Un aptamero pegilado o no pegilado para su uso en un metodo para tratar, estabilizar y/o prevenir la degeneracion macular asociada a la edad (DMAE) de tipo no exudativo, comprendiendo el metodo la etapa de administrar un aptamero pegilado o no pegilado a un sujeto que lo necesite, en donde el aptamero pegilado o no pegilado se une al complemento de C5 y tiene la secuencia de SEC ID No: 4.

Los oligonucleótidos al azar pueden sintetizarse a partir de oligonucleótidos unidos mediante enlaces fosfodiester usando técnicas de síntesis de oligonucleótidos de fase sólida bien conocidos en la técnica. Véase, por ejemplo, Froehler et al., Nucl. Acid Res., 14:5399-5467 (1986) y Froehler et al., Tet. Lett. 27:5575-5578 (1986). Los oligonucleótidos al azar también pueden sintetizarse usando métodos en fase de solución, tales como métodos de síntesis de triester. Véase, por ejemplo, Sood et al., Nucl. Acid Res., 4:2557 (1977) y Hirose et al., Tet. Lett., 28:2449 (1978). La biblioteca de partida de oligonucleótidos puede generarse mediante síntesis química automatizada en un sintetizador de ADN. Para sintetizar secuencias aleatorizadas, se añaden mezclas de los cuatro nucleótidos en cada etapa de adición de nucleótido durante el proceso de síntesis, permitiendo la incorporación de nucleótidos al azar. Como se afirma anteriormente, en una realización, los oligonucleótidos al azar comprenden por completo secuencias al azar; sin embargo, en otras realizaciones, los oligonucleótidos al azar pueden comprender porciones de secuencias no aleatorias o parcialmente aleatorias. Las secuencias parcialmente aleatorias pueden crearse mediante la adición de los cuatro nucleótidos en diferentes proporciones molares en cada etapa de adición.
La biblioteca de oligonucleótidos de partida puede ser ARN, ADN, ARN o ADN sustituido o combinaciones de los mismos. En aquellos casos en los que se va a usar una biblioteca de ARN como librería de partida, se genera normalmente sintetizando una biblioteca de ADN, opcionalmente amplificando mediante PCR, después transcribiendo la biblioteca de ADN in vitro usando ARN polimerasa T7 o ARN polimerasas T7 modificadas, y purificando la secuencia transcrita. En aquellos casos donde se estén seleccionando aptámeros de ARN, el método SELEX™ comprende las etapas de: (i) retrotranscribir los ácidos nucleicos disociados de los complejos ácido nucleico-diana antes de la etapa de amplificación (d); y (ii) transcribir los ácidos nucleicos amplificados de la etapa (d) antes de volver a comenzar el proceso.
Dentro de una mezcla de ácidos nucleicos que contiene un gran número de secuencias y estructuras posibles, hay un gran intervalo de afinidades de unión para una diana dada. Aquellos que tengan la mayor afinidad de unión (menores constantes de disociación) por la diana son los que son más probables que se unan a la diana. Después del particionamiento, disociación y amplificación, se genera una segunda mezcla de ácido nucleico, enriquecida para los candidatos de mayor afinidad de unión. Los ciclos adicionales de selección favorecen de manera progresiva a los mejores ligandos hasta que la mezcla resultante de ácido nucleico esté compuesta predominantemente de solo una o unas pocas secuencias. Estas pueden clonarse, secuenciarse y ensayarse adicionalmente como ligandos o aptámeros para 1) afinidad de unión por la diana; y 2) capacidad para afectar a la función de la diana.
Los ciclos de selección y amplificación se repiten hasta que se logra una meta deseada. En el caso más general, la selección/amplificación continua hasta que no se logra una mejora significativa en la fuerza de unión tras la repetición del ciclo. El método se usa normalmente para muestrear aproximadamente 1014 especies diferentes de ácidos nucleicos, pero puede usarse para muestrear hasta aproximadamente 1018 especies de ácido nucleico deferentes. En general, las moléculas de aptámero de ácido nucleico se seleccionan en un procedimiento de 5 a 20 ciclos. En una realización, se introduce heterogeneidad solo en las etapas iniciales de selección y no sucede durante el proceso de replicación.
En una realización del método SELEX™, el proceso de selección es tan eficiente para aislar aquellos ligandos de ácido nucleico que se unen con más fuerza a la diana seleccionada, que solo se necesita un ciclo de selección y amplificación. Puede suceder una selección así de eficiente, por ejemplo, en un proceso de tipo cromatográfico en el que la capacidad de los ácidos nucleicos para asociarse con dianas unidas sobre una columna funciona de tal modo que la columna es suficientemente capaz de permitir la separación y aislamiento de los ligandos de ácido nucleico de la mayor afinidad.
En muchos casos, no es necesariamente deseable efectuar las etapas iterativas de SELEX™ hasta que se identifica un solo ligando de ácido nucleico. La solución de ligando de ácido nucleico específico de diana puede incluir una familia de estructuras de ácido nucleico o motivos que tengan un número de secuencias conservadas y un número de secuencias que pueden sustituirse o añadirse sin afectar de manera significativamente la afinidad de los ligandos

oligonucleótidos que son resistentes a nucleasas también pueden incluir una o más uniones internucleótido sustitutas, azúcares alterados, bases alteradas, o una de sus combinaciones. Dichas modificaciones incluyen, pero sin limitación, modificaciones de azúcar en posición 2', modificaciones de pirimidina en posición 5, modificaciones de purina en posición 8, modificaciones en aminas exocíclicas, sustitución de 4-tiouridina, sustitución de 5-bromo o 5yodo-uracilo; modificaciones de estructura, modificaciones de fosforotioato o fosfato de alquilo, metilaciones, y combinaciones de emparejamiento de bases inusuales tales como las isobases isocitidina e isoguanosina. Las modificaciones también pueden incluir modificaciones 3' y 5', tales como taponado, por ejemplo, adición de un tapón 3'-3'-dT para aumentar la resistencia a exonucleasas (véase, por ejemplo, las Patentes de Estados Unidos 5.674.685; 5.668.264; 6.207.816; y 6.229.002).
En una realización, se proporcionan oligonucleótidos en los que el grupo P(O)O se sustituye por P(O)S ("tioato"), P(S)S ("ditioato"), P(O)NR2 ("amidato"), P(O)R, P(O)OR’, CO o CH2 ("formacetal") o 3’-amina (-NH-CH2-CH2-), en los que cada R o R' es independientemente H o alquilo sustituido o sin sustituir. Las modificaciones después del proceso SELEX™ efectuadas sobre ligandos de ácido nucleico pueden dar como resultado estabilidad mejorada, por ejemplo, estabilidad in vivo sin afectar de manera negativa a la capacidad de unión del ligando de ácido nucleico. El método SELEX™ abarca combinar oligonucleótidos seleccionados con otros oligonucleótidos seleccionados y unidades funcionales no de oligonucleótido como se describe en la Patente de los Estados Unidos Nº 5.637.459 y la Patente de los Estados Unidos Nº 5.683.867. El método SELEX™ abarca además combinar ligandos de ácido nucleico seleccionados con compuestos de elevado peso molecular lipófilos o no inmunogénicos en un complejo diagnóstico o terapéutico, como se ha descrito, por ejemplo, en la patente de los Estados Unidos nº 6.011.020, La patente de Estados Unidos Nº 6.051.698, y en la publicación PCT Nº WO 98/18480. Estas patentes y solicitudes enseñan la combinación de un amplio abanico de formas y otras propiedades, con las eficientes propiedades de amplificación y replicación de oligonucleótidos, y con las propiedades deseables de otras moléculas.
También se ha explorado la identificación de ligandos de ácido nucleico a péptidos pequeños y flexibles mediante el método SELEX™. Los péptidos pequeños tienen estructuras flexibles y existen normalmente en solución en un equilibrio de múltiples confórmeros, y por lo tanto se pensó inicialmente que las afinidades de unión pueden estar limitadas por la entropía conformacional perdida tras la unión a un péptido flexible. Sin embargo, la posibilidad de identificar ligandos de ácido nucleico a péptidos pequeños en solución se demostró en la Patente de los Estados Unidos Nº 5.648.214. En esta patente, se identificaron ligandos de ácido nucleico de ARN de alta afinidad a sustancia P, un péptido de 11 aminoácidos.
Los aptámeros con especificidad y afinidad de unión a la diana (o dianas) de complemento de la presente invención se seleccionan normalmente mediante el proceso SELEX™ como se describe en el presente documento. Adicionalmente, pueden efectuarse selecciones con secuencias que incorporan nucleótidos modificados para estabilizar las moléculas de aptámero contra la degradación in vivo.
SELEX™ 2' modificado
Para que un aptámero sea útil para su uso como agente terapéutico o diagnóstico, es preferentemente no costoso sintetizar, seguro y estable in vivo. Los aptámeros de ARN y ADN de tipo silvestre son normalmente no estables in vivo debido a su susceptibilidad a degradación mediante nucleasas. La resistencia a la degradación por nucleasa puede aumentarse en gran medida mediante la incorporación de grupos modificantes en la posición 2'.
Los grupos 2'-fluoro y 2'-amino se han incorporado con éxito en grupos de oligonucleótidos a partir de los cuales a continuación se han seleccionado aptámeros. Sin embargo, estas modificaciones aumentan en gran medida el coste de la síntesis del aptámero resultante, y pueden introducir preocupaciones respecto a la seguridad en algunos casos porque la posibilidad de que los nucleótidos modificados puedan reciclarse en ADN huésped mediante degradación
