Abstract
Ultramikroemulsionen aus spontan dispergierbaren Konzentraten mit pharmakologisch wirksamen Estern von Apocarotinolen, deren Verwendung als Arzneimittel mit Wirksamkeit gegen Tumore, Psoriasis und Ekzeme, neue Ester mit diesen Apocarotinolen, sowie Verfahren zu ihrer Herstellung werden beschrieben.

EINLEITUNG
Die vorliegende Erfindung betrifft Ultramikroemulsionen aus spontan dis- pergierbaren Konzentraten mit Estern von Apocarotinolen, neue Ester mit Apocarotinolen, Verfahren zu ihrer Herstellung, sowie ihre Verwendung zur Behandlung von Tumoren, Psoriasis und Ekzemen.
Ausgewählte Ester von Apocarotinolen haben überraschenderweise eine sehr gute Wirkung gegen Tumore, Psoriasis und Ekzeme, insbesondere dann, wenn sie in spontan dispergierbare Konzentrate eingearbeitet worden sind, welche mit Wasser verdünnt thermodynamisch stabile Ultramikro¬ emulsionen ergeben mit Mizellen, die einen hydrodynamischen Radius von 1,5 bis 3 nm aufweisen. Messungen wurden durchgeführt mit (dynamischer) quasielastischer Lichtstreuung am Institut für Polymere an der E.T.H., Zürich (Prof.Dr. Pier Luigi LUISI und Prof.Dr. Peter SCHURTENBERGER).

c) Herstellung von Fettsäureestern von 8'-Apo-ß-carotin-8'-ol im Wege des
DIBAH-Verfahrens.
Die Verbindung (XVII), d.h. der C3Q-Alkohol:
(XVII) wurde bislang hergestellt durch Reduktion von 8'-Apo-ß-carotin-8'-al mit LiAIH4 in Ether. [R. Rüegg, M. Montavon, G. Ryser, G. Saucy, G. Schwieter und O. Isler, Helv.Chim.Acta 1959, 42., 854]. Der C30-Alkohol wurde wie folgt charakterisiert: instabile orange Kristalle mit Smp. 148-149 °C; λmaχ
(Petrolether): 426, 453 nm; O-Acetylverbindung: orange Blättchen, Smp.130- 132 °C, λ aχ (Petrolether): 426, 452 nm; leicht erfolgende Eliminations¬ reaktion bei Chromatographieversuchen an AI2O3. Andere, insbes. höhere Ester sind bisher nicht beschrieben worden.
Das neue, ergiebige Herstellverfahren für den C30-Alkohol führt über die Reduktion des 8'-Apo-ß-carotin-8'-säuremethylesters

mittels DIBAH (Diisobutylaluminiumhydrid).
C.3Q-Alkohol.8'-Apo-ß-carotin-8'-ol
In einem trockenen 1 liter Dreihalskolben mit Einleitrohr für Schutzgas, Kühler, Magnetrührer und Septum werden 3.0 g Methyl-8'-Apo-ß-carotin-8'- oat und 300 ml absoluter Diethylether vorgelegt. Hierauf wird unter 2 auf -30°C gekühlt und dann 3,0 ml einer ca. 4,5 molaren Lösung von DIBAH in Hexan mithilfe einer Spritze langsam zugetropft. Es tritt keine sichtbare Reaktion ein. Nun lässt man das Gemisch Ingsam auf RT kommen und kontrolliert die Vollständigkeit der Reduktion mit UV/VIS-Spektren und DC. Wenn noch nicht aller Ester reduziert ist, wird erneut auf -30 °C gekühlt und die benötigte Menge DIBAH zugefügt; etc. Ein Überschuss an DIBAH ver¬ ringert die erreichbare Ausbeute an Cßo-Alkohol beträchtlich. Zur Aufarbeitung wird der Ansatz auf 0 CC gekühlt, dann mit 5 ml Essigester versetzt und schliesslich vorsichtig solange mit kleinen Eisstückchen versetzt, bis kein Aufschäumen mehr eintritt. Hierauf wird das KUhlbad entfernt, das Gemisch mit Celite versetzt und dann abgenutscht. Der Filterkuchen wird mit Et2θ/AcOEt/lsopropanol 10:4:1 farbstofffrei gewa¬ schen und das klare, rote Filtrat i.V. zur Trockene eingedampft. Der bereits kristalline, rote Rückstand lässt sich entweder aus heissem Toluol/Hexan oder Essigester/Hexan Umkristallisieren.
Ausbeute an C3o-Alkohol 2,2 bis 2,4 g (80-90%). Orangerote Nadeln, Smp. 148-149 °C; UV/VIS (Et2θ, qualitativ): λ a : 416, 425, 452 nm; IR (KBr): 2,77 (3603 cm-1, OH frei), keine Banden im Carbonylbereich; CI-MS (NH3): 419 [M + 1,(16)] 401 [M + I-H2O, (100)], 402 [M + 2-H20, (30)], 309 [M + 1-Tθluol, (5%)]
Herstellung und Charakterisierung von Uπdec-10-ensäure -r8'-Apo-ß-carotin- 8'-yl-ester1
Eine Lösung von 400 mg C30-Alkohol in 20 ml abs. Et2θ und 1,5 ml abs. Pyridin wird in einem Dreihalskolben mit Magnetrührer, N2-Einleitrohr, Rückflusskühler und Septum unter 2 auf -30 CC gekühlt und hierauf unter Rühren mit 200 ml Undec-10-enoylchlorid tröpfchenweise versetzt. Nach beendeter Zugabe lässt man die Temperatur langsam auf 20 °C steigen und kontrolliert die Vollständigkeit der Veresterung durch DC auf Kiesel¬ gelplatten mit dem Fliessmittel Benzol/Petrolether 1:1. Der Ester weist einen bedeutend höheren Rf.-Wert als der Alkohol auf. Nun wird das Gemisch i.V. eingedampft, der Rückstand in Benzol aufgenommen und das Gelöste rasch durch eine kurze Säule von CaC03 (Merck Art.2066) filtriert. Das Filtrat wird eingedampft und der Rückstand an 40 g Kieselgel (Merck, 15-40 μm) mit Benzol chromatographiert. Die Substanz aus der roten Zone wird aufgefangen, i.V. eingedampft und der erhaltene Ester aus t-Butylmethyl- ether/Pentan bei -20 °C kristallisiert. Man erhält 450 mg hellrote Kristalle, nach Trocknen bei 40 CC/0,01 Torr; Ausbeute 80%. Smp. 76-77 °C, nach Sintern ab 75 °C. Analytische Details vgl. die technische Beilage 1/2.
In analoger Weise werden die Ester mit Laurinsäure-, Palmitinsäure- und Stearinsäurechlorid hergestellt. Vgl. die technische Beilage 1/2.
d) Herstellung von Azafrinol und von Azafrinylestern Azafrinol
Die Verbindung
(XXI) Azafrinol; (5R,6R)-5,6-Dihydro-10'-apo-8-ß-carotin-5,6,10'-triol) wurde erstmals von W. Eschenmoser und CH. Eugster durch Reduktion von Azafrinmethylester
hergestellt und als labiles Oel besc hrieben ; s. Helv.Ch im.Acta 1975, 58. 1722.
Durch eine modifizierte Reduktion und Aufarbeitung kann Azafrinol als wohlkristallisierte Verbindung isoliert und anschliessend auch eingehend charakterisiert werden. Azafrinol wird so durch Kristallisation aus Benzol/ Hexan in leu chtend hellorangegelben Nadeln erhalten. Die Verbindung ist besonders gegen Säuren sehr empfindlich und verliert dabei unter Ring¬ aufspaltung Wasser. Diese Reaktion tritt be reits bei einer Chromatographie an Kieselgel ein.
Herstellung und Charakterisierung
In einem sorgfältig getrockneten Dreihalskol ben, versehen mit Rück¬ flusskü hler, Gaseinleitrohr, Septum und Mag netrü hrer werden 3 g Azafrin- methylester mit 300 ml absolutem Ether überg össen und hierauf unter Schutzgas und g utem Rühren auf -30 °C gekü hlt. Dan n werden d urch das Septum 3 ,5 m l einer ca. 4,5 molaren Lösu ng von DIBAH (Diisobutyl- alumiumhydrid) in Hexan mittels eine r S pritze tropfenweise zugegeben ; es tritt keine sichtbare Reaktion ein. Während einer langsamen Temperatur¬ erhöhung bis auf 0 °C wird der Verlauf der Reaktion mithilfe von UV/VIS- S pektren und DC überprüft.
Zeigt sich, dass d ie Red uktion noch unvollständig ist, wird erneut auf -30 °C gekühlt und eine entsprechende Menge von DIBAH-Lösu ng zugegeben. Ein geringer Überschuss an DIBAH wirkt sich auf die Ausbeute nicht wesentlich aus, doch muss ein grösserer Überschuss unbedingt vermieden werden. Nach Beendig ung der Reaktion wird die Lösung sehr vorsichtig solange mit kleinen Eisstückchen versetzt, bis kein Aufschäumen mehr eintritt und das Ausfallen von Alumium hydroxid beendet ist. Nach Zugabe von genügend Celite und von 0,2 ml Ethyldiisopropylamin wird abgenutscht und der Fi lterkuchen mit einem Gem isch von Ether/Isopropylal kohol 5: 1 farbstoffrei gewaschen. Das gelborange gefärbte Filtrat wird hierauf i.V. eingedampft und der kristalli ne Rückstand in Tol uol/Isopropylal kohol 99 : 1 heiss gelöst und im Falle einer Trübung die Lösung rasch durch eine kleine, breite Säule
von CaCθ3 filtriert. Es folgt ein erneutes Eindampfen des Filtrates und die Umkristallisation des Rückstandes aus heissem Bezol/Hexan oder Essig¬ ester/Hexan. Nach Trocknen bei 40 °C/0,01 Torr erhält man 2,2 g leuchtend hellorange Nadeln. Smp. 133-134 °C (Vakuumröhrchen). Für die analytischen Einzelheiten vgl. die technische Beilage 2/2.
Herstellung von Undec-10-ensäure[azafrinyl]ester
In einer geeigneten, gut getrockneten Schliffapparatur mit Magnetrührer, Schutzgaseinleitrohr, Kühler und Septum werden 180 mg frisch um¬ kristallisiertes Azafrinol unter N2 in 15 ml Et2θ und 0,5 ml abs. Pyridin gelöst und hierauf auf - 30 °C gekühlt. Dann werden mit einer Spritze 100 ml 10-Undenoylchlorid tröpfchenweise zugegeben. Nach einer h Rühren bei einer Temperatur von -30 °C lässt man auf RT kommen. Anschliessend wird das Gemisch i.V. eingedampft, der Rückstand in Hexan/Ether 3:1 auf¬ genommen und das Lösliche durch eine kurze und breite Säule aus CaCOß (Merck, Art. 2066) filtriert. Das zitronengelbe Eluat wird erneut eingedampft und mit Et2θ/Hexan/lsopropanol 50:49:1 + einer Spur Ethyldiisopropylamin an einer Säule aus Sephadex LH-20 (im selben Lösungsmittel gequollen) chromatographiert. Aus der orangen Hauptzone wurden 161 mg vakuum¬ trockenes, zähes Oel erhalten, das allmählich durchkristallisierte. Es konnte kein signifikanter Smp. erhalten werden. UV/VIS (Et2θ, quäl.): scharfes Dreibandenspektrum mit λmax 375, 394,5, 419 nm; (Benzol quäl.): 377, 402, 428 nm. Für die übrigen analytischen Daten vgl. die technische Beilage 2/2.
In analoger Weise wurden die Ester
n = 9 ; n = 13 ; n = 15 , bzw. Azafrinyl-Laurat, Azafrinyl-Palmitat und Azafrinyl-Oleat hergestellt. Für die analytischen Daten vgl. die techn. Beilage 2/2.
Die Apocarotinester der Formeln (I) bis (IX) haben überraschenderweise eine ausgezeichnete antitumorale Wirkung, insbesondere dann, wenn sie in spontan dispergierbare Konzentrate eingearbeitet worden sind, welche mit Wasser verdünnt thermodynamisch stabile Ultramikroemulsionen mit einem
hydrodynamischen Radius von 1,5 bis 3 nm ergeben. (Messungen mit dynamischer, quasi-elastischer Lichtstreuung QLS). Gegenstand der vor¬ liegenden Erfindung sind deshalb wesentlich auch spontan dispergierbare Konzentrate mit den Apocarotinestern der Formeln (I) bis (IX).
Die Apocarotinester der Formeln (I) bis (IX) sind nahezu wasserunlösliche und polymer agglomerierte Verbindungen. Damit die Moleküle dieser Ver¬ bindungen aber durch die Membranbarriere der Tumorzellen diffundieren und im Innern der Zelle wirksam werden können, müssen derartige Ver¬ bindungen vorerst in geeigneter Weise im wässrigen Medium solubilisiert werden. Im Wege der Bildung von thermodynamisch stabilen Oel-in-Wasser Ultramikroemulsionen mithilfe von besonders ausgewählten Cotensiden oder Lösungsvermittlern einerseits und geeigneten Tensiden anderseits gelingt es, einen optimalen Solubilisierungsgrad der Apocarotin-Ester zu erzielen.
Alle experimentellen Beobachtungen an dergestalt ausgebildeten Ultra¬ mikroemulsionen lassen sich einheitlich durch die Annahme deuten, dass die ausgewählten Tenside und Cotenside als ausgewogenes System genommen in der wässrigen Phase organisierte Aggregate, sog. Mizellen bilden. Sie besitzen mehr oder weniger kugelförmige Gestalt, mit einem hydrodynamischen Radius von 1,5 bis 3 nm. Die Tenside und Hydrotrope (Cotenside) lassen zwischen der äusseren, wässrigen Phase und der inneren, öligen Phase der Mikroemulsion [enthaltend die pharmazeutischen Wirksubstanzen gelöst im biotensiden Lösungsvermittler] eine Grenzschicht entstehen, wodurch die Mischung dieser beiden Phasen unterbleibt. In der öligen, inneren Phase liegen die Wirksubstanz-Moleküle in monomerer oder in oligomer agglomerierter Form vor.
Die Mizellen der erfindungsgemässen Ultramikroemulsionen mit der Wirksubstanz-haltigen inneren Phase sind an ihrer Grenzschicht mit einem Tensidmantel versehen, was sie instand setzt, leicht durch die Zellmembran ins Innere der Zelle zu diffundieren. Die Diffusion durch die Plasmamembran der Zellen erfolgt ausschliesslich aufgrund thermischer
Molekularbewegungen.
Die Richtung, die ein konkreter Diffusionsvorgang einschlägt, wird vom Konzentrationsunterschied bestimmt, welcher an der Plasmamembran zwi¬ schen ausserhalb und innerhalb der einzelen Zelle besteht. Die Diffusion
verläuft solange entlang dem Konze ntrationsgefäl le , bis es abgebaut ist. Zwischen der extrazellulären Zone und dem Innere n der einzelnen Zelle wird die Konzentration an Wirksubstanz, bzw. am Wirkstoffsystem ("multiple drug system") ausgeglichen, wobei auch verzögerte Abgabeeffekte ("slow release effects") auftreten können. Derartige Diffusionsvorgänge verlaufen unab¬ hängig von jeglicher Energiezufuhr. S ie haben keinen Bezug auf die zelluläre Stoffwechselenergie.

Die erfindungsgemäss einzusetzenden Tenside oder Tensid gemische können anionaktiv, kationaktiv, amphoter oder nicht-ionogen sei n. Bevorzugt sind sie am photer oder nicht-ionogen u nd haben ein HLB-Verhältnis (d.h. eine "hydrophilic-lipophil ic-balance") zwischen 2 und 1 8 ; vorzugsweise liegt er zwischen 2 und 6 einerseits und 1 0 und 15 anderseits . HLB-Werte geben Ausku nft über die hydrophi len und lipophiler Eigenschaften eines Emul- gators. Geeignete an ionische Tenside kön nen sowoh l sog. wasserlösliche Seifen als auch wasserlösliche synthetische Verbindungen sein .
Als Seifen eig nen sich die Al kali-, Erdalkali- oder gegebenenfal ls sub¬ stituierten Ammoniumsalze von höheren Fettsäu ren (C-i o bis C22) . wie z.B. die natürlichen Na- oder K-Salze der Oel- oder Stearinsäure, oder von natürlichen Fettsäuregemischen, welche sich u.a. aus Kokosnuss- oder Talgöl gewinnen lassen. Ferner sind als Tenside auch die Fettsäure-Methyl- taurinsalze, sowie modifizierte und nicht-modifizierte Phospholipide zu erwähnen.
Häufiger werden jedoch sogenannte synthetische Tenside verwendet, ins¬ besondere Fettsu lfonate, Fettsu lfate, su lfonierte Benzimidazol-Derivate oder Alkylarylsulfonate.
Die Fettsulfonate und -sulfate liegen in der Regel als Al kali-, Erdalkali- oder gegebenenfalls su bstituierte Ammoniumsalze vor u nd weisen im al lgemeinen einen Alkylrest mit 8 bis 22 C-Atomen auf, wobei Alkyl auch den Alkylteil von Acylresten einschliesst. Beispiele hiefür sind das Na- oder Ca-Salz der Lig ninsulfosäu re,. des Dodecylschwefelsäureesters und Sulfonsäuren von
Fettalkohol-Äthylenoxyd-Add u kten . Die su lfon ierten Benzimidazol-Derivate enthalten vorzugsweise zwei S u lfonsäuregru ppen und einen Fettsäurerest m it etwa 8 bis 22 C-Atomen. Alkylaryl-S ulfonate sind z.B. die Na-, Ca- oder Triäthanolaminsalze der Dodecyl-benzolsulfonsäure, der Dibutylnaphtha- linsulfonsäure oder eines Naphtha-Iinsulfonsäure-Formaldehyd-
Kondensationsprodu ktes.
Als nichtionische Tenside stehen in erster Linie zur Wahl die Polygly- kolätherderivate von aliphatischen oder cycloaliphatischen Alkoholen, gesät-tigten oder ungesättigten Fettsäuren und Alkylphenolen, welche 3 bis 30 Glykolätherg ruppen und 8 bis 20 C-Atome im (aliphatischen) Kohlen¬ wasserstoffrest und 6 bis 1 8 C-Atome im Alkylrest enthalten können. Weiterhin kommen als nichtionisc he Tenside in Frage die wasserlöslichen, 20 bis 250 Äthylenglykoläthergruppen und 1 0 bis 1 00 Propylenäthergru ppen enthaltenden Polyäthylenoxydadd ukte an Polypropylenglykol und Alkyl- polypropylenglykol mit 1 bis 10 C-Atomen in der Alkylkette. Die genannten Verbindungen enthalten üblicherweise pro Propyleng lykoleinheit 1 bis 5 Äthyleng lykoleinheiten.
Als Beispiele nicht-ionischer Tenside seien erwähnt:
Nonyl phenolpolyäthoxyäthanole, Ricinusöl polyglykoläther, Polypropylen- polyäthylenoxyd-Addukte, Tributylphenoxypolyäthoxyäthanol, Polyäthylen- g lykol u nd Octyl phenoxypolyäthoxyäthanol. Ueberdies kommen auch Fett¬ säu reester von Polyoxyäthylensorbitan, wie das Polyoxyäthylensorbitan- Trioleat, in Betracht.
Bei den kationischen Tensiden handelt es sich vor allem um quaternäre Ammoni umsalze, welche als N-Substituenten mindestens einen Alkylrest mit 8 bis 22 C-Atomen enthalten u nd als weitere Substituenten niedrige, gege¬ benenfalls halogenierte Alkyl-, Benzyl- oder niedrige Hydroxy-alkylreste aufweisen. Die Salze liegen vorab als Halogenide, Methylsulfate oder Äthylsulfate vor, z.B. das Stearyltrinethylammoniumchlorid oder das Benzyl- di(2-chloräthyl)äthylammoniumbromid.
In hohem Masse bevorzugt zur Herstellung von erfind ungsgemässen, spontan disperg ierbaren Konzentraten sind einerseits Phosphorsäure- estertenside, wie z.B. Tristyrylphenolpolyoxyäthylen-18-mono/d imethylphosphorsäureester

Alle technischen Tenside wurden vor dem Eintrag in die spontan disper- gierbaren Konzentrate mittels Filtration, bzw. Chromatographie über neutralem Al um iniumoxyd mit einem inerten Lösungsmittel wie z.B. Tetra- hydrofuran, Äthylal kohol oder Methylench lorid gereinigt.
Als Zusätze in d ie erfindungsgemässen spontan dispergierbaren Konzentrate eignen sich Vitamine und Provitamine (wie z.B. Vitamin A, Retinol, Tocopherole), sowie auch ausgewählte Penetrationsverbesserer ("Flux enhan ers") und Radi kalfänger.
Die zur pharmazeutischen Anwendung erforderliche Tagesdosis beträgt 0,001 bis 25 mg/kg Körpergewicht, wenn möglich verteilt auf 2 bis 3 Einzeldosen. Hiefür lassen sich die neuen Ester mit Apocarotinolen oder die spontan dis-pergierbaren Konzentrate mit diesen Verbind ungen in die gängigen pharmazeutischen Zubereitungen und Darreichungsformen wie Dragees, Tabletten, Kapseln , Pulver, Granulate, Pellets , Lösungen, Ampullen, Emu lsionen, Cremes oder Zäpfchen zusammen mit den üblichen Trägerstoffen und/oder Verdünnungs- und Stabilisierungsmitteln ein¬ arbeiten.
Die Gegenstand der Erfind ung bildende n Wirkstoffe oder Wirkstoffmi¬ schungen , sowie die spontan dispergierbaren Konzentrate, welche diese Wirkstoffe oder Wi rkstoffm ischungen enthalten, können dem Menschen oral, durch Injektion (intravenös, su bkutan oder intramuskulär) oder in anderer Weise verabreicht werden. Wenn sie als feste Darreichungsformen für die orale Verwendu ng aufbereitet werden, kan n dies in Form von Tabletten, G ranulaten, Pellets, Pulvern oder Kapseln, usw. geschehen. Die Auf¬ bereitungen können Zusatzstoffe enthalten, z.B. einen Arzneimittelträger wie eine Saccharid- oder Cellulose-Grundlage, ein Bindemittel wie Stärkepaste oder Methylcellulose, ein Füllmittel oder ein Desintegriermittel, usw. - wobei Zusatzstoffe eingesetzt werden , wie sie üblicherweise bei der Herstellung medizinischer oder paramedizinischer Formulierungen verwendet werden. Wenn die erfind ungsgetreuen Wirkstoffe oder Wirkstoffmischungen als fl üssige Darreic h u ngsformen zu oraler Ver-abreic hung gelangen, so können sie irgend ei ne aus wässrigen Zu bereitungen für i nnere Verwendung, aus S uspensionen, Emulsionen und S irups usw. ausgewählte Form haben, und sie können ausserdem in der Form Vacu um-getrockneter Präparate vorliegen, welc he vor ih rer Ver-wendung in Lösung oder Em ulsion gebracht werden.
Wenn die erfind u ngsgemässen Wirkstoffe oder Wirkstoffmischungen in der Form wässriger Lösu ngen , S uspensionen oder öliger, bzw. wässriger Emul¬ sionen, vorzugsweise als Mikroemulsionen aus den erfindungskonformen, spontan disperg ierbaren Konzentraten aufbereitet sind , können sie auch injiziert werden . Die Injektionslösungen werden jedoch üblicherweise kurz vor der Anwend ung hergestellt, indem man d ie Extrakte oder Konzentrate in wässrigen, flüssigen Medien wie sterilem Wasser, Glucoselösung oder physiolog ischer Kochsalzlösung auflöst oder suspendiert. Falls erforderl ic h, kön nen zu einem Injektionspräparat üblicherweise ver¬ wendete Lösungsmittel, Stabi lisierungsm ittel, Konservierungsmittel und Zu¬ sätze für die Herstellu ng isotonischer Lösungen hinzugegeben werden. Die auf diese Weise erhaltenen Injektions-Präparate werden intravenös, intramus-ku lär, subkutan oder in einer anderen geeigneten Weise ver¬ abreicht.
Die vorliegen de Erfi nd ung betrifft ebenfalls pharmazeutische Präparate, welche die Wirkstoffe, bzw. Wi rkstoffgemische, und/oder die beschriebenen , spontan disperg ierbaren Konzentrate zur Bekämpfung des Wachstums von Tumorzellen enthalte n. Bei den der Erfindung entsprechenden pharma-
zeutischen Präparaten handelt es sic h um solche zur enteralen (wie oralen oder rektalen), sowie zur parenteralen oder topischen Verabreichung an Warmblüter, welche das spontan disperg ierbare Konzentrat allein oder zusammen mit einem pharmazeutisch anwend bare n Trägermaterial enthalten.
Die Dosierung der erfindungsgemässen Konzentrate hangt von der Warm¬ blüterspezies, dem Alter und dem ind ividuellen Zustand, sowie von der Verabreichungsart ab. So werden z.B. zur Erzielung eines Abtötungseffektes von Tumorzellen an Warmblütern mit geringem Körpergewicht, wie z.B. Mäusen, Ratten und Hamstern, bei sukutaner Verabreichung Dosen im Bereich von ca. 0,1 bis 50 mg/kg Körpergewicht, und bei intraperitonealer Verabreichu ng Dosen im Bereich von 0,05 bis 5 mg/kg Körpergewicht angewandt.
Die oralen u nd rektalen Formen der neuen pharmazeutischen Präparate enthalten zwischen 1 - 95 %, vorzugsweise zwischen 1 0 - 95 %, insbesondere zwisc hen 20 - 95 % des erfind ungsgemässen , spontan dispergierbaren Konzentrates. Sie können z.B. in Dosis-Einheitsform vorl iegen, also als Dragees, Micropellets, Tabletten, S uppositorien oder Am pullen und vor allem als Kapsel n.
Geeignete pharmazeutisch anwe nd bare Trägerstoffe für d ie oralen Formen sind hauptsäch lich Füllstoffe, wie Zucker (z.B. Lactose , Saccharose, Mannit oder Sorbit), Cellulosepräparate und/ode r Calciumphosphate (z.B. Tri- calcium- oder Calciumhydrogenphosphat), ferner Bindemittel wie Stärke¬ kleister u nter Verwend ung von u.a. Mais-, Weizen-, Reis- oder Kartoffel¬ stärke , Gelatine, Traganth, Methylcellulose, Hydroxylmethylcellulose, Hydroxypropyl-Methylcellulose, Natriumcarboxymethylcellulose und/oder Polyvinylpyrrolidon u nd/ oder Sprengmittel (wenn erwünscht), wie die obgenannten Stärken, ferner Carboxymethylstärke, q uervernetztes Poly¬ vinylpyrrolidon, Agar, Alginsäure oder ein Salz davon, wie z.B. Natrium- alginat.
Als Fliessreguliermittel sind z.B. die Polyäthylenglykole Nr. 200 bis 600 und höher geeignet.
Die beim Menschen als Darreich ungsform bevorzugten Gelatinekapseln werden mit geeig neten Ueberzügen versehen , wobei man u.a. konzentrierte Zuckerlösungen - welche gegebenenfalls arabischen G ummi , Talk, P c .yvinyl- pyrrol idon , Polyäthylenglykol und/oder Titandioxid enthalten - Lack¬ lösungen (wässrige oder solche, die unter Verwend ung organischer Lösungsmittel aufbereitet worden si nd ) , oder magensaftresistente Ueber-
züge aus Lösungen von geeigneten Cellulosepräparaten, wie mikro¬ kristalliner Cellulose (Avicel®), Acetylcellulosephthalat, Hydroxylmethyl- cellulosephthalat (Metolose®), Hydroxylpropylmethylcellulose-Acetat- succinat (AQOAT®) oder einem Copolymerisat wie Eudragit® L30D ver¬ wendet.
Als erfindungsgemäss besonders geeignete, oral anwendbare pharmazeu¬ tische Darreichungsform eignen sich Steckkapseln aus Gelatine und einem Weichmacher, wie Glyzerin oder Sorbitol. Die Weich- bzw. Hartgelatine- Kapseln, sowie solche aus AQOAT® (Hydroxypropyl-Methylcellulose-Acetat- Succinat) können das der Erfindung entsprechende, spontan dispergierbare Konzentrat im Gemisch mit Füllstoffen, wie Laktose, Bindemitteln wie Stärke und/oder Gleitmitteln wie Talk oder Magnesium-Stearat und gegebenenfalls mit Stabilisatoren und Antioxydantien wie z.B. α-, ß- oder γ-Tocopherol enthalten. Der Einsatz von geeigneten Flüssigkeiten wie flüssigen Poly- äthylenglykolen No. 200 bis 600 als Verdünnungsmittel kann sich empfehlen, wobei sich ebenfalls Stabilisatoren und Antioxydantien zufügen lassen.
Zur parenteralen Verabreichung werden die erfindungsgemässen Konzentrate mit destilliertem Wasser versetzt. Der entstehenden wässrigen Injektions-Mikroemulsion können Viskositätserhöhende Stoffe, wie z.B. Na- Carboxymethylcellulose, Sorbit, Mannit und/oder Dextran, und gegebenen¬ falls auch Stabilisatoren und Antioxydantien zugefügt werden. Die pharmazeutischen Präparate für die parenterale Anwendung enthalten vorzugsweise zwischen 0,1 bis 60 %, und hauptsächlich zwischen 1 bis 40 % des erfindungskonformen, spontan dispergierbaren Konzentrates. Als topisch anwendbare Präparate, welche sich vornehmlich zur Prophylaxe und Therapie von Hautkrebsarten eignen, kommen z.B. Cremen, Salben, Pasten, Pomaden, Schäume, Tinkturen und Lösungen in Betracht, welche zwischen 0,01 bis 70 % des erfindungsgemässen Konzentrates enthalten. Für Cremen und Oel-in-Wasser-Emulsionen, welche mehr als 50 % Wasser aufweisen, verwendet man alt oelige Grundlage in erster Linie Fettalkohole, z.B. Lauryl-, Cetyl- oder Stearylalkohol, flüssige bis feste Wachse, z.B. Isopropylmyristat, Woll- oder Bienenwachs und/oder Kohlenwasserstoffe wie z.B. Vaseline (Petrolatum) oder Paraffinoel. Zur Emulgierung dieser öligen Grundlagen eignen sich in erster Linie oberflächenaktive, pharma- verträgliche Substanzen mit vorwiegend hydrophilen Eigenschaften, wie z.B. nicht-ionogene Emulgatoren, vorab Fettsäureester von Polyalkoholen oder Äthylenoxydaddukten (etwa Polyglycerinfettsäureester oder Polyäthylen-
sorbitan-Fettsäureester) mit einem H LB-Wert von u nter 8. Zusätze zur Wasserphase sind u.a. Mittel, welche die Austrockn ung der Cremen ver¬ mindern, z.B. Polyalkohole wie G lyzerin , Sorbit, Propylenglykol und/oder Polyäthylenglykole No. 200 bis 600, ferner Konservierungsmittel, Riechstoffe, etc.
Salben sind Wasser-in-Oel Em ulsionen , die bis zu 70 %, vorzugsweise jedoch zwischen 20 und 50 % Wasser oder wässrige Phasen enthalten. Als Fettphase kommen in erster Linie Koh lenwasserstoffe, z.B. Vaselin, Paraffinoel und/oder Hartparaffine in Frage, welche zur Verbesserung des Wasserbindungsvermögens geeignete Hydroxydverbindungen, wie z.B. Fett¬ alkohole oder Ester, davon etwa Cetylal kohol oder Wollwachsalkohole, ent¬ halten .
Fal lweise werden noch Emu lgatoren m it einem HLB-Wert von 8 bis 16, wie z.B. Sorbitan-Fettsäureester (etwa Sorbitanisostearol) zugesetzt. Zusätze zur Wasserphase sind u.a. Feuchthaltungsmittel, wie Polyalkohole (G lycerin, Propylenglykol, Sorbit und/oder Polyäthylenglykole No. 200, 400, 600); ferner Konservierungsmittel , Riechstoffe, etc.
Fettsalben sind wasserfrei und enthalten als G rund lage vornehmlich Kohlenwasserstoffe, z.B. Paraffin, Vaseli n und/oder flüssige Paraffine ; über¬ dies natürliche oder partial synthetische Fette wie z.B. Kokosfettsäure- triglycerid, ferner: Fettsäurepartialester des Glycerins, wie z.B. die im Zusammen hang mit den Sal ben erwähnten, die Wasser-Aufnahmefähigkeit steigernden Fettal kohole, Emulgatoren und/oder Zusätze.
Pasten sind Cremen und Salben mit sekretabsorbierenden Puderbe¬ standteilen , wie beispielsweise Metalloxide (etwa Titanoxid oder Zinkoxid), ferner Talk und/oder Aluminiumsilikate, welche die Aufgabe haben , vorhan¬ dene Feuchtigkeit oder Sekrete zu bi nden.
Schäume werden aus Druckbehältern verabreicht und sind in Aerosolform vorliegende Oel-in-Wasser Emulsionen der erfindungsgemässen, spontan dispergierbaren Konzentrate, wobei halogenierte Kohlenwasserstoffe (wie z.B. Chlorfluorniederaikane : etwa Dichlordifluormethan und Dichlortetra- fluoräthan) als Treibmittel verwendet werden. Dazu kommen gegebenenfalls die üblichen Zusätze wie Konservieru ngsm ittel, usw.
VERFAHRENSBEISPIELE zur Herstellung von Estern mit Apocarotinolen der Formeln (I) bis (IX) :
a) Herstel lung von ß-Apo-8'-carotinol-10-undecenoat
Zu 85 mg ß-Apo-8'-carotinol (Synthese vgl. R. Rüegg et al., Helv.Chim Acta 42,847-864 )1959), 100 mg Pyridin und 100 mg Dimethylformamid in 20 ml Chloroform werden bei 0 °C und unter Schutzgaszuleitung 100 mg 10- Undecenoylchlorid in 10 ml Chloroform zugetropft. Nach einstündigem Rühren bei 0 CC und zweistündigem Rühren bei 20 bis 40 °C wird mit Wasser verd ünnt und ausgeschüttelt. Der Rückstand wird schonend getrocknet und anschliessend auf einer S ilicagelsäule m it Hexan/Essigester (90 : 1 0) als Eluiermittel chromatog raph iert.
Man erhält das ß-Apo-8'-Carotinol-1 0-undecenoat mit einem S c hmelzpunkt von 76-77 °C, nach S intern ab 75 "C; UV λmax. 425 nm (in Methylenchlorid); Rf-Wert im DC mit Hexan/Essigester (80 : 20) 0,92.

b) Herstell ung von ß-Apo-8'-carotinol-10-undecenoat
Ausgangsmaterial : 8'-Apo-ß-caroti n-8'-säure-äthyl u n d/oder -methylester. In einer geeigneten Apparatur mit Mag netrührer, N2-Einleitrohr, Rück¬ flusskühler u nd Septum werden 2 g 8'-Apo-ß-carotin-8'-säu remethylester zu 25 ml Et2θ gegeben, das Gemisch unter N2 und Rühren auf -35 °C gekühlt und m it der berechneten Menge DIBAH (Diisobutylalumin ium hydrid) in He¬ xan tropfenweise versetzt. Nach beendeter Zugabe wird das Kühlbad
entfernt. Man lässt auf RT kommen. Nac h vollständ iger Red uktion wird sehr vorsichtig wen ig Eis zugegeben und damit solange fortgefahren, bis eine Trübung eintritt. Jetzt kühlen. AI(OH)3 fällt langsam aus. Nach Beendigung der Fällung wird über genügend Celite filtriert, mit MeOH nachgewaschen und das Filtrat eingedampft. Wenn der Rückstand zu kristallisieren beginnt, kann ohne weitere Rei nig ung aus heissem Benzol und unter Zugabe von Hexan umkristallisiert werden. Ausbeute 1 ,2 g braunrote Kristalle; UV/VIS λmax. 425, 452 nm (in Et2θ)
Veresterung von 8'-Apo-ß-carotin-8'-ol mit Undecenoylchlorid.
400 mg C30-Al kohol, sowie 20 ml Et2θ und 1 ml Pyrid in werden unter Rühren u nd unter 2 bei -30 °C tropfenweise mit 0,22 ml Undecenoylchlorid ver¬ setzt.
Man lässt langsam auf RT kommen, verdü nnt mit Petroläther und sch üttelt nacheinander m it Wasser und Sole aus. Dann Trocknen über MgS04, Eindampfen un d Chromatographie an 40 g Kieselgel (Merck, 15 bis 40 μm Korngrösse) mit Benzol/Hexan 2: 1 . Nach dem Verwerfen einer gelben Vorzone wird die rote Hauptzone aufgefangen , eingedampft und bei 0,01 Torr bei 40 °C getrocknet. Dabei kristallisiert das Produkt vollständig durch. Smp. 76-77 °C nach Sintern ab 75 "C UV/VIS λmax. 425,5, 452,5 nm (in Et20). CI-MS (mit seh r schonender chem ischer Ion isation ) mit peaks bei 575 und
401 nm; m/z. Mw. (M+ +1 ); IR-Spektrum Ester-peak bei 1724 cm-1. 1 H-NMR Spektrum entspricht.
c) Herstellu ng von Azafrinyl-10-U ndecenoat
Ausgangsmaterial Azafrin , isoliert aus Rhizomen von Escobedia scrabifolia. Herstellung des Methylesters aus der C27-Säure mit Diazomethan/Ether, vgl. W. Eschenmoser und CH. Eugster, Helv.Chim.Acta, 58., 1722 (1975), Reduktion mit DIBAH analog dem Verfahren b); kein Ausschütteln, sondern direkte Filtration unter Zugabe von 0,5 % Hünig base. Das Filtrat eindampfen, den Rückstand aufnehmen in Et2θH u nd vorhandenes Wasser abscheiden, trocknen über MgSθ4, filtieren, eindampfen, den Rückstand mit Toluol/Hexan an einer CaCθ3-Säule 4,3 x 20 cm Chromatographieren. Die Hauptzone wird mit Toluol/Isopropylalkohol (99 : 1 ) eluiert, das Filtrat i.V. zur Trockene gebracht. Nach dem Um kristallisieren aus Benzol/Hexan und AcOEt/Hexan erhält man leuchtend gelbe, sehr säureem pfind liche Nädelchen mit einem Smp. von 135 °C.
Bildung von Azafrin-10'-ol[undec-10-enoyl]ester.
Apparatur wie beim Verfahren b). Ansatz von 450 mg der Verbindung mit

1,5 ml Pyridin, 2 ml DMF, 20 ml Et2θ; N2, Rührer, Septum, Temperatur -40 bis -50 °C, Zugabe von 0,20 ml Undecenoylchlorid. Auf 0 °C kommen lassen, dann nochmals 0,20 ml Undecenoylchlorid zugeben und 3'Λ h bei RT rühren. Verdünnen mit Et2θ und H2O, Phasentrennung im Scheidetrichter, nochmals H2O zufügen, dann Sole, Trocknen über MgSθ4; eindampfen. Den Rückstand in wenig CH2Cl2/Hexan (10:15) + 0,25 % Hünigbase (N,N-Diisopropyl- äthylamin) lösen und auf einer Sephadex LH-20-Säule 3,6 x 34,5 cm chro- matographieren. Die breite, gelborange Zone auffangen und eindampfen. Zweite Chromatographie-Passage auf einer Kieselgelsäule 3,3 x 17,5 cm mit Toluol/Isopropylalkohol (99:1 bis 98:2), was eine gute Trennung ergibt. Die hellorange Fraktion auffangen, eindampfen, trocknen 0,002 Torr bei 42 °C. Man erhält das Produkt: 376 mg als ein viskoses, hellorangerotes Öl.

Biologische Prüfungen.
Die antitumorale Wirkung von spontan dispergierbaren Konzentraten mit Wirkstoffen gemäss den Herstellungsbeispielen a) bis d), sowie den Auf¬ arbeitungsbeispielen a) bis d) wird anhand folgender Prüfungsergebnisse bestätigt:
1. In vitro-Tests mit geeigneten Tumorzell-Linien
Es wurde ein biologisches Assay-System entwickelt, das mit Mikro- titerplatten und Verdünnungsreihen arbeitet. Angesetzt werden je 1θ4/ml Tumorzellen in Kulturmedium RPMI 1640 mit 10 % fötalem Kalbserum inak¬ tiviert (GIBCO); sie werden so undicht ausgesät, dass sie während des Assays wachsen können, in sog. nichtkonfluenten Monolayers. Die Probenzugabe erfolgt nach 6 bis 24 Stunden, mit 100 μl pro Reihe, die man im 1. Loch mit 100 μl Medium versetzt. Davon wird die Hälfte entnommen und in das folgende Loch eingebracht, wieder mit 100 μl Medium versetzt, usf. Es entsteht eine geometrische Verdünnungsreihe n1/s.
Die Proben werden im Plaque Assay während 3 bis 5 Tagen bei 37° C mit 3Λ % CO2 inkubiert. Anschliessend färben/fixieren mit 0,1 % Kristallviolett (Fluka, Buchs) in einer Lösung von 70 % Methanol, 1 % Formaldehyd, 29 % Wasser. Die Auswertung wird am Mikroskop vorgenommen, Vergrösserung 300-fach. Man bestimmt die grösste zytotoxische Verdünnung. Die quanti¬ tative Auswertung lässt sich auch mittels Scanning und Absorptions¬ messung am Spektrophotomerer vornehmen.

LANGZEITVERSUCH
Um herauszufinden, ob bei längerer Exposition signifikante Veränderungen an der Tumorzelle - sei es in Richtung Regression oder Redifferenzierung - festzustellen seien, wurde mit Py6-Zellen auch ein Langzeitversuch an¬ gestellt, der über 41 Tage geführt wurde. (Experiment vom 24.05.1994 bis zum 29.06.1994). Eingesetzt wurden als Kontrollen nicht transformierte 3T3 Mausfibroblasten, sowie die Polyomavirus-transformierten 3T3 MT 18 Zellen. Als Leitpräparate wurden geprüft: ein 2%-PHYSALIEN-Konzentrat (Zea- xanthin-di-Palmitat) und ein 2 % C11:1-8'-APOCAROTINYLESTER-Konzen- trat.
Einzelheiten zum Prüfprotokoll: Nach der Aussaat ist das Zellwachstum anfäng lich langsam. Nach 12 Tagen treten, Dosis-abhängig , viele nekro- tische Zel len auf. Auswaschen de r Ku lturen mit EDTA. Neuzusatz von Medi um und wirkstoffhaltiger Mikroemulsion in je 3 verschiedenen Konzen¬ trationen (wie zu Beginn des Versuches) nach 1 3 und 20 Tagen. Wiedereinsetzen des Zellwachstums bei allen überlebenden Gruppen. Nach 34 Tagen erneutes Auswaschen , sowie Zugabe von frischem Medium und Konzentrat. Abbruch des Versuchs nach 41 Tagen.
Ergebnis: Die morphologischen Unterschiede zwischen den transformierten und den nicht-transformierten Zellen sind im in-vitro Test selbst bei Versuchsende an den überlebenden Populationen immer noch feststellbar. Die transformierten Zellen besitzen längere Filopodien und behalten diese Charakteristik bei.

G leiche Methodik wie 1 ). Der charakteristische Peak für die Apocarotin-Ester erscheint nach ca. 8 Minuten ; die Verzögerung gegenüber der reinen W.S. -Messung ist eine Folge der Ummantelung der inneren Phase der Konzentrate mit Tensiden. Sie gibt einen wichtigen Hinweis auf das Diffusionsverhalten der Ultramikro¬ emulsion.
3) Nachweis de r Membran-Penetration an der Tumorzelle
Am Lichtmikroskop (wie auc h am Elektronenmi kroskop) lässt sich aufzeigen, dass wenige Stunden nach der Inkubation (Beispiel 3T3 Py6-Zellen der Maus; d ü nn ausgesät, mittlere Verdün n u ng der Wirkstoff-Konzentrate) sich ein Kranz von Vakuolen um den Zellkern herum ausbildet.

Der Wirksubstanz-Peak erscheint nach ca.4 Minuten.
Auflösung bis 0,5 ppm.
D) Chromatographische Identifikation der Konzentrat-Mizellen in der wässrigen Mikroemulsion/bzw. im Zellplasma der Tumorzellen.
Gleiche Methodik wie C). Der charakteristische Peak für die Apocarotin-Ester erscheint nach ca. 8 Minuten; die Verzögerung gegenüber der reinen W.S. -Messung ist eine Folge der Ummantelung der inneren Phase der Konzentrate mit Tensiden. Sie gibt einen wichtigen Hinweis auf das Diffusionsverhalten der Ultra¬ mikroemulsion.
E) Nachweis der Membran-Penetration an der Tumorzelle
Am Lichtmikroskop (wie auch am Elektronenmikroskop) lässt sich aufzeigen, dass wenige Stunden nach der Inkubation (Beispiel 3T3 Py6-Zellen der Maus, d.h. transformierte Fibroblasten; dünn ausgesät, mittlere Verdün¬ nung der Wirkstoff-Konzentrate) sich ein Kranz von Vakuolen um den Zellkern herum ausbildet.
Der analytische Nachweis, dass diese Vakuolen die Apocarotinester- Wirksubstanzen enthalten, kann sehr deutlich dadurch erbracht werden, dass man den gereinigten inkubierten Tumorzellen das Zellplasma entnimmt, es zentrifugiert und mit einer 0,05 %-Lösung von Uvitex® CF conc. (CIBA- GEIGY) in Aceton/Wasser (85:15) oder von Uvitex® EBF (CIBA-GEIGY) oder von Tinopal® GS (CIBA-GEIGY) versetzt.
Die eingesetzten erfindungsgetreuen Apocarotinester löschen die durch den Marker Uvitex CF conc, bzw. Uvitex EBF, bzw. Tinopal GS hervorgerufene Fluoreszenz im langwelligen UV-Lichtbereich aus; auf der Dünnschicht- Platte entsteht eine blaue Färbung. UV-Scanning bei 366 nm. Kontrolle mithilfe von mizellarer Kapillar-Zonen-Electrophorese, (Beckman Instru¬ ments, P/ACE System 2100).

Durchführung der Proben: Prof. Dott. Guido FORNI, Dotta Stefanie VAI, Universitä di Torino, Dipartimento di Scienze Cliniche e Biologiche, Ospe- dale San Luigi Gonzaga, 1-10*043 ORBASSANO (TO), August/September 1993.
Test mit normalen 8-wöchigen weiblichen BALB/c nAncr (H-2d)-Mäusen, geliefert von Charles River Laboratories, Calco (Italien). Während 4 Wochen täglich zweimalige Injektion i.v. von je 0,250 ml wässriger Mikroemulsion, gebildet aus den angegebenen Konzentraten, bzw. mit physiolgischem Puffer für die Kontrollen. Färbung mit May Grünwald-Giemsa.
Zeit der Behandlung: 28 Tg.
Blutanalyse nach der letzten Injektion
Anzahl Tiere: 13 Gruppen zu 5 je Tieren
RESULTAT: Es treten keine signifikanten Unterschiede auf zu den Kontrollen. Es konnte keine Toxizität der Konzentrate auf die Leukozyten- Population festgestellt werden. Auch die Erythrozyten-Population zeigte normale Werte. Alle Tiere waren und blieben gesund bis zum Schluss der Versuche.
Claims
PA TE N TA N S P R Ü C H E
1) Spontan dispergierbare Konzentrate, welche mit Wasser oder 5%-
Glucoselösung verdünnt, thermodynamisch stabile Ultramikroemulsionen ergeben mit Mizellen, die einen hydrodynamischen Radius von 1,5 bis 3 nm aufweisen, dadurch gekennzeichnet, dass folgende Komponenten zu einem
System zusammengefügt sind:
0,001 bis 30 Gewichts-% einzelner Ester mit Apocarotinolen der Formeln (I) bis (IX), bzw. eine Kombination solcher Ester

(IX) 0 bis 40 Gewichts-% eines als Hydrotrop oder Co-Emulgator dienenden pharmazeutisch verträglichen Lösungsmittels oder Lösungsmittelgemisches,

0 bis 10 Gewichts-% eines Vitamins oder Provitamins,
0 bis 10 Gewichts-% eines Stabilisators oder eines Penetrationsverbes- serers und Trägerstoffe und/oder Verdünnungsmittel.
2) Spontan dispergierbares Konzentrat, enthaltend Apocarotinylester der Formeln (I) bis (IX) gemäss Anspruch 1, zur Verwendung als Arzneimittel mit Wirksamkeit gegen Tumore, Psoriasis und Ekzeme.
3) Spontan dispergierbares Konzentrat gemäss Anspruch 1, dadurch ge¬ kennzeichnet, dass folgende Komponenten zusammengefügt sind:
0,5 bis 30 Gewichts-% eines oder mehrerer Wirkstoffe der Formeln (I) bis
(IX),
1 bis 40 Gewichts-% Isopropylmyristat, Isopropylpalmitat oder Oleum neutrale,
0 bis 45 Gewichts-% eines Phosphorsäureester-Tensides,
5 bis 90 Gewichts-% des wasserfreien Octylphenylpolyoxyäthylenäther- Tensids mit 9 bis 10 Oxyäthylengruppen.
4) Spontan dispergierbares Konzentrat gemäss Anspruch 1, dadurch ge¬ kennzeichnet, dass folgende Komponenten zusammengefügt sind:
0,5 bis 30 Gewichts-% eines oder mehrerer Wirkstoffe der Formeln (I) bis
(IX),
1 bis 40 Gewichts-% eines oder mehrerer biotensider Ester der allgemeinen Formel (XXIV),
R4 COO— R5
(XXIV) worin R4 Citronellyl, Geranyl, Farnesyl, Phytyl oder Isophytyl und R5 eine C1- bis C32-Akyl, bzw. C2- bis C32-Alkenyl- oder Alkapolyengruppe bedeuten,
5 bis 90 Gewichts-% eines pharmaverträglichen Tensides oder Tensidge- misches,
0 bis 10 Gewichts-% eines Vitamins oder Provitamins,
0 bis 10 Gewichts-% eines Stabilisators oder eines Penetrationsver- besserers und Trägerstoffe und/oder Verdünnungsmittel.
5) Spontan dispergierbares Konzentrat gemäss Anspruch 1 , dadurch ge¬ kennzeichnet, dass folgende Komponenten zu einem Systempräparat zusam¬ menfügt sind:
10 Gewichts-% eines öligen Wirkstoffes der Formeln (I) bis (IX), 0 bis 20 Gewichts-% Isopropylmyristat, Isopropylpalmitat, Oleum neutrale oder eines/bzw. mehrerer biotensider Ester der allgemeinen Formel (XXIV)
R4 COO R5 (XXIV) worin R4 Citronellyl, Geranyl, Farnesyl, Phytyl oder Isophytyl und R5 eine
C1 - bis C32-Akyl, bzw. C2- bis C32-Alkenyl- oder Alkapolyengruppe bedeuten,
35 bis 45 Gewichts-% des wasserfreien Octylphenylpolyoxyäthylenäther-
Tensids mit 9 bis 10 Oxyäthylengruppen,
35 bis 45 Gewichts-% des Mischemulgators, bestehend aus je 50 % der beiden Verbindungen mit Formeln:
(-CH2-CH2-O 7T?J-P— OCH3
OCH3 oder des Tristyrylphenolpolyoxyäthylen-18-mono/dimethyl-phophorsäure- ester Tensids.
6) Spontan dispergierbares Konzentrat gemäss Anspruch 1 , dadurch ge¬ kennzeichnet, dass folgende Komponenten <.u einem Systempräparat zusammenfügt sind:
2 bis 5 Gewichts-% eines kristallinen Wirkstoffes der Formeln (I) bis (IX), 10 bis 20 Gewichts-% Isopropylmyristat, Isopropylpalmitat, Neutralöl oder eines/bzw. mehrerer biotensider Ester der allgemeinen Formel (XXIV)
R4 C OO R5 (XXIV) worin R4 Citronellyl, Geranyl, Farnesyl, Phytyl oder Isophytyl und R5 eine C1 - bis C32-Akyl, bzw. C2- bis C32-Alkenyl- oder Alkapolyengruppe bedeuten,
35 bis 45 Gewichts-% des wasserfreien Octylphenylpolyoxyäthylenäther- Tensids mit 9 bis 10 Oxyäthylengruppen,
35 bis 45 Gewichts-% des Mischemulgators, bestehend aus je 50 % der beiden Verbindungen mit Formeln:

oder des Tristyrylphenolpolyoxyäthylen-18-mono/dimethyl-phophorsäure- ester Tensids.
7) Neue Ester mit Apocarotinolen der Formeln (I) bis (IX)

worin R für eine C4- bis C β-Alkyl-, eine C2- bis Cis-Alkenyl-, bzw. Alkapolyen-, eine C2- bis C-| 8-Alkinylgruppe oder für ein Radikal der Formel

8) Das ß-Apo-8'-carotinyl-10-undecenoat, ß-Apo-8'-carotinyl-laurat, ß-Apo-8'- carotinyl-palmitat, ß-Apo-8'-carotinyl-stearat, sowie das Azafrinyl-10-unde- cenoat, Azafrinyl-Iaurat, Azafrinyl-palmitat und das Azafrinyl-stearat gemäss Anspruch 7.
9) Verfahren zur Herstellung von Estern mit Apocarotinolen gemäss An¬ spruch 7, dadurch gekennzeichnet, dass man ein Apocarotin der Formeln (XV) bis (XXXIII) :

wiedergibt, bei einer Temperatur von 0 bis 60 °C unter Schutzgaszuleitung in einem indifferenten Lösungsmittel und in Gegenwart eines Katalysatoren umsetzt.
10) Verfahren zur Herstel lung von ß-Apo-δ'-carotinylestern, dadurch gekenn¬ zeichnet, dass man den δ'-Apo-ß-carotin-δ'-säuremethylester bei -30 °C mit DIBAH (Diisobutylaluminiumhydrid) red uziert, um den C30-Alkohol als Zwischenstufe zu gewinne n , der sodann mit einem aliphatischen Säure¬ ch lorid in Et2θ und Pyridin bei - 30 °C and anschl iessend bei Raum¬ tem peratur verestert wird.
1 1 ) Verfahren zur Herstel l ung von Azafrinylestern, dadurch gekennzeichnet, dass man zu nächst Azafrinol aus Azafrinmethylester m ittels DIBAH-Reaktion herstellt und diese Verbind ung unmittelbar anschl iessend mit einem ali phatischen Säurechlorid i n Et2θ und Pyridin bei - 30 °C and anschlies¬ send bei Raumtemperatur verestert.
