Claims (11)
1. Kimær, adenoviral ekspressionsvektor til beskyttelse mod pandemisk H1N1-influenza, hvilken vektor omfatter en ekspressionskassette omfattende: (a) en første promoter, der er operativt forbundet med en nukleinsyre, der koder for en toll-like receptor-3- (TLR-3) agonist, hvor TLR-3-agonisten er et dobbeltstrenget RNA (dsRNA); og (b) en anden promoter, der er operativt forbundet med en nukleinsyre, der koder for hæmagglutininpolypeptid fra A/CA/04/2009 H1N1-influenza.
2. Kimær, adenoviral ekspressionsvektor ifølge krav 1, hvor nukleinsyren, der koder for TLR-3-agonisten, omfatter et kort hårnåle-RNA.
3. Kimær, adenoviral ekspressionsvektor ifølge krav 1, hvor nukleinsyren, der koder for TLR-3-agonisten, omfatter en sekvens, der er udvalgt fra gruppen bestående af SEQ ID NO: 1-6.
4. Kimær, adenoviral ekspressionsvektor ifølge et hvilket som helst af de foregående krav, hvor den første promoter og den anden promoter er ens.
5. Kimær, adenoviral ekspressionsvektor ifølge krav 4, hvor den første promoter og den anden promoter begge er en CMV-promoter.
6. Farmaceutisk sammensætning omfattende den kimære, adenovirale ekspressionsvektor ifølge et hvilket som helst af de foregående krav og en farmaceutisk acceptabel bærer, hvor den farmaceutiske sammensætning er formuleret til oral, mukosal eller intranasal administration.
7. Farmaceutisk sammensætning til oral indgivelse, hvilken sammensætning omfatter den kimære, adenovirale ekspressionsvektor ifølge et hvilket som helst af kravene 1-5 og en farmaceutisk acceptabel bærer.
8. Immunogent effektiv mængde af en kimær, adenoviral ekspressionsvektor ifølge et hvilket som helst af kravene 1 -5, eller sammensætning ifølge krav 6 eller 7, til anvendelse i en fremgangsmåde til fremkaldelse af et immunrespons på pandemisk H1N1-influenza hos et pattedyr, hvor immunresponset er rettet mod hæmagglutininpolypeptidet, og hvor administrationsvejen er udvalgt fra gruppen bestående af: oral, intranasal og mukosal.
9. Ekspressionsvektor eller sammensætning til anvendelse i fremgangsmåden ifølge krav 8, hvor administrationsvejen er oral.
10. Ekspressionsvektor eller sammensætning til anvendelse i fremgangsmåden ifølge krav 8 eller 9, hvor immunresponset er et mukosalt immunrespons.
11. Ekspressionsvektor eller sammensætning til anvendelse i fremgangsmåden ifølge et hvilket som helst af kravene 8-10, hvor pattedyret er et menneske.
