Claims (10)
1
1. Plasm idvektor, omfattende: (a) en RNA-polymerasefremmer, som er virksomt forbundet med en DNA, 5 der koder et infektiøst RNA-molekyle, og (b) en poly-A-hale nedstrøms for nævnte DNA, hvori (i) RNA-molekylet koder en svækket gul feber (YF) virus, og (ii) RNA-polymerasefremmeren er egnet til eksprimering i pattedyrsceller. 10
2. Vektor ifølge krav 1, hvori fremmeren er placeret fra ca. 12 til ca. 18 nukleoti-der opstrøms for 5’-enden for nævnte DNA.
3. Vektor ifølge krav 2, hvori fremmeren er placeret 15 nukleotider opstrøms for 15 5’-enden af nævnte DNA.
4. Vektor ifølge ethvert foregående krav, hvori poly-A-halen er placeret fra 0 til 500 nukleotider nedstrøms for nævnte DNA.
5. Vektor ifølge ethvert foregående krav, hvori fremmeren er en CMV-RNA- polymerasefremmer.
6. Vektor ifølge ethvert foregående krav, hvori nævnte DNA, der koder RNA-molekylet, omfatter en YF17D cDNA (nukleotider 703 til 11564 i Figur 8) eller en 25 polynukleotidsekvens med mindst 95% sekvensidentitet med nævnte YF17D cDNA over sidstnævntes samlede længde.
7. Vaccine omfattende en terapeutisk effektiv mængde af vektoren ifølge ethvert af de foregående krav. 30
