Abstract
Un proceso para la producción de ácido polisiálico con una pureza superior al 95 % y un peso molecular de 70.000 Da a 100.000 Da, en que el proceso comprende: (a) la obtención de un medio nutriente que comprende sorbitol en una cantidad de 8 g/l a 12 g/l, extracto de levadura en una cantidad de 0,4 g/l a 0,6 g/l, casaminoácidos en una cantidad de 8 g/l, hidrogenofosfato de potasio en una cantidad de 8 g/l a 12 g/l, sulfato de cobre en una cantidad de 0,001 g/l a 0,003 g/l y sulfato de magnesio en una cantidad de 0,4 g/l a 0,6 g/l; (b) la inoculación de bacterias productoras de ácido polisiálico pertenecientes al serogrupo O7:K1 de E. coli, que no muestran O-acetilación del ácido polisiálico en dicho medio nutriente, opcionalmente donde la bacteria productora de ácido polisiálico es E. coli LP 1674; (c) la incubación del medio inoculado a una temperatura de 30 °C a 40 °C, a un pH de 6-8, un porcentaje de oxígeno disuelto del 35 % al 45 % y una agitación de 100 a 150 rpm; (d) la recogida del ácido polisiálico producido en la etapa (c); (e) la purificación del ácido polisiálico obtenido en la etapa (d); y (f) opcionalmente la concentración del ácido polisiálico obtenido en la etapa (e).
