Abstract
Un procedimiento para producir {b -Galactosidasa recombinante de Themus SP. (Cepa T2) en células hospedantes, y purificarla en un solo paso cromatográfico. Secuencia de nucleótidos inédita que corresponde a un gen de Thermus sp. (Cepa T2: ATCC 27737) y que codifica para una bota-galactosidasa termoestable. Obtención de construcciones genéticas para la expresión a alto nivel de dicha secuencia de nucleótidos en células de Eacherichia coli mediante su clonación en vectores de expresión. Purificación de una proteína híbrida con actividad beta-galactosidasa termoestable producida en E. coli en un sólo paso cromatográfico. Esta proteína quimérica termoestable tiene aplicación en hidrólisis de leche y productos lácteos, reducción del contenido en lactosa en los vertidos de sueros de lechería, marcaje frío de sondas de ácidos nucléicos por conjugación con los mismos, síntesis de oligosacáridos por transglicosilación y evaluación rápida de tratamientos térmicos moderados.
