Se apreciará que la invención no se restringe a una secuencia codificante de proteína particular de una de estas proteínas a co-expresar. Además, y en particular con respecto a factores de coagulación sanguínea, se han descrito en la técnica numerosas formas mutantes de las proteínas. La presente invención es igualmente aplicable a estas formas mutantes, incluyendo variantes alélicas de origen natural, de las proteínas, como a la secuencia de tipo silvestre. En una realización, la invención puede emprenderse con cualquier protrombina de tipo silvestre o una con al menos un 90%, preferiblemente al menos un 95% de identidad de secuencia con la misma.
La identidad de secuencia entre dos secuencias puede determinarse por análisis de alineación informática por pares, usando programas tales como, BestFit, PILEUP, Gap o FrameAlign. La herramienta de alineación preferida es BestFit. En la práctica, en la búsqueda de secuencias similares/idénticas para la búsqueda de consulta, desde dentro de una base de datos de secuencias, generalmente es necesario realizar una identificación inicial de secuencias similares usando algoritmos adecuados, tales como Blast, Blast2, NCBI Blast2, WashU Blast2, FastA, o Fasta3, y una matriz de valores tal como Blosum 62. Dichos algoritmos tratan de aproximarse estrechamente al algoritmo de alineación "de norma de oro" de Smith-Waterman. Por tanto, el software/programa motor de búsqueda preferido para su uso en la evaluación de la similitud, es decir, el modo en que dos secuencias polipeptídicas primarias se alinean es Smith-Waterman. Identidad se refiere a apareamientos directos, similitud permite sustituciones conservativas.
La expresión "γ-glutamil carboxilasa" o "GGCX", como se usa en este documento, se refiere a una enzima dependiente de vitamina K que cataliza la carboxilación de restos de ácido glutámico.
Las enzimas GGCX están ampliamente distribuidas, y se han clonado a partir de diferentes especies como la ballena beluga Delphinapterus leucas, el pez sapo Opsanus tau, el pollo (Gallus gallus), los mixinos (Myxine glutinosa), el cangrejo de herradura (Limulus polyphemus), y el caracol cónico Conus textile (Begley et al., 2000, ibid; Bandyopadhyay et al. 2002, ibid). La carboxilasa del caracol cónico es similar a la carboxilasa bovina y se ha expresado en células COS (Czerwiec et al. 2002, ibid). Pueden encontrarse proteínas adicionales similares a GGCX en insectos y procariotas tales como Anopheles gambiae, Drosophila melanogaster y Leptospira con los números de acceso NCBI: gi 31217234, gi 21298685, gi 24216281, gi 24197548 y (Bandyopadhyay et al., 2002, ibid), respectivamente. La enzima carboxilasa presenta conservación evolutiva notable. Varias de las enzimas no humanas han mostrado, o puede predecirse que tengan, actividad similar a la de GGCX humana que hemos usado, y por lo tanto pueden usarse como alternativa a la enzima humana.
La Tabla 3 identifica secuencias representativas de proteínas predichas homólogas a GGCX humana (ordenados por origen de la especie) que pueden usarse en la presente invención.

Uno modo para efectuar la expresión diferencial de las dos proteínas co-expresadas es usar diferentes promotores como parte de las respectivas secuencias de control de la expresión. La técnica está repleta de ejemplos de diferentes promotores y otras secuencias de control de la expresión que son capaces de expresar proteínas heterólogas en diferentes grados o niveles. La tecnología de expresión recombinante está adecuadamente
5 avanzada de modo que un especialista en la técnica arte de la expresión de proteínas es capaz de seleccionar promotores y otras secuencias de control para lograr la co-expresión de la proteína que requiere carboxilación y la carboxilasa en la proporción deseada. La selección de los cuales de los promotores particulares y otras secuencias de control de la expresión usar es una cuestión de elección individual.
10 Un promotor fuerte se define aquí como un promotor que da lugar a más de 1000 transcritos/célula. Un promotor débil se define aquí como un promotor que da lugar a menos de 1000 transcritos/célula.
La invención se ha ejemplificado mediante el uso del promotor de CMV fuerte (Boshart et al. Cell 41: 521-530, 1985) para sobre-expresar Factor IX o protrombina y el promotor de SV40 más débil (Wenger et al. Anal Biochem 221: 15 416-418, 1994) para controlar la expresión de GGCX. Otro promotor fuerte que podría usarse incluye, aunque sin limitación, pEF-1α [gen de la subunidad 1α del factor de elongación humano) (Mizushima y Nagata, Nuc Acids Res
18: 5322, 1990; Goldman et al., BioTechniques 21: 1013-1015, 1996), pRSV [virus del sarcoma de Rous (Gorman et al., Proc Natl Acad Sci USA 79: 6777-6781, 1982)] y pUbC [ubiquitina humana (Schorpp et al., Nuc Acids Res 24: 1787-1788, 1996)].
20 Es importante asegurarse de que la proteína a producir (proteína que requiere carboxilación) está en exceso en comparación con la enzima de modificación, dando una proporción de al menos 10:1. Las maneras de conseguir un bajo nivel de expresión de la enzima de modificación (γ-glutamil carboxilasa) incluyen:
25 1) Usar un promotor débil para controlar la expresión de la enzima de modificación incluyendo, aunque sin limitación, el promotor temprano inmediato de SV40, el promotor FIX minimizado (Rouet et al., The Journal of Biological Chemistry 267: 20765-20773, 1992) o el promotor de timidina quinasa de HSV (Wenger et al., 1994, ibid). 2) Mutar las secuencias promotoras o potenciadoras de un promotor fuerte para reducir la fuerza del promotor.

35 proteína a producir. 5) Clonar el ARN que codifica la proteína a producir y el ADN que codifica vectores de modificación de GGCX en vectores diferentes, co-transfectar o transfectar por separado, y usar un sistema de amplificación para amplificar la expresión de la proteína a producir. 6) Aislar una línea celular estable que expresa de forma recombinante bajos niveles de GGCX (pero por encima
40 de los niveles endógenos) y el usar como línea de células hospedadoras para la expresión de proteínas en necesidad de γ-carboxilación. 7) Introducir una o más mutaciones en la GGCX para disminuir la afinidad por sustrato de GGCX.
Además de éstos, los especialistas en la técnica de la expresión de proteínas recombinantes serán conscientes de 45 otros métodos que podrían usarse para generar una célula hospedadora que exprese la proteína que requiere carboxilación y la proteína carboxilasa en una proporción de al menos 10:1.
La sobre-expresión de proteínas dependientes de carboxilación anteriormente ha provocado generalmente productos su-carboxilados. Esto se debe a que se está limitando la capacidad endógena de carboxilación de la 50 célula hospedadora. Por otro lado una inmensa (16 a 70 veces) sobre-expresión de la actividad GGCX no mejora el rendimiento del producto (Rehemtulla et al., Proc Natl Acad Sci USA 90: 4611-4615, 1993), (Berkner y Pudota, Proc Natl Acad Sci USA. 95: 446-471, 1998), (Hallgren et al., Biochemistry 41 (50): 15045-15055, 2002). La razón de esto no se entiende completamente. Nuestra invención requiere una moderada sobre-expresión de GGCX. Esto asegura que se expresen niveles mayores que los endógenos de GGCX a partir de la célula, por ejemplo, donde el nivel de
55 actividad GGCX se eleva sólo de 1,5 a 5 veces. A este nivel moderadamente elevado, se obtuvieron niveles sorprendentemente altos de rhFII completamente carboxilado como se muestra en el Ejemplo 1.
Por lo tanto se apreciará que la proporción de expresión de la proteína que requiere carboxilación y la carboxilasa que distingue a esta invención de las enseñanzas previas de co-expresión, excluye niveles de GGCX que se 60 producen de manera endógena. Para cumplir con la alta productividad requerida es necesario expresar la carboxilasa y la proteína que requiere carboxilación a niveles por encima de los encontrados en células normales.
En una realización preferida, se usa una célula o una línea celular que tiene poco o nada de carboxilasa expresada constitutivamente y/o proteína que requiere carboxilación. 65

pSV40-GGCX-pA clonado en el sitio DraIII romo de pCDNA3.1. Se seleccionó un clon con el fragmento que contiene GGCX en orden inverso en comparación con el Ej. 1. Clonación del fragmento FII se hizo después de la misma manera que en el Ej. 1. La construcción final PP6 se muestra en la Figura 2 y la secuencia de nucleótidos de PP6 se muestra en la SEC ID Nº 13.
5 Se obtuvieron dos líneas celulares de producción de protrombina, B2F4 y H3B 10 transfectando células CHO-S como se ha descrito en el Ej. 1. Se purificó la protrombina de estas dos líneas celulares y se caracterizó como en el Ej. 1. Los cultivos de B2F4 dieron productividades que varían de 30-70 mg/l y el porcentaje de completamente carboxilado del 55-87% (cuanto más rhFII menos completamente carboxilado). La adición de butirato dio
10 productividad un poco más alta, pero disminuyó el porcentaje de rhFII completamente carboxilado y no se consideró beneficioso. H3B10 es de crecimiento lento y dio una productividad de aproximadamente 50 mg/l, que fue elevada en relación con la cantidad de células en el cultivo, y el porcentaje de rhFII completamente carboxilado fue de aproximadamente el 60%. En comparación con la línea celular obtenida en el ejemplo 1, se produjo menos rhFII completamente carboxilado usando la construcción PP6 para una línea celular CHO. La producción de protrombina

Análisis por RT-PCR a tiempo real de la expresión de γ-carboxilasa y protrombina en líneas celulares CHO-S 20 midiendo la cantidad de ARNm
Se analizaron las líneas celulares B2F4 y H3B10 del ejemplo 4 por análisis de PCR a tiempo real por el mismo método y los mismos cebadores que en el Ej. 3. Se recogieron muestras de cultivo de 10 ml en la máxima productividad para que fueran equivalente a las muestras del Ej. 3. Para el clon H3B10 las muestras eran del día 10
25 debido al lento crecimiento de este clon, y para el clon B2F4 las muestras eran del día 6.
