Description
CHINOXALIN-DERIVATE, MIT AFFINITÄT AN DIE QUISQUALAT-REZEPTOREN
Die Erfindung betrifft Chinoxalindion-carbonsäure- und -phosphonsäurederivate, deren Herstellung und Verwendung in Arzneimitteln.
Es ist bekannt, daß Chinσxalinderivate Affinität an die Quisqualat-Rezeptoren besitzen und sich auf Grund der Affinität als Arzneimittel zur Behandlung von Krankheiten des zentralen Nervensystems eignen.
Es wurde nun gefunden, daβ sich die erfindungsgemäβen Verbindungen im Vergleich mit den aus EP-A-315 959 und WO 91/138 78 bekannten Chinoxalinen durch ihr gutes Bindungsvermögen an die Quisqualat-Rezeptoren auszeichnen.
Die erfindungsgemäßen Verbindungen haben die Formel I
worin mit R2 substituiertes C1-12-Alkyl, mit R2 substituiertes
C2-12-Alkenyl, mit R2 substituiertes C2-12-Alkinyl, mit R2 substituiertes C3-7-Cycloalkyl, - (CH2)n-C6-12-Aryl, das im Aryl- oder im
Alkyl-Rest mit R2 substituiert ist oder -(CH2)n-Hetaryl, das im Hetaryl- oder Alkylrest mit R2 substituiert ist,
Wasserstoff, mit R2 substituiertes C2-12-Alkyl, mit R2 substituiertes C2-12-Alkenyl, mit R2 substituiertes C2 -12-Alkinyl. (CH2)n-C6-12-Aryl, das im Aryl- oder Alkylrest mit R2 substituiert ist oder -(CH2)n-Hetaryl, das im Hetaryl- oder Alkylrest mit R2 substituiert ist,
R5, R6, R7 und R8 gleich oder verschieden sind und Wasserstoff, Halogen, Nitro, NR9R10, NHCOR11, SO R12, C3-7-Cycloalkyloxy, COR13, Cyano, CF3, C1-6-Alkyl, C1-4-Alkoxy oder gegebenenfalls mit Cyano, C1-4-Alkyl oder
-COO-C1-6-Alkyl substituiertes Imidazol bedeuten oder
R5 und R6 oder R7 und R8 einen ankondensierten Benzolring darstellen, wobei
R2 -CO-R3 oder -PO-XY und R2 ein- bis zweifach gleich oder verschieden steht und n 0, 1, 2, 3, 4 oder 5 ist und
R3 Hydroxy, C1-6-Alkoxy oder NR9R10,
X und Y gleich oder verschieden sind und Hydroxy, C1-6-Alkoxy, C1-4-Alkyl oder NR9 R10 bedeuten und
R 9 und R10 gleich oder verschieden sind und Wasserstoff, C1-4-Alkyl oder gemeinsam mit dem Stickstoffatom einen gesättigten 5- oder
6-gliedrigen Heterocyclus bilden, der ein weiteres Sauerstoff,
Schwefel oder Stickstoffatom enthalten kann,
R11 C1-6-Alkyl oder Phenyl,
R12 Wasserstoff, C1-4-Alkyl, NH-, N{C1-4-Alkyl), und
R13 Hydroxy, C1-6-Alkoxy, C1-6-Alkyl oder NR9R10 bedeuten
sowie deren Isomeren oder Salze, wobei, falls R4, R5, R6, R7 und R8 Wasserstoff bedeuten, kann R1 nicht Carbamoylmethyl, 1-Carboxy-1-phenylmethyl oder geradkettiges C1-6-Alkyl sein, das in 1-Stellung mit -COOH oder -COO-C1-6-Alkyl substituiert ist, und falls R geradkettiges C1-6-Alkyl, das in 1 -Stellung mit -COOH oder
-COO-C1-6-Alkyl substituiert ist, bedeutet, können R6 und/oder R7 bzw. R6 und R8 nicht Fluor, Chlor oder Brom und R4-R8 jeweils
Wasserstoff sein, und
falls R1 -CH2-COOH ist, können
a) R 6 und R7 nicht gleichzeitig Methyl oder
b) R6 oder R7 nicht NO2 und R4-R8 jeweils Wasserstoff sein.
Die Verbindungen der allgemeinen Formel I beinhalten auch die möglichen tautomeren Formen und umfassen die E- oder Z-Isomeren oder, falls ein chirales Zentrum vorhanden ist, die Razemate oder Enantiomeren.
Die Substituenten stehen bevorzugt in 6- und/oder 7-Stellung. Der Substituent R2 steht ein- bis zweifach gleich oder verschieden in beliebiger Position am Alkyl-, Alkenyl-, Alkinyl-, Cycloalkyl-, Hetaryl- oder Arylrest.
Unter Alkyl ist jeweils ein geradkettiger oder verzweigter Alkylrest zu verstehen wie beispielsweise Methyl, Ethyl, Propyl, Isopropyl, Butyl, Isobutyl, sek. Butyl, tert. Butyl, Pentyl, Isopentyl, Hβxyl, Heptyl, Octyl, Nonyl, Decyl, wobei C1-6-Alkylreste bevorzugt werden.
Alkenyl beinhaltet insbesondere C2-6-Alkenylreste, die geradkettig oder verzweigt sein können wie beispielsweise 2-Propenyl, 2-Butenyl,
3-Methyl-2-propenyl 1-Propenyl, 1-Butenyl, Vinyl.
Als Alkinylreste sind insbesondere Ethinyl, 1-Propinyl, 2-Propinyl, 1-Butinyl mit 2-4 Kohlenstoffatomen geeignet.
Unter C3-7-Cycloalkyl ist jeweils Cyclopropyl, Cyclobutyl, Cyclopentyl,
Cyclohexyl und Cycloheptyl gemeint insbesondere C3-5-Cycloalkyl.
Als Arylrest seien beispielsweise Phenyl, Naphthyl, Biphenylyl und Indenyl genannt, insbesondere (CH2)n-Phenyl mit n = 0,1 oder 2.
Als Hetarylrest sind 5- oder 6-gliedrige Heteroaromaten mit 1-3 Stickstoffatomen geeignet wie beispielsweise Pyrazol, Imidazol, Pyrazin, Pyridin, Pyrimidin, Pyridazin, Triazin.
Unter Halogen ist Fluor, Chlor, Brom und Jod zu verstehen.
Bilden R9 und R10 gemeinsam mit dem Stickstoffatom einen gesättigten Heterocyclus, so ist beispielsweise Piperidin, Pyrrolidin, Morpholin, Thiomorpholin oder Piperazin gemeint.
Bedeutet R1 C1-12-Alkyl und R2 COR3 , so sind R5-R8 insbesondere Substituenten wie NO2, NR9R10, NHCOR11, SO2R12, C3-7-Cycloalkyloxy, COR13, Cyano, CF3, C1-4-Alkoxy, gegebenenfalls substituiertes Imidazol oder ein ankondensierter Benzolring. Die Verbindungen der Formel I mit R2 = -PO-XY zeichnen sich durch sehr gute Wasserlöslichkeit aus.
Unter physiologisch verträglichen Salzen sind Salze organischer und anorganischer Basen wie beispielsweise die gut löslichen Alkali-und Erdalkalisalze sowie N-Methyl-glukamin, Dimethyl-glukamin, Ethyl-glukamin, Lysin, 1,6-Hexandiamin, Ethanolamin, Glukosamin, Sarkosin, Serinol, Tris-hydroxy-methyl-amino-methan, Aminopropandiol, Sovak-Base, 1-Amino- 2,3,4-butantriol zu verstehen.
Die Verbindungen der Formel I sowie deren physiologisch verträglichen Salze sind auf Grund ihrer Affinität zu den Quisqualat-Rezeptoren als Arzneimittel verwendbar. Auf Grund ihres Wirkprofils eignen sich die erfindungsgemäßen Verbindungen zur Behandlung von Krankheiten, die durch Hyperaktivität excitatorischer Aminosäuren wie Glutamat oder Aspartat hervorgerufen werden. Da die neuen Verbindungen als Antagonisten excitatorischer Aminosäuren wirken und eine hohe spezifische Affinität zu den AMPA-Rezeptoren zeigen, indem sie den radioaktiv markierten spezifischen Agonisten (RS)α-Amino-3-hydroxy-5-methyl-4-isoxazolpropionat (AMPA) von den AMPA-Rezeptoren verdrängen, eignen sie sich insbesondere zur
Behandlung von solchen Krankheiten, die über die Rezeptoren excitatorischer Aminosäuren, insbesondere den AMPA-Rezβptor, beeinflußt werden können, wie beispielsweise Morbus Parkinson, Morbus Alzheimer, Morbus Huntington, Epilepsie, Hypoglykämie, Psychosen, Muskelsteifigkeit,
Emesis, Schmerzzustände, Anoxie und Defizite nach Ischämie.
Die Erfindung umfaßt auch die Kombination der erfindungsgemäθen Verbindungen mit Dopaminagonisten wie Lisurid, Tergurid, Bromokriptin, Amantadin-Derivaten, Memantin und dessen Derivate und den in EP-A-351 352 beschriebenen Verbindungen sowie die Kombination mit L-DOPA bzw. L-DOPA und Benserazid. In der Kombination wird die zu verabfolgende Dosis des herkömmlichen Arzneimittels verringert und deren Wirkung synergistisch gesteigert.
Die Affinität der erfindungsgemäβen Verbindungen zu zentralen AMPA-Rezeptoren wurde in klassischen Bindungsuntersuchungen in-vitro geprüft. Sie binden mit hoher Affinität an mit 3H-AMPA markierten Bindungsstellen.
Zur Prüfung der Wirkqualität und Wirksamkeit in-vivo wurden die Verbindungen nach intravenöser Applikation an Mäusen geprüft. Nach Vorbehandlung mit den erfindungsgemäβen Verbindungen werden die durch
intracerebroventrikulärβ Injektion von AMPA ausgelösten Krämpfe dosisabhängig antagonisiert.

Diese Befunde zeigen, daß es sich bei den Verbindungen um potente zentral wirksame AMPA-Antagonisten handelt. Damit sind sie für die Behandlung von Krankheitsszuständen geeignet, die mit einer Störung des Glutamatstoffwechsels einhergehen. Insbesondere eignen sie sich zur Behandlung der cerebralen Ischämie unterschiedlicher Genese, des Morbus Parkinson und auch der anderen im vorigen Absatz genannten Erkrankungen.
Zur Verwendung der erfindungsgemäßen Verbindungen als Arzneimittel werden diese in die Form eines pharmazeutischen Präparats gebracht, das neben dem Wirkstoff für die enterale oder parenterale Applikation geeignete pharmazeutische, organische oder anorganische inerte Trägermaterialien, wie zum Beispiel Wasser, Gelatine, Gummi arabicum, Milchzucker, Stärke, Magnesiumstearat, Talk, pflanzliche Öle, Polyalkylenglykole usw. enthält. Die pharmazeutischen Präparate können in fester Form, zum Beispiel als Tabletten, Dragees, Suppositorien, Kapseln oder in flüssiger Form, zum Beispiel als Lösungen. Suspensionen oder Emulsionen vorliegen. Gegebenenfalls enthalten sie darüber hinaus Hilfsstoffe wie Konservierungs-, Stabilisierungs-, Netzmittel oder Emulgatoren, Salze zur Veränderung des osmotischen Drucks oder Puffer.
Für die parenterale Anwendung sind insbesondere Injektionslösungen oder Suspensionen, insbesondere wässrige Lösungen der aktiven Verbindungen in polyhydroxyethoxyliertem Rizinusöl, geeignet.
Als Trägersysteme können auch grenzflächenaktive Hilfsstoffe wie Salze der Gallensäuren oder tierische oder pflanzliche Phospholipide, aber auch Mischungen davon sowie Liposome oder deren Bestandteile verwendet werden.
Für die orale Anwendung sind insbesondere Tabletten, Dragees oder Kapseln mit Talkum und/oder Kohlenwasserstoffträger oder -binder, wie zum Beispiel Lactose, Mais- oder Kartoffelstärke, geeignet. Die Anwendung kann auch in flüssiger Form erfolgen, wie zum Beispiel als Saft, dem gegebenenfalls ein Süßstoff beigefügt wird.
Die Dosierung der Wirkstoffe kann je nach Verabfolgungsweg, Alter und Gewicht des Patienten, Art und Schwere der zu behandelnden Erkrankung und ähnlichen Faktoren variieren. Die tägliche Dosis beträgt 0,5 - 1000 mg, vorzugsweise 50 - 200 mg, wobei die Dosis als einmal zu verabreichende Einzeldosis oder unterteilt in 2 oder mehrere Tagesdosen gegeben werden kann.
Die Herstellung der erfindungsgemäβen Verbindungen erfolgt nach an sich bekannten Methoden. Beispielsweise gelangt man zu Verbindungen der Formel I dadurch, daß man
a)eine Verbindung der Formel II
worin R 1, R5, R6, R7 und R8 die obige Bedeutung haben, mit reaktiven
Oxalsäuredβrivaten cyclisiert und gegebenenfalls mit R4 '-X worin X
Halogen, Tosylat, Mesylat oder Triflat bedeutet und R4' die Bedeutung von R4 mit Ausnahme von Wasserstoff hat, umsetzt oder b) eine Verbindung der Formel III

R4, R5, R6, R7 und R8 die obige Bedeutung haben, mit R1-X umsetzt zu Verbindungen der Formel I und gewünschtenfalls die Estergruppe verseift oder die Säuregruppe verestert oder amidiert oder die
Nitrogruppe reduziert zur Aminogruppe oder die Aminogruppe alkyliert oder acyliert oder die Aminogruppe gegen Halogen oder Cyano austauscht oder die Aminogruppe mit einem 2-Azabutadien der Formel IV
worin U und V Fluchtgruppen darstellen und R11 Wasserstoff, Cyano oder COOC1-6-Alkyl ist und R12 Wasserstoff oder C1-6-Alkyl ist, umsetzt zu einem Imidazolderivat oder die Isomeren trennt oder die Salze bildet.
Die Cyclisierung der Verbindungen der Formel II mit einem reaktiven Oxalsäurederivat erfolgt einstufig oder auch zweistufig. Als bevorzugt ist das Zweistufenverfahren zu betrachten, bei dem das Diamin mit einem Oxalsäurederivat wie dem Oxalesterhalbchlorid in polaren Lösungsmitteln wie Dimethylfσrmamid oder cyclischen oder acyclischen Ethern oder halogenierten Kohlenwasserstoffen beispielsweise Tetrayhdrofuran, Diethylether oder Methylenchlorid in Gegenwart einer Base wie organischen Aminen beispielsweise Triethylamin, Pyridin, Hünig-Base oder Diethylaminopyridin umgesetzt wird. Die anschlieβende Cyclisierung kann basisch oder auch sauer, vorzugsweise aber in saurem Milieu durchgeführt werden, wobei dem Lösungsmittel Alkohol zugesetzt werden kann. Die Einführung der Substituenten R1 und R4 erfolgt nach den üblichen Alkylierungsmethoden, indem man das Chinoxalindion mit R1- oder R4 '-X, worin X Tosylat, Mesylat oder insbesondere Triflat oder Halogen bedeutet, in Gegenwart von Basen bei Raumtemperatur oder erhöhter Temperatur in aprotischen Lösungsmitteln umsetzt. Das Anion kann auch erzeugt werden, bevor R1- oder R4 'X zugegeben werden.Als Basen sind beispielsweise
Alkaliverbindungen wie Kaliumcarbonat, Natriumhydroxid, Alkalialkoholate
und insbesondere Metallhydride wie Natriumhydrid geeignet. Eventuell können die Alkaliverbindungen auch unter Phasentransferbedingungen umgesetzt werden. Erhält man Gemische von Verbindungen mit dem
Substituenten R1 bzw. R4, so werden diese in üblicher Weise getrennt.
Für die Umsetzung geeignete Lösungsmittel sind aprotische polare Lösungsmittel wie Dimethylformamid, N-Methylpyrrolidon aber auch cyclische Ether wie Dioxan oder Tetrahydrofuran. Wird in der Verfahrensvariante b) mit 2 Mol R -X unter sonst analogen Reaktionsbedingungen umgesetzt, so werden gleichzeitig die Substituenten R1 und R4 eingeführt.
Die sich gegebenfalls anschließende Verseifung einer Estergruppe kann basisch oder vorzugsweise sauer erfolgen, indem man bei erhöhter Temperatur bis zur Siedetemperatur des Reaktionsgemisches in Gegenwart von Säuren wie hoher konzentrierter wässriger Salzsäure in Lösungsmitteln wie beispielsweise Trifluoressigsäure oder Alkoholen hydrolysiert. Phosphonsäureester werden bevorzugt durch Erhitzen in hochkonzentrierten wässrigen Säuren wie zum Beispiel konzentrierter Salzsäure oder durch Behandlung mit Trimethylsilylbromid und anschließende Behandlung mit Wasser hydrolysiert.
Die Veresterung der Carbonsäure oder Phosphonsäure geschieht in an sich bekannter Weise mit dem entsprechenden Alkohol in Säure oder in Gegenwart eines aktivierten Säurederivats. Als aktivierte Säurederivate kommen zum Beispiel Säurechlorid, -imidazolid oder -anhydrid in Frage. Bei den
Phosphonsäuren ist die Umsetzung mit Orthoestern des entsprechenden Alkohols möglich. Auch die Umsetzung mit dem Anlagerungsprodukt von Dicyclohexylcarbodiimid und dem entsprechenden Alkohol führt zu dem Ester. Methylester können durch Reaktion mit Diazomethan dargestellt werden.
Die Amidierung erfolgt an den freien Säuren oder an deren reaktiven Derivaten wie beispielsweise Säurechloriden, gemischten Anhydriden, Imidazoliden oder Aziden durch Umsetzung mit den entsprechenden Aminen bei Raumtemperatur.
Die Reduktion der Nitrogruppe zur Aminogruppe erfolgt katalytisch in polaren Lösungsmitteln bei Raumtemperatur oder erhöhter Temperatur unter
Wasserstoffdruck. Als Katalysatoren sind Metalle wie Raney-Nickel oder Edelmetallkatalysatσren wie Palladium oder Platin gegebenenfalls auf
Trägern geeignet. Statt Wasserstoff kann auch Ammoniumformiat in bekannter Weise benutzt werden. Reduktionsmittel wie Zinn- II-Chlorid oder Titan-III-chlorid können ebenso verwendet werden wie komplexe Metallhydride evtl. in Gegenwart von Schwermetallsalzen. Es kann vorteilhaft sein, vor der Reduktion die Estergruppe einzuführen.
Wird eine Alkylierung der Aminogruppe gewünscht, so kann nach üblichen Methoden beispielsweise mit Alkylhalogeniden alkyliert werden. Auch die reduktive Aminierung mit einem Aldehyd und Reduktionsmitteln wie Natriumcyanoborhydrid ist möglich. Die Acylierung erfolgt nach den bekannten Methoden. Beispielsweise wird in wässrigem Milieu in Gegenwart einer Base mit den entsprechenden Säureanhydriden oder Säurehalogeniden umgesetzt.
Die Einführung der Cyanogruppe kann mit Hilfe der Sandmeyer-Reaktion erfolgen; beispielsweise kann man die aus den Aminoverbindungen mit Nitriten intermediär gebildeten Diazoniumsalze mit Cyaniden in Gegenwart von Cu-I-cyanid oder mit K2Ni(CN)4 umsetzen.
Die Einführung der Halogene Chlor, Brom oder Jod über die Aminogruppe kann nicht wässrig oder wässrig erfolgen; beispielsweise nach Sandmeyer wässrig, indem man die mit Nitriten intermediär gebildeten Diazoniumsalze mit Cu(I) Chlorid oder Cu(I)bromid in Gegenwart der entsprechenden Säure Salzsäure oder Bromwasserstoffsäure umsetzt oder mit Kaliumiodid umsetzt. Nicht wässrig wird das Hydrochlorid in bekannter Weise mit i-Amylnitrit und beispielsweise Methyleniodid oder Bromoform in aprotischen Lösungsmitteln wie Dimethylformamid umgesetzt. Die Einführung von Fluor gelingt beispielsweise durch die Balz-Schiemann-Reaktion des Diazoniumtetrafluorborates.
Die Umsetzung der Aminogruppe mit 2-Azabutadienen der Formel IV zu den Imidazolderivaten erfolgt in Gegenwart von Säuren bei Temperaturen von 0 bis 150 °C. Die Fluchtgruppen U und V können gleich oder verschieden sein; insbesondere geeignet sind C1-3-Dialkylamine, wie Dimethyl-,
Diethyl-und Dipropylamin, und cyclische Amine, wie Pyrrolidin.
Die Umsetzung wird beispielsweise so ausgeführt, daβ das Aminderivat und das Azadien in einer organischen Säure, wie zum Beispiel Ameisensäure, Essigsäure, Propionsäure oder Trifluoressigsäure, zunächst bei
Raumtemperatur gerührt und dann bis zur Siedetemperatur des
Reaktionsgemisches (bis etwa 120 °C) erhitzt wird.
Die Säure kann gleichzeitig als Reaktionsmittel und auch als
Lösungsmittel dienen. Es können aber auch Lösungsmittel wie zum Beispiel Alkohole, Ether, Ketone, Ester, wie Ethylacetat, Kohlenwasserstoffe, wie Toluol, oder Halogenkohlenwasserstoffe, wie Tetrachlorkohlenstoff, zugesetzt werden.
Die Menge der Säure kann in weiten Grenzen variiert werden, sie wird jedoch im Überschuß angewendet. Vorzugsweise wird ein 3 - 10facher Säureüberschuß, bezogen auf das Amin und das Azadien, gewählt.
Die Isomerengemische können nach üblichen Methoden wie beispielsweise Kristallisation, Chromatographie oder Oberführung in Diastereomere wie z.B. Salzbildung in die Enantiomeren bzw. E/Z-Isomeren aufgetrennt werden.
Die Herstellung der Salze erfolgt in üblicher Weise, indem man eine Lösung der Verbindung der Formel I mit der equivalenten Menge oder einem Überschuß einer Alkali- oder Erdalkaliverbindung, die gegebenenfalls in Lösung ist, versetzt und den Niederschlag abtrennt oder in üblicher Weise die Lösung aufarbeitet.
Soweit die Herstellung der Ausgangsverbindungen nicht beschrieben wird, sind diese bekannt oder analog zu bekannten Verbindungen oder hier beschriebenen Verfahren herstellbar.
Beispielsweise können die Verbindungen der Formel II dargestellt werden, indem man 2,4-Dinitroarylamine nach der Methode von Sanger darstellt, dadurch daß man o-Halogen-nitroaromaten vorzugsweise o-Fluor-nitroaromaten wie beispielsweise Dinitrofluorbenzol in wässriger Lösung mit Aminosäurederivaten in Gegenwart einer Base wie Natriumcarbonat oder
Natriumhydrogencarbonat bei Temperaturen zwischen 0 °C bis Rückfluß umsetzt und anschließend reduziert. Diese Reaktion läßt sich auch auf andere substituierte 2-Nitrohalogenverbindungen übertragen. Auch durch Ulimann Reaktion von Dinitrochlorbenzol mit einem aromatischen Amin können Diarylaminoverbindungen erhalten werden. Für diese Reaktion werden hochsiedende Lösungsmittel wie Dimethylformamid oder Collidin und festes Kaliumcarbonat und Kupferpulver als Base verwendet. Es ist auch möglich, die entsprechenden o-Nitroaniline durch Alkylierung oder mit
substituierten Aldehyden durch reduktive Alkylierung herzustellen. Die sich anschließende Reduktion der o-Nitrogruppe erfolgt bei Anwesenheit mehrerer Nitrogruppen selektiv durch Natriumsulfid in Gegenwart von Ammoniak sowie Ammoniumchlorid in polaren Lösungsmitteln bei Raumtemperatur oder erhöhter Temperatur. In einigen Fällen ist es vorteilhaft, die Umsetzung mit Estern vorzunehmen und im letzten Schritt diese zu hydrolysieren.
Die Enantiomerentrennung kann auf der Endstufe oder in den Zwischenstufen durch optisch aktive Hilfsbasen wie z.B. Brucin oder 1-Phenethylamin erfolgen oder aber auch durch Chromatographie über optisch aktive Trägermaterialien. Die Enantiomeren können aber auch synthetisch durch
Umsetzung der entsprechenden optisch aktiven Aminosäuren mit den entsprechenden Fluornitroaromaten nach der Sanger-Methode und Weiterverarbeitung der Aminonitroaromaten wie oben beschrieben hergestellt werden.
Die nachfolgenden Beispiele sollen das erfindungsgemäβe Verfahren erläutern:
Beispiel 1
3-(6-Nitro-2,3-dioxo-1,2,3,4-tetrahydrochinoxalin-1-ylmethyl)-benzoesäuremethylester und
3-(7-Nitro-2,3-dioxo-1,2,3,4-tetrahydrochinoxalin-1-ylmethyl)-benzoesäuremethylester
1,03 g (5 mM) 6-Nitro-2,3-dioxo-1,2,3,4-tetrahydrochinoxalin werden unter N2-Strom und Feuchtigkeitssausschluβ in 50 ml Dimethylformamid bei Raumtemperatur vorgelegt. 330 mg (11 mM) Natriumhydrid (80Zig) werden in 3 Portionen zum Ansatz gegeben. Es wird dann 1 Stunde bei Raumtemperatur gerührt. 1,26 g (5,5 mM) 3-Brommethylbenzoesäuremethylester in 5 ml Dimethylformamid werden zum Ansatz getropft und 3 1/2 Stunde nachgerührt. Nach Einengen wird der Rückstand in essigsaurem Wasser/Essigester verteilt. Die organische Phase wird abgetrennt, getrocknet, filtriert und eingeengt. Der Rückstand wird über Kieselgel mit Dichlormethan : Ethanol = 95 : 5 chromatographiert. Man erhält neben 211 mg 3-(6-Nitro-2,3-dioxo-1,2,3,4-tetrahydrochinoxalin-1-ylmethyl)-benzoesäure-methylester, der nicht weiter gereinigt wird, 222 mg 3-(7-Nitro-2,3-dioxo-1,2,3,4-tetrahydrochinoxalin-1-ylmethyl)-benzoesäure-methylester vom Schmelzpunkt: 265 - 267 °C.
In analoger Weise werden hergestellt:

Beispiel 3
A) 4-(2,4-Dinitrophenyl)-aminobenzoesäure-ethylester
1,01 g (5 mM) 1-Chlor-2,4-dinitrobenzol, 1,01 g (6 mM) 4-Aminobenzoesäureethylester, 13 mg (0,2 mM) Kupferpulver und 961 mg (7 mM) Kaliumcarbonat (gepulvert) werden in 5 ml absolutem Dimethylformamid für 25 Minuten bei 180 °C Badtemperatur unter Argon und Feuchtigkeitsausschluß gerührt.
Nach Einengen wird in Wasser gegeben, mit Ammoniak alkalisch gestellt, mit Essigester ausgeschüttelt sowie die organische Phase getrocknet, filtriert und eingeengt. Der Rückstand wird über Kieselgel mit Cyclohexan : Essigester = 8 : 2 chromatographiert. Man erhält: 768 mg 4-(2,4-Dinitrophenyl)-aminobenzoesäure-ethylester vom Schmelzpunkt 99 - 102 °C.
In analoger Weise werden hergestellt:

566 mg (1,7 mM) 4-(2,4-Dinitrophenylamino)-benzoesäureethylester, 761 mg (12,2 mM) Ammoniumchlorid, 0,68 ml conc. Ammoniak, 15 ml Ethanol und 6 ml dest. Wasser werden bei 78 °C Innentemperatur (90 °C Badtemperatur) zusammen vorgelegt. In 3 Portionen werden 1,27 g (5,68 mM) Natriumsulfid
(35Zig) hinzugefügt und 1 Stunde nachgerührt. Der Ansatz wird bei Raumtemperatur abgesaugt und zunächst mit Wasser und dann unter Äther gewaschen. Man erhält 535 mg 4-(2-Amino-4-nitrophenylamino)-benzoesäureethylester als Rohprodukt (ohne weitere Aufreinigung verarbeitet).
In analoger Weise werden hergestellt:
3-(2-Amino-4-nitrophenylamino)-benzoesäureethylester vom Schmelzpunkt 145 - 150 °C.

C) 4-(6-Nitro-2.3-dioxo-1,2,3,4-tetrahydrochinoxalin-1-yl)-benzoesäureethylester
582 mg (2,3 mM) 4-(2-Amino-4-nitrophenylamino)-benzoesäureethylester werden mit 488 mg (4,8 mM) Triethylamin in 27 ml trockenem Tetrahydrofuran bei + 4 °C Badtemperatur unter Argon und Feuchtigkeitsausschluß vorgelegt. Eine Lösung aus 659 mg (4,8 mM) Oxalsäureethylesterchlorid und 8 ml trockenem Tetrahydrofuran wird zum Ansatz getropft und 2 Stunden bei Raumtemperatur gerührt. 0,2 ml Triethylamin und 0,1 ml Oxalsäureethylesterchlorid werden nachgegeben und 1 Stunde bei Raumtemperatur gerührt. Der Ansatz wird filtriert und das Filtrat eingeengt. Man verteilt in Wasser / Essigester. Die organische Phase wird eingeengt. Der Rückstand wird in 25 ml 1-N Salzsäure und 25 ml Ethanol für 2 Stunden bei Badtemperatur am RύckfluB gekocht. Das ausgefallene Produkt wird abgesaugt, mit Wasser gewaschen und getrocknet. Man erhält: 220 mg
4-(6-Nitro-2,3-dioxo-1,2,3,4-tetrahydrochinoxalin-1-yl)-benzoesäureethylester (ohne Aufreinigung weiterverarbeitet).

1,37 g (10 mM) 2-Aminobenzoesäure werden mit 2 g (18,7 mM) Natriumcarbonat in 40 ml Wasser bei 40 °C Badtemperatur unter kräftigem Rühren mit 1,86 g (10 mM) 2,4 Dinitrofluorbenzol versetzt und 2 Stunden gerührt. Der Ansatz wird ca. 400 ml Wasser verdünnt und mit 4-N HCl gefällt. Das Produkt wird abgesaugt,mit Wasser gewaschen und getrocknet. Man erhält 2,8 g 2-(2,4-Dinitrophenyl)-aminobenzoesäure vom Schmelzpunkt 266 - 270°C.

600 mg 5-Fluor-2,2-dinitrofluorbenzol werden in 30 ml Wasser und 10 ml Ethanol bei 40 °C vorgelegt und mit einer Lösung aus 376 mg rac.
Aminoethylphoshonsäure in 10 ml Wasser und 600 mg Natriumcarbonat tropfenweise versetzt. Es wird 1,5 Stunden bei Temperatur gerührt. Nach Abdestillieren des Ethanols wird gegen Essigsäure extrahiert. Die wassrige Phase wird mit 200 mg Imidazol versetzt und 2 Stunden auf 110 °C erwärmt. Danach werden nochmals 200 mg Imidazol zugegeben und 2 Stunden auf 110 °C erwärmt. Es wird mit 4-N-Salzsäure angesäuert, vom Ungelösten abgesaugt und das Filtrat mit Essigester gewaschen. Die wassrige Phase wird eingeengt und mit Ethanol ausgekocht. Der Ethanolextrakt wird eingeengt und über Kieselgel mit Methanol: Butanol:Wasser: Ammoniak = 75:25:17:3 chromatographiert. Man erhält 300 mg 5-(Imidazol-1-yl-)-2,4-dinitrophenyl-(1aminoethylphosphonsäure).
C) 2-(2-Amino-4-nitrophenylamino)-benzoesäure
1,80 g (6 mM) 2-(2,4-Dinitrophenylamino)-benzoesäure, 2,66 g (42,6 mM) Ammoniumchlorid, 2.4 ml Ammoniak conc, 52 ml Ethanol und 21 ml dest. Wasser werden bei 78 °C Innentemperatur (90 °C Badtemperatur) vorgelegt. In 3 Portionen werden 4.44 g (20 mM) Natriumsulfid (35Zig) zum Ansatz gegeben und 1 Stunde nachgerührt. Der Ansatz wird bei Raumtemperatur abgesaugt und nacheinander mit Wasser und Äther gewaschen. Das Filtrat wird bis zur wäßrigen Phase eingeengt und mit Essigester ausgeschüttelt. Die organische Phase wird getrocknet, filtriert und eingeengt. Die wäßrige Phase wird mit 1-N Salzsäure angesäuert und abgesaugt. Man erhält: 1,1 g 2-(2-Amino-4-nitrophenylamino)-benzoesäure (ohne Aufreinigung verarbeitet).

1-[5-(Imidazol-1-yl)-2-amino-4-nitrophenylamino]-methylphosphonsäure
D) (2-Amino-4-trifluormethylphenylamino)-methanphosphonsäure
894 mg (2-NO2-4-trifluormethylphenylamino)-methanphosphonsäure werden in 180 ml Ethanol mit 3 g Raney-Nickel versetzt und 3 Stunden bei Raumtemperatur unter Wasserstoffnormaldruck hydriert. Der Ansatz wird vom Katalysator abgesaugt und das Filtrat eingeengt. Es wird ohne weitere Reinigung in die Stufe E eingesetzt.

E) 3-(6-Nitro-2.3-dioxo-1,2,3,4-tetrahvdrochinoxalin-1-yl)-propionsäure 211 mg (0,9 mM) 3-(2-Amino-4-nitrophenylamino)-propionsäure werden mit 200 mg (2 mM) Triethylamin in 20 ml trockenem Tetrahydrofuran bei + 4 °C Badtemperatur unter Argon unter Feuchtigkeitsausschluß vorgelegt. Eine Lösung aus 270 mg (2 mM) Oxalsäureethylesterchlorid und 5 ml trockenem Tetrahydrofuran wird zum Ansatz getropft und 2 Stunden bei Raumtemperatur gerührt. 0,05 ml Triethylamin und 0,05 ml Oxalsäureethylesterchlorid werden nachgegeben und 1 Stunde bei Raumtemperatur gerührt. Der Ansatz wird filtriert und das Filtrat eingeengt. Man verteilt in Wasser und Essigester. Die organische Phase wird eingeengt. Der Rückstand wird in 15 ml
Ethanol und 15 ml 1-N Salzsäure aufgenommen und für 2 Stunden bei 110 °C Badtemperatur am Rückfluß gekocht. Der Ansatz wird eingeengt, in etwas Wasser aufgenommen und abgesaugt. Man erhält 120 mg 3- (6-Nitro-2,3-dioxo-1,2,3,4-tetrahydrochinoxalin-1-yl)-propionsäure vom Schmelzpunkt: 148 -156 °C und Zers.

211 mg (0,6 mM) 3-(6-Nitro-2,3-dioxo-1,2,3,4-tetrahydrochinoxalin-l-ylmethyl)-benzoesäuremethylester werden in 4 ml 4-N Salzsäure vorgelegt, mit 4 ml Trifluoressigsäure versetzt und 3 1/2 Stunden bei 110 °C Badtemperatur gerührt. Der Ansatz wird nach Abkühlen auf Raumtemperatur mit Wasser verdünnt und abgesaugt. Der Filterkuchen wird mit Wasser und Ethanol gewaschen und getrocknet. Man erhält 179 mg 3-(6-Nitro-2,3-dioxo- 1,2,3,4-tetrahydrochinoxalin-1-ylmethyl)-benzoesäure vom Schmelzpunkt: > 330 °C.

Nach Abkühlen wird der Ansatz mit Wasser versetzt und abgesaugt. Der Filterkuchen wird getrocknet. Man erhält: 253 mg 4-(6-Nitro-2,3-dioxo-1,2,3,4-tetrahydrochinoxalin-1-yl-methyl)- phenylphoshonsäure vom Schmelzpunkt: 253 - 265 °C unter Zersetzung.

(6-Nitro-2,3-dioxo-1,2,3,4-tetrahydro-chinoxalin-1-yl)-methanphosphonsäuremonoethyl sowie diethylester
Zu 300 mg (6-Nitro-2,3-dioxo-1,2,3,4-tetrahydro-chinoxalin-1-yl)-methanphosphonsäure in 5 ml absolutem Dimethylformamid werden bei - 15 °C 0,29 ml (476 mg) Thionylchlorid getropft. Nach beendeter Zugabe wird der
Ansatz 20 Minuten bei + 4 °C Badtemperatur gerührt. Anschließend setzt man 0,35 ml (276 mg) Ethanol zu dem Ansatz und rührt 1,5 Stunden bei Raumtemperatur.
Nach Einengen unter Vakuum wird über Kieselgel mit Toluol:Eisessig:Wasser = 10:10:1 chromatographiert. Man erhält zunächst 100 mg (6-Nitro-2,3-dioxo-1,2,3,4-tetrahydro-chinoxalin-1-yl)-methanphosphonsäurediethylester vom Schmelzpunkt 220 - 260 °C und anschließend 36 mg (6-Nitro-2.3-dioxo-1,2,3,4-tetrahydrochinoxalin-1-yl)-methanphosphonsäuremonoethylester vom Schmelzpunkt 197 °C.
In analoger Weise werden hergestellt:
(6-Nitro-2,3-dioxo-1,2,3,4-tetrahydro-chinoxalin-1-yl)-methanphosphonsäure-mono-N,N-dimethylamid sowie bis-N,N-dimethylamid.
Beispiel 8
1-(6-Amino-2,3-dioxo-1,2,3,4-tetrahydro-chinoxalin-1-yl)-methanphosphonsäure
300 mg 1-(6-Nitro-2,3-dioxo-1,2,3,4-tetrahydro-chinoxalin-1-yl)methanphosphonsäure werden in 60 mg Methanol gelöst und unter Stickstoff nacheinander mit 50 mg Pd/C (10%), 300 mg Ammoniumformiat und 18 ml Wasser versetzt und 1 Stunde auf 80 °C erhitzt. Nach dem Abkühlen wird vom Katalysator abfiltriert, das Filtrat eingedampft und der Rückstand gefriergetrocknet. Man erhält 200 mg 1-(6-Amino-2,3,-dioxo-1 ,2,3,4-tetrahydrochinoxalin-1-yl)-methanphosphonsäure in Form eines weiβen Feststoffes.
In analoger Weise wird hergestellt:
1-(6-Amino-2,3-dioxo-1,2,3,4-tetrahydrochinoxalin-1-yl)-ethanphosphonsäure.
Beispiel 9
1[6-(4-Carbethoxy-imidazol-1-yl)-2,3-dioxo-1,2,3,4-tetrachinoxalin1-yl1-methanphosphonsäure
200 mg 1,4-Bis-dimethylamino-3-carbethoxy-2-azabutadien-1,4 werden mit 3
ml Eisessig unter gelinder Kühlung versetzt und 10 Minuten bei Raumtemperatur gerührt. Anschließend werden 180 mg 1-(6-Amino-2,3-dioxo-1,2,3,4- tetrahydro-chinoxalin-1-yl)-methanphosphonsäure gelöst in 3 ml Eisessig zum Ansatz gegeben und über Nacht bei Raumtemperatur gerührt. Es wird dann 4 Stunden auf 100 °C Badtemperatur erhitzt. Nach Einengen erhält man 50 mg 1[6-(4-Carbethoxy-imidazol-1-yl)-2,3-dioxo-1,2,3,4-tetrachinoxalin- 1-yl]-methanphosphonsäure als Öl.
In analoger Weise werden hergestellt:
1-[6-(4-Cyano-5-methyl-imidazol-1-yl)-2,3-dioxo-1,2,3,4-tetrahydro-chinoxalin-1-yl)-methanphosponsäure
Beispiel 10
1-(6-Jod-2,3-dioxo-1,2,3,4-tetrahydrochinoxalin-1-yl)-methanphosphonsäure
180 mg 1-(6-Amino-2,3-dioxo-1,2,3,4-tetrahydrochinoxalin-1-yl)-methanphosphonsäure werden in 10 ml 25Ziger Schwefelsäure eingetropft. Nach 5 Minuten Rühren bildet sich eine Suspension des Salzes, die auf 0 °C gekühlt wird. Dazu wird eine Lösung von 60 mg Natriumnitrit in 2 ml Wasser getropft. Nach 15 Minuten Rühren bei 0 °C ist die Reaktionsmischung fast gelöst. Man tropft dazu eine Lösung von 180 mg Kaliumjodid in 2 ml Wasser. Das Eisbad wird entfernt und 2 Stunden auf 100 °C erwärmt. Die abgekühlte Reaktionsmischung neutralisiert man mit konz. Ammoniaklösung und dampft zur Trockene ein. Der Rückstand wird mit
Ethanol und wenig Wasser ausgekocht, filtriert und das Filtrat eingeengt. Nach Chromatographie über silanisiertes Kieselgel mit Wasser:Methanol = 4:1 erhält man 40 mg 1-(6-Jod-2,3,-dioxo-1,2,3,4-tetrahydrochinoxalin-1-yl-methanphosphonsäure vom Schmelzpunkt 295 - 297 °C.
In analoger bzw. literaturbekannter analoger Weise werden gewonnen:

6-Jod-2,3-dioxo-1,2,3,4-tetrahydrochinoxalin-1-yl-methanphosphonsäure
100 mg 6-Amino-2,3-dioxo-1,2,3,4-tetrahydrochinoxalin-1-yl-methanphosphonsäure werden in konz. Salzsäure gelöst und zur Trockene eingeengt.
Das Hydrochlorid wird gut getrocknet in 10 ml Dimethylformamid vorgelegt und nacheinander mit 4 ml Methylenjodid und 0,6 ml i-Amylnitrit versetzt. Nach 2 Stunden auf 80 °C Badtemperatur hat sich alles gelöst. Es wird am Kugelrohr im Vakuum eingeengt und der Rückstand über silanisiertes
Kieselgel 60 (Reversed Phase) mit Wasser :Methanol = 4:1 als Laufmittel chromatographiert. Man erhält 20 mg 6-Jod-2,3-dioxo-1,2,3,4-tetrahydrochinoxalin-1-yl-methanphosponsäure vom Schmelzpunkt 295 - 297 °C.
In analoger Weise werden hergestellt:
6-Brom-2,3-dioxo-1,2,3,4-tetrahydrochinoxalin-1-yl-methanphosphonsäure

Beispiel 12
100 mg 1-(6-Amino-2,3-dioxo-1,2,3,4-tetrahydrochinoxalin-1-yl)-methanphosphonsäure werden in 20 ml Wasser mit einer gesättigten Natriumcarbonatlösung von auf einen pH von 9,5 eingestellt und mit 0,2 ml Acetanhydrid versetzt. Nach einer Stunde Rühren wird eingeengt, in möglichst wenig Wasser gelöst, auf einen Ionenaustauscher (IR 120. stark sauer) gegeben und mit Wasser eluiert: Die entsprechenden Fraktionen werden zusammengefasst, eingeengt und getrocknet. Man erhält 110 mg 1-(6-Acetylamino-2,3-dioxo-1,2,3,4-tetrahydrochinoxalin-1-yl)-methanphosphonsäure vom Schmelzpunkt 120 °C.

C1-6-Alkyl, C1-4-Alkoxy oder gegebenenfalls mit Cyano, C1-4-Alkyl oder
-COO- C1-6-Alkyl substituiertes Imidazol bedeuten oder
R 5 und R6 oder R7 und R8 einen ankondensierten Benzolring darstellen, wobei
-CO-R 3,oder -PO-XY und R2 ein- bis zweifach gleich oder verschieden steht und n 0 , 1 , 2 , 3 , 4 oder 5 ist und
R3 Hydroxy, C1-6-Alkoxy oder NR9R10,
X und Y gleich oder verschieden sind und Hydroxy, C1-6-Alkoxy,
C1-4-Alkyl oder NR9R10 bedeuten und R 9 und R 10 gleich oder verschieden sind und Wasserstoff, C1-4-Alkyl oder gemeinsam mit dem Stickstoffatom einen gesättigten 5- oder
6-gliedrigen Heterocyclus bilden, der ein weiteres Sauerstoff,
Schwefel oder Stickstoffatom enthalten kann,
R11 C1-6-Alkyl oder Phenyl,
R12 Wasserstoff, C1-4-Alkyl, NH2, N( C1-4-Alkyl), und
R13 Hydroxy, C1-6-Alkoxy, C1-6-Alkyl oder NR9R10 bedeuten
sowie deren Isomeren oder Salze, wobei, falls R4, R5, R6, R7 und R8 Wasserstoff bedeuten, kann R 1 nicht Carbamoylmethyl, 1-Carboxy-1-phenylmethyl oder geradkettiges C1-6-Alkyl sein, das in 1-Stellung mit -COOH oder -COO- C1-6-Alkyl substituiert ist, und falls R1 geradkettiges C1-6-Alkyl, das in 1-Stellung mit -COOH oder
-COO-C1-6-Alkyl substituiert ist, bedeutet, können R 6 und/oder R 7 bzw. R 6 und R 8 nicht Fluor, Chlor oder Brom und R4-R8 jeweils
Wasserstoff sein, und falls R1 -CH2-COOH ist, können
a) R6 und R7 nicht gleichzeitig Methyl oder
b) R6 oder R7 nicht NO und R4-R8 jeweils Wasserstoff sein.

3. Arzneimittel auf Basis der Verbindungen nach Anspruch 1 und 2.
4. Verwendung von Verbindungen nach Anspruch 1 und 2 zur Herstellung von Arzneimitteln.
5. Verfahren zur Herstellung von Verbindungen der Formel I, dadurch gekennzeichnet, daβ man a) eine Verbindung der Formel II
worin R1, R5, R6, R7 und R8 die obige Bedeutung haben, mit reaktiven Oxalsaurederivaten cyclisiert und gegebenenfalls mit R4'-X, worin X Halogen, Tosylat, Mesylat oder Triflat bedeutet und R4' die Bedeutung von R4 mit Ausnahme von Wasserstoff hat, umsetzt oder b) eine Verbindung der Formel III
worin
R4, R5, R6, R7 und R8 die obige Bedeutung haben, mit R1-X umsetzt zu
Verbindungen der Formel I und gewünschtenfalls die Estergruppe verseift oder die Säuregruppe verestert oder amidiert oder die Nitrogruppe reduziert zur Aminogruppe oder die Aminogruppe alkyliert oder acyliert oder die Aminogruppe gegen Halogen oder Cyano austauscht oder die Aminogruppe mit einem 2-Azabutadien der Formel IV
worin U und V Fluchtgruppen darstellen und R11 Wasserstoff, Cyano oder COO C 12
1-6-Alkyl ist und R Wasserstoff oder C1-6-Alkyl ist, umsetzt zu einem Imidazolderivat oder die Isomeren trennt oder die Salze bildet.
6.) Arzneimittel nach Anspruch 5 in Kombination mit L-DOPA und/oder Dopaminagonisten und/oder Benserazid.
