Den foreliggende opfindelse angår en vandig injektionsopløsning indeholdende cholintheophy11 i nat, der er ejendommelig ved det i kravets kendetegnende del angivne.
5 2-hydroxy-N,Ν,Ν-trimethylethanaminium-saltet af 3,7-dihydro-l,3-dimethyl-lH-purin-2,6-dion (1:1) (cholintheophyllinat) er kendt som værende et farmakologisk virksomt stof, der er indeholdt som aktivt stof i bronkolytisk virksomme lægemidler ("Rote Liste 1980" - Editio Cantor Verlag, Aulendorf, Wurttem-10 berg DT, nr. 27 042). Dette theophyllinsalt har i forhold til andre salte af dette stof, som f.eks. theophyllin-ethylen-diamin (=aminophyllin) det fortrin, at det bedre kan tåles. (Arzneim. Forsch., 31, 1981, 1503 ff og 32, 1982, 409 ff).
De cholintheophyllinatholdige præparater indgives oralt. Dette 15 har til følge at koncentrationen af det aktive stof i blodplasma først når sit maksimum efter ca. to timer (Brit. J. Pharmacol., 3, 1976, side 194 - 196), hvilket navnlig er en ulempe ved behandling af svære krampetilstande. Der foreligger således et behov for et injektionsmiddel, der indholder dette 20 aktive stof.
Vandige opløsninger af cholintheopyllinat er ikke stabile og er således uegnede som injektionsmidler. Dersom man forsøger at stabilisere disse opløsninger ved tilsætning af i den ga-25 leniske teknik konventionelt anvendte syrer eller antioxidanter (myresyre, eddikesyre, phosphorsyre, benzoesyre, glutarsyre, adipinsyre, sorbinsyre, æblesyre, vinsyre, ascorbinsyre, citronsyre etc.) udfældes cholintheophyllinat eller man opnår lige så ustabile opløsninger, der misfarves og ikke er steriliserbare. 30
Det har nu vist sig, at man opnår overraskende stabile vandige opløsninger af cholintheophyllinat, når man til denne opløsning, regnet på saltet, tilsætter 5 til 8, fortrinsvis 6 til 7, og navnlig 6,5 til 6,8 vægt% propionsyre. De vundne opløs-35 m'nger har en pH-værdi på 9 til 10 - fortrinsvis 9,4 til 9,8.
De kan på sædvanlig måde fyldes i ampuller eller lignende og steriliseres.

I disse vandige injektionsopløsninger udgør koncentrationen af cholintheophyllinat 1 til 25, fortrinsvis 2 til 20, og navnlig 3 til 15 vægt%.
5 De vandige injektionsopløsninger ifølge opfindelsen anvendes blandt andet til behandling af akutte og svære krampetilstande i bronkialvejen ved kronisk bronkitis, lungeemfysem og bron-kialastma - navnlig ved status astmatikus. Den indgivne startdosis bør fortrinsvis udgøre 5 til 10 mg/kg, og navnlig 7 10 til 9 mg/kg legemsvægt, og den indgivne dagsdosis bør ligge inden for grænserne fra 15 til 40 mg/kg og navnlig inden for grænserne fra 19 til 31 mg/kg legemsvægt.
De efterfølgende eksempler tjener til illustration af den 15 vandige injektionsopløsning ifølge opfindelsen.
Eksempel 1
Man tilbereder ud fra 20 300.00 g cholintheophyllinat 20,00 g propionsyre og 9730.00 g redestilleret vand 25 10,00 liter injektionsopløsning, der sterilfiltreres og under sterile betingelser fyldes i ampuller med 10,00 ml pr. ampul.
Eksempel 2 30 Man tilbereder ud fra 150.00 g cholintheophyllinat 10.00 g propionsyre og 875.00 g redestilleret vand 35 1.00 liter injektionsopløsning, der sterilfiltreres og fyldes i ampuller med 1,00 ml pr. ampul.
