Un proceso para la preparación de un compuesto intermedio sintético, proceso que comprende a) tratar un compuesto de fórmula P-2**Fórmula** con bis(trimetilsilil)amida de litio y benzaldehído para formar un compuesto de fórmula P-3**Fórmula** tratar el compuesto de fórmula P-3 con trietilamina y cloruro de metanosulfonilo, seguido de N-metil-2-pirrolidona y 1,8-diazabiciclo[5.4.0]undec-7-eno para formar un compuesto de fórmula P-4**Fórmula** y tratar el compuesto de fórmula P-4 con RuCl3 y NalO4 para formar un compuesto de fórmula P-5**Fórmula** o b) tratar un compuesto de fórmula P-5**Fórmula** con 3-amino-propano-1,2-diol para formar un compuesto de fórmula P-6**Fórmula** tratar el compuesto de fórmula P-6 con NaHCO3 y (MeO)2SO2 o Mel para formar un compuesto de fórmula P-7**Fórmula** y tratar el compuesto de fórmula P-7 con NalO4 para formar un compuesto de fórmula P-8**Fórmula** o c) metilar un compuesto de fórmula P-6 con NaHCo3 y (MeO)2SO2 o Mel**Fórmula** para formar un compuesto de fórmula P-7**Fórmula** y tratar el compuesto de fórmula P-7 con NalO4 para formar un compuesto de fórmula P-8 como se ha definido anteriormente.

c) Carbonato de {[(3S,11aR)-8-({[(2,4-difluorofenil)metil]amino}carbonil)-3-metil-5,7-dioxo-2,3,5,7,11,11ahexahidro[1,3]oxazolo[3,2-a]pirido[1,2-d]pirazin-6-il]oxi}metilo y 4-nitrofenilo. El derivado del carbonato de nitrofenilo se preparó a partir de sal sódica de (3S,11aR)-N-[(2,4-difluorofenil)metil]-6-hidroxi-3-metil-5,7-dioxo-2,3,5,7,11,11a-hexahidro[1,3]oxazolo[3,2-a]pirido 5 [1,2-d]pirazina-8-carboxamida (50 mg, 0,117 mmol), carbonato de yodometilo y 4-nitrofenilo (76 mg, 0,234 mmol), carbonato potásico (49 mg, 0,351 mmol), e hidrogenosulfato de tetrabutilamonio (40 mg, 0,117 mmol) de una forma similar a la que se ha descrito en el ejemplo 1, etapa c. El compuesto se aisló en una mezcla impura que se usó más adelante sin purificación adicional. La mezcla resultante se agitó a ta durante 3 h. El procesamiento acuoso convencional seguido de purificación mediante HPLC en fase inversa proporcionó el compuesto del título en forma de la sal de TFA que se suspendió en diclorometano y se lavó con bicarbonato sódico acuoso y se secó sobre sulfato sódico. El producto preparado como se ha descrito en la etapa (a) anterior (173 mg, 0,239 mmol) se disolvió en diclorometano, se lavó con solución de bicarbonato sódico, y se secó sobre sulfato sódico. Este aislado 5 se disolvió en dioxano, se enfrió en un baño de hielo-agua y se añadió HCl (0,24 ml, solución 1 normal) y la reacción se agitó 15 minutos a temperatura ambiente. Ejemplo 26: Farmacocinética en rata
Ratas macho CD en ayuno recibieron el compuesto del Ejemplo 12 en forma de una dosis en suspensión oral (5 mg de
15 equivalente precursor/kg en hidroxipropilmetilcelulosa al 0,1 %/Tween-80 al 0,1 %) administrada por vía oral mediante una sonda nasogástrica. Se extrajeron muestras de sangre (0,2 ml cada una) desde una cánula implantada quirúrgicamente en la vena femoral a intervalos de tiempo durante 24 h después de la administración de la dosis; todas las muestras se extrajeron usando jeringas tratadas con AEDT . Cada muestra de sangre se combinó con 0,02 ml de una solución inhibidora de proteasa [ácido e-amino-n-caproico, benzamida HCl, y fluoruro de
20 4-(2-aminoetil)bencenosulfonilo HCl en agua] para inhibir ex vivo la conversión del profármaco en el compuesto precursor, se agitó vorticialmente para mezclar, y se centrifugó (4000 x g, 4 ºC, 20 min) para obtener el plasma.
