SELECTIVIDAD DE E. COLI C600 (PGL4) PARA MERCURIO. TASA DE LUMINISCENCIA CONTROL NEGATIVO. SE DESCRIBE UN PROCEDIMIENTO, EN EL QUE SE PUEDEN EMPLEAR MICROORGANISMOS, EQUIPADOS CON VECTOR PLASMIDO, PGL4, CONSTRUIDO MEDIANTE TECNICAS BIOLOGICAS MOLECULARES, COMO BIOSENSORES PARA COMPUESTOS DE MERCURIO. LA BASE DE LA INVENCION ES LA COMBINACION DE UN COMPLEJO-GEN (MER-OPERON) INDUCIBLE MEDIANTE IONES DE MERCURIO CON EL SISTEMA DE GEN LUCEFERASA (LUX) DE VIBRIO-HARVEYI MEDIANTE LA TECNICA DE CRONACION MOLECULAR. EN ESTE CASO, LA DISPOSICION DE AMBAS UNIDADES DE GEN ESTA CONCEBIDA DE FORMA QUE EL PROMOTOR DEL MER-OPERON REGULADO POSITIVAMENTE MEDIANTE COMPUESTOS DE MERCURIO CONTROLA TAMBIEN LA TRANSCRIPCION DE LOS GENES-LUX. COMO CONSECUENCIA, LOS MICROORGANISMOS TRANSFORMADOS CON PLASMIDO PGL4 RESPONDEN AL INCREMENTO DE LAS CANTIDADES DE CATIONES DE MERCURIO EN EL MEDIO CIRCUNDANTE CON UN AUMENTO EN SU BIOLUMINISCENCIA EMITIDA. EL PROCESO ES ABSOLUTAMENTE ESPECIFICO DEL MERCURIO, RAPIDO Y ALTAMENTE SENSIBLE: EL LIMITE PROBADO PARA IONES DE MERCURIO ES DE APROXIMADAMENTE 0,2 PPB, ES DECIR, 5 VECES INFERIOR AL UMBRAL DE DETECCION DE LOS METODOS MAS POTENTES ESTABLECIDOS (ESPECTROMETRIA DE ABSORCION ATOMICA), QUE DETECTAN DE 1 A 5 PPB DE MERCURIO Y REQUIEREN UNA CANTIDAD CONSIDERABLE DE APARATOS. OTRA VENTAJA DE LA INVENCION ES LA POSIBILIDAD DE DETERMINAR VALORES DE MEDICION (SEÑALES DE BIOLUMINISCENCIA EMITIDAS POR MICROORGANISMOS TRANSFORMADOS POR PLASMIDO PGLK4) SIN CONTACTO DIRECTO ENTRE EL BIOSENSOR Y LA UNIDAD AMPLIFICADORA DE LAS SEÑALES (APARATO DE BIOLUMINISCENCIA).
