Abstract
Die Erfindung betrifft die Herstellung von Naphthalinderivaten der allgemeinen Formel I und pharmazeutisch annehmbaren Salzen davon. Die Naphthalinderivate (I) und deren Salze weisen eine ausgezeichnete hypolipaemische Aktivitaet auf und sind fuer die Behandlung oder Prophylaxe von Hyperlipidaemie und/oder Arteriosklerosis geeignet. 5. Verfahren nach Anspruch 4, gekennzeichnet dadurch, daß es zur Herstellung von i-iSADimethoxyphenylJ-S-hydroxymethyl^-hydroxy-y-methyl^-naphtoesäure-lacton oder pharmazeutisch annehmbaren Salzen davon angewendet wird.
Die Erfindung betrifft ein Verfahren zur Herstellung von Naphthalinderivaten, die wertvolle hypolipämische Mittel sind.
Hyperlipämie, wie Hypercholesterolämie, sind bekanntlich die Hauptrisikofaktoren für Arteriosklerose, einschließlich Atherosklerose, und als hyperlipämische Mittel sind schon Arzneimittel, wie Clofibrate (chemische Bezeichnung: 2-(4-Chlorophenoxy)-2-methylpropansäureethylester), Probucol (chemische Bezeichnung:
4,4'-((1-Methylethyliden)bis-(thio)bis(2,5-bis(1,1-dimethylethyl)-phenol)) und Cholestyraminharz, verwendet werden. Es ist bekannt, daß Cholesterin im Blutserum in unterschiedlichen Formen vorliegt, wie einem sehr niedrig-dichten Lipoprotein (VLDL)-cholesterin, niedrig-dichten Lipoprotein (LDL)-cholesterin und hoch-dichtem Lipoprotein (HDL)-cholesterin. In diesem Zusammenhang ist es auch bekannt, daß HDL eine therapeutische oder prophylaktische Wirkung auf Arteriosklerose hat, weil es die Wirkung, daß sich Cholesterin an der Arterienwand ablagert, während VLDL und LDL die Ablagerung von Cholesterin induzieren und dadurch ursächlich Arteriosklerose verursachen (Annals of Internal Medicine, Bd. 90, Seiten 85 bis 91 [1979]).
Es besteht deshalb auf dem Gebiet der Therapie oder Prophylaxe von Arteriosklerosis ein Bedürfnis, hypolipämische Mittel zur Verfugung zu stellen, welche das Gesamtserumcholesterinniveau erniedrigen und gleichzeitig das Serum-HDL-Cholesterinniveau erhöhen.
Der Erfindung liegt die Aufgabe zugrunde, ein Verfahren zur Herstellung von Verbindungen zu finden, die als hypolipämische Mittel zur Therapie oder Prophylaxe von Arteriosklerosis eingesetzt werden können.
Erfindungsgemäß wird die Aufgabe dadurch gelöst, daß zur Herstellung von neuen N phthalinderivaten oder eines pharmazeutisch annehmbaren Salzes davon der allgemeinen Formel I

R5 und R' Niedrigalkyl und R3, R4 und Ring A die vorher angegebenen Bedeutungen haben, reduktiv lactonisiert wird. Die reduktive Lactonisierung der Verbindung (II) oder einem Salz davon (z. B einem Alkalisalz, Erdalkalisalz oder quaternären Ammoniumsalz) kann auf übliche Weise erfolgen, z. B. indem man die Verbindung mit einem Reduktionsmittel in einem Lösungsmittel behandelt {z. B. Tetrahydrofuran, Ethylether, Dimethoxyethan) und zwar bei einer Temperatur zwischen O0C und Rückflußtemperatur, und dann.das Produkt mit einer katalytischer! Menge einer Säure behandelt. Als Reduktionsmittel kann man beispielsweise Borankomplexe (z. B. Boran-Methylsulfid-Komplex, Boran-Tetrahydrofuran-Komplex), Boran-Pyridin-Komplex), Natriumborhydrid-Bortrifluoridetherat-Komplex, Calciumborhydrid, Lithiumborhydrid und Lithiumalurriniumhydrid verwenden. Eine geeignete Säure ist beispielsweise eine anorganische Säure, wie Salzsäure oder Schwefelsäure, oder eine organische Säure, wie Ameisensäure, Essigsäure und Methansulfonsäure.
Wenn das so erhaltene Produkt wenigstens eine Pl' π Ί-niedrig-alkoxygruppe im Ring A hat, kann man gewünschtenfalls die Phenyl-niedrigalkoxygruppe durch katalytische Hydrie ung in eine Hydroxygruppe überführen. Die katalytische Hydrierung kann beispielsweise in Gegenwart eines Katalysators (wie Palladiumauf-Kohle) bei 10 bis 50 °C in einer Wasserstoff atmosphäre durchgeführt werden.
Das erfindungsgemäße Naphthalinderivat (I) kann als Arzneimittel entweder in freier Form oder in Form eines pharmazeutisch annehmbaren Salzes verwendet werden. Beispiele für pharmazeutisch annehmbare Salze sind Alkalisalze (beispielsweise Natriumsalz, Kaliumsalz), Erdalkalisalze (z. B. Calciumsalz), quPternäre Ammoniumsalze (z. B. Tetramethylammoniumsalz,
Tetraethylammoniumsalz) und dergleichen. Solche Salze erhält man einfach auf übliche Weise, z. B. indem man das Naphtalinderivat (I) mit einem alkalischen Mittel, wie einem Alkalihydroxid (z. B. Natriumhydroxid, Kaliumhydroxid) oder Erdalkakihydroxid (z. B. Calciumhydroxid) oder einem quaternären Ammoniumhydroxid (z. B. Tetrarnethylammoniumhydroxid oder Tetraethylammoniumhydroxid) und dergleichen in einem Lösungsmittel umgesetzt.
Typische Beispiele für Naphthalinderivate der vorliegenden Erfindung sind solche der Formel (I), in denen jeder der Reste R1 und R4 Niedrigalkoxy (z. B. Methoxy, Ethoxy, Propoxy, Isopropoxy, Butoxy) oder einer der Reste R3 und R4 ist ein Wasserstoffatom und der andere Niedrigalkoxy (z. B. Methoxy, Ethoxy. Propoxy, Isopropxy, Butoxy) bedeutet, und der Ring A ist ein unsubstituierter Benzolring, ein Benzolring, der mit einer Niedrigalkoylendioxygruppe (z. B. Methylendioxy, Ethylendioxy) substituiert ist oder ein Benzolring mil 1 bis 3 Substituenten, ausgewählt aus der Gruppe, bestehend aus Niedrigalkyl (z. B. Methyl, Ethyl, Propyl, Isopropyl, Butyl), Niedrigi Ikoxy (z. B. Methoxy, Ethoxy, Propoxy, Isopropoxy, Butoxy), Phenylniedrigalkoxy (beispielsweise Benzyloxy, Phenethyloxy), Hydroxy und Halogenatomen (z. B. ein Fluoratom, ein Chlorate.,,, ein Bromatom oder ein Jodatom). Eine besonders bevorzugte Verbindung ist i-fS^-Dimethoxyphcnyl^-Hydroxymethyl^-hydroxy-V-methyl^-naphtosäurelacton. Die Naphthalinderivate (I) oder pharmazeutisch annehmbare Salze davon weisen eine starke hypolipämische Aktivität auf und haben die besondere Eigenschaft, daß sie das Serum HDL-Cholesterin-Niveau erhöhen und gleichzeitig das Gesamtcholesterinniveau erniedrigen. Beispielsweise wurde die Wirkung einer Testverbindung (Dosis 100 mg-% in der Nahrung) auf das Gesamtcholesterinniveau und das Serum-HDL-Cholesterinniveau untersucht, indem man Ratten mit einer Diät ernährte, welche mit Cholesterin und Natriumcholat angereichert war. Dabei zeigte
!•(S^-Dimethoxiphenylj-S-Hydroxymethyl^-hydroxyJmethyl^naphtoesäurelacton der vorliegenden Erfindung eine 63%ige
und weisen im wesentlichen keine unerwünschten Nebenwirkungen, wie eine hepatitische Disfunktion, auf. Bei oraler

Deshalb sind die Naphthalinderiv&'o I) und deren Salze wirksame Mittel für die Be'.nndlung oder Prophylaxe von Hyperlipidämie (z. B. Hypercholesterolämie) oder / V skleross (einschließlich Atherosklerose, Mönkeberg's Sklerosis und Arteriolosklerosis) bei Warmblütern, einschließlich Mensci
Dio tägliche Dosis des Naphthalindei. a (I) oder eines pharmazeutisch annehmbaren Salzes davon kann in einem weiten Bereich variieren und hängt von der Schwere ( - Krankheit, dem Alter, dem Gewicht und dem Zustand des Patienten usw., ab, wobei bevorzugte tägliche Dosen im allgemein „n im Bereich von 1,5 bis 35 mg, insbesondere 5 bis 25 mg pro kg Körpergewicht liegen. Das Naphthalinderivat (I) oder ein pharmazeutisch annehmbares Salz davon kann oral oder parentheral Warmblütern, einschließlich Menschen, verabreicht werden, wobei man vorzugsweise die orale Route wählt. Man kann ein Naphthalinderivat (I) oder ein Salz davon in einer pharmazeutischen Zusammensetzung in Mischung mit einem pharmazeutisch annehmbaren Adjuvants oder Träger verwenden. Beispielsweise kann eine pharmazeutische Zusammensetzung für eine orale Verabreichung in fester Dosierungsform in Form von Tabletten, Pillen, Pulvern, Kapseln oder Granulaten vorliegen und es kann als pharmazeutisch annehmbares Adjuvants oder als Träger Calciumcarbonat, Calciumphosphat, Maisstärke, Kartoffelstärke, Zucker, Lactose, Talkum, Magnesiumstearat und dergleichen enthalten. Außerdem kann die pharmazeutische Zusammensetzung der festen Form auch noch Bindemittel, Verdünnungsmittel, Zerfallsmittel, Befeuchtungsmittel und dergleichen enthalten. Alternativ kann eine pharmazeutische Zusammensetzung für eine orale Verabreichung auch in einer flüssigen Dosierungsform, wie einer wäßrigen oder öligen Suspension, Lösung, als Sirup, Elexier oder dergleichen vorliegen. Geeignete Adjuvantien für eine flüssige Dosierungsform schließen flüssige Träger, Suspendiermittel, oberflächenaktive Mittel, sowie nicht-flüssige Träger und dergleichen ein. Weiterhin kann mon^die pharmazeutischen Zusammensetzungen für parenteral Verabreichungen als Injektionen oder Suppositorien formulieren. Die Injektionen können als Lösung oder Suspension vorliegen und können einen pharmazeutisch annehmbaren Träger, wie ein essentielles öl (z. B. Erdnußöl, Maisöl) oder ein aprotisches Lösungsmittel (z. B. Polyethylenglykol, Polypropylenglykol, Lanolin oder Kokosnußöl) enthalten.

Die Ums6tzung der Verbindung (III) mit der Verbindung (IV) oder (VII) kann in Gegenwart eines Alkyllithiums in einem Lösungsmittel bei einer Temperatur zwischen - 1000C und Rückflußtemperatur durchgeführt werden. Die Umwandlung des Diniedrigalkylacetals der Verbindung (V) in die freie Aldehydverbindung (V) kann durchgeführt werden, indem man das Di niedrigalkylacetal mit einer Säure (z. B. Salzsäure oder Trifluoressigsäure) in einem Lösungsmittel (z. B. wäßrigem Methanol oder wäßrigem Ethanol) bei 0 bis 300C behandelt.
Die nachfolgende Kondensationsreaktion des erhaltenen Produktes mit der Verbindung (Vl) kann in Gegenwart einer Säure in oder ohne einer.': Lösungsmittel erfolgen. Geeignete Säuren für diese Umsetzung sind beispielsweise anorganische Säurer, wie Salzsäure oder Schwofelsäure, und organische Säuren, wie Ameisensäure, Essigsäure, Paratoluolsulfonsäure oder Methansulfonsäure. Als Lösungsmittel sind Benzol, Toluol, Xylol oder Dimethylformamid beispielsweise geeignet. Vorzugsweise wird die Umsetzung bei einer Temperatur zwischen 0 und 15O0C, insbesondere bei 50 bis 1000C durchgeführt.
Praktische und derzeit bevorzugte Ausführungsformen der Erfindung werden in den nachfolgenden Versuchen und Beispielen gezeigt.
In der Beschreibung und in den Ansprüchen bedeuten „Niedrigalkyl", „Niedrigalkoxy" und „Niedriyalkylendioxy" jeweils Alkylgruppen mit 1 bis 4 Kohlenstoffatomen, Alkoxygruppen mit 1 bis 4 Kohlenstoffatomen und Alkylendioxygruppen mit 1 oder 2 Kohlenstoffatomen.
Wirkung auf das Gesamtcholesterinniveau und Serum-HDL-Cholesterinniveau Männliche SD-Ratten (Körpergewicht 110 bis 170 5, eine Gruppe bestand jeweils aus G Ratten) wurden ad libitum für 4 Tage mit einer Diät aus 2 W/W-% Cholesterin und 0,5 W/W-% Natriumcholat gefüttert. Dann wurden die Ratten ad libitum mit der gleichen Diät, enthaltend 100 mg-% bzw. 20 mg-% der Testverbindung, gefüttert. Weiterhin wurde eine Kontrollgruppo der Ratten mit einer Diät, welche die Testverbindung nicht enthielt, gefüttert. Drei Tage später wurden die Ratten mit Ether anästhesiert. Nachdem das Körpergewicht der Ratten gemessen war, wurde Blut aus der abaominalen Aorta gesammelt. Man ließ das Blut bei Raumtemperatur 1 Stunde stehen und zentrifugierte es dann. Dann wurde das Gesamtcholesterinniveau in dem so erhaltenen Serum enzymatisch nach der Methode, die in Clinical Chemistry, Bd. 20, Seite 470 (1974) beschrieben wird, gemessen. Serum-HDL-Cholesterin wurde von den anderen Arten des Cholesterin nach der Lipoproteinsedimentationsmethode unter Verwendung von Dextransulf.it (Canadian Journal of Biochemistry, Bd. 47, Seite 1043 (196bj) gesammelt und enzymatisch nach der vorerwähnten Methode gemessen Auf Basis der erzielten Ergebnisse wurden dann die Wirkungen der Testverbindungen auf das Serum-Gesamtcholesterinniveau und auf das Serum-HDL-Cholesterinniveau nach der folgenden Formel berechnet: (prozentuale Abnahme des Serum-Gesamtcholesterinniveaus)

Weiterhin wurde unmittelbar nach der Blutsammlung bei den vorerwähnten Versuchen die Leber der einzelnen Ratten herausgenommen und deren Gewicht gemessen. Das relative Lebergewicht wurde nach der nachfolgenden Formel untersucht und das durchschnittliche relative Lebergewicht wurde mit dem der Kontrollgruppe verglichen. Die bei den vorerwähnten Versuchen verwendeten Testverbindungen verursachten keine wesentliche Erhöhung des relativen Lebergewichtes.

0,7 g 1-(3,4-Dimethoxyphenyl)-2,3-bis(methoxycarbonyl)-4-hydroxy-7-methylnaphthalin wurden in 30 ml Tetrahydrofuran gelöst. 0,24 ml Boran-Methylsulfid-Komplex wurden zu der Lösung gegeben. Die Mischung wurde 4 Stunden unter Rückfluß behandelt. Nach dem Abkühlen wurde das Reaktionsgemisch unter vermindertem Druck bis zur Trockne eingedampft. Der Rückstand wurde in 300 ml Methanol, enthaltend Trifluoressigsäure, gelöst. Die Lösung ließ man bei Raumtemperatur 12 Stunden stehen und der kristalline Niederschlag wurde abfiltriert, wobei man 450 m!
i-(3,4-Dimethoxyphenyl)-3-hydroxymethyl-4-hydroxy-7-methyl-2-naphthoesäure-lacton als farblose Nadeln in einer Ausbeute von 76% erhielt.

4,5 g 1(3,4Dimethoxyphonyl)-3hydroxymethyl-4 hydroxy-6methoxy-7benzyloxy-2naphthoesäure lacton, erhalten gemäß Beispiel 6, werden in einer Mischung aus 200 ml Tetrahydrofuran und 200 ml Methanol gelöst. Die Lösung wird in Gegenwart von 3 g 10%igem Palladium auf-Kohle in einer Wasserstoffatmosphäro von 2,8 bar gerührt. Nach der Umsetzung wird der Katalysator abfiltriert und das Filtrat wird unter vermindertem Druck eingedampft. Die erhaltenen rohen Kristalle werden aus Methanol umkristallisiert, wobei man 3,2 g !-(SADimethoxyphenylJ-S-hydroxymethyl^J-dihydroxy-ö-methoxy^-naphthoesäure-lacton als farblose Mikronadeln erhält.

Referenzbeispiel
(1) Eine Lösung von 1,55 M n-Butyllithium in 430 ml Hexan wurde tropfenweise zu einer Lösung von 204,0 g 2-Bromo-3,4,5-trimethoxybenzadehyddimethylacetal in 800 ml Tetrahydrofuran gegeben. Die Zugabe erfolgte bei -70 bis -5O0C unter Rühren während etwa 15 Minuten. Die Mischung wurde 15 Minuten bei -70 bis -600C gerührt. Dann wurde eine Lösung «on 105,5 g 3,4-Dimethoxybenzaldehyd in 300 ml Tetrahydrofuran zu der Mischung bei -70 bis -500C während 15 Minuten zugegeben. Die Mischung wurde bei der gleichen Temperatur während 15 Minuten gerührt und dann zu 2 I Wasser gegossen. Zu der wäßrigen Mischung wurden 4 I Ethylacetat gegeben. Nach dem Schütteln wurde die organische Schicht von der Mischung abgetrennt, mit Wasser gewaschen, getrocknet und zur Entfernung von anorganischen Materialien filtriert. Die organische Schicht wurde unter vermindertem Druck eingedampft und das Lösungsmittel entfernt, wobei man 266 g '•(S^-Dimethoxy-a-hydroxybenzylJ-SAS-trimethoxybenzaldehyd-dirnethylacetal als gelben Sirup erhielt.

(2) 266 g 2-(3,4-Dimetho".<y-ahydroxybenzyl)-3,4,5-trimetnoxybenzaldehyddimethylacetal wurden in 95 ml Benzol gelöst. 95 ml Dimethylacetylendicarboxylat und 300 mg p-Toluolsulfonsäuremonohydrat wurden zu der Lösung gegeben. Ncch 2stündigem Rückfluß wurde die Mischung gekühlt und unter vermindertem Druck zum Entfernen des Lösungsmittels eingedampft. Zu dem Rückstand wurden 600 ml Methanol gegeben und die Mischung ließ man über Nacht bei -300C stehen. Der kristalline Niederschlag wurde abfiltriert und aus Ethyl/icetat umkristallisiert, wobei man 202 g i-IS^-Dimethoxyphenyl^.S-bislmethoxycarboiiyl^-hydroxy-ßJ.e-trimethoxynaphthalin als farblose Prismen erhielt.

Referenzbeispiel 9
(1) Eine Lösung aus 7 g 3,4-Dihydroxybenzaldehyd in 20 ml Dimethylformamid wurde tropfenweise zu einer Mischung aus 4,8 g 60%igem Natriumhydrid in 70 ml Dimethylformamid während eines Zeitraums von 15 Minuten unter Eiskühlung gegeben. Die Mischung wurde bei der gleichen Temperatur 15 Minuten gerührt. Dann wurden 50 g n-Propyljodid zugegeben. Nach 12stündigem Rühren wurde die Mischung unter Entfernung des Lösungsmittels eingedampft. Der Rückstand wurde mit Ether extrahiert und das Extrakt wurde mit Wasser gewaschen, getrocknet und anorganische Materialien wurden abfiltriert. Dann wurde das Lösungsmittel abgedampft, wobei man 8,5 g 3,4-Dipropoxybenzaldehyd als hellgelbes öl erhielt.

(1) Eine Lösung aus 1,55 M n-Butyllithium in 20 ml Hexan wurde tropfenweise zu einer Lösung von 9,24 g
N-(3,4-Methylendioxybenzyliden)cyclohexylamin in 100 ml Tetrahydrofuran gegeben. Die Zugabe erfolgte bei -70 bis -6O0C unter Rühren während 15 Minuten. Dann wurde die Mischung weitere 15 Minuten bei der gleichen Temperatur gerührt und es wurden 6,85 g einer Lösung aus 3,4-Dimethoxybenzaldehyd in 15 mi Tetrahydrofuran tropfenweise bei dergleichen Temperatur während eines Zeitraums von 15 Minuten zugegeben. Die Mischung wurde weitere 15 Minuten bei der gleichen Temperatur gerührt und dann zu einer Mischung aus 300 ml Wasser und 300 ml Ethylether gegossen. Din organische Schicht wurde von der Mischung abgetrennt, mit Wasser gewaschen, getrocknet und unter vermindertem Druck unter F^tfernung des Lösungsmittels eingedampft. Der erhaltene gelbe Sirup wurde säulenchromatisch über Kieselgel (Lösungsmittel: Benzol-Ethylether [4:1 ], gesättigt mit Wasser) behandelt und das Eluat wurde unter vermindertem Druck eir gedampft. Die erhaltenen farblosen Kristalle wurden aus einer Mischung aus Ethylacetat und Hexan umkristallisiert, wobei r.ian 8,7 g 3,4-Methy : 'idioxy-2-(3,4-dimethoxy-a-hydroxybenzyl)benzüldehyd als farblose Nadeln in einer Ausbeute von 69% erhielt.

Der kristalline Niederschlag wurde abfiltriert und aus einer Mischung aus
Tetrahydrofuran und Methanol umkristallisiert, wobei man 7,5 g

von 83% erhielt.

Claims (3)
- Patentansprüche:worin jedes von R3 und R4 ein Niedrigalkoxy oder eines von R3 und R4 ein Wasserstoffatom und das andere ein Niedrigalkoxy ist und Ring A ein substituierter oder unsubstituierter Benzolring ist, mit der Ausnahme, daß wenn der Ring A ein Bf>nzolring der Formelnoderist, jedes von R3 und R4 Niedrigalkoxy mit wenigstens 2 Kohlenstoffatomen bedeutet oder eines von R3 und R4 ein Wasserstoffatom und das andere Niedrigalkoxy bedeutet, oder eines pharmazeutisch annehmbaren Salzes davon, gekennzeichnet dadurch, daß man eine Verbindung der Forme; il..000 R-OORworin jedes R5 und RB ein Niedrigalkyl ist und R3, R4 und Ring A die vorher angegebenen Bedeutungen haben oder ein Salz davon, einer reduktiven Lactonisierung unterwirft und erforderlichenfalls das Produkt weiter in ein pharmazeutisch annehmbares Salz überführt.

Verfahren nach Anspruch 1, gekennzeichnet dadurch, daß (a) jedes von R3 und R4 ein Niedrigalkoxy oder eines von R3 und R4 ein Wasserstoffatom und das andere ein Niedrigalkoxy und Ring A ein Benzolring der Formeln-CC·ROR9OoderX0ist, worin R* ein Wasserstoffatom, ein Niedrigalkyl oder ein Niedrigalkoxy, Rb ein Niedrigalkyl, jedes von Rd, R", R' Niedrigalkyl, Rfl ein Wasserstoffatom oder ein Phenyl-niedrigalkyl und jedes Xa und Xb ein Halogenatom bedeuten; oder(b) jedes von R3 und R4 Niedrigalkoxy mit wenigstens 2 Kohlenstoffatomen oder eines von R3 und R4 ein Wasserstoffatom und das andere Niedrigalkoxy und der Ring A ein Benzolring der Formelnoderist, worin Rh ein Niedrigalkyl darstellt und Rb die vorher angegebene Bedeutung hat. 3. Verfahren nach Anspruch 2, gekennzeichnet dadurch, daß (a) jedes von R3 und R4 Niedrigalkoxy und der Ring A ein Benzolring der FormelnCH3OHOCHCH3OCH7OCH3OoderClClist, oder(b) jedes von R3 und R4 Niedrigalkoxy mit wenigstens 2 Kohlenstoffatomen oder eines von R3 und R4 ein Wasserstoffatom und das andere Methoxy und Ring A ein Benzolring der Formelnoder
- 4. Verfahren nach Anspruch 2, gekennzeichnet dadurch, daß sowohl R3 und R4 Methoxy und Ring A ein Benzolring der Formeln
