Abstract
Un derivado de heteroarilamida que tiene la fórmula (I) **(Ver fórmula)** y N-óxidos, estereoisómeros y sales farmacéuticamente aceptables de los mismos; en la que R' y R" se seleccionan cada uno de ellos independientemente del grupo que consiste en H y alquilo; z es un número entero de 0 a 3; X se selecciona del grupo que consiste en O, S, S(O), SO2, NR12, en el que R12 se selecciona del grupo que consiste en H, alquilo, y COR, en el que R es hidrógeno o alquilo; CH2 y CO; o cuando z es 0, entonces X puede ser N y forma junto con R11 un anillo heterocíclico seleccionado del grupo que consiste en morfolina, 1,2,4-triazol, imidazol e imidazol N-sustituido; y R11, cuando no forma un anillo con X, tal como se ha definido, se selecciona del grupo que consiste en alquilo, alquenilo, (per)haloalquilo, haloalquenilo, CN-alquilo y un arilo, heteroarilo, un anillo alifático o heteroalifático, de 3 - 7-miembros opcionalmente sustituido con 1 - 5 sustituyentes seleccionados del grupo que consiste en alquilo, alcoxi, hidroxi, halógeno, (per)haloalquilo, CN, NO2, COR, COOR, CONHR, NR2, NHCOCF3, NHCOR, NHCONHR, NHCOOR, OCONHR, NHSO2R, NHCSCH3, SR, SOR y SO2R, en las que R es tal como se ha definido anteriormente; RB es un anillo heteroaromático que tiene 6 miembros de anillo y que opcionalmente está sustituido en uno o más de los átomos de carbono, en el que 1 o 2 miembros del anillo son átomos N y los otros miembros del anillo son átomos de carbono, o RB es un grupo fenilo opcionalmente sustituido, seleccionándose el (los) sustituyente(s) en RB del grupo que consiste en alquilo, alcoxi, hidroxi, halógeno, (per)haloalquilo, CN, NO2, COR, COOR, CONHR, NR2, NHCOCF3, NHCOR, NHCONHR, NHCOOR, OCONHR, CONHR, en el que R es tal como se ha definido anteriormente; NHCSCH3, SR, SOR y SO2R, en el que R es tal como se ha definido; o, cuando RB es un grupo fenilo opcionalmente sustituido, entonces dos sustituyentes adyacentes también pueden formar con los átomos de carbono a los que están unidos un anillo alifático, heteroalifático o heteroaromático sustituido o sin sustituir; y RA es un sistema de anillo mono- o bicíclico heteroaromático que tiene de 6 a 10 miembros de anillo y que está opcionalmente sustituido en uno o más átomos de carbono del anillo, en el que 1 a 4 miembros del anillo son átomos N y los demás miembros del anillo son átomos de carbono, de manera que, cuando RB es un grupo fenilo opcionalmente sustituido, entonces el anillo unido al grupo NH contiene al menos un átomo N como miembro del anillo, o cuando RB es un anillo heteroaromático opcionalmente sustituido, entonces RA puede ser también un grupo fenilo opcionalmente sustituido, seleccionándose el (los) sustituyente(s) en RA del grupo que consiste en alquilo, alcoxi, halógeno, (per)haloalquilo, hidroxi, (CH2)nCHO, en el que n es un número de entero de 0 a 6; CN, NO2, COR, COOH y CONHR, en el que R es tal como se ha definido anteriormente, o cuando RA es un grupo fenilo opcionalmente sustituido, entonces dos sustituyentes adyacentes pueden formar también con los átomos de carbono a los que están unidos un anillo alifático, heteroalifático o heteroaromático sustituido o sin sustituir.

Asimismo son preferentes aquellos en los que R11 es alquilo, especialmente metilo, etilo, isopropilo, isopentilo o terc-butilo, más preferentemente metilo, etilo o isopropilo, siendo sobre todo preferente metilo o etilo, o fenilo opcionalmente sustituido con halo, especialmente cloro. Son preferentes además los compuestos de fórmula (I) en la que R11 es ciclopropilo.

Descripción general de las propiedades farmacológicas de los compuestos de la presente invención
Los derivados de arilamida de la presente invención presentan una alta actividad antagonista en RA. La actividad antagonista en RA se refiere a la potencia del compuesto para inhibir o competir con la actividad de ligandos para
5 RA naturales como dihidrotestosterona (DHT) y testosterona. La presente invención proporciona compuestos que tienen actividad antagonista en RA para inhibir y/o competir con la actividad de ligandos de RA no naturales, como andrógenos sintéticos o antiandrógenos utilizados como medicamentos (pero que pueden ejercer efectos secundarios perjudiciales).
Asimismo, la presente invención proporciona compuestos que demuestran tener una potente actividad antiandrógina
10 de forma dependiente de la dosis. Una de los principales inconvenientes de bicalutamida es el antagonismo de RA incompleto. En el caso de bicalutamida, el aumento de la concentración no proporciona ningún beneficio extra (véase Tabla 2). Es posible que se necesiten antiandrógenos más potentes que bicalutamida para tratar CaP en fase avanzada caracterizado por la elevación de los niveles de RA, por lo tanto, existe la necesidad de contar con antiandrógenos potentes que puedan compensar los elevados niveles de RA de forma dependiente de la dosis. La
15 presente invención proporciona compuestos que ejercen efectos agonistas mínimos en RA.
Los compuestos de la presente invención se pueden utilizar para tratar enfermedades relacionadas con RA, tales como HBP y CaP. Los compuestos se pueden utilizar asimismo para tratar CPRC. Por otra parte, los compuestos se pueden utilizar en combinación con otros tratamientos antiandrógenos.
Los compuestos de la presente invención no ganan actividad agonista en las mutaciones relacionadas con CPRC.
20 Se entiende por mutaciones relacionadas con CPRC todas las mutaciones que afectan al desarrollo, avance o gravedad de la enfermedad. La mutación relacionada con CPRC puede derivarse del enriquecimiento de células de cáncer de próstata inducido por la privación de andrógeno que albergan dicha mutación. Por ejemplo, se hace referencia a triptófano 741 para la mutación de leucina en cisteína y también treonina 877 para la mutación de alanina.
25 Los compuestos de la presente invención retienen sus actividades antagonistas cuando se elevan los niveles de RA.
