Den foreliggende opfindelse angår hidtil ukendte LHRH-analoge peptider og deres ikke-toksiske salte, som hos pattedyr, herunder mennesker, inhiberer hypofysens frigørelse af gonadotro-piner. Den foreliggende opfindelse angår nærmere bestemt pep-5 tider, som inhiberer gonadalfunktion og frigørelsen af steroidhormonerne, progesteron og testosteron. Opfindelsen angår også et farmaceutisk middel indeholdende peptiderne eller deres salte.
10 Et af de hypothalamiske hormoner virker som en faktor, som udløser frigørelsen af de gonadotrope hormoner, især LH, og dette hormon betegnes heri som LHRH eller GnHR, selv om det også er blevet betegnet som LRF. GnRH er blevet isoleret og karakteriseret som et decapeptid med følgende opbygning: 15 p-Glu-His-Trp-Ser-Tyr-Gly-Leu-Arg-Pro-Gly-NH2
Peptider er forbindelser, der indeholder to eller flere aminosyrer, og hvor den ene syres carboxylgruppe er forbundet med 20 den anden syres aminogruppe. Disse aminosyrer sammen med valin, isoleucin, threo-nin, lysin, asparaginsyre, asparagin, glutamin, cystein, me-thionin, phenylalanin og prolin antages almindeligvis at være de almindelige, naturligt forekommende eller protein-afledte 35 aminosyrer. Den en grunde til at ønske at hindre ovulation i hun-pattedyr, og indgift af GnRH-analoge, som er antagonistiske til GnRH's normal funktion, er blevet anvendt til at hindre ovulation. Af denne grund er analoge til GnRH, som er antagonistiske til 5 GnRH, blevet undersøgt med henblik på deres mulige anvendelse til antikonception eller til regulering af konceptionsperioder. Sådanne antagonister har også vist sig anvendelige til at regulere sekretion af gonadotropiner i hanpattedyr og kan med henblik på antikonception anvendes til hanner. Det er ønske-10 ligt at tilvejebringe peptider, som er kraftigt antagonistiske over for endogent GnRH, og som hindrer sekretion af LH og pattedyrs kønskirtlers frigørelse af steroider.
Fra EP offentliggørelsesskrift nr. 81.877 kendes en række 15 LHRH-analoge peptider med LHRH-antagonistisk virkning. Disse LHRH-antagonister har L-tyrosin i 5-sti11 ingen, og D-Arg, D-His eller D-Lys eller lignende i 6-stillingen. Skønt disse analoger har antiovulatoriske egenskaber, har de vist sig uacceptable, fordi de fremkalder frigivelse af store mængder hi-20 stamin, som kan give anledning til ødem. Fra DK patentskrifterne nr. 150.206 og 151.033 kendes tilsvarende LHRH-antago-nister, der har tyrosin i 5-stillingen. Disse forbindelsers antiovulatoriske virkning er imidlertid begrænset, idet der behøves doser på mindst 10 mikrogram pr. rotte, svarende til 25 40 mikrogram pr. kg legemsvægt, for at sådanne forbindelser skal være effektive.
Den foreliggende opfindelse tilvejebringer peptider, som inhi-berer frigørelsen af gonadotropiner i pattedyr, herunder men-30 nesker. De forbedrede GnRH-analoge er agonistiske til GnRH og har en hæmmende virkning på pattedyrs reproduktionsprocesser. Disse analoge kan anvendes til at inhibere dannelsen af gonadotropiner og kønshormoner under forskellige omstændigheder, herunder tidligt udviklet pubertet, hormonbetinget vævsnydan-35 nelse, menstruationsbesvær og endometriosis.

Disse peptider er analoge til GnRH, hvori der er en 1-stilling susbti tution, såsom β-(2-naphthy1) D-alanin (i det følgende 0-D-2NAL), 5 en 3 stilling substitution, såsom D-Trp, en 5-stilling substitution med Arg eller D-Arg i stedet for Tyr, en 6-stilling D-isomer substitution, som det er almindeligt kendt i teknikken, og en specifik substituent i 2-sti11 ingen. 1-stilling er sub-stituenten modificeret således, at dens α-aminogruppe indehol-10 der acetyl(Ac). Modificeret D-Phe findes i 2-stillingen og giver forøget antagonistisk aktivitet som et resultat af de specifikke modifikationer, som findes i benzenringen, specielt når α-carbonatomet også er methyleret. 20 På grund af at disse peptider er kraftigt virkende med hensyn til at inhibere frigørelse af LH, betegnes de ofte som GnRH-antagonister. Peptiderne inhiberer ovulation hos hunpattedyr, når de indgives i meget små mængder før oestrus, og bevirker også resorption af befrugtede æg, hvis de indgives kort tid 25 efter konception. Disse peptider er også effektive til opnåelse af antikonception ved behandling af hanpattedyr.

Med β-D-Nal menes D-isomeren af alanin, som er substitueret med naphthyl på jS-carbonatomet, hvi lket også kan betegnes 3-D-NAL. /3-D-2NAL anvendes fortrinsvis, hvilket betyder, at β-carbonatomet er bundet til naphthalenet ved 2-stillingen på 5 ringstrukturen. /3-D-1NAL betragte imidlertid som værende en ækvivalent.
Peptiderne ifølge den foreliggende opfindelse kan syntetiseres ved hjælp af klassiske opløsningssynteser eller ved hjælp af 10 en fastfaseteknik under anvendelse af en chlormethyleret harpiks, en methylbenzydrylaminharpiks (MBHA) eller en benzydryl-aminharpiks (BHA). Fastfasesyntesen gennemføres på en sådan måde, at man trinvis tilføjer aminosyrerne i kæden på den måde, som er angivet detaljeret i US patentskrift nr. 4.211.693.

Kriteriet for valget af s idekædebeskyttelsesgrupper for X2-X5 er, at beskyttelsesgruppen skal være stabil i nærværelse af reagenset under de valgte reaktionsbetingelser til fjernelse af α-aminobeskyttelsesgruppen ved hvert af syntesens trin. Be-5 skyttelsesgruppen må ikke blive afspaltet under koblingsbetingelserne, og beskyttelsesgruppen skal kunne fjernes efter endt syntese af den ønskede aminsyresekvens under reaktionsbetingelser, som ikke vil ændre peptidkæden.
10 Når X6-gruppen er Gly-0-CH2~[harpikskæder], D-Ala-0-CH2~[harpiksbærer] eller 0-CH2-[harpiksbærer], præsenteres esterdelen af én af de mange funktionelle grupper af polystyrenharpiks-bæreren. Gruppen Ac i den endelige formel kan tjene som Xl-beskyttel-sesgruppen for D-NAL's α-aminogruppe ved at tilsætte den før koblingen af denne sidstnævnte aminosyre til peptidkæden. En 20 reaktion gennemføres imidlertid fortrinsvis med pept i det på harpiksen (efter afbeskytte!se af α-aminogruppen medens side-kædegrupperne forbliver beskyttede) f.eks. ved omsætning med eddikesyre i nærværelse af dicyklohexylcarbodiimid (DCC) eller fortrinsvis med eddikesyreanhydrid eller med en anden egnet i 25 teknikken kendt reaktion.
Det fuldstændigt beskyttede peptid kan spaltes fra en chloro-methyleret harpiksbærer ved hjælp af ammonolyse, således som det er velkendt i teknikken, til dannelse af det fuldstændigt 30 beskyttede amidmellemprodukt. Afbeskyttelse af peptidet, såvel som spaltning af peptidet fra en benzhydrylaminharpiks, kan finde sted ved 0°C med hydrogenf 1uoridsyre (HF). Anisol sættes fortrinsvis til peptidet før behandling med HF. Efter fjernelsen af HF, under vakuum, bliver det spaltede, afbeskyttede 35 peptid hensigtsmæssigt behandlet med ether, dekanteret, optaget i fortyndet eddikesyre og lyofiliseret.

Rensning af peptidet sker ved hjælp af ionbytterkromatografi på en CMC-kolonne, efterfulgt af fordelingskromatografi under anvendelse af elueringssystemet: n-butanol, 0,1N eddikesyre (lige volumendele) på en kolonne pakket med Sephadex 6-25 el-5 ler ved anvendelse af højtryksvæskekromatografi (HPLC), således som det er kendt i teknikken.
Peptiderne ifølge opfindelsen er effektive i mængder på under 100 mikrogram pr kg legemsvægt, når de indgives omkring kl 12 10 dagen før oestrus, for at hindre ovulation i hunrotter. Til langtids undertrykkelse af ovulation kan det være nødvendigt at anvende dosismængder i intervallet fra ca. 0,1 til ca. 2,5 mg pr kg legemsvægt. Disse antagonister er også effektive som antikonceptionsmidler, når de gives til han-dyr regelmæssigt.
15 Da disse forbindelser vil reducere testosteronmængden (en uønsket konsekvens på den normale, seksuelt aktive han) kan det være rimeligt at give erstatningsdoser af testosteron sammen med GnRH-antagonisten. Disse antagonister kan også som tidligere anført anvendes til med andre formål at regulere produk-20 tionen af gonadotropiner og kønshormoner.

Efter trin 13 udtages, hvis syntesen gennemføres manuelt, en prøve til en ninhydrintest. Hvis testen er negativ, går man tilbage til trin 1 til kobling af den næste aminosyre.
Hvis testen er positiv eller svagt positiv, går man tilbage 2 5 og gentager trinene 9-13.
Den ovennævnte plan anvendes til kobling af hver af aminosy-rerne i peptidet ifølge opfindelsen, efter at den første a-minosyre er blevet vedføjet. NaBoc-beskyttelse anvendes for hver af de resterende aminosyrer under syntesen. NaBoc-/3-D-2NAL fremstilles ved hjælp af en i teknikken kendt metode, f.eks. som beskrevet detaljeret i US-PS 4.234.571. Arg's sidekæde er beskyttet med Tos. OBzl anvendes som en sidekædebe- skyttelsesgruppe for Ser's hydroxyl gruppe, og 2-6 dichloro- 3 5 benzyl anvendes som sidekædebeskyttelsesgruppe for D-Tyrs hydroxylgruppe ved syntetisering af peptid nr. 2. Trp holdes ubeskyttet. NaBoc-/3-D-2NAL indføres som den sidste aminosyre. Boc-Arg(Tos) og Boc-D-Trp, som har lav opløselighed i CH2C12, kobles under anvendelse af DMF:CH2Cl2-blandinger.

Efter afbeskyttelse af α-aminogruppen ved N-terminalen, opnås acetylering under anvendelse af et stort overskud af ed-dikesyreanhydrid i dichlormethan. Spaltningen af peptidet fra harpiksen og fuldstændig afbeskyttelse af sidekæderne 5 sker meget let ved 0°C med HF. Anisol tilsættes som rense middel før HF-behandling. Efter HF-fjernelse under vakuum ekstraheres harpiksen med 50% eddikesyre, og vaskevæskerne lyofiliseres til opnåelse af et råt peptidpulver.
10 Rensning af peptidet gennemføres derpå ved ionbytterkromato- grafi på CMC (Whatman CM 32, under anvendelse af en gradient af 0,05-0,3M NH^OAc i 50/50 methanol/vand) efterfulgt af fordelingskromatografi i en gelfiltreringskolonne under anvendelse af elueringssystemet: n-Butanol, 0,1N eddikesyre (1:1 15 - volumenforhold).
Det bedømmes om peptidet er homogent ved anvendelse af tyndt-lagskromatografi og flere forskellige opløsningsmiddelsystemer, såvel som ved anvendelse af omvendtfase højtryksvæske-20 kromatografi og en vandig triethylammoniumphosphatopløsning plus acetonitril. Aminosyreanalyse af det resulterende, rensede peptid stemmer overens med formlen for den fremstillede struktur, hvilket viser i alt væsentligt hele-talsværdier for hver aminosyre i kæden. Den optiske drejning måles på 25 et fotoelektrisk polarimeter, som [α]ρ^=-2,2°+1(σ=1, 50% ed dikesyre) .
De ovenfor beskrevne peptider undersøges in vivo til bestemmelse af deres effektivitet med hensyn til at hindre ovula-30 tion i hunrotter. I dette forsøg indsprøjtes der i en række fuldt udviklede Sprague-Dawley hunrotter, dvs. ti, hver med en legemsvægt fra 225-250 g', 10 mikrogram peptid i majsolie omkring klokken 12 på dagen før oestrus (pro-oestrus). Prooestrus er eftermiddagen før oestrus (ovulation). En særlig 35 gruppe hunrotter anvendes som kontrol, der ikke indgives pep tid. Hver af kontrol-hunrotterne har ovulation ved oestrus.

Ingen af de behandlede rotter har ovulation. Peptidet anses derfor som værende signifikant effektivt til at hindre ovulation hos hunrotter i en meget ringe dosis, og peptidet anses for at være fuldstændigt effektivt i en dosis på ca. 10 5 mikrogram. Yderligere afprøvning kan også gennemføres ved lavere eller højere doser, idet resultaterne er anført i den efterfølgende tabel II.

Alle peptiderne anses for at være effektive til at hindre ovulation hos hunpattedyr i meget lave doser. Disse peptider kan indgives i form af farmaceutisk acceptable ikke-toksi-ske salte, såsom syreadditionssalte eller metalkomplekser, 35 f.eks. med zink, barium, calcium, magnesium, aluminium eller lignende (hvilket anses for at være additionssalte), eller kombinationer af disse to. phater, nitrater, oxalater, fumarater, gluconater, tannater, maleater, acetater, citrater, benzoater, succinater, algina-ter, maleater, ascorbater, tartrater og lignende. Hvis den aktive bestanddel skal indgives i tabletform, kan tabletten 5 indeholde et farmaceutisk acceptabelt fortyndingsmiddel, som omfatter et bindemiddel, såsom tragant, majsstivelse eller gelatine, et desintegrationsmiddel, såsom alginsyre, og et smøremiddel, såsom magnesiumstearat. Hvis man ønsker at indgive det i flydende fom, kan der anvendes sødestoffer og/-10 eller aromastoffer som en del af det farmaceutisk acceptab le fortyndingsmiddel, og intravenøs indgift i isotonisk saltvand, phosphatpufferopløsninger eller lignende kan gennemføres .
15 De farmaceutiske midler vil sædvanligvis indeholde peptidet sammen med en sædvanlig farmaceutisk acceptabel bærer. Dosen vil sædvanligvis være fra ca. 1 til ca. 100 mikrogram af peptidet per kg af værtens legemsvægt, når der anvendes intravenøs indgift. Orale doser vil være højere. Overalt gennem-20 føres behandlingen af patienter med disse peptider alminde ligvis på samme måde, som den kliniske behandling, med andre antagonister af GnRH.
Disse peptider kan gives intravenøst, subkutant, intramus-25 kulært, oralt, intranasalt eller intravaginalt til pattedyr for at opnå frugtbarhedsinhibering og/eller regulering, og ligeledes vil anvendelser, som kræver reversibel undertrykkelse af kønskirtelaktivitet, f.eks. til styring af tidligt udviklet pubertet eller under strålings- eller kemoterapi.
30 Effektive doser vil variere med indgivelsesformen og af hvil ken type pattedyr, der behandles. Et eksempel på en typisk dosisform er en fysiologisk saltvandopløsning indeholdende peptidet, hvilken opløsning indgives til frembringelse af en dosis i intervallet fra ca. 0,1 til 2,5 mg/kg legemsvægt. 35 Oral indgift af peptidet kan ske i enten fast form eller fly dende form.

LHRH-analogt peptid eller et ikke-toksisk salt deraf, 5 kendetegnet ved, at peptidet har formlen: Ac-R1-R2-R3-Ser-R5-R6-Leu-Arg-Pro-R10 I hvori Rj er β-Ο-2-NAL, dehydroPro eller 4C1-D-Phe, R2 4C1-D-10 Phe, 4F-D-Phe eller CaMe-4Cl-D-Phe, R3 er D-Trp eller /S-D-2NAL, R5 er Arg eller D-Arg, Rg er D-Tyr, J3-D-2NAL eller D-Arg og R^g er Gly~NH2 eller D-Ala-NH2.
