CONSISTE EN PONER EN UNA MEZCLA REACTIVA UN PRIMER LIGANTE CON UN SEGUNDO LIGANTE, QUE SE ENLAZA COVALENTEMENTE A UNA MARCA PROTEINICA ELEGIDA ENTRE PROTEINAS DERIVADAS DE COELEUTERATO, LUCIFERASA O APOFOTOPROTEINAS, PARA FORMAR UN COMPLEJO DE ENLACE ENTRE AMBOS LIGANTES; SEPARAR DICHO COMPLEJO DE LA MEZCLA DE REACCION; PONER EN CONTACTO UN REACTIVO DE DISPARO DE LUZ CON DICHO COMPLEJO; Y DETECTAR LA LUZ EMITIDA. CUANDO LA MARCA PROTEINICA ES UNA FOTOPROTEINA QUE CONTIENE LUCIFERINA O SIMILAR SE PONEN EN CONTACTO IONES CALCIO CON EL COMPLEJO; CUANDO ES UNA LUCIFERASA SE PONE EN CONTACTO UNA MOLECULA DE LUCIFERINA O SIMILAR; Y CUANDO ES UN APOFOTOPROTEINA, SE CONVIERTE EN UNA FOTOPROTEINA Y SE PONEN EN CONTACTO IONES CALCIO CON EL COMPLEJO.
