Las bases representativas que se pueden usar en la preparación de sales farmacéuticamente aceptables incluyen las siguientes: amoníaco, L-arginina, benetamina, benzatina, hidróxido de calcio, colina, deanol, dietanolamina, dietilamina, 2-(dietilamino)-etanol, etanolamina, etilendiamina, N-metil-glucamina, hidrabamina, 1H-imidazol, L-lisina, hidróxido de magnesio, 4-(2-hidroxietil)morfolina, piperazina, hidróxido de potasio, 1-(2-hidroxietil)pirrolidina, amina secundaria, hidróxido de sodio, trietanolamina, trometamina e hidróxido de zinc.
Los ésteres farmacéuticamente aceptables de los compuestos de la presente invención se seleccionan de alquilo de C1-7, cicloalquilo de C5-7, fenilo, fenilo substituido, y ésteres de fenil-alquilo de C1-6. Los ésteres preferidos incluyen ésteres metílicos.
Si R7 es el grupo de fórmula (II), n es preferentemente 1. En una realización preferida, R8 y R9 son átomos de hidrógeno, mientras que R10 es un grupo metilo.
Si R7 es el grupo de fórmula (III), las combinaciones preferidas de q y r son, por ejemplo q = 1 y r = 1, q = 2 y r = 0, y q = 3 y r =0.
Una clase preferida de compuestos es los compuestos de quinazolina de la invención, en la que X es N e Y es CR3. Los vehículos fisiológicamente aceptables son bien conocidos en la técnica. Los ejemplos de vehículos líquidos son disoluciones en las que los compuestos según la invención forman disoluciones, emulsiones, y dispersiones. Los antioxidantes compatibles tales como metilparabeno y propilparabeno pueden estar presentes en composiciones sólidas y/o líquidas, como lo pueden estar los endulzantes.
Las composiciones farmacéuticas según la invención pueden incluir emulsionantes apropiados típicamente usados en composiciones de emulsión. Los agentes gelificantes se pueden añadir también a composiciones según la invención. Los derivados de poli(ácido acrílico) tales como carbómeros, son ejemplos de agentes gelificantes, y más particularmente, varios tipos de carbopol. Las suspensiones se pueden preparar en forma de una crema, una pomada, incluyendo una pomada libre de agua, una emulsión de agua en aceite, una emulsión de aceite en agua, y emulsión de gel, o un gel.
Los compuestos según la presente invención se pueden administrar por rutas orales o parenterales, incluyendo administración intravenosa, intramuscular, intraperitoneal, subcutánea, rectal, intracisternal, intravaginal, intravesical, tópica o local, y por inhalación (bucal o nasal, preferentemente en la forma de una pulverización). Para administración oral, los compuestos según la invención se proporcionarán generalmente en la forma de comprimidos, cápsulas, o en forma de una disolución o suspensión. Otros métodos de administración incluyen formulaciones de liberación controlada, tales como implantes subcutáneos y parches dérmicos.
Las dosis farmacéuticamente efectivas de los compuestos según la invención se pueden determinar por métodos convencionales. El nivel de dosificación específica requerido para cualquier sujeto particular dependerá de varios factores, que incluyen la severidad del estado, tipo de los síntomas que necesitan tratamiento, la ruta de administración, el peso, edad, y estado general del sujeto, y la administración de otros medicamentos.
En general, la dosis diaria (administrada como dosis unitaria o como dosis divididas) estará en el intervalo de alrededor de 0,01 mg a alrededor de 1.000 mg por día, más usualmente de alrededor de 1 mg a alrededor de 500 mg por día, y lo más usualmente de alrededor de 10 mg a alrededor de 200 mg por día. Expresada como dosificación por unidad de peso corporal, se esperará que una dosis típica esté entre alrededor de 0,0001 mg/kg y alrededor de 15 mg/kg, especialmente entre alrededor de 0,01 mg/kg y alrededor de 7 mg/kg, y lo más especialmente entre alrededor de 0,15 mg/kg y 2,5 mg/Kg.
Los intervalos de dosis oral incluyen de alrededor de 0,01 a 500 mg/kg, diariamente, más preferentemente de alrededor de 0,05 a alrededor de 100 mg/kg, tomadas en 1-4 dosis separadas. Algunos compuestos de la invención se pueden dosificar oralmente en el intervalo de alrededor de 0,05 a alrededor de 50 mg/kg diariamente, mientras que otros se pueden dosificar a de 0,05 a alrededor de 20 mg/kg diariamente. Ejemplo 10
3-(6-cloro-2-(4-metilpiperazin-1-il)quinazolina-4-amino)-N-metilpropanosulfonamida
Se añadió 2,4,6-tricloroquinazolina (200 mg) a una disolución de DIPEA (0,46 ml) e hidrocloruro de 3-aminopropanoN-metilsulfonamida (162 mg) en THF (3,0 ml) y la mezcla se agitó durante la noche a temperatura ambiente. La disolución se diluyó con EtOAc y se lavó con agua y salmuera. El secado sobre Na2SO4 y la retirada del disolvente dio un sólido que se purificó sobre SiO2 (EtOAc:Hex, 1:1) para dar el intermedio 3-(2,6-dicloro-quinazolina-4-amino)N-metilpropanosulfonamida. Este intermedio se añadió a un tubo de microondas que contiene N-metilpiperazina (1,0 ml) y THF (3,0 ml) y esta disolución se calentó a 130ºC. Después de 15 minutos la mezcla obtenida se diluyó con agua y se extrajo con EtOAc. Los extractos orgánicos combinados se secaron sobre Na2SO4 y se evaporaron para dar el producto en bruto. La purificación sobre SiO2 (EtOAc:MeOH:Et3N, 90:5:5) dio el compuesto del título en forma de un sólido. Rendimiento: 104 mg (30%). 1H-RMN (CDCl3) δ (ppm) 7,48-7,32 (m, 3H), 5,91 (m, 1H), 5,91 (m, 1H), 3,88 (t, J = 5,0 Hz, 4H), 3,78 (q, J = 6,2 Hz, 2H), 3,12 (t, J = 7,2 Hz, 2H), 2,78 (s, 3H), 2,45 (t, J = 5,1 Hz, 4H), 2,32 (s, 3H), 2,22 (p, J = 7,1 Hz, 2H). 13C-RMN (CDCl3) δ (ppm); 158,88, 158,39, 150,28, 132,30, 126,76, 123,78, 121,81, 111,00, 54,39, 47,14, 45,64, 43,08, 28,34, 22,61; MS (ESI) m/z 413 (M + H)+.

Preparado según el método general E a partir de 2,4-dicloro-5-trifluorometilquinazolina (200 mg) y 2-aminoetano-Nfenilsulfonamida (164 mg).
