CONSISTE EN: A) PONER EN CONTACTO CON UN MEDIO REACCIONAL EL PRIMER LIGANTE CON UNA CANTIDAD DEL SEGUNDO LIGANTE QUE ES IGUAL AL LIGANTE QUE HA DE DETECTARSE, SIENDO EL SEGUNDO LIGANTE ENLAZADO COVALENTEMENTE CON UNA MARCA PROTEINICA ELEGIDA ENTRE FOTOPROTEINAS, LUCIFERASA Y APOFOTOPROTEINAS DERIVADAS DE COELENTERATO, Y CON LA MUESTRA CON LA QUE SE FORMA UN PRIMER COMPLEJO ENTRE EL PRIMER Y SEGUNDO LIGANTES, Y FORMAR UN SEGUNDO COMPLEJO ENTRE EL PRIMER LIGANTE Y EL LIGANTE QUE HA DE DETECTARSE; B) SEPARAR DE LA MEZCLA REACCIONAL AMBOS COMPLEJOS; C) PONER EN CONTACTO UN REACTIVO DISPARADOR DE LUZ CON AMBOS COMPLEJOS PARA EMITIR LUZ; Y D) DETECTAR LA LUZ EMITIDA POR LA MOLECULA DE LUCIFERINA.
