethyl)-guaninderivaten der Formel IX und deren Salzen, in welcher R eine Acylgruppe ist,z.B. durch Dearalkylierung von Derivaten, in denen die freien Hydroxylgruppen des in 9-Stellung stehenden Restes benzyliert sind.

j 15 VERFAHREN ZUR HERSTELLUNG VON 9-(1,3-DIHYDROXY-2-PROPOXY- ! METHYD-GUANINDERIVATEN.
Die vorliegende Erfindung betrifft Verfahren zur Herstellung von 9-(1,3-Dihydroxy-2-propoxymethyl)-guaninderivaten, die Zwischenprodukte zur Herstellung des antiviralen Mittels!

9-(1,3-Dihydroxy-2-propoxymethyl)-guanin sind. j
VirusInfektionen sind weitverbreitet und führen zu vielen verschiedenartigen Symptomen. Einige Virusinfektionen können leicht durch den Abwehrmechanismus des Körpers überwunden werden, während andere ernsthafterer Natur sind und zu permanenten Schäden, wie Blindheit f oder sogar zum -Tode führen. Eine Virenfamilie, die erhebliche Infektionen verursachen kann, ist die Gruppe der Herpes-Viren.
. Die derzeit zur Behandlung von Virusinfektionen verwendeten Arzneimittel sind in vielen Fällen unwirksam, oder, falls sie wirksam sind, werden sie in hohen und/oder ständigen Dosen benötigt, die ernste Nebenwirkungen und/ oder Toxizität ergeben. Es besteht daher das Bedürfnis nach einem wirksamen antiviralen Mittel, das in niedrigeren Dosen als die derzeit verfügbaren Arnzelmittel wirksam ist und damit die Gefahr möglicher Nebenwirkungen und Toxizität verringert.
Die US PS 4 199 574 beschreibt Verbindungen der folgenden allgemeinen Formel

Von der durch die obige Formel dargestellten Klasse von Verbindungen und deren pharmazeutisch annehmbaren Säureadditionssalzen wird gesagt, daß sie eine antivirale Aktij 20 vität zeigen (vgl. auch Tetrahedron Letters 2A_, 327-30

! Ziel der Erfindung ist die Bereitstellung von Zwischen-
produkten zur Herstellung von wirksamen, noch unbekannten antiviralen Arzneistoffen.
Darlegung des Wesens der Erfindung:
Aufgabe der Erfindung ist es, ein Verfahren zur Herstellung von 9-(1,3-Dihydroxy-2-propoxymethyl)-guaninderivaten anzugeben, die Zwischenprodukte für die Herstellung des besonders aktiven antiviralen Mittels 9-(1,3-Dihydroxy-2-propoxymethyl)-guanin und dessen Salzen sind.

Die Erfindung betrifft ein Verfahren zur Herstellung von 9-(1,3-Dihydroxy-2-propoxymethyl)-guaninderivaten der Formel:

a) eine Verbindung der Formel ο

in welcher R wie oben definiert ist und R' eine Schutz-
gruppe, z.B. Benzyl, ist, die gegebenenfalls mit 1 oder 2 Alkoxygruppen oder 1 oder 2 niedrigen Alkylgruppen substituiert sein kann, in welchen Alkyl eine gerade oder verzweigte Kohlenwasserstoffkette mit 1 bis 4 Kohlenstoffatomen ist,dearalkyliert; oder
b) eine Verbindung der Formel (VIII) mit einer Lewis-Säure umsetzt; oder
c) eine Verbindung der Formel (VIII) mit Essigsäure umsetzt; oder
d) eine Verbindung der Formel (IX) in deren Salz umwandelt; oder
e) das Salz in die entsprechende freie Verbindung der Formel (IX) umwandelt.
Die Verbindungen der Formel (IX) dienen als Zwischenprodukte zur Herstellung von 9-(1,3-Dihydroxy-2-propoxy-5 methyl)-guanin, das daraus durch Deacylierung erhalten wird.
Die Salze können entweder Säure-Additionssalze oder anorganische Metallsalze, vorzugsweise Erdalkalimetallsalze, i 10 sein. Geeignete Säuren zur Salzbildung sind anorganische j Säuren, wie Salzsäure, Bromwasserstoffsäure, Schwefel-I säure, Salpetersäure, Phosphorsäure usw., und organische
Säuren, wie Trifluoressigsaure, Methansulfonsäure, Ethanj sulfonsäure, p-Toluolsulfonsäure usw. Geeignete Basen zur i 15 Salzbildung sind organische Basen, anorganische Metallbasen, wie Kupfer- oder Silberbasen, und Alkalimetallj basen, wie Alkalimetallhydroxide, z.B. Natriumhydroxid, ] Kaliumhydroxid usw.

Die Verbindung der Formel (III) wird hergestellt, indem man Epichlorhydrin (II) tropfenweise zu einer Lösung eines Alkalimetallsalzes, vorzugsweise des Natriumsalzes, von Benzylalkohol, wahlweise gemäß obiger Definition substituiert, in einem Lösungsmittel, -wie Dimethylformamid, Dimethylacetamid, Hexamethylphosphoramid, Dimethylsulfoxid, SuIfolan, Tetrahydrofuran und Dioxan, bei einer Temperatur von etwa 0 bis 1000C, vorzugsweise etwa 15 bis 400C, zufügt. Die Reaktionsmischung wird etwa 10 bis Stunden, vorzugsweise etwa 12 bis 18 Stunden, bei einer Temperatur von etwa 0 bis 1000C, vorzugsweise etwa 20 bis 5O0C, gerührt. .
Die Verbindung der Formel (III) wird zur Verbindung der Formel (IV) chlormethyliert, indem man trockenen gasförmigen Chlorwasserstoff in eine Lösung aus der Verbindung und Paraformaldehyd, gelöst in einem halogenierten Kohlenwasserstoff lösungsmittel, wie Dichlorethan, Chloroform, Dichlormethan und 1,1,2-Trichlorethan, die auf eine Temperatur von etwa 0 bis 250C, vorzugsweise auf eine Temperatur
von etwa O0C, abgekühlt ist, einleitet. Der gasförmige Chlorwasserstoff wird innerhalb von 30 Minuten bis 3 Stunden, vorzugsweise innerhalb von 1 bis 2 Stunden, zugefügt,
bis sich der Paraformaldehyd löst. Die Lösung wird etwa ί 12 bis 48 Stunden auf einer Temperatur von etwa 0 bis 10 C, vorzugsweise etwa 16 bis 24 Stunden auf einer Temperatur . von etwa 0 bis 5°C, gehalten.
Die Verbindung der Formel (V) wird hergestellt, indem man ein Alkalimetallacetat, wie Natriumacetat, mit einer Verbindung der Formel (IV), gelöst in einem Lösungsmittel, wie Dimethylformamid, Tetrahydrofuran, Dimethylacetamid, Hexamethylphosphoramid, Dirnethylsulfoxid, SuIfolan und Dioxan, bei einer Temperatur von etwa 0 bis 45°C, vorzugsweise von etwa 0 bis 25°C, umsetzt. Die Lösung wird etwa 5 bis etwa 24 Stunden, vorzugsweise etwa 10 bis etwa 18 Stunden, bei einer Temperatur von etwa 10 bis etwa 300C, Vorzugs-
weise etwa 15 bis 25°C, gerührt. j
Die Verbindung der Formel (VII) wird hergestellt, indem j man Guanin (VI) mit einem .Carbonsäureanhydrid mit 2 bis 20 j Kohlenstoffatomen, wie Essigsäureanhydrid, Buttersäureanhydrid, Valeriansäureanhydrid, Caprylsäureanhydrid, vorzugsweise Essigsäureanhydrid, etwa 10 bis 24 Stunden·.. j
vorzugsweise etwa 12 bis 18 Stunden zum Rückfluß erhitzt. j Sin Lösungsmittel braucht nicht verwendet zu werden.
j Das N ,9-Diacylguanin der Formel (VII) wird mit der Verbindung der Formel (V) unter Bildung einer Verbindung der Formel (VIII) mit oder ohne ein Lösungsmittel, wie Dioxan, Sulfolan usw., in Anwesenheit einer katalytischer! Menge einer Säure, wie Bis-(p-nitrophenyl)-phosphat, Toluolsulfonsäure, Methylphosphonsäure oder Dichloressigsäure, vorzugsweise Bis-(p-nitrophenyl.)-phosphat, bei einer Temperatur von etwa 75 bis 2000C, vorzugsweise etwa 110 bis 1800C, umgesetzt. Die Reaktion erfolgt gewöhnlich unter Verwendung von 0,8 bis 1,2 Mol Verbindung der Formel (V) pro Mol Verbindung der Formel (VII).
Die Benzyl-Schutzgruppen (R1) können in an sich bekannter Weise, z.B. durch katalytische Hydrierung mit Lewis-Säuren oder Acetolyse unter Bildung der Verbindung der Formel (-IX) aus der Verbindung der Formel (VIII) entfernt werden. Die katalytische Hydrierung erfolgt gewöhnlichin Anwesenheit eines Hydrierungskatalysators, wie j Palladium, Platin, Rhodium oder Nickel, auf einem Träger, j j wie Kohle, und in Anwesenheit eines inerten Lösungsmittels j j für die Verbindung der Formel (IX). Wasserstoff wird der Lösung bei einem Druck von etwa 1 bis 14, vorzugsweise etwa 2 bis; j 5,5 bar, zugefügt. Die Reaktionstemperatur liegt gewöhnlich j zwischen O bis 5O0C. Unter bevorzugten Bedingungen wird 15 Palladium-auf-Kohle in einer"Aufschlämmung zur Lösung der Verbindung von Formel (VIII) in einem Lösungsmittel, wie ein wässriger niedriger Allylalkohol (wobei Alkyl als gerader oder verzweigtkettiger Kohlenwasserstoff mit 1 bis 4 Kohlenstoffatomen definiert ist), vorzugsweise wäss-
riges Methanol, zugefügt.
9-(l,3-Dihydroxy-2-propoxymethyl)-guanin kann hergestellt j werden, indem man die Verbindung (IX) durch Entfernung der ! Schutzgruppen R in an sich bekannter Weise deacyliert. ! 25 Diese Entfernung kann unter solvolytischen Bedingungen durchgeführt werden, wodurch ein geeignetes Lösungsmittel die Aminofunktion in 2-Stellung des Guaninringsystems wiederherstellt. Diese Lösungsmittel sind z.B. Wasser oder niedrige Alkenole mit 1 bis 4 Kohlenstoffatomen. Die Solvolyse kann unter sauren oder basischen Bedingungen erfolgen. Die Reaktionstemperaturen liegen gewöhnlich zwischen 0 bis 500C, vorzugsweise 10 bis 300C. Besonders bevorzugt wird der Bereich zwischen 15 bis 250C. Die basische Solvolyse wird bevorzugt. Geeignete Basen sind z.B.
Alkalihydroxide, Alkalicarbonate oder Ammoniak. Niedrige Alkenole sind die bevorzugten Lösungsmittel, insbesondere Methanol. Unter den bevorzugten Bedingungen wird eine Lösung der Verbindung von Formel (IX) und der Base etwa 1 bis 36 Stunden, vorzugsweise etwa 10 bis 24 Stunden, bei Ij
einer Temperatur von etwa 10 bis 300C, vorzugsweise etwa 15 bis-25°C, gerührt.
Die Salze der Verbindung (IX) werden hergestellt, indem man die Verbindung (IX) mit einer geeigneten Base oder Säure gemäß obiger Definition umsetzt. Diese Umwandlung kann in einem Lösungsmittel erfolgen, wobei etwa stöchiometrisehe Äqivalente der Base oder Säure zur Verbindung der Formel (IX) in einem geeigneten inerten Lösungsmittel bei einer Temperatur zwischen 0 und 5O0C zugefügt werden.
In ähnlicher Weise können die Verbindungen der Formel {ix) is aus Salzen der Verbindungen von Formel (IX) freigesetzt werden, indem man ein stöchiometrisches Äquivalent einer Base zu einem Säureadditionssalz oder Säure zu einem Salz der Verbindung (ix) mit einer Base in einem Lösungsmittel j bei einer Temperatur zwischen 0 und 500C zufügt.
ί Ein Salz der Verbindung von Formel (IX) kann auch in ein j
Salz z.B. durch übliche doppelte Umsetzung umgewandelt werj den, wodurch das Anion oder Kation des Ausgangssalzes durch j das pharmazeutisch annehmbare Anion oder Kation ersetzt wird ! 25 Die doppelte Umsetzung bedient sich gewöhnlich der Tatsache, daß sich das weniger lösliche Salz von dem stärker löslichen Salz bei einer Umsetzungstemperatur von 0 bis 500C abtrennt.
Ausführungsbeispiele
(Die Versuche beschreiben die Herstellung von Ausgangs» materialien.)
- 10.-Versuch I
100 g Natriumhydrid (50 %ige Dispersion in Mineralöl; 2,08 Mol) wurden zweimal mit 1 1 Hexan gewaschen und dann unter Stickstoff getrocknet. 1,5 1 trockenes Dimethylformamid wurden zugefügt und dann 400 ml Benzylalkohol mit solcher Geschwindigkeit zugegeben, daß die Temperatur unter 500C blieb. Die Zugabe erforderte 2 Stunden. 92,5 g (1 Mol) Epichlorhydrin wurden anschließend innerhalb einer halben Stunde unter Kühlen mit Eis eingetropft, um die Temperatur unter 400C zu halten. Dann wurde die Lösung 16 Stunden bei 210C und anschließend·2 1/2 Stunden bei 5O0C gerührt. Das Dimethylformamid wurde durch Abdampfen bei vermindertem Druck entfernt. Der ölige Rückstand wurde in.2,5 1 Diethyl ether gelöst. Die organische Lösung wurde mit 2 1 Wasser, . 2 1 2-Joiger Salzsäure, 2 1 1-tigern Natriumbicarbonat und 1 1 Kochsalzlösung gewaschen, über Natriumsulfat getrocknet und zu einem brauen Öl konzentriert. Nach Destillation erhielt man 147,8 g 1,3-Di-O-benzylglycerin (Kp, 170-1800C/ 1 ,3 mbar. ' Versuch II
Trockener gasförmiger Chlorwasserstoff wurde 11/2 Stunden in eine Lösung aus 15g (55 Milliinol) 1,3-Di-O-benzylglycerin aus Versuch-1 und 3,3 g- (1-10 Millimol) Paraformaldeiryd in 175 ml 1,2-Dichlorethan bei O0C eingeleitet._ Dann wurde die Lösung 21 Stunden bei 40C in einem verschlossensn. Kolben gelagert. Anschließend wurde die Lösung über Magnesiumsulfat unter Erwärmen auf 210C getrocknet, filtriert und zu 17,5 g 1,3-Di-0-benzyl-2-0-chlormethylglycerin konzentriert
Versuch III
Zu einer Lösung aus 17,5 g (55 Millimol) 1,3-Di-O-benzyl-2-0-chlormethylglycerinaus Versuch II in 400 ml Dimethylformamid bei O0C unter einem Trocknungsrohr wurden 6 g Natriumacetat zugefügt. Dann wurde die Lösung aus 21 C
erwärmt und magnetisch 15 Stunden gerührt. Das Lösungsmittel wurde durch Abdampfen unter vermindertem Druck entfernt und der ölige Rückstand in 454 g Diethylether gelöst. Die Etherlösung wurde einmal mit 750 ml Wasser, zweimal mit 250 ml Wasser und einmal mit 250 ml Kochsalzlösung gewaschen, über Natriumsulfat getrocknet und zu 19 g 2-0-Acetoxymethyl-1,3-di-O-benzylglycerin (Öl) konzentriert.
Versuch IV
Herstellung_von_N_i9-DiacetYlguanin .20 g (0,132 Mol) Guanin wurden mit 300 ml Essigsäureanhydrid vsr-ds ci.it - und die Mischung 16 Stunden zum Rückfluß erhitzt. Dann wurde die Mischung abgekühlt und das überschüssige Essigsäureanhydrid durch Abdampfen bei vermindertem Druck entfernt. Der Rückstand wurde aus Dimethylsulfoxid umkristallisiert und lieferte 25,6 g N2,9-Diacetylguanin.

15,61 g (66 Millimol) ]>r ,9-Diacetylguanin aus Versuch IV, 19 g (55 Millimol) 2-0-Acetoxymethyl-i,3-di-0-benzylglycerin aus Versuch III und 0,5g Bis-(p-nitrophenyl)-phosphat wurden zusammen mit 150 ml Diethylether gerührt. Das Lösungsmittel wurde durch Abdampfen entfernt und der Rückstand in einem Ölbad von 1750C 1 1/2 Stunden unter einem Stickstoffstrom erhitzt. Kolonnenchromatographie unter Eluieren mit 1:9 Methanol/Methylehchlorid und anschließende Umkristallisation aus Sthylacetat lieferten 4,76 g N2-Acetyl-9-(1,3-dibenzyloxy-2-propoxyme-thyl)-guanin;

Zu einer Lösung aus 4,62 g (9,67 Millimol) ir-Acetyl-9-(1,3-dibenzyloxy-2-propoxymethyl)-guanin in 150 ml Methanol plus 40 ml Wasser wurde 20-%iges Palladiumhydroxid-auf-Kohle als Aufschlämmung in 10 ml Wasser zugefügt. Die Mischung wurde in einer Parr-Hydriervorrichtung bei etwa 4,1
bar Wasserstoff 38 Stunden hydriert, dann durch Celite filtriert und zu einem weißen Feststoff konzentriert. Durch Umkristallisation aus Methanol/Ethylacetat erhielt man 1,4 g N2-Acetyl-9-(1 ,^-dihydroxy^-propoxymethyl)-guanin F. 205-2080C.
Die Mutterlauge wurde mit 10 % Palladium-auf-Kohle (1 g) in 150 ml Methanol oIus 50 ml Wasser bei etwa 4,1 bar 47

Stunden weiter reduziert. Die Gesamtausbeute an N~-Acetyl- i 9-(1,3-dihydroxy-2-propoxymethyl)-guanin betrug 2,11 g. I C) Herstellung_von_9-(1i3-DihYdroxY::2=2ro2OXYmethYl2-guanin
721,9 mg (2,4 Millimol) N2-Acetyl-9-(i,3-dihydroxy-2-prop- ! ox3?rnethyl)- guanin wurden mit 50 ml methanol is eher Ammoniaklösung (mit Ammoniak bei O0C gesättigtes Methanol) 17 Stunden bei 210C gerührt. Die Lösung wurde zu einem weißen Feststoff konzentriert und der Rückstand aus Methanol umkristallisiert; er lieferte 582,3 mg 9-0,3-Dihydroxy-2-

propoxymethyl)-guanin; F. 250 C (Zers.).
Claims (1)
- Erfindungsanspruch(VIII)in welcher R wie oben definiert ist und R' eine Schutzgruppe, wie eine Benzylgruppe, ist, die gegebenenfalls mit 1 oder 2 Alkoxygruppen, in denen Alkyl wie unten definiert ist, vorzugsweise Methoxy, oder 1 oder 2 niedrigen Alkylgruppen, wobei Alkyl eine gerade oder verzweigte Kohlenwasserstoffkette mit 1 bis 4 Kohlenstoffatomen ist, vorzugsweise Methyl, substituiert ist, dearalkyliert; oder- 14 -b) eine Verbindung der Formel (VIII) mit einer Lewis-Säure umsetzt; oderc) eine Verbindung der Formel (VIII) mit Essigsäure umsetzt; oderd) eine Verbindung der Formel (IX) in deren Salz um- j10 jwandelt; oder je) das Salz in die entsprechende freie Verbindung der Formel (IX) umwandelt.
