Abstract
Un método mejorado para la purificación de un polipéptido del factor VIII que comprende: a) añadir una mezcla que contiene el polipéptido del factor VIII que se va a purificar a una matriz de inmunoafinidad que se une por atracción hidrofóbica al polipéptido del factor VIII; b) eluir el polipéptido del factor VIII desde la matriz de inmunoafinidad con una solución de desorción que causa la desorción del polipéptido del factor VIII, que se libera a una solución de elución; c) diluir la solución de elución con una solución que comprende mayor fuerza iónica que la de la solución de elución, dando como resultado una solución diluida del factor VIII, por lo que la solución de elución se diluye de 3 veces a 5 veces con respecto a la solución de elución inicial; d) hacer pasar la solución diluida de factor VIII a través de una columna de intercambio iónico capaz de unirse al polipéptido del factor VIII, por lo que se une el polipéptido del factor VIII mientras que se permite que los contaminantes pasen a través de la columna de intercambio iónico; y e) eluir el polipéptido del factor VIII purificado de la columna de intercambio iónico.
