Description

ducerer human-parathyroid hormon (PTH), kan (A) fremstilles ved (a) isolering af mRNA fra porcine parathyroider, (b) kloning af den isolerede mRNA som ds-cDNA ved hjælp af en vektor i E. coli, (c) isolering af hybridvektor-DNA fra pools af resulterende kloner, (d) fiksering af hybridvektor-DNA til en bærer for hver pool, hybridisering af mRNA isoleret ifølge (a) og dernæst fjernelse deraf igen, udførelse af forsøg på at translatere mRNA, der er blevet fjernet, til porcint parathyroid hormon (PTH), der er blevet identificeret ved antistof- -fældning, hvorved der bestemmes kloner, der er opnået ifølge (b), som har porcine parathyroid hormon (PTH)-gensekvenser, sekvensering af hybridvektor-DNA af bestemte kloner med porcine parathyroid hormon(PTH)-gensekvenser, og identifikation af den klon eller de kloner, der har hybridvektor-DNA, med en DNA-sekvens, der koder for porcint parathyroid hormon (PTH), (e) radioaktiv mærkning af hybridvektor-DNA af de ifølge (d) identificerede kloner, (f) screening af en human genbank med resulterende radioaktivt mærket hybridvektor-DNA og (g) isolering af det bestemte human-parathyroid hormon (PTH)-gen (DNA-sekvensen mellem EcoRI-restrik-tionsstederne ifølge (B) og overføring af dette til en ekspressions-hybridvektor, der kan klones i eukaryotiske celler, (B) og denne vektor er karakteriseret ved en DNA-sekvens lokaliseret mellem to EcoRI-restriktions-steder, der inkluderer en specifik DNA-sekvens, eller en subregion af DNA-sekvensen lokaliseret mellem EcoRI-restriktionsstedeme, især en subregion , der udtrykker human-parathyroid hormon (PTH) eller angonister eller antagonister deraf.
