Abstract
Un anticuerpo modificado con cisteína, en el que el anticuerpo modificado con cisteína comprende una sustitución de uno o más aminoácidos a un resto de cisteína en la región 131-139 de la cadena pesada de un anticuerpo como se define por el sistema de numeración del índice EU, en el que el anticuerpo modificado con cisteína comprende al menos un grupo tiol libre.

132-134-136Cys (D). El trazado de puntos representa un conjunto de marcadores de peso molecular definidos usados para establecer el peso molecular aparente de los anticuerpos. Como se demuestra en (A), el anticuerpo 1C1 WT eluye de la columna de SEC con un peso molecular correspondiente a un anticuerpo monomérico. El análisis de SEC de los anticuerpos manipulados con cisteína 1C1 Ser134Cys (B), 1C1 Ser132Cys (C) y 1C1 Ser131-132-134-136Cys (D) demostró que estos anticuerpos también existen en forma monomérica, similar al anticuerpo no mutante.
La FIGURA 5 representa los resultados de un ensayo de unión al antígeno basado en ELISA realizado en anticuerpos purificados, concretamente 1C6 WT y 1C6 Ser131Cys. Estos anticuerpos reconocen específicamente el receptor de EphB4. Como se demuestra en la figura, el perfil de afinidad de unión medido en un formato de ELISA del anticuerpo WT y el anticuerpo Ser131Cys manipulado con cisteína son muy similares.
La FIGURA 6 representa los resultados de un ensayo de unión al antígeno basado en ELISA realizado en anticuerpos purificados, concretamente 1C1 WT y 1C1 Ser131Cys. Estos anticuerpos reconocen específicamente el receptor de EphA2. Como se demuestra en la figura, el perfil de afinidad de unión medido en un formato de ELISA del anticuerpo WT y el anticuerpo Ser131Cys manipulado con cisteína son muy similares. Además, la inclusión de DTT 1 mM no tuvo un efecto medible sobre el perfil de unión.
La FIGURA 7 representa termogramas de calorimetría diferencial de barrido (DSC) del anticuerpo 1C6 WT (A) y el anticuerpo 1C6 Ser131Cys (B). Ambos anticuerpos presentan temperaturas de fusión (Tm) muy similares de 70 ºC y 69 ºC, respectivamente.

La FIGURA 10 representa los resultados de un ensayo BIACORE® que mide las afinidades relativas para los anticuerpos 1C1 WT y 1C1 Ser131Cys para diversos receptores de Fcγ. Los diversos receptores de Fcγ estudiados fueron FcγRI (A), FcγRIIIA (B), FcγRIIA (C), FcγRIIB (D). Los anticuerpos 1C1 WT y 1C1 Ser131Cys presentan afinidades de unión muy similares para diversos receptores de Fcγ.
La FIGURA 11 representa los resultados de un ensayo BIACORE® que mide las afinidades relativas para los anticuerpos 1C1 WT y 1C1 Ser131Cys para el receptor FcRn a pH 6,0 y pH 7,4. El anticuerpo 1C1 Ser131Cys se une al receptor FcRn con un perfil de unión similar al anticuerpo 1C1 WT a tanto pH 6,0 como pH 7,4.

También se conoce en la técnica que la región Fc puede modificarse para aumentar las semividas de proteínas. El aumento en la semivida permite la reducción en la cantidad de fármaco administrada a un paciente, además de reducir la frecuencia de administración. Por consiguiente, los anticuerpos de la invención con semividas elevadas pueden generarse modificando (por ejemplo, sustituyendo, delecionando o añadiendo) restos de aminoácidos identificado como implicados en la interacción entre Fc y el receptor FcRn (véanse, por ejemplos, las publicaciones PCT N.º 97/34631 y 02/060919 e). La presente invención engloba proteínas de variantes de Fc que tienen propiedades de unión alteradas para un ligando de Fc (por ejemplo, un receptor de Fc, C1q) con respecto a una molécula comparable (por ejemplo, una proteína que tiene la misma secuencia de aminoácidos, excepto que tiene una región Fc no mutante). Generalmente se entiende que una molécula de unión (por ejemplo, una proteína de variantes de Fc tal como un anticuerpo) con una KD baja puede ser preferible a una molécula de unión con una KD alta. Sin embargo, en algunos casos, el valor de kas o kdis puede ser más relevante que el valor de KD. Un experto en la materia puede determinar qué parámetro cinético es el más importante para una aplicación de anticuerpo dada.
Las afinidades y propiedades de unión de una región Fc por su ligando pueden determinarse mediante una variedad de métodos de ensayo in vitro (ensayos bioquímicos o basados en inmunología) conocidos en la técnica para determinar interacciones Fc-FcγR, es decir, unión específica de una región Fc con un FcγR que incluye, pero no se limita a, métodos de equilibrio (por ejemplo, enzimoinmunoanálisis de adsorción (ELISA) o radioinmunoensayo (RIA)), o cinética (por ejemplo, análisis BIACORE®), y otros métodos tales como ensayos de unión indirecta, ensayos de inhibición competitiva, transferencia de energía por resonancia de fluorescencia (FRET), electroforesis en gel y cromatografía (por ejemplo, filtración en gel). Estos y otros métodos pueden utilizar una marca en uno o más de los componentes que se examinan y/o emplear una variedad de métodos de detección que incluyen, pero no se limitan a, marcas cromogénicas, fluorescentes, luminiscentes o isotópicas. Células efectoras útiles para tales ensayos incluyen células mononucleares de sangre periférica (CMSP) y linfocitos citolíticos espontáneos (NK). Ejemplos específicos de ensayos ADCC in vitro se describen en Wisecarver et al., 1985 79:277-282; Bruggemann et al., 1987, J Exp Med 166:1351-1361; Wilkinson et al., 2001, J Immunol Methods 258:183-191; Patel et al., 1995 J Immunol Methods 184:29-38. La actividad de ADCC de la proteína de variantes de Fc de interés también puede evaluarse in vivo, por ejemplo, en un modelo animal tal como aquél desvelado en Clynes et al., 1998, Proc. Natl. Acad. vector que expresa el anticuerpo es amplificable, el aumento en el nivel de inhibidor presente en el cultivo de célula huésped aumentará el número de copias del gen marcador. Como la región amplificada está asociada al gen de anticuerpo, también aumentará la producción de anticuerpo (Crouse et al., 1983, Mol. Cell. Biol. 3:257).
La célula huésped puede co-transfectarse con dos vectores de expresión de la invención, el primer vector que codifica un polipéptido derivado de cadena pesada y el segundo vector que codifica un polipéptido derivado de cadena ligera. Los dos vectores pueden contener marcadores de selección idénticos que permiten la expresión igual de polipéptidos de cadena pesada y ligera. Alternativamente, puede usarse un único vector que codifica, y es capaz de expresar, tanto polipéptidos de cadena pesada como ligera. En tales situaciones, la cadena ligera debe ponerse antes de la cadena pesada para evitar un exceso de cadena pesada libre tóxica (Proudfoot, 1986, Nature 322:52; y Kohler, 1980, PNAS 77:2197). Las secuencias codificantes para las cadenas pesadas y ligeras pueden comprender ADNc o ADN genómico.
Una vez se ha producido un anticuerpo modificado con cisteína de la invención por expresión recombinante, puede purificarse por cualquier método conocido en la técnica para la purificación de una molécula de inmunoglobulina, por ejemplo, por cromatografía (por ejemplo, intercambio iónico, afinidad, particularmente por afinidad por el antígeno específico después de Proteína A, y cromatografía de exclusión molecular), centrifugación, solubilidad diferencial o por cualquier otra técnica estándar para la purificación de proteínas. Además, los anticuerpos de la presente invención o fragmentos de los mismos pueden fusionarse con secuencias de polipéptidos heterólogas descritas en el presente documento o conocidas de otro modo en la técnica para facilitar la purificación.
Producción escalable de anticuerpos manipulados con cisteína
En un esfuerzo por obtener grandes cantidades de los anticuerpos manipulados con cisteína de la invención, pueden producirse por un proceso escalable (denominado en lo sucesivo "proceso escalable de la invención"). En algunas realizaciones, los anticuerpos manipulados con cisteína pueden producirse por un proceso escalable de la invención en el laboratorio de investigación que puede aumentarse de escala para producir las proteínas de la invención en biorreactores a escala analítica (por ejemplo, pero no se limitan a, biorreactores de 5 l, 10 l, 15 l, 30 l o 50 l) mientras que se mantiene la actividad funcional de las proteínas. Por ejemplo, en una realización, las proteínas producidas por procesos escalables de la invención presentan niveles de bajos a indetectables de agregación como se mide por HPSEC o rCGE, es decir, no superiores al 5 %, no superiores al 4 %, no superiores al 3 %, no superiores al 2 %, no superiores al 1 %, o no superiores al 0,5 % de agregado en peso de proteína, y/o niveles de bajos a indetectables de fragmentación, es decir, 80 % o más alta, 85 % o más alta, 90 % o más alta, 95 % o más alta, 98 % o más alta, o 99 % o más alta, o 99,5 % o más alta del área de pico total en el (los) pico(s) que representa(n) anticuerpos manipulados con cisteína intactos.
En otras realizaciones, los anticuerpos manipulados con cisteína pueden producirse por un proceso escalable de la invención en el laboratorio de investigación que puede aumentarse de escala para producir las proteínas de la invención en los biorreactores a escala de producción (por ejemplo, pero no se limitan a, 75 l, 100 l, 150 l, 300 l, o 500 l). En algunas realizaciones, el proceso escalable de la invención produce poca o ninguna reducción en la eficiencia de producción en comparación con el proceso de producción realizado en el laboratorio de investigación. En otras realizaciones, el proceso escalable de la invención produce anticuerpos manipulados con cisteína a eficiencia de producción de al menos aproximadamente 10 mg/l, al menos aproximadamente 20 mg/l, al menos aproximadamente 30 mg/l, al menos aproximadamente 50 mg/l, al menos aproximadamente 75 mg/l, al menos aproximadamente 100 mg/l, al menos aproximadamente 125 mg/l, al menos aproximadamente 150 mg/l, al menos aproximadamente 175 mg/l, al menos aproximadamente 200 mg mg/l, al menos aproximadamente 250 mg/l, al menos aproximadamente 300 mg/l, o más alta.
