Den foreliggende opfindelse angår en analogifrem-gangsmåde til fremstilling af hidtil ukendte N-phenyl-ethanolaminoalkyl-substituerede nitrogenholdige hetero-cycliske forbindelser med den almene formel (I):

Cl·) V— CH-CH-NH-C -(CHj)n-Het (X) R3 10 hvor symbolerne er som defineret i krav 1, i form af racemater, enantiomere eller diastereomere antipodepar, eller syreadditionssalte deraf, hvilken fremgangsmåde er ejendommelig ved det i krav 1's kendetegnende del anførte.

De ved den omhandlede fremgangsmåde fremstillede forbindelser har navnlig broncholytisk, spasmolytisk og antiallergisk aktivitet, som nedenfor nærmere omtalt.
I tysk fremlæggelsesskrift nr. 1.643.224 er beskrevet broncholytisk aktive 1-phenyl-2-aminoethanolderi-vater, blandt hvilke kan fremhæves den til handelsprodukt udviklede forbindelse salbutamol (2-(tert.-butyl-amino)-1-(4-hydroxy-3-hydroxymethylphenyl) ethanol).
De ved den foreliggende fremgangsmåde fremstillede forbindelser udmærker sig sammenlignet med nævnte kendte forbindelser blandt andet ved et bedre resorptionsforhold mellem intravenøs og oral aktivitet, således som det fremgår af nedenstående forsøgsdata med salbutamol som sammenligningsforbindelse. Et særligt gunstigt resorptionsforhold udvises af den ifølge nedenstående eksempel 5 fremstillede forbindelse 1-(2-fluor- 4-hydroxyphenyl)-2-[4-(1-benzimidazolyl)-2-methyl-2-butylamino]-ethanol eller syreadditionssalte deraf.
Ved fremgangsmåden a) til fremstilling af de om-handlede forbindelser anvendes opløsningsmidler,

2 der under reaktionsbetingelserne er tilstrækkeligt indifferente, f.eks. alkoholer, såsom methanol eller ethanol, og sædvanlige hydrogeneringskatalysatorer, f.eks. palladium, platin eller Raney-nikkel. Som reduktionsmiddel anvendelige hydrider er natriumborhydrid og andre komplek-

se hydrider eller boran. Reaktionstemperaturen ligger mellem ca. 0°C og kogetemperaturen for reaktionsblandingen. Hvis den centrale (i sidekæden) indeholdte amino- 2 gruppe eller phenylgruppe-substituenterne OH og/eller R i udgang s forbindelsen udviser en hydrogenolytisk fraspaltelig be-10 skyttelsesgruppe, f.eks. en eventuelt substitueret ben-zylgruppe, bliver denne fjernet under eller om nødvendigt efter reduktionen af CO-gruppen.
Udgangsforbindelserne II vindes for eksempel ved omsætning af aminer iv med bromketoner III i opløsnings-15 midler, såsom acetonitril eller ethylacetat, i nærværelse af et syrebindende middel, såsom natriumcarbonat eller aminoverskud.

De eventuelt som mellemproduktforbindelser optræ-igdende Schiff'ske baser VIII: H0 *5 (T )V-CO-CH=N-C - (CH„) -Het (VIII) r2 ^ψΓ l6 15 r3 hvori de enkelte symboler er som ovenfor defineret, kan også isoleres og derefter underkastes reduktion.
Som reduktionsmiddel anvendes komplekse hydrider, fortrinsvis natriumborhydrid, eller hydrogen i nærværelse 20 af hydrogeneringskatalysatorer, såsom platin, palladium eller Raney-nikkel.
Hvis phenoliske OH-grupper foreligger beskyttet af en hydrogenolytisk fraspaltelig gruppe, såsom benzyl, bliver disse grupper om nødvendigt efter reduktionen fjernet på 2 5 sædvandlig made.
Aminerne IV kan for eksempel vindes ved, at de hete-rocycliske forbindelser Het-H ved aminogruppen alkyleres med forbindelser med formlen X: 30 e5 1 I 1 B C - (CH2)n - X1 (X) R6 hvor: 35X er chlor, brom, en methyl- eller tolylsulfonylsyregruppe,

4 B er en funktionel gruppe, såsom NC^, dibenzylamino eller benzalamino, der ved katalytisk hydrogenering eller hydrolyse kan omdannes til aminogruppen, i nærværelse af natriumhydrid i opløsningsmidler, såsom 5 hexamethylphosphorsyretriamid, hvorefter den funktionel-1 le gruppe B overføres i aminogruppen.
De som udgangsforbindelser anvendte phenylglyoxa-ler VI eller de tilsvarende halvacetaler VII kan for eksempel vindes ved oxidation af acetophenoner IX: 10 c°-ch3 <ix) R3 15 2 3 hvor R og R er som ovenfor defineret, med selendi-oxid i vandig dioxan og krystallisation af vand eller alkoholer.

lysatorer, såsom palladium, platin eller Raney-nikkel, ved temperaturer mellem 0°C og kogetemperaturen for reaktionsblandingen. Som opløsningsmiddel tjener fortrinsvis lavere alkoholer, navnlig methanol.

De ved den her omhandlede fremgangsmåde fremstillede forbindelser er anvendelige som lægemidler. De har navnlig broncholytisk, spasmolytisk og antiallergisk aktivitet og kan derfor anvendes ved bronchitis og 15 astma, ved urticaria, conjunktivitis, høfeber og forkølelsessygdomme, endvidere som relaxationsmidler for uterusmuskulaturen, f.eks. ved vanskeligheder forud for fødsel. Endvidere egner de omhandlede forbindelser sig til behandling af cardiovaskulære sygdomme, såsom forhøjet blodtryk, perifere karsygdomme og hjertearytmi-er.
Endvidere skal nævnes den hæmmende aktivitet på mavesekretionen og den, navnlig antidepressive, aktivitet ^ på centralnervesystemet.
Der skal fremhæves den stærke og langvarige bron-cholytiske aktivitet, der er forbundet med kun ringe bivirkning på hjerte og skeletmuskulatur.

De forbindelser med formlen I, hvor R er CONH2, ^ virker endvidere blodtrykssænkende, mens de øvrige forbindelser udviser de andre anførte aktiviteter.
Den terapeutiske dosis afhænger af den anvendte forbindelse, arten af sygdomstilstanden, anvendelsesmåden og legemsvægt, såfremt der ikke er tale om lokal an- vendelse.

Til en voksen person kommer følgende doseringer pr.dag i betragtning:
Til broncholyse: oralt 2-20 mg, inhalativt 0,1-1,5 mg, subcutant 0,2-1,5 mg. Til uterusspasmolyse: oralt 5 10-50 mg, som infusionsopløsning 0,1-1 mg i ampuller med 10 ml opløsning. Til karudvidelse kan der oralt gives 20-100 mg, og i fom af opløsninger til intramuskulær injektion 20-40 mg, og til blodtryksænkning oralt 200-1800 mg.
.jQ Med henblik på anvendelse bliver der ud fra de om handlede forbindelser fremstillet de sædvanlige galeni-ske kompositioner, f.eks. kapsler, tabletter, dragée, opløsninger, suspensioner, pulvere, cremer, salver, emulsioner og sprayprodukter. Ved pulmonal anvendelse bliver 15 fortrinsvis pulver med en partikeldiameter på 0,5-7 um indført i bronchialområdet som aerosol sammen med åndingsluften, eventuelt også med yderligere drivgasser.
Den parenterale anvendelse foregår fortrinsvis i fom af sterile isotoniske vandige opløsninger, mens der 2Q til lokal anvendelse navnlig anvendes lotioner, cremer, salver, emulsioner og sprayprodukter.
De gunstige aktiviteter af de omhandlede forbindelser fremgår af nedenstående data.
1. Bronchospasmolyse.
25 Aktiviteten blev undersøgt på marsvineunger i ure- than-narkose. Ved legemsplethysmografi bestemtes indvirkningen på acetylcholin-bronchospasmus efter intravenøs og oral anvendelse. Endvidere blev hjertefrekvensen kontrolleret. Som sammenligningsforbindelse anvendtes salbutamol.

De omhandlede forbindelser udviser et udmærket forhold mellem intravenøs og oral aktivitet. Indvirkningen på hjertefrekvensen er samtidigt ringe. Endvidere er to-xiciteten så lav, at der foreligger en betydelig terapeu-20 tisk bredde. For eksempel er for forbindelsen ifølge Eksempel 5 LD50 på mus i.v. 29 mg/kg, p.o. 330 mg/kg.
2. Uterus relaxation (Tokolyse).
Uterusrelaxationen blev undersøgt på narkotiserede rotter. Der bestemtes den intravenøse ED^ for uterusre-25 laxation i pg/kg (50% af de undersøgte dyr reagerede), samt den (uønskede) forøgelse af hjertefrekvensen ved ED s o-værdien. Som sammenligningsforbindelse anvendtes fenoterol (1-(3,5-dihydroxyphenyl)-1-hydroxy-2-[(4-hydroxyphenyl)isopropylamino]ethan).

De omhandlede aktive stoffer kan også kombineres med kendte aktive stoffer. Til broncholytisk anvendelse f.eks. med theophylliner, parasympatolytica (f. eks. ipratropiumbromid), sekretolytica (f.eks. bromhex-30 in), muskulotrope spasmolytica (f.eks. papaverin), cor-ticosteroider og/eller antiallergica. Ved uterusrelaxa-tionsmidler er blandt andet kombinationer me d corticoi-der mulig.

Til fremgangsmåde a:’
Eksempel 1 5 HO—/ CH-CH2-NH-CH2-CH2-CH2- / 2HC1 30.5 g 2-Brom-p-benzyloxyacetophenon og 35 g 1-aminopropylbenzimidazol blev i 150 ml acetonitril omrørt 1 time ved 30-40°C. Efter fraskillelse af hydrobromidet 10 blev moderluden syrnet med 12 g maleinsyre, og det udfældede a-[3-(1-benzimidazolyl)-propyl-amino]-4-benzyloxy-acetophenon-maleinat (smp. 145-148°C) blev frasuget. Med vandig ammoniak blev der derudfra fremstillet basen, der i 200 ml alkohol blev reduceret med natriumborhydrid til 15 1-(4-benzyloxyphenyl)-2-[3-(1-benzimidazolyl)-propylami- no]-ethanol (smp. 83-85°C).
Ved katalytisk hydrogenering af 7 g af denne forbindelse i 100 ml methanol med 1 g palladiumkul som katalysator vandtes 4,5 g 1-(4-hydroxyphenyl)-2-[3-(l-benz-20 imidazolyl)-propylamino]-ethanol (smp. 146-148°C, udbytte 83% af det teoretiske), hvis dihydrochlorid havde smp. 184-185°C.

17.5 g 2-Benzyloxy-5-bromacetyl-salicylamin, 17,6 g 1-(3-aminorpopyl)-lH-benztriazol, 6 g natriumcarbonat 30 og 150 ml ethylacetat blev tilbagesvalet i 1,5 timer.

Efter fraskillelse af de uorganiske bestanddele blev moderluden inddampet, inddampningsresten blev opløst i 100 ml acetonitril og syrnet med 5 g oxalsyre. Det udfældede 1-[3-(3-carbamoyl-4-benzyloxy-3-oxo-phenethylamino)-pro-5 pyl]-lH-benztriazol-oxalat blev frasuget, med vandig ammoniak overført i basen (smp. 186-188°C) og i 100 ml ethanol reduceret med natriumborhydrid til l-[3-(3-car-bamoyl-4-benzyloxy-3-hydroxyphenethylamino)-propyl]-1-H-benztriazol.
10 Ved hydrogenering af 6 g af denne forbindelse i 100 ml methanol ved et tryk på 6 bar og 40°C under tilsætning af palladiumkul vandtes 3 g 1-[3-(3-carbamoyl-4,3-dihydroxyphenethylamino)-propyl]-1-H-benztriazol (smp. 154-155°C, udbytte 77,5% af det teoretiske), hvis 15 cyclamat havde smp. 165°C.

0 ch3 12,9 g 5-Bromacetylsalicylamid, 15,45 g l-(3-benz-ylaminopropyl)-3-methyl-quinazolin-2,4-dion, 6 g natri-umcarbonat og 300 ml acetonitril blev kogt 1,5 timer un-25 der tilbagesvaling. Efter frasugning af de uorganiske bestanddele blev moderluden inddampet, inddampningsresten blev opløst i 500 ml methanol og efter tilsætning af 12 ml benzylchlorid hydrogeneret ved 6 bar og 60°C med palladiumkul som katalysator. Efter optagning af 2 30 mol hydrogen blev hydrogeneringsproduktet oparbejdet, og det dannede 1- [3- (3-carbamoyl-|3-oxo-hydroxyphenethylami-no)-propyl]-3-methyl-quinazolin-2,4-dion-hydrochlorid (smp. 253°C (dek.)) blev isoleret.
Ved katalytisk hydrogenering af 13 g af denne for-35 bindelse i 250 ml methanol/vand-blanding 1:1 ved 6 bar, 50°C med palladium som katalysator vandtes 8 g l-[3-car-

12 bamoyl-4-(3-dihydroxyphenethylamino)-propyl]-3-methyl-quinazolin-2,4-dion-hydrochlorid, smp. 220-221°C. Udbytte: 61,5% af det teoretiske.

14,4 g 2' -Benzyloxy-5 '- (l-oxo-2-hydroxy-2-ethoxy- 10 ethyl)-methansulfonanilid, 7 g 1-(3-amino-3-methyl-bu-ty 1) -benzimidazol og 150 ml alkohol blev opvarmet 3 timer ved 50°C, og der blev portionsvis tilsat 9,2 g na-triumborhydrid. Opløsningen blev holdt 12 timer ved stuetemperatur, hvorefter alkoholen blev fjernet ved redu-15 ceret tryk på rotationsfordamper, og inddampningsresten blev opløst med 200 ml vand og 500 ml ethylacetat. Efter dekomponering af natriumborhydridet under omrøring med koncentreret saltsyre under isafkøling blev der indstillet alkalisk med vandig ammoniak, ethylacetatfasen blev 20 fraskilt, tørret med natriumsulfat og inddampet på rotationsfordamper. Inddampningsresten blev opløst i 200 ml alkohol, syrnet med 6,3 g oxalsyre, og det udfældede 2'-benzyloxy-5'-[l-hydroxy-2-[4-(1-benzimidazolyl)-2-methyl- 2-butylamino]-ethyl]-methansulfonanilid-dioxalat (smp.
25 o 185-187 C) blev frasuget. Med vandig ammoniak blev der ud fra denne forbindelse fremstillet basen (smp. 65-70°C).
Ved katalytisk hydrogenering af denne forbindelse (smp.
65-70°C) i 250 ml methanol under normalbetingelser med palladiumkul som katalysator vandtes 8 g 2'-hydroxy-5'- 30 [l-hydroxy-2-[4-(1-benzimidazolyl)-2-methyl-2-butylami-no]-ethyl]-methansulfonanilid (smp. 170-173°C, udbytte: 81% af det teoretiske), hvis formiat havde smp. 161-164° C.

Til 15,2 g 1-(2-Fluor-4-benzyloxyphenyl)-l-oxo-2-hydroxy-2-ethoxy-ethan, opløst i 200 ml alkohol, blev der sat 8,2 g 1-(3-amino-3-methyl-butyl)-benzimidazol og om-10 rørt 3 timer ved 50°C. Derefter blev opløsningen afkølet, der blev tilsat 4 g natriumbordydrid og omrørt 6 timer ved stuetemperatur. Reaktionsblandingen blev oparbejdet, som beskrevet i Eksempel 3. Inddampningsresten blev opløst i 200 ml alkohol og syrnet med 7,2 g oxalsyre, og 15 det udfældede 1-(2-fluor-4-benzyloxyphenyl)-2-[4-(l-benz-imidazolyl)-2-methyl-2-butylamino]-ethanol-dioxalat (smp. 184-188°C) blev frasuget. Ud fra dioxalatet blev der med vandig ammoniak fremstillet basen (smp. 85-88°C).
Ved katalytisk hydrogenering af denne forbindelse 20 i 250 ml methanol ved 6 bar og med palladiumkul som ka-• talysator ved ~ 30°C vandtes 1-(2-fluor-4-hydroxyphenyl)- 2-[4-(1-benzimidazolyl)-2-methyl-2-butylamino]-ethanol (smp. 151-153°C), hvis formiat havde smp. 157-159°C.
Ved tilsætning af den beregnede mængde methansul-25 fonsyre til opløsningen af basen i ethanol vandtes me-thansulfonatet, smp. 178-179°C. På tilsvarende måde blev der fremstillet hydrochloridet, smp. 205°C.
Ud fra basen i vandig acetonitril og den beregnede mængde koncentreret saltsyre vandtes dihydrochlorid-te-30 trahydratet, smp. 177°C.
Analogt vandtes i vandig ethanol sulfat-hydratet, smp. 207-208°C.

15 g 2'-Benzyloxy-5'-[ l-hydroxy-2-[ 4-(1-imidazoly 3) -2-methyl-2-butylamino]-ethyl]-acetanilid (smp. af dioxa-lat 160-163°C), 18,5 g KOH, 80 ml alkohol og 15 ml vand blev kogt 24 timer under tilbagesvaling, og derefter blev 1Q det dannede 1-(3-amino-4-benzyloxyphenyl)-2-[4-(1-imida-zolyl)-2-methyl-2-butylamino]-ethanol isoleret som trio-xalat (smp. 95-100°C). Med vandig ammoniak blev der herudfra fremstillet basen, der med kaliumcyanat blev omsat til l-{2'-benzyloxy-5[l-hydroxy-2-[4-(1-imidazolyl)-2-15 methyl-2-butylamino]-ethyl]-phenyl}-urinstof (smp. 142-143°C). Ved katalytisk hydrogenering af 5,1 g af denne forbindelse i 100 ml methanol med palladiumkul som katalysator vandtes 3,5 g 1-{2'-hydroxy-5'-[l-hydroxy-2-[4-(1-imidazolyl)-2-methyl-2-butylamino]-ethyl]-phenyl}-u-20 rinstof, hvis sulfat havde smp. 243-244°C. Udbytte: 70% af det teoretiske.
Eksempel 7 oc- CH0OH CH, i H0~O“ ^H"CH2"NH“<|"CH2"CH2"NL_~jl »C6H5C00h OH CH3 13,5 g 2'-Benzyloxy-5'-[l-hydroxy-2-[4-(1-imidazol·· 30 yl)-2-methyl-4-butylamino]-ethyl]-benzoesyremethylester (smp. af dioxalat 156-158°C) blev i 200 ml tetrahydrofu-ran reduceret med 6 g lithiumaluminiumhydrid til l-(3-hydroxymethyl-4-benzyloxyphenyl)-2-[4-(1-imidazolyl)-2-

Med vandig ammoniak blev der ud fra 10 g dioxalat fremstillet basen, og denne blev med palladium som katalysator hydrogeneret i 100 ml methanol. Der vandtes 5 4,5 g 1-(3-hydroxymethyl-4-hydroxyphenyl)-2-[4-(1-imida- zolyl)-2-methyl-2-butylamino]-ethanol (smp. 135-137°C), hvis benzoat havde smp. 150-152°C. Udbytte: 83% af det teoretiske.
