Opfindelsen angår en særlig fremgangsmåde til spaltning af peptider og proteiner ved raethionylbindingen.
Inden for rammerne af den genteknologiske udvinding af peptider og proteiner (f.eks. insulin, proinsulin, 5 præproinsulin og analoge deraf, interferon, væksthormon) har det ofte vist sig at være fordelagtigt først at isolere produkterne i form af højeremolekylære "fusionsproteiner" .
De således dannede fusionsproteiner er mere sta-10 bile end de ønskede produkter og kan isoleres i højere udbytter ved udnyttelse af induktionssystemer. Til sådanne fusioner kommer i princippet alle proteiner, der syntetiseres af værtsorganismerne i større mængder, på tale, fremfor alt, når deres syntese er højinducerbar; ofte 15 beskrevne er β-galactosidasen, β-lactamaseri, dele af cro-proteinet, af C^j-proteinet og af tryptofanstofskiftets enzymer (fremfor alt Trp D, Trp E) . Hertil er fusioner af det ønskede genprodukt med β-galactos idase i besiddelse af særlig popularitet, fordi de derved først 20 dannede proteiner hyppigt er tungtopløselige og følgelig kan højkoncentreres (A.V. Fowler og I. Zabin, J. Biol.
Chem. 258 (1983) 14354-58.
Ud fra de resulterende fusionsproteiner kan man fraspalte de ønskede produkter ved kemiske og enzymatiske 25 fremgangsmåder, idet disse spaltningsreaktioner skal være højspecifikke for at undgå beskadigelser af produkterne.
Kemiske fremgangsmåder til proteinspaltning er for det meste refereret i sammenhæng med peptid-sekvensana-lytikken af proteiner (jfr. eksempelvis Handbook of 30 Protein Sequence Analyses, 2nd Edition, L.R. Croft,
John Wiley a. Sons; B. Witkop, Advan. Prot. Chem. 16 (1961) , 221 samt E. Gross, Methods in Enzymology, Vol.
XI, 238 (1967).
Særligt hyppigt er refereret en spaltning ved O c methionyl-bindinger med bromcyanid (B. Witkop, Adv. Prot.

Chem. 16 (1961) 221), da man hermed i almindelighed opnår høje spaltningsudbytter uden beskadigelse af andre peptidbindinger eller aminosyres'idek'æder.
For at opnå høje spaltningsudbytter anvender man 5 på gængs måde store bromcyanidoverskud (op til 250 gange, jfr. B. Witkop-artiklen), beregnet på den methionyl-binding, der skal spaltes, i forbindelse med lange reaktionstider op til 30 timer og stuetemperatur. Reaktionen gennemføres i stærkt surt miljø, og et foretrukket opløsningsmiddel er 10 myresyre i et koncentrationsområde fra 70 til 88 rumfangsprocent.
Bromcyanid (BrCN) er et fast stof (smp. 52°C, kp. 62°C) og en højtoksisk forbindelse. Den dødelige dosis for et normalt menneske er 92 ppm ved en virketid på 10 15 minutter (Gmelin Syst. Nr. 14 C, Fl. D 3 1976, S. 217-241; Petty's Industrial Hygene and Toxicologiy, 3. Auflage 1982,
Bd. 2 e, S. 4861). Ved urene præparater er beskrevet en spontan eksplosion af emballeret stof (se brochuren:
Sicherheit mit Merck, E. Merck, Darmstadt 5/13090/60/1282 R).
20 Håndteringen af store mængder bromcyanid ved den højttekniserede proteinspaltning er således særdeles problematisk; foruden sikkerhedsaspekterne spiller også tekniske problemstillinger (f.eks. transport af faste stoffer) en rolle.
25 Det har nu overraskende vist sig, at methionyl-bin- dingen lader sig spalte med chlorcyanid (C1CN).
Opfindelsen angår således en fremgangsmåde til spaltning af peptider og proteiner ved methionyl-bindingen, og denne fremgangsmåde er ejendommelig ved, at man gennem-30 fører spaltningen med chlorcyanid anvendt i et molært overskud fra 2 til 30 gange pr. methionyl-binding, der skal spaltes.
Proteinspaltninger med C1CN er ikke beskrevet i litteraturen.

forudseelig, fordi C1CN har en reaktivitet, der er tydeligt forskellig fra BrCN's (jfr. Houben-Weyl, Bd. II, s.545; Ullmann Bd. 9, s. 668, B.S. Thyagarajan, The Chemistry of Cyanogen Halides 2, (1968); R.T. Parfitt, 5 J. Chem. Soc. (C), 140, (1967)).Foruden den forskellige reaktivitet og aktivitet hos BrCN og C1CN iagttager man også forskellig reaktionsadfærd i forhold til samme reaktionspartnere.
C1CN er ved stuetemperatur en gas (smp. -6,9°C, 10 kp. 13,0°C); det er ikke så giftigt som BrCN (jfr. Gmelin Syst. Nr. 14 C, F 1, D 3, 1976, s. 185; Petty's Industrial Hygiene and Toxicology, 3. Auflage 1982, Bd. 2 c, s. 4859).
Den dødelige dosis for et menneske er 159 ppm ved en virketid på 10 minutter (jfr. 92 ppm for BrCN) .. Da irri- 3 15 tationstærskelværdien for C1CN med 2,5 mg pr. m ligger 3 tydeligt lavere end værdien for BrCN (6,0 mg pr. m ), har C1CN en større "selv-varsling". Det har således for en teknisk anvendelse vigtige sikkerhedsfordele.
Desuden har anvendelsen af C1CN også vigtige 20 fordele med hensyn til den tekniske fremgangsmåde: C1CN kan som gas doseres og transporteres simplere og mere ufarligt end det faste stof BrCN.
Efter at reaktionen er tilendebragt, kan man fjerne chlorcyanid-overskuddet fra reaktionsblandingen ved ind-25 blæsning af en egnet gas. Egnede er indifferente gasser såsom nitrogen. Til undertrykkelse af skumdannelsen kan man anvende egnede skumdæmpende midler. Velegnede er eksempelvis typerne SE 3, 6 og 9 fra firmaet Wacker Chemie. I tilslutning dertil oparbejder man reaktionsblan-30 dingen på gængs måde.
Den CICN-holdige gasstrøm kan ledes gennem en gasvasker, der er fyldt med vandig NaOH- og/eller NaOCl-opløs-ning. Derved nedbrydes C1CN til NaCl og NaOCN (jfr. Gmelin. Syst. Nr. 14 C, F 1, D 3, 1976, s. 184; Petty's Industrial 35 Hygiene and Toxikology, 3. Auflage 1982, Bd. 2 c, s. 4859).

Ved denne fremgangsmåde kan man bortskaffe spaltningsreagenset (C1CN) væsentligt mere sikkert end det mere giftige BrCN. Bortskaffelsen er dermed tidsligt og rumligt adskilt fra proteinisoleringen, der kan gennemfø-5 res ved kendte fremgangsmåder (inddampning, frysetørring).
Man anvender fortrinsvis chlorcyanid i et molært overskud fra 5 til 8 gange pr. methionyl-binding, der skal spaltes. Reaktionstiden er sædvanligvis fra 1 til 10 timer, fortrinsvis fra 3 til 6 timer.
10 Som reaktionsmedium egner sig en blanding af vand og en mineralsyre eller en med vand blandbar organisk syre. Organiske syrer, såsom myresyre, foretrækkes. Indholdet af myresyre i reaktionsmediet kan være fra 50 til 95 rumfangsprocent, fortrinsvis fra 65 til 88 rumfangsprocent.

Eksempel 1 (sammenligningseksempel').
1 g β-Galactosidase fra E. Coli (indhold ca. 70%, 20 methionin-indhold 0,16 mmol pr. g, bestemt ved aminosyre-analyse) røres sammen med 7,5 ml 70 volumenprocents HCOOH, og der tilsættes 0,135 g (1,28 mmol) bromcyanid.
Reaktionsblandingen omrøres i 6 timer ved stuetemperatur, fortyndes med 100 ml H20 og frysetørres.
25 Remanensen udgør 0,97 g spaltningsproteinblanding.
Ved gelelektroforesen (SDS-PAGE) kan man ikke længere se nogen (3-galactosidase. Man finder fortrinsvis proteinfragmenter i et molekylvægtsområde fra 5.000 til 20.000 Dalton. Ifølge aminosyreanalyse og gaskromatografisk 30 methylrhodani-dbestemmelse udgør spaltnings graden 94%.
Eksempel 2 1 g /?-Galactosidase fra E. coli (indhold ca. 70%, methionin—indhold 0,16 mmol pr. g, bestemt ved aminosyreana-35 lyse) røres sammen med 7,5 ml 70 volumen-%'s HCOOH, og der tilsættes 0,079 g (1,28 mmol) C1CN.

Efter at reaktionen er tilendebragt, tilsætter man fra 1 til 5 yul skumdæmpende middel (SE 9 fra firmaet 5 Wacker Chemie, Burghausen), og det overskydende chlorcyanid fjernes ved indblæsning af N2 (15 1 pr. time) i ca.
1,5 timer. Den chlorcyanid-holdige N2-strøm ledes til en vaskeflaske, fyldt med vandig natriumhydroxid-opløsning.
Efter uddrivelsen af chlorcyanidet fortyndes opløs-10 ningen med 100 ml H20 og frysetørres.
Remanensen udgør 1 g.
Ved SDS-PAGE kan man ikke længere se nogen β-galacto-sidase. Man finder fortrinsvis proteinfragmenter i et molekylvægtsområde fra 5.000 til 20.000 Dalton. Ifølge 15 aminosyreanalyse og gaskromatografisk methylthiodanidbe-stemmelse udgør spaltningsgraden 93%.
Eksempel 3 1 g β-Lactamase, isoleret fra E. Coli-celler, som 20 indeholder plasmidet pBR 322 (indhold ca. 75%, methioninind-hold 0,28 mmol pr. g) røres sammen med 10 ml 70 volumen-%'s HCOOH, og der tilsættes 0,236 g (2,28 mmol) CICN.
Reaktionsblandingen omrøres i 6 timer ved stuetemperatur .

Remanensen udgør 0,99 g.

kylvægtsområde fra 3.000 til 10.000 Dalton. Ifølge amino-syreanalyse og gaskromatografisk methylrhodanidbestemmelse udgør spaltningsgraden 92%.

Claims (9)
1. Fremgangsmåde til spaltning af peptider og proteiner ved methionyl-bindingen, kendetegnet ved, at man gennemfører spaltningen med chlorcyanid anvendt i et 5 molært overskud fra 2 til 30 gange pr. methionyl-binding, der skal spaltes.
2. Fremgangsmåde ifølge krav 1, kendetegnet ved, at man, efter at reaktionen er tilendebragt, fjerner chlorcyanidoverskuddet fra reaktionsblandingen med en 10 egnet gas og derpå oparbejder reaktionsblandingen på gængs måde.
3. Fremgangsmåde ifølge krav 2, kendetegnet ved, at man fjerner chlorcyanid-overskuddet fra reaktions-blandingen med nitrogen.
4. Fremgangsmåde ifølge ét af kravene 1-3, ken detegnet ved, at man anvender chlorcyanid i et molært overskud fra 5 til 8 gange pr. methionyl-binding, der skal spaltes.
5. Fremgangsmåde ifølge ét af kravene 1-4, k e n- 20 detegnet ved, at man gennemfører reaktionen i løbet af 1 til 10 timer.
6. Fremgangsmåde ifølge ét af kravene 3-5, k e n-detegnet ved, at man gennemfører reaktionen i løbet af 3 til 6 timer.
7. Fremgangsmåde ifølge ét af kravene 3-6, k e n- detegnet ved, at man som reaktionsmedium anvender en blanding af vand og en med vand blandbar syre.
8. Fremgangsmåde ifølge krav 7, kendetegnet ved, at man som reaktionsmedium anvender en blanding af 30 vand og myresyre.
9. Fremgangsmåde ifølge krav 8, kendetegnet ved, at myresyreindholdet i reaktionsmediet udgør fra 50 til 95 rumfangsprocent. 35
