Claims (12)
1. Effektiv mængde af et isoleret antistof eller et antigenbindende fragment deraf, der binder specifikt til semaphorin-4D (SEMA4D), til anvendelse til behandling til reducering af permeabiliteten af blod-hjerne-barrieren (BBB) hos et individ med forøget BBB-permeabilitet og en neuroinflammatorisk forstyrrelse, hvor det isolerede antistof eller det antigenbindende fragment deraf specifikt hæmmer semaphorin 4D (SEMA4D)'s interaktion med SEMA4D-receptoren plexin Bl.
2. Antistof eller antigenbindende fragment deraf til anvendelse ifølge krav 1, hvor individet er et pattedyr.
3. Antistof eller antigenbindende fragment deraf til anvendelse ifølge krav 2, hvor pattedyret er et menneske.
4. Antistof eller antigenbindende fragment deraf til anvendelse ifølge et hvilket som helst af kravene 1 til 3, hvor antistoffet eller det antigenbindende fragment deraf hæmmer kompetitivt et reference-monoklonalt antistof, der omfatter 1) den variable tunge kæde (VH) med aminosyresekvensen ifølge SEQ ID NO: 9 og den variable lette kæde (VL) med aminosyresekvensen ifølge SEQ ID NO: 17 eller 2) VH-aminosyresekvensen ifølge SEQ ID NO: 10 og VL-aminosyresekvensen ifølge SEQ ID NO: 18.
5. Antistof eller antigenbindende fragment deraf til anvendelse ifølge et hvilket som helst af kravene 1 til 4, hvor antistoffet eller det antigenbindende fragment deraf binder specifikt til den samme SEMA4D-epitop som et reference-monoklonalt antistof, der omfatter 1) VH-aminosyresekvensen ifølge SEQ ID NO: 9 og VL-aminosyresekvensen ifølge SEQ ID NO: 17 eller 2) VH-aminosyresekvensen ifølge SEQ ID NO: 10 og VL-aminosyresekvensen ifølge SEQ ID NO: 18.
6. Antistof eller antigenbindende fragment deraf til anvendelse ifølge et hvilket som helst af kravene 1 til 5, hvor antistoffet eller det antigenbindende fragment deraf er humant eller humaniseret.
7. Antistof eller antigenbindende fragment deraf til anvendelse ifølge et hvilket som helst af kravene 1 til 6, hvor antistoffet eller det antigenbindende fragment deraf omfatter en VH, der omfatter VH-CDR'erne 1-3, der omfatter henholdsvis SEQ ID NO: 6, 7 og 8, og en VL, der omfatter VL-CDR'erne 1-3, der omfatter henholdsvis SEQ ID NO: 14, 15 og 16.
8. Antistof eller antigenbindende fragment deraf til anvendelse ifølge krav 7, hvor antistoffet eller det antigenbindende fragment deraf omfatter 1) VH-aminosyresekvensen ifølge SEQ ID NO: 9 og VL-aminosyresekvensen ifølge SEQ ID NO: 17 eller 2) VH-aminosyresekvensen ifølge SEQ ID NO: 10 og VL-aminosyresekvensen ifølge SEQ ID NO: 18.
9. Effektiv mængde af et isoleret antistof eller et antigenbindende fragment deraf, der binder specifikt til semaphorin-4D (SEMA4D), til anvendelse ifølge krav 1, og en effektiv mængde af et isoleret antistof eller et antigenbindende fragment deraf, der binder specifikt til plexin-Bl, til anvendelse til behandling af et indvid med forøget BBB-permeabilitet og en neuroinflammatorisk forstyrrelse, hvor SEMA4D- og plexin-Bl-antistofferne eller de antigenbindende fragmenter deraf kan reducere permeabilitetet af BBB.
10. Antistof eller antigenbindende fragment deraf til anvendelse ifølge et hvilket som helst af kravene 1 til 9, hvor den neuroinflammatoriske forstyrrelse er valgt fra gruppen, der består af multipel sklerose, amyotrofisk lateral sklerose, epilepsi, Alzheimers sygdom, Parkinsons sygdom, meningitis, hjerneødem og hjernetraume.
11. Antistof eller antigenbindende fragment deraf til anvendelse ifølge et hvilket som helst af kravene 1 til 10, hvor den effektive mængde af antistoffet eller det antigenbindende fragment deraf fremkalder en forøgelse af den specifikke modstand i BBB'en, en forøgelse af antallet, densiteten eller koncentrationen af endotelceller, der er til stede på BBB'en, en ændring i morfologien eller funktionen af endotelcellerne, en ændring i interaktioner mellem endotelceller eller astrocytter, en ændring i interaktioner mellem endotelceller og astrocytter, der danner BBB'en.
12. Antistof eller antigenbindende fragment deraf til anvendelse ifølge et hvilket som helst af kravene 1 til 11, hvor antistoffet eller det antigenbindende fragment deraf anvendes i kombination med en eller flere behandlinger mod neuroinflammatoriske forstyrrelser, hvor den yderligere terapi administreres før, under eller efter brugen af antistoffet eller det antigenbindende fragment deraf.
