Abstract
Azetidine der allgemeinen Formel * in der R3 ein Amino-Radikal, ein Alkylamino-Radikal, ein Dialkylamino-Radikal, ein Cycloalkylamino-Radikal, ein Acylamino-Radikal, ein Alkylacylamino-Radikal, ein Aminomethyl-Radikal, ein Alkylaminomethyl-Radikal, ein Acylaminomethyl-Radikal oder ein Alkylacylaminomethyl-Radikal darstellt, worin jedes Acyl-Bruchstueck durch ein oder mehrere Halogenatome, insbesondere Fluor, substituiert sein kann; R1, R2, R4, R5 und R6 Wasserstoffatome sind oder ein niederes Alkyl-Radikal bedeuten, unter der Bedingung, dasz mindestens eines von ihnen ein niederes Alkyl-Radikal ist, sowie ihre Salze. Die Verwendung der Verbindung der allgemeinen Formel (I) erfolgt als Zwischenprodukte fuer die Herstellung von nuetzlichen pharmazeutischen Verbindungen, speziell mit antimikrobieller Wirkung. ihro Herstellung und Verwendung als Zwischenprodukte für dio Herstellung von biologisch wirksamem Substanzon. Es sind Vcrbinduntl&n dor allgomoinon Formol (II)

bekannt. Sie find in dem europäischen Patont EP 88/403352.3 und in den französischen Potenten FR 89/03459 und FR 89/05129 beschrieben. Einige von ihnen sind im Begriff, wegen ihror ausgezeichneten antimikrobiollen Eigenschaften weitorentwickelt zu worden.
Aus der wissenschaftlichen Literatur sind einigo Derivate der allgemeinen Formol (I) bekannt, in der Π, ein Amino-Radikal, ein Alkylamino-Radikal, ein Qialkylamino-Radikal. ein Aminomothyl-Radikal odor oin Alkylaminomothyl-Radikal darstellt, jodoch existiert kein Beispiel bol don Verbindungen dor Formel (I), worin Hj dio erwähnte Bodoutung besitzt und mindestens einer der anderen Substiluenten gleichzeitig ein niederes Alkyl-Radikal ist. Die in der Wissenschaft bis houte beschriebenen aminosubstituierten Verbindungen der allgemeinen Formol (I) finden sich in Chem. Der Erfindung llogt rllo Aufgabe zugrunde, Aiotldlne der allgemeinen Formol (I), ihro Horstollung und Vorwondung als Zwischenprodukte für die Horstollung von biologisch wirksamem Substanzen so auszubilden, daß olnfacho und offoktivo Zwischenprodukte sowio Synlhosowogo für dio Vorbindung dor allgomoinon Formol (II) orhalton worden, dio dio 1-Azotidinyl-Gruppe clor Formol (III)

Gemäß dor vorliogündon Erfindung wurdon Azetidine clor allgomoinon Formol (I)

Dio neuon Azetidine der allgomeinon Formel (I) können je nach Anzahl, Boschaffonhoit und relativor Position dor Sustituontcn bis zu drei chiralo Zentren, jeweils mit einer „R"- und ,^"-Konfiguration, aufwoison.
Die Verbindungen der allgemeinen Formol (I), in der Ri, R2, R3, R4, R» und R0 die obonorwähnto Bodoutung besitzen, sind Produkto, dio durch Substitution von R3 mit oinem adäquaten Nucleophil orhalton werden, wonn R3 eine guto „Abgangs-Gruppo" darstellt. Dioso Substitution ist hinsichtlich dor Ausbouto sehr vorteilhaft wcgon der Anwesenheit der Bonzhydryl-Gruppe als Schutzgruppe für das Azotidin in Position 1. Außerdem ist die Benzhydryl-Gruppe obonfalls sehr vorteilhaft, wenn es um das Abspalton dor Schutzgruppe geht, was zu don Azetidinon dor allgemeinen Formel (IV)

führt, die ihrerseits notwendig sind für die Herstellung der Verbindungen der allgomoinon Formol (II), wio in den Patonten EP 324298, EP 90/400684.8 und EP 90/401036.0 boschriobon.
Ein weiterer zusätzlicher Vorteil der Verwendung dos Benzhydryl-Radikals als Schutzgruppe bei den Verbindungen der allgemeinen Formel (I) besteht in dem Fall, wo es sich um chiralo Kohlonstoffo handelt, da man eine Retontion der Konfiguration beobachtet, die die Bildung von storeoisomeren Mischungen verhindert, wonn man später die obenerwähnten Substitutions-Reaktionen vornimmt. Außerdem eignen sich die Verbindungen der allgemeinen Formel (I) sohr gut, wonn man die Spaltung racemischer Mischungen mit Hilfo von Enzymen oder enantiomerisch roinen organischen Säuren durchführen will. Die vorliegende Erfindung bezieht sich auch auf die Horstollung der Verbindungen der allgemeinen Formel (I). Die neuen, den Gegenstand der Erfindung bildonden Derivate können gemäß dem Schema 1 hergestellt werden, das auf allen Wegen zu den Zwischenprodukten der allgemeinen Formel (Xl) führt, dio ihrerseits durch geeignete Reaktionon dio Verbindungen der allgemeinen Formel (I) ergeben. In den Verbindungen der Formel (V) bis (XIII) des Schemas 1 besitzen die Radikale R,, R2, R4, R5 und Re die obenerwähnten Bedeutungen, X ist oin Chloratom, ein Bromatom, oin Hydroxyl-Radikal, oin Acotyloxy-Radikal, oin Methylsulfonyloxy-Radikal, ein para-Toluensulfonyloxy-Radikal, ein para-Nitrobonzoyloxy-Radikal, ein 2-Naphthalonsulfonyloxy-Radikal, ein tert-Butyldiphonylsilyloxy-Radikal oder ein tert-Butyldimothylsilyloxy-Radikal, Y stellt oin Chloratom odor ein Bromatom dar und Z bedeutet ein Methylsulfonyloxy-Radikal, ein para-Toluensulfonyloxy-Radikal oder ein Cyano-Radikal.

h 1) Die Vorbindungon dor allgomoinon Formol (XII) in dor Z oin Mothylsulfonyl-Radikal oder oin paro-Toluensulfonyloxy-Rndikal darstellt, ergeben sich durch Roaktion der Verbindungen dor allgomoinon Formel (Xl) mit dem Chlorid von jowoils Mothansulfcnsäuro odor para-Toluensulfonsäuro. Dio Roaktion orfolgt vorzugsweise in Anwosonhoit von chlorierten Lösungsmitteln wie Mothylonchlorid odor Chloroform, tortiäronAminon wio Pyridin odor Triethylamin odor auch in Mischungen dieser Lösungsmittel. Dio Roaktion findot boi Tomporaturon zwischon -30°C und 40°C statt und während einor Dauer zwischon 1 und 24 Stunden.
h 2) Die Verbindungen der allgemeinen Formol (XII), in dor Z ein Cyano-Radikal darstellt, orgobcn sich durch Roaktion der Vorbindungon dor allgemeinen Formel (XII), in denen Z ein Mothylsulfonyloxy-Radikal odor ein para-Toluonsulfonyloxy-Radikal ist, mit Natriumcyanid. Dio Roaktion orfolgt unter Verwendung oinos gooigneton Lösungsmittels wio Dimethylformamid oder Dimcthylsulfoxid. Man rührt dazu 3 bis 8 Stunden lang bei oinor zwischon 50cC und 100°C schwankenden Temperatur.
i 1) Die Verbindungen der allgemeinen Formol (I), in dor Rj oin Amino-Radikal, oin Alkylamino-Radikal, oin Dialkylamino-Radikal oder oin Cycloalkylamino-Radikol darstellt, ergobon sich durch Roaktion oinor Verbindung dor allgemeinen Formol (XII), in dor Z oin Methylsulfonyloxy-Radikal oder oin para-Toluensulfonyloxy-Radikal ist, mit jowoils Ammoniak, oinom Alkylamin, oinom Dialkylamin oder oinom Cycloalkylamin. Dio Reaktion orfolgt In einem geeigneten Lösungsmittel bei Temperaturen zwischen 4O0C und 120°C und einer Dauer zwischen 1 und 48 Stundonsowio toils unter atmosphärischem Druck und teils in einem geschlossenen Behälter. Dio geeigneten Lösungsmittel sind Wassor, aprotische dipolaro Lösungsmittel wio Dimethylformamid odnrDimethylsulfoxid, Alkohole wie ,.thanoloderlsopropanol. Ether wio Totrahydrofuran oder Dioxan oder auch oino Mischung von zwoi der genannten Lösungsmittel.
12) Die Verbindungen dor allgemeinen Formol (I), in dor Rs oin Aminomothyl-Radikol darstellt, orgobon sich durch Reduktion einer Verbindung der allgemeinen Formel (XII), in der Z oin Cyano-Radikal ist. Dio Reduktion orfolgt vorzugsweise mit Hilfo eines Motall-Hydridos wie Aluminiumhydrid odor Lilhiumhydrid in EtIiC-, wie Diethylether oder Tetrahydrofuran. Die Temperatur stellt sich während der Zugabe des Nitrils zwischen 250C und 40°C oin und später rührt man noch 12 bis 24 Stundon lang bei Umgebungstemperatur (25°C).
13) Die Verbindungen der allgemeinen Formel (I), in der R3 oin Acylamino-Radikal, oin Alkylacylamino-Radikal oder oin Acylaminomothyl-Radikal darstellt, ergeben sich durch Reaktion der Verbindungen der allgemeinen Formol (I), in der Rj jeweils ein Amino-Radikal, ein Alkylamino-Radikal oder ein Aminomethyl-Radikal ist, mit einom Carbonsäuro-Chlorid oder einem Carbonsäure-Chlorid oder einem Carbonsäure-Anhydrid. Die Roaktion erfolgt vorzugsweise in einem aromatischen Kohlenwasserstoff wie Benzen oder Toluon, in einer chlorierton Verbindung wio Methylonchlorid odor Chloroform odor auch in einer Mischung dieser Lösungsmittel. Außerdem ist die Anwesenheit einer anorganischen Base wio Natriumcarbonat oder oinor organischen wie Pyridin oder Triethylamin wünschenswert. Dio geeigneten Temperaturen schwankon zwischen O0C und 30'C und die Dauer zwischen 1 und 4 St jndon.
ί<1) Dio Vorbindungon dor allgomoinon Formol (I), in dor Rj ein Alkylocylaminomothyl-Radikel darstellt, orgobon sich durch Alkylierung dor Verbindungen dor tllgemolnon Formol (I), in dor Rj oin Acylaminomothyl-Radikal ist, untor Vorwondung oinos Alkyl-Halogonidos als Alky liorungsmitlol. 15) Dio Verbindungen der ollgomoinen Formol (I), in dor R1 oin Alkylominomothyl-Radikal darstellt, orgobon sich durch Hydrolyse) dor Vorbindungon der ollgomoinon Formol (I), in oor Rj oin Alkylacylaminomothyl-Radikel ist. Dioso Hydrolyso orfolgt im bnsischon Modium, vorzugsweise mit Hilfo oinor Alkalibnso wio Natriumhydroxid odor Kaliumhydroxid boi Tomporaturon zwischon 40 und 100"C.
Dio don Gogonstand dor vorliogondon Erfindung bildondon Vorbindungon dor allgomoinon Formol (I) woison clon Vortoil auf, sohr stabil zu so in und siel) einfach in dio Azotidino dor ollgomoinon Formol (IV) umwdndoln zu lassen, dio ihrorsoits notwondigo Produkte für dio Synthoso dor Verbindungen djr allgomoinon Formol (II) sind, dio oino antimikrobiollo Wirkung aufwoison. Die Roaktion, dio die Umwandlung dor Vorbindungon dor ollgomoinon Formol (I) in dio Vorbindungon dor allgomoinon Formol (IV) ermöglicht, bostoht in oinor Hydrogenolyse untor Vorwendung oinos Palladium-Katalysators, vorzugsweise Palladium-Hydroxid. Dio Roaktion orfolgt in oinom Alkohol wio Mothanol odor Ethanol boi oinom Wnssorstoff-Druck zwischon 1.1 at und 20at und gooignoton Tomporaturon, dio zwischon 200C und 7O0C schwankon. Dio Erfindung wird nachstohond on oinigon Boispiolon näher orläutort.
In don folgondon Boispiolon wird dio Horstollung der orfindungsgomäßon nouon Dorivato boschriobon. Dio angogobonon Goispielo solion jedoch in koinor Woiso don Rnhmon dor Erfindung oinschränkon.
α) i-Chlor^.Sepoxy^-mothylpropan
von 1O0C ab und setzt dann 50%igo Natronlauge (3MoI) mit oinor solchon Geschwindigkeit hinzu, daß dio Tomporatur boi etwa250C bleibt. Anschließend rührt man noch 2 Stundon lang, tronnt dio untoro organische Phaso ab, trocknot mit Magnesiumsulfat(2Og), dampft oin und erhält 266g (84%) dos rohen Produktos. Die Extraktion dor wa' ßrigon Phaso mit Chloroform (250 ml) ergibtzusätzlich 50g (12%). Das Produkt kann direkt in dor nächsten Stufo vorwondot worden, es is' jodoch vorteilhaft zu dostillioron,damit dio letzten Spuren dos Succinimide (Siedepunkt 65°C/40 Torr) ontfornt wordon.

erhält 28,8g (85%) I-Dihonylmothyl-Shydroxy-Smothylozotidin-Hydrochlorid vom Schmelzpunkt 187-1970C.

|) 1-Diphenylmothyl-3-hydroxy-3-mothylazotldin
Zu einer Lösung von 3,05g (12,8mMol) 1-Diphonylmothyl-3-azotidinon in 61 ml auf oino Tomporatur von O0C gokühltom wasserfreiem Ether gibt man tropfenweise 1,41 g Mothyllitr ium (64mMol) in 140ml wassorfroion Ether und hält dio Reaktion 2 Stunden lang boi O0C aufrecht. Dann gibt man tropfonwoisn Wasser hinzu. Man wäscht die organische Phase mit Wasser, trocknet mit wassorfreiom Natriumsulfat und dampft bis zur Trockne oin. Anschließend löst man don ontstandonen Foststoff in Methanol, gibt gesättigtos Chlorwasserstoff/Mothanol hinzu und dampft bis zur Trockno ein. Man wäscht don orhaltonon Feststoff mit Aceton, filtriert und erhält 3,12g (84%) 1 -Diphonylmothyl-S-hydroxy-S-methylazotidin-Hydrochlorid vom Schmelzpunkt 187-1970C.
h) 1-Diphonylmethyl-3-mothyl-3-mothylsulfonyloxyazotldin
gekühltem Pyridin gibt man tropfenweise 5,3g (46,8mMol) Mothansulfonylchlorid. Man hält die Temperatur eino Stunde langauf -2O0C und läßt sio dann für 12 Stunden auf 40C ansteigen. Dann gießt man auf oino Wasscr-Eis-Mischung, filtriert dengebildeton Niederschlag ab und wäscht ihn mit Wasser. Man löst don Foststoff in Mothylenchlorkl, dokantiert das verbliebene
rekristallisiert man don erhaltenen Foststoff mit oinor Mcthylonchlorid-Hoptan-Mischung und erhält 7,2g (70%) 1-

Man gibt 0,16g (3,6 mMol) Natriumhydrid (55%ig) zu oinor Lösung von 1,3g (3,6 mMol) 1-Diphonylmothyl-3-mothyl-3-trifluoracetylamino-mothylazetidin in 40ml Dioxon und 10ml Dimethylformamid und rührt zwei Stundon lang boi einer Temperatur zwischon 60 und 70"C. Dann kühlt man ilio Lösung auf Umgobungstomporatur ab, gibt 0,73g (4,6 mMol) Ethyliodid hinzu, rührt 4 Stundon lang bei oinor Temperatur von 700C, dampft bis zur Trockno oin, löst don Rückstand in Chloroform, wäscht mit Wassor, trocknot mit wasserfroiom Natriumsulfat, dampft oin und erhält 1,1g (79%) 1 -Diphonylmothyl-3-mothyl-3-|(N-olhyl)· trifluoracotylaminomothyll-azotidin. Man löst dioso Vorbindung in Ethanol und gibt gesättigten Chlorwassorstoff/Ethylothor hinzu. Dann läßt man kristallisieren, filtriert und orhält 1-Diphonylmolhyl-3-mothyl-37(N-othyl)-trilluorncotylaminor"notliyl/· azotidin-Hydrochlorid vom Schmolzpunkt 191 bis 1940C

Claims (5)
- in dorR3 ein Amino-Radikal, oin Alkylamino-Radikal, oin Diolkylamino-Radikal, oin Cycloalkylamino-Radikal, oin Acylamino-Radikal, oin Alkylacylamino-Radikat, oin Aminomothyl-Radikat, oin Alkylaminomethyl-Radikal, oin Acylaminomothyl-Radikal odor oin Alkylacylaminomothyl-Radikal darstellt, worin jedes Acyl-Bruchstück durch ein odor mohroro Halogonatomo, insbosondero Fluor, substituiert sein kann;Ri, R2, FU/ R5 und R6 Wasserstoffatome sind odor ein niederes Alkyl-Rodikal bedeuton, unter der Bedingung, daß mindestens eines von ihnen ein niederes Alkyl-Radikal ist, sowie ihre Salze. :?. Verbindungen der «ilgemoinen Formel (I) gemäß Anspruch 1, ausgowählt aus dor folgondon Gruppe:- 3-Amino-1-diphonylmethyl-3-mothylazotidin;- trans-3-Amino-1-diphenylmethyl-2-methylazetidin;- 1-Diphenylmethyl-3-othylarninomethyl-3-mothylazotidin;- 1-Diphenylmothyl-3-/N-(othyl)-trifluoracotylaminomothyl/-3-mothylazetidin;- i-Diphonylmothyl-S-methyl-S-trifluoracotylamino-mothylazetidin;- 3-Aminomethyl-1-diphenylinothyl-3-methylazetidin;- 1-Diphenylmethyl-3-methyl-3-methylaminoazotidin;- S-Dimethylamino-i-diphonylmothyl-S-mothylazetidin;- trans-1-Diphenylmothyl-2-mothyl-3-methylaminoazotidin;- trans-3-Dimethylamino-1-diphonylmothyl-2-rnothylazotidin;- trans-3-Aminomethyl-1-diphonylmethyl-2-methylazotidin;- trane-i-Diphenylmethyl^-methyl-S-trifluoracetylamino-mothylazetidin;- trans-1-Diphenylmethyl-3-/N-(othyl)-trifluoracetylamino-methyl/-2-methylazetidin;- cls-S-Amino-i-diphenylmethyl^-methylazetidin;- r-3-Amino-3,trans-2-dimethyl-1-diphenyl-methylazetidin;- i-Diphenylmethyl-S-methyl-S-trifluoracetylaminoazetidin;- S-Acetylaminomethyl-i-diphenylmothyl-S-methylazetidin;- cls-S-Amino-i-diphenylmethyl^-ethylgzetidin;- 3-Amino-2,2-dimethyl-1-diphenyl-methylazetidin;- (2R,3R)-3-Amino-1-diphenylmethyl-2-methylazetidin;- (2S,3S)-3-Amino-1-diphenylmethyl-2-methylazetidin;- (2R,3S)-3-Amino-1-diphenylmethyl-2-methylazetidin;- (2S,3R)-3-Amino-1-diphenylmethyl-2-methylazetidin.3. Verfahren zur Herstellung der Verbindungen der allgemeinen Formel (I) nach einem der Ansprüche 1 und 2, dadurch gekennzeichnet, daß man eine der folgenden Operationen durchführt:- Substitutions-Reaktion einer Verbindung der allgemeinen Formel (XII) in der Z ein Mothylsulfonyloxy-Radikal oder ein p-Toluonsulfonyloxy-Radikal darstellt, mit Ammoniak, einem Alkylamin, einem Dialkylamin oder einem Cycloalkylamin;- Reduktion einer Verbindung der allgemeinen Formel (XII), in der Z eine Cyano-Gruppe bedeutet, insbesondere mit Hilfe eines Metall-Hydrides;- Acylierung einer Verbindung der allgemeinen Formel (I), in der R3 ein Amino-Radikal, ein Alkylamino-Radikal odor ein Aminomethyl-Radikal darstellt, mit einem Carbonsäure-Chlorid oder-Anhydrid;- Alkylierung der Vorbindungen dor allgemeinen Formel (I), in dor R3 einAcylaminomothyl-Radikal bodoutot, durch Reaktion mit oinom Alkyl-Hologonid;Hydrolyse der Verbindungen der allgemeinen Formel (I), in der R3 oin Alkylacylaminomothyl-Radikal ist, vorzugsweise im basischen Medium.
- 4. Verfahren nach Anspruch 3, dadurch gekennzeichnet, duß die Verbindungen dor allgomoinon Formel (XII), in der Z ein Methylsulfonyloxy-Radikal oder oin p-Toluonsulfonyloxy-Radikal darstellt, durch Reaktion dor Vorbindungen der allgemeinen Formel (Xl) mit Mothansulfonsäure-Chlorid odor p-Toluensulfonsäure-Chlorid hergestellt werden.
- 5. Vorfahren nach Anspruch 3, dadurch gekennzeichnet, daß dio Verbindungen der allgemeinen Formel (XII), in der Z ein Cyano-Radikal bedeutet, durch Reaktion der Verbindungen der allgemeinen Formel (XII), in der Z ein Mothylsulfonyloxy-Radikal oder ein p-Toluensulfonyloxy-Radikal ist, mit Natriumcynnid hergestellt werden.
- 6. Vorwendung dor Verbindungen der allgemeinen Formel (I), gomäß Anspruch 1 und 2, als Zwischenprodukte für die Herstellung von nützlichon pharmazoutischon Verbindungen.
- 7. Verwendung der Verbindungen der allgemeinen Formol (I) gomäß Anspruch 6, als Zwischenprodukte für die Hnrstollung von Dorivaton dor Chinolone, Naphthyridine, Pyridobonzoxozino, Isothiazolochinolino, Thiozotochinolono, Benzochinolizine, Bonzothiazolochinolono, Pyridobonzothiazino, Bonzoxazolochinolono und Epoxymothanothiazolochinolone.
