Abstract
Proteína LAG-3 recombinante LAG-3, o derivado de la misma que es un mutante o fragmento de LAG-3 que mantiene la capacidad de LAG-3 para unirse a moléculas MHC de clase II, que provoca una respuesta inmunitaria mediada por monocitos, y un agente de quimioterapia, para su uso en el tratamiento del cáncer mediante la administración de una pluralidad efectiva de dosis de la proteína LAG-3 recombinante o derivado de la misma, donde la proteína LAG-3 recombinante o derivado de la misma se debe administrar antes de, con, o posteriormente a la administración del agente de quimioterapia.
Description
Se predice que el régimen posológico óptimo para IMP321 será de entre 6 y 30 mg/inyección.
Estas dosis han demostrado ser seguras y proporcionar una exposición sistémica aceptable en base a los resultados de los datos farmacocinéticos obtenidos en pacientes con cáncer de células renales metastásico (Figura 5). Podría 5 obtenerse una concentración en sangre de IMP321 superior a 1 ng/ml durante al menos 24 horas después de la inyección s.c. en pacientes inyectados con dosis de IMP321 de más de 6 mg (Figura 5).
10 Los pacientes, que reciben como una primera línea de quimioterapia para el cáncer de páncreas avanzado (o pacientes no elegibles para la extirpación quirúrgica del tumor) 6 ciclos de gemcitabina estándar (1 gm/m2 administrada por vía i.v. durante 30 min) en los días 1, 8 y 15 de un ciclo de 28 días, reciben además de 6 a 30 mg de IMP321 s.c. en los días 2 y 16 de cada ciclo de 28 días. O bien, IMP321 se administra en los días 3 o 17.
15 Por consiguiente, cada paciente recibe un ciclo estándar de 6 meses de gemcitabina con 12 inyecciones s.c. de IMP321, administrándose cada inyección uno a dos días después de la administración de gemcitabina en un esquema cada dos semanas.
20 El número de monocitos se analiza mediante clasificación de células activadas por fluorescencia (FACS) como en el ejemplo 1.
