Opfindelsen angår en analogifremgangsmåde til fremstilling af hidtil ukendte N-dihydrothiazolyl-3-quinolincar-boxamidderivater.
Fransk patentskrift nr. 2.340.735 beskriver en stor 5 familie af n-substituerede derivater af 4-hydroxyquinolin-3--carboxamid, hvis quinolinkerne kan være substitueret i 5-, 6-, 7- eller 8-stillingen med forskellige substituenter. Desuden kan carboxamidgruppen bære forskellige heterocykliske grupper, f.eks. thiazolyl og 4,5-dihydrothiazolyl.
10 Det eneste derivat, som beskrives specifikt i dette patentskrift, og hvor nitrogenet i amidgruppen er substitueret med en 4»5-dihydrothiazolylgruppe, omfatter en trifluorme thylgruppe i 8-stillingen i quinolinet. Dette derivat har smertestillende virkning, men har ikke nogen angstdæmpende 15 virkning.
Ved· undersøgelser vedrørende denne række har det nu vist sig, at en gruppe af derivater af 4-hydroxyquinolin-3--carboxamid, hvor amidgruppen er substitueret med 4,5-dihydrothiazolyl, og hvor quinolinkemen ikke er substitueret eller 20 er substitueret i 6- eller 7-stillingett, hvilke derivater hverken er beskrevet eller specifikt foreslået i det nævnte franske patentskrift, ikke har nogen smertestillende virkninger, men derimod har en udtalt angstdæmpende virkning. Disse derivater udmærker sig ved en kraftig affinitet til benzo-25 diazepinreceptorerne« Således angår opfindelsen en analogifremgangsmåde til fremstilling af hidtil ukendte N-dihydrothiazolyl-3-quinolincarboxamidderivater samt deres famaceutisk acceptable additionssalte med uorganiske eller organiske syrer,

Blandt de forbindelser, som fås ved fremgangsmåden ifølge opfindelsen, skal især nævnes derivaterne svarende til formlen I ovenfor samt deres farmaceutisk acceptable additionssalte med uorganiske eller organiske syrer, og som udmærker sig ved, at R betegner hydrogen eller methyl, R^ betegner hydrogen eller chlor eller methoxy, eller i 25 6-stillingen betegner fluor eller methyl, ethyl, isopropyl, methylthio eller trifluormethyl.
Blandt disse sidste skal nævnes de derivater, som udmærker sig ved, at R betegner hydrogen, og specielt N-(4,5-dihydrothiazol-2-yl)-4-hydroxy-3-quinolincarboxamid, 30 N-(4,5-dihydrothiazol-2-yl)-6-ethyl-4-hydroxy-3-quinolin- carboxamid, N-(4,5-dihydrothiazol-2-yl)-6-chlor-4-hydroxy--3-quinolincarboxamid og N-(4,5-dihydrothiazol-2-yl)-4--hydroxy-2-methyl-3-quninolincarboxamid samt deres farmaceutisk acceptable additionssalte med uorganiske eller or-35 ganiske syrer.

hvor R-og R^ har samme betydning som ovenfor, som man isolerer og om ønsket omdanner til et farmaceutisk acceptabelt addi-20 tionssalt.
Omsætningen af et acylchlorid eller -bromid med formlen II med 2-aminothiazolin kan udføres i indifferente opløsningsmidler eller suspensionsmilieuer såsom lavmolekylære aliphatiske ketoner, dioxan, dimethylformamid, benzen, toluen 25 eller alkylhalogenider. Man kan med fordel arbejde i nærværelse af hydrogenhalogenidbindende midler såsom alkalimetalhydrox- . ider som kaliumhydroxid, alkalimetalcarbonater som kaliumcar-bonat, alkalimetalbicarbonater som natrium- eller kaliumbicar-bonat, alkalimetalalkoholater som natriumethylat eller for-30 trinsvis tertiære aminer såsom trialkylaminer eller pyridin.
Omsætningen af en syre med formlen II med 2-aminothia-zolyl udføres med fordel i nærværelse af dicyclohexylcarbo-diimid i dimethylformamid.
Man arbejder med fordel ud fra en alkylester med form- 35 150070 4 len II, navnlig en ethylester. Man arbejder da under tilbagesvaling af et opløsningsmiddel såsom de ovennævnte med et højt kogepunkt på f.eks, 50-150°c, idet tilbagesvalingen normalt opretholdes i 12-48 timer, 5 Seaktionen udføres fortrinsvis i nærværelse af et kon densationsmiddel såsom et aluminiumalkylderivat, f,eks. tri-methylaluminium og navnlig triisobutylaluminium.
Man kan ligeledes med fordel arbejde i nærværelse af spor af et alkalimetalhydrid såsom natriumhydrid, , 10 Forbindelserne med formlen II er kendte eller kan fremstilles som angivet i fransk patentskrift nr, 2,340,735.
De hidtil ukendte derivater af H-dihydrothiazolyl-3--quinolincarboxamid med formlen I har basisk karakter. Man kan med fordel fremstille deres additionssalte, idet man i 15 praktisk taget støkiometriske mængder omsætter en uorganisk eller organisk syre med disse derivater. Saltene kan fremstilles uden isolering af de tilsvarende baser.
De ved fremgangsmåden ifølge opfindelsen fremstillede derivater har meget interessante farmakologiske egenskaber.
20 le har navnlig en bemærkelsesværdig affinitet til benzodiaze-pinreceptoreme.
De har en kraftig angstdæmpende virkning.
Disse egenskaber er illustreret nedenfor i den eksperimentelle del.
25 Disse egenskaber retfærdiggør anvendelsen af deriva terne af R-dihydrothiazolyl-3-quinolincarboxamid med formlen I samt deres farmaceutisk acceptable salte som lægemidler.
Blandt lægemidlerne skal især nævnes de hidtil ukendte derivater af N-dihydrothiazolyl-3-quinolinearboxamid med 30 formlen I, hvor R betegner hydrogen eller methyl, R-^ betegner hydrogen, chlor eller methoxy, eller R^ i 6-stillingen betegner fluor eller methyl, ethyl, isopropyl, methylthio eller trifluormethyl, samt deres additionssalte med farmaceutisk acceptable syrer, 35 Blandt disse skal især nævnes sådanne med formlen I, hvor R betegner hydrogen, samt deres additionssalte med farmaceutisk acceptable syrer.

Blandt disse sidste skal især nævnes følgende: - N-(4,5-dihydrothiazol-2-yl)-4-hydroxy-3-quinolin-carboxamid, - N-( 4 ,5-dihy d rothiaz ol-2-y1)-6-e thy1-4-byd roxy-3-5 -quinolincarboxamid, - N-(4»5-dihydrothiazol-2-y1)-6-chlor-4-hydroxy-3--quinolincarboxamid samt deres additionssalte med farmaceutisk acceptable syrer. Især skal nævnes ligeledes - N-(4f5-dihydrothiazol-2-yl)-4-hydroxy-2-methyl-3-10 -quinolincarboxamid samt additionssaltene deraf med farmaceutisk acceptable syrer.
De omhandlede lægemidler finder f.eks. anvendelse ved behandlingen af ængstelsestilstande såsom kronisk ængstelse, eventuelt i forbindelse med søvnløshed eller adfærd s -15 forstyrrelser, angst hos voksne og børn eller som kompletterende middel ved behandling med neuroleptiske midler eller antidepressionsmidler i tilfælde af psykotiske eller depressive tilstande.
Ben normale dosis, som varierer efter det benyttede 20 derivat, patienten og den pågældende lidelse, kan f.eks. være fra 1 mg til 100 mg pr, dag ad oral vej hos mennesker af derivatet ifølge eksempel 1 til behandling af kronisk ængstelse,
Forbindelserne med formlen I samt deres additionssal-25 te med farmaceutisk acceptable syrer kan benyttes til fremstilling af farmaceutiske præparater, der indeholder dem som aktiv bestanddel.
Som lægemidler kan derivaterne med formlen I og deres additionssalte med farmaceutisk acceptable syrer inkorporeres 30 i farmaceutiske præparater beregnet til oral eller parenteral indgift.

Eksempel 1, U-(4.5-dihydrothia2ol-2-yl)-4-hydroxy-3-quinolincarboxamid og dets hydroohlorid.
Man omrører 37,4 g 2-aminothiazolin i 800 ml methylen-5 chlorid, tilsætter i løbet af 20 minutter ved 8-10°0 167 ml 25$'s toluenopløsning af triisobutylaluminium, omrører yderligere 30 minutter ved 8°C, tilsætter 15,89 g 3-carbethoxy-4--hydroxyquinolin, omrører 30 minutter, tilbagesvaler i 16 timer, inddamper til tørhed under formindsket tryk, optager i 10 1 liter 1 li saltsyre, filtrerer, isafkøler i 16 timer, frafiltrerer udfældningen og vasker den med vand. Man optager moder-ludene, gør dem alkaliske med natriumcarbonat, filtrerer udfældningen, vasker den med vand og tørrer den. Man får 11,7 g af den forventede base.
15 Dannelse af hydroohloridet i Man opløser forbindelsen ovenfor i 600 ml ethanol, filtrerer, tilsætter 8 ml ethanolisk 5,3 N saltsyre, isafkøler, filtrerer, tørrer og får 8,5 g af den forventede forbindelse. Smp. ca, 300°c.

Eksempel 2.
Hydroohlorid et af N-(4.5-dih.vdrothiazol-2-yl)-6-eth.vl-4-25 -hydroxy-3-quinolinoarboxamid.
Man omrører 10,2 g 2-aminothiazolin i 500 ml methy-lenchlorid, tilsætter i løbet af 10 minutter ved 7-10°C 45,36 ml 1,1 M toluenopløsning af triisobutylaluminium, omrører yderligere 1 time ved 7-10°C, tilsætter 4,88 g 6-ethyl-30 -4-hydroxy-3-quinolincarboxylsyreethylester, opvarmer til tilbagesvaling i 16 timer, inddamper til tørhed under formindsket tryk, optager i 100 ml 1 I saltsyre, opvarmer 2 timer til 70°0 og behandler med aktivkul. Filtratet krystalliserer, og man suger fra, vasker med 1 li saltsyre, tørrer under for-35 mindsket tryk, omkrystalliserer af eddikesyre og får 2,2 g af den forventede forbindelse. Trin A: 4-ethylphenyIaminomethylenpropandisyreeth.vlester.
5 Man blander under omrøring og tinder en indifferent gasstrøm 50 g 4-ethylanilin og 89 g ethoxymethylenmalonsyre-ethylester. Man opvarmer blandingen til 160°C, idet man afdestil-lerer det dannede ethanol. Ved afkøling får man 4-ethylphe-nylaminometbylenpropandisyreethylester som ventet i form af 10 en olie, som man umiddelbart benytter i det følgende trin. frin B: 4-hyd roxy-6-ethyl-5-quinolinoarboxylsyreethyle ster.
Man blander under omrøring og under en strøm af indifferent gas den i trin A fremstillede forbindelse med 70 ml phenyloxid. Man opvarmer den opnåede blanding til 250°0 i 1 ti-15 me, idet man afdestillerer det dannede ethanol. Man afkøler blandingen, tilsætter 30 ml acetone og får således en udfældning, som man suger fra, udriver med 30 ml acetone og tørrer.
Man omkrystalliserer af ethanol og får således 36 g 4-hy-droxy-6-ethyl-3-quinolincarboxylsyreethylester med smp. over 20 350°0.

Farmakologisk undersøgelse.
1) Affinitet til benzodiazepinreceutorerne.
Forsøgsteknikken er inspireret af teknikken ifølge MOhler og Okada, Science nr. 198, side 849-851 (1977).
5 Man homogeniserer til 1/20 (vægt/rumfang) i 0,52 M
saccharose cortex udtaget af hjernen på hanrotter på 150 g i gennemsnit. Efter centrifugering af den homogeniserede blanding ved 1000 g i 10 minutter ved 0°0 centrifugeres den overliggende væske ved 30.000 g i 20 minutter ved 4°C. Bund-10 stoffet suspenderes i 20 rumfang 50 mM Tris-HCl-stødpude pH
7,4 og centrifugeres ved 3000 g i 20 minutter ved 4°0. Det nye bundstof suspenderes i 50 ml Krebs’ Tris-HCl-stødpud.e pH 7,4.
Derpå lader man inkubere i 30 minutter ved 0°C 2 ml 15 suspension i nærværelse af H-diazepam i en koncentration

på 10 I alene med voksende koncentrationer af den forbindelse, som skal undersøges, eller - til bestemmelse af den ikke-specifikke fiksering - med ikke-radioaktivt diazepam i en koncentration på 10 M, 20 De inkuberede suspensioner filtreres på Whatman GF/C, og filtrene vaskes med 2 gange 5 ml Krebs’ Tris-HCl-stød-pude pH 7,4 ved 0°C.
^Radioaktiviteten af filtrene måles ved væskescin- ti11ation.
25 Aktiviteten af forbindelsen udtrykkes ved 0150, dvs. den koncentration, som inhiberer 50$ af den specifikke binding af -^H-diazepam, bestemt grafisk.

Teknik: Det benyttede apparat (Apelab) og den benyttede protokol er beskrevet af Boissier m.fl., European J.
Pharmacol. 1968, £, 145. Pladerne forbindes med en stimulator 5 (U. Sachs, Roucaire), som gør det muligt at give elektriske stød på 120 volt i 0,5 s. Forsøgene udføres på hold på 10 mus 30 minutter efter oral indgift af den undersøgte forbindelse. Hvert dyr anbringes enkeltvis på apparatet. Efter 15 sekunders ophold udsættes musen for et elektrisk stød, hver gang 10 den passerer fra en plade til en anden, idet der er mindst 3 sekunder mellem hvert stød. Antallet af stød i 1 minut tæl les. De opnåede resultater sammenlignes med resultaterne fra kontroldyr ved et Dunnett-forsøg.
Nedenfor er angivet den indgivne orale dosis, hvor 15 antallet af elektriske stød forøges maksimalt.

Forbindelserne ifølge eksempel 1, 2, 4, 6, 8 og 9 har altså en god angstdæmpende virkning.
25 3) Virkning på støjstress udtr.vkt ved corticosteronindholdet i blodplasmaet (angstdæmpende virkning).
Teknik: Forsøgene udføres på hold på 5 rotter. Den undersøgte forbindelse indgives oralt mellem kl. 8 og 9 om morgenen, idet dyrene har været anbragt i grupper på 5 et døgn i forvejen.
30 En time efter påfører man stress, hvilket består i at lade dyrene skifte bur og at anbringe dem på et sted,hvor en kraftigt støjende radio arbejder, og 30 minutter senere at give dem et intraperitonealt stik. 10 minutter efter bedøves rotterne med halothan, og oa. 3 ml blod udtages efter dekapitation.
35 Serumindholdet af corticosteron bestemmes ved en ra- dioimmunologisk metode. I betragtning af de betydelige serumkoncentrationer af corticosteron hos hanrotter udføres analysen direkte uden forudgående ekstraktion af hormonet på 17 150070 0,1 ml prøve fortyndet til 1/100. Til prøveudtagningen sættea der i hemolyseglas (12 x 65 mm) 0,1 ml vandig %-cortico te-ron (5000 cpm)og 0,5 ml phosphatstødpude (0,02 M; pH 6,9) tilsat 0,02/¾ gelatine og 0,02$ natriumnitrid, indeholdende i 5 suspension antiserumet anticorticosteron (fortynding 1/300.000), fåreantiserum mod kaninanti-y-glubulin (fortynding 1/50) og normalt kaninserum behandlet med kuldextran (fortynding 1/500).
Bumfanget indstilles på 1 ml, og inkubationen foretages en nat ved 4°0. Efter centrifugering (3000 omdrejninger 10 pr. minut) i 20 minutter elimineres den overliggende væske, og radioaktiviteten i udfældningen måles i et tælleapparat efter tilsætning af 2 ml scintillationsvæske indeholdende 5$ Soluen 350, Koncentrationerne af corticosteron bestemmes ved hjælp af en standard kurve (0-4000 pg), som er lavet under 15 betingelserne for behandlingen af prøverne.
Eet individuelle corticosteronindhold i serumet, udtrykt i μg/l00 ml, er gennemsnittet af tre bestemmelser. Re-sultaterne sammenlignes derpå ved Student's prøve med de resultater, som fås med kontroldyr, som har været udsat for 20 samme stress.
Resultaterne udtrykkes i EE 50 (effektiv dosis 50), dvs. den oralt indgivne dosis, som reducerer de behandlede dyrs corticosteronindhold med 50$.

Eer er mindre stress hos de behandlede dyr. Eisse dyr udviser altså en lavere grad af ængstelse end kontroldyrene.
4) Undersøgelse af den akutte toksicitet.
Man undersøger den letale dosis ELq for forskellige 35 undersøgte forbindelser efter oral indgift på mus.
Med ELq betegner man den maksimale dosis, som ikke fremkalder nogen dødelighed i løbet af 8 dage.
