Claims (9)
1. Fremgangsmåde til fremstilling af et protein i en cellekultur omfattende: (a) at dyrke celler i cellekulturen ved en reduceret temperatur, hvor den reducerede temperatur ligger i området fra 27,0°C til mindre end 30,0°C; og (b) at dyrke celler i cellekulturen ved en reduceret pH-værdi, hvor den reducerede pH-værdi ligger i et område på 6,80 til mindre end 7,00; for at reducere produktionen af forkert foldede proteiner og/eller aggregerede proteiner, hvor cellekulturen er en pattedyr-cellekultur, hvor det producerede protein er en opløselig receptor, hvor den opløselige receptor er TNFR-Fc eller slL-13R.
2. Fremgangsmåde ifølge krav 1, hvor cellekulturen er en CHO-cellekultur.
3. Fremgangsmåde ifølge krav 1 eller 2, hvor den opløselige receptor er TNFR-Fc.
4. Fremgangsmåde ifølge krav 3, hvor den reducerede temperatur er 29,5 °C.
5. Fremgangsmåde ifølge krav 3 eller 4, hvor den reducerede pH-værdi er 6,95.
6. Fremgangsmåde ifølge krav 1, hvor den opløselige receptor er sll_-13R
7. Fremgangsmåde ifølge et af kravene 1-6, yderligere omfattende et trin, hvor proteinet isoleres fra cellekulturen.
8. Fremgangsmåde ifølge krav 7, hvor proteinet endvidere oprenses eller forarbejdes til formulering.
9. Fremgangsmåde ifølge krav 8, hvor proteinet formuleres til en farmaceutisk sammensætning.
