Claims (40)
1. Komplex,
dadurch gekennzeichnet,
daß er ein für Impfzwecke geeignetes Immunogen umfaßt, das an ein Trägermolekül gebunden ist, welches zur-spezifischen Bindung an die epitheliale Schleimhaut eines Wirbeltierwirts geeignet ist, worin sowohl die immunologische Aktivität des Immunogens als auch die Eignung des Trägers, spezifisch an das Schleimhautepithel zu binden, im wesentlichen erhalten bleibt, und das Trägermolekül ein zur Bindung an das Schleimhautepithel geeignetes Molekül ist, ausgewählt aus dem Ganzen, einem Teil, einem Analogon, einem Homologen, einem Derivat oder einer Kombination hiervon eines bakteriellen Adhesins, eines viralen Hämagglutinins, eines Toxins oder einer Bindungsuntereinheit eines Toxins oder eines Lectins, wobei jedoch Immunogen-Trägerkomplexe ausgenommen sind, worin das Immunogen natürlicherweise an das Trägermolekül gebunden ist.
2. Komplex nach Anspruch 1, worin das Immunogen ausgewählt ist aus einem Antigen oder Hapten, oder dem Ganzen, Teil(en), einem Analogon, einem Homologen, einem Derivat oder einer Kombination hiervon eines Hormons oder eines therapeutischen Mittels.
3. Komplex nach Anspruch 2, wobei das Immunogen das Ganze, ein Teil, ein Analogon, ein Homologes, ein Derivat oder eine Kombination hiervon eines Hormons ist, ausgewählt aus FSH, HGH, LHRH oder Inhibin.
4. Komplex nach Anspruch 1 oder 2, worin das Immunogen ein Allergen ist.
5. Komplex nach Anspruch 4, worin das Allergen von Gräserpollen, Unkrautpollen, Baumpollen, Pflanzenpollen, Katzenhaar, Hundehaar, Schweinehaar oder anderen Epithelien oder Hausstaub, Weizenspreu oder Kapok stammt.
6. Komplex nach Anspruch 1 oder 2, worin das Immunogen ein Oberflächenprotein ist, welches aus einem Influenza, Masern, Rubella, Pocken, Gelbfieber, Diphtherie, Tetanus, Cholera, Pest, Typhus oder BCG verursachenden Mittel, Haemophilus influenza, Neisseria catarrhalis, Klebsiella pneumoniae, einem Pneumococcus oder einem Streptococcus stammt; oder es aus einem Pilus stammt von E. coli, N. gonorrheae, N. meningitidis, N. catarrhalis, einer Yersinia-Art, Pseudomonas aeruginosa, einer Pseudomonas-Art, Moraxella bovis, Bacteroides nodosus, einer Staphylokokken-Art, einer Streptokokken-Art oder einer Bordetella-Art.
7. Komplex nach Anspruch 1 oder 2, worin das Immunogen ein Oberflächenpolysaccharid ist, das aus einem Diphtherie, Tetanus, Cholera, Pest, Typhus oder BCG verursachenden Mittel, Haemophilus influenza, Neisseria catarrhalis, Klebsiella pneumoniae, einem Pneumococcus oder einem Streptococcus stammt.
8. Komplex nach Anspruch 1 oder 2, worin das Immunogen ein sekretorisches Produkt ist, das aus einem Diphtherie, Tetanus, Cholera, Pest, Typhus, oder BCG verursachenden Mittel, Haemophilus influenza, Neisseria catarrhalis, Klebsiella pneumoniae, einem Pneumococcus oder einem Streptococcus stammt.
9. Komplex nach einem der Ansprüche 6 bis 8, worin das Oberflächenprotein, das Oberflächenpolysaccharid oder das sekretorische Produkt aus Streptococcus mutans stammt.
10. Komplex nach Anspruch 1, worin das Trägermolekül ein hitzelabiles Toxin des enterotoxigenen E. coli ist.
11. Komplex nach Anspruch 1, worin das bakterielle Adhesin der K99- oder 987P-Pilus von E. coli ist.
12. Komplex nach Anspruch 1, worin das bakterielle Adhesin CFAI, CFAII, K88 oder F41 ist.
13. Komplex nach Anspruch 1, worin das Lectin Concanavalin A, Pokeweed Mitogen oder das Lectin aus Lens culinaris, Helix pomatia, Glycine max, Arachis hypogea oder Ulex europeus ist.
14. Komplex nach Anspruch 1, worin das Lectin Abrin-, Asparagus-Erbsen-, Saubohnen-, Kamelfußbaum-, Caster-Bohnen-, Fava-Bohnen-, marine Grünalgen-, haarige Wicken-, Pferdekichererbsen- (horse gram), Hufeisenkrabben-, Jack-Bohnen-, Japanische Glyzinen-, Jequirity-Bohnen-, Schottischer Goldregen-, Lima-Bohnen-, Limulin-, Lotus-, Europäische Mistel-, Mungbohnen-, Osage-Orange-, Pagodenbaum-, Gartenbohnen-, Kartoffel-, Rote Kidney-Bohnen-, marine Rotalgen-, Sibirischer Erbsenbaum-, eßbare Schnecken-, Gartenschnecken-, Spindelbaum-, süße Erbsen-, Tomaten-, Weizenkeim- oder Wingederbsen-Lectin ist.
15. Komplex nach Anspruch 1, worin das virale Hämagglutinin ein Hämagglutinin ist, das aus Influenza-, Masern-, Rubella-, Pocken- oder Gelbfiebervirus stammt.
16. Komplex nach einem der Ansprüche 1 bis 3, worin das Immunogen das Ganze, ein Teil, ein Analogon, ein Homologes, ein Derivat, oder eine Kombination hiervon des Luteinisierungshormon-freisetzenden Hormons und das Trägermolekül das Ganze, ein Teil, ein Analogon, ein Homologes, ein Derivat oder eine Kombinatin hiervon von LTB ist.
17. Verfahren zur Herstellung eines Komplexes nach einem der Ansprüche 1 bis 16, wobei das Verfahren die folgenden Schritte aufweist:
(a) Umsetzung des Immunogens mit dem Trägermolekül, um den Komplex zu bilden;
(b) chemische Modifikation des Immunogens, um wenigstens eine funktionelle Gruppe bereitzustellen, die in der Lage ist, eine chemische Bindung auszubilden, und Umsetzung des Immunogens und des Trägermoleküls, um den Komplex zu bilden;
(c) chemische Modifikation des Trägermoleküls, um wenigstens eine funktionelle Gruppe bereitzustellen, die in der Lage ist, eine chemische Bindung auszubilden, und Umsetzung des Immunogens und des Trägermoleküls, um den Komplex auszubilden;
(d) chemische Modifikation des Immunogens und des Trägermoleküls, um funktionelle Gruppen bereitzustellen, die in der Lage sind, eine chemische Bindung auszubilden, und Umsetzung des Immunogens und des Trägermoleküls, um den Komplex auszubilden;
(e) Umsetzung des Immunogens mit wenigstens einem Bindungsmittel, und Umsetzung des Immunogens und des Trägermoleküls, um den Komplex auszubilden;
(f) Umsetzung des Trägermoleküls mit wenigstens einem Bindungsmittel, und Umsetzung des Immunogens und des Trägermoleküls, um den Komplex auszubilden;
(g) Umsetzung des Immunogens und des Trägermoleküls mit wenigstens einem Bindungsmittel, und Umsetzung des Immunogens und des Trägermoleküls, um den Komplex auszubilden; oder
(h) eine Kombination aus einem der oben genannten Verfahrensschritte.
18. Verfahren zur Herstellung eines Komplexes nach einem der Ansprüche 1 bis 3, wobei das Verfahren die nachfolgenden Verfahrensschritte aufweist:
Bereitstellen eines rekombinanten DNA-Moleküls, das eine erste DNA-Sequenz aufweist, die bei Expression für die Aminosäuresequenz des Immunogens codiert, eine zweite DNA-Sequenz, die bei Expression für die Aminosäuresequenz des Trägermoleküls codiert und eine Vektor-DNA; Transformation eines Wirts mit dem rekombinanten DNA-Molekül, so daß der Wirt in der Lage ist, ein Hybrid-Proteinprodukt zu exprimieren, das den Komplex umfaßt; Kultivierung des Wirts, um die Expression zu erhalten und Sammeln des Hybrid-Proteinprodukts.
19. Verfahren zur Herstellung eines Komplexes nach einem der Anprüche 1 bis 16, wobei das Verfahren die nachfolgenden Verfahrensschritte aufweist:
(a) chemische Synthese des Immunogens und/oder des Trägermoleküls und Ausbildung des Komplexes durch Reaktionen nach dem Verfahren des Anspruchs 17; oder
(b) Synthese eines Hybridpeptids, das Aminosäuresequenzen des Immunogens und des Trägermoleküls aufweist.
20. Verfahren nach Anspruch 19, worin das Peptid durch Festphasen-, enzymatische oder manuelle Peptidsynthese hergestellt wird.
21. Verfahren nach Anspruch 20, worin das synthetisierte Immunogen oder Trägermolekül an das Trägermolekül oder Immunogen gekoppelt ist, während es an ein Harz eines Festphasen-Peptidsyntheseapparats gebunden ist.
22. Verfahren nach einem der Ansprüche 19 bis 21, worin das Immunogen das Ganze, ein Teil, ein Analogon, ein Homologes, ein Derivat oder eine Kombination hiervon des Luteinisierungshormon-freisetzenden Hormons (LHRH) ist.
23. Verfahren nach Anspruch 17, worin das Trägermolekül LTB und das Verbindungsmittel Isoglutaraldehyd ist.
24. Komplex nach einem der Ansprüche 1 bis 3, hergestellt durch ein Verfahren nach einem der Ansprüche 17 bis 21.
25. Expressionsprodukt eines transformierten Wirts, dessen Produkt einen Komplex nach einem der Ansprüche 1 bis 3 umfaßt.
26. Wirt, transformiert mit einem rekombinanten DNA-Molekül, das eine erste DNA-Sequenz umfaßt, die bei Expression für die Aminosäuresequenz eines für Impfzwecke geeigneten Immunogens codiert und eine zweite DNA-Sequenz, die bei Expression für die Aminosäuresequenz eines Trägermoleküls codiert, das zur Bindung an die epitheliale Schleimhaut in der Lage ist, wobei das Trägermolekül ein zur Bindung an die epitheliale Schleimhaut geeignetes Molekül ist, ausgewählt aus dem Ganzen, einem Teil, einem Analogon, einem Homologen, einem Derivat oder einer Kombination hiervon eines bakteriellen Adhesins, eines viralen Hämagglutinins, eines Toxins oder einer Bindungsuntereinheit eines Toxins, oder eines Lectins, worin das Immunogen und das Trägermolekül ein Fusionspolypeptid bilden, ausgenommen ein DNA-Molekül, das für einen Immunogen-Trägerkomplex codiert, worin das Immunogen an das Trägermolekül natürlicherweise gebunden ist.
27. Transformierter Wirt nach Anspruch 26, worin der Wirt ein Gram-negatives oder ein Gram-positives Bakterium, eine Hefe, ein Pilz oder eine höhere eukaryontische Zelle ist.
28. Transformierter Wirt nach Anspruch 26 oder 27, worin der Wirt E. coli ist.
29. Rekombinantes DNA-Molekül, das eine DNA-Sequenz aufweist, die bei Expression für ein Fusionspolypeptid codiert, das die Aminosäuresequenz eines für Impfzwecke geeigneten Immunogens umfaßt, und die Aminosäuresequenz eines zur Bindung an die epitheliale Schleimhaut geeigneten Trägermoleküls, wobei das Trägermolekül ein zur Bindung an die epitheliale Schleimhaut geeignetes Molekül ist, ausgewählt aus dem Ganzen, einem Teil, einem Analogon, einem Homologen, einem Derivat oder einer Kombination hiervon, eines bakteriellen Adhesins, eines viralen Hämagglutinins, eines Toxins oder einer Bindungsuntereinheit eines Toxins oder eines Lectins, ausgenommen ein DNA-Molekül, das für einen Immunogen-Trägerkomplex codiert, worin das Immunogen an das Trägermolekül natürlicherweise gebunden ist.
30. Rekombinantes DNA-Molekül nach Anspruch 29, das ebenfalls eine Vektor-DNA aufweist.
31. Rekombinantes DNA-Molekül nach Anspruch 30, worin die Vektor-DNA aus Plasmid-, Virus-, Bakteriophagen- oder Cosmid-DNA ausgewählt ist.
32. Rekombinantes DNA-Molekül nach Anspruch 29, worin die DNA-Sequenz als eine codierende Sequenz für ein Antigen oder ein Hapten wirkt, das mit der Aminosäuresequenz des Trägermoleküls fusioniert ist.
33. Rekombinantes DNA-Molekül nach Anspruch 29, worin die DNA-Sequenz als codierende Sequenz des Ganzen, eines Teils, eines Analogons, eines Homologen, eines Derivats von LHRH wirkt, fusioniert mit der Aminosäuresequenz des Trägermoleküls.
34. Arzneimittel, welches ein für Impfzwecke geeignetes Immunogen umfaßt, das an ein Trägermolekül gebunden ist, welches zur spezifischen Bindung an die epitheliale Schleimhaut eines Wirbeltierwirts in der Lage ist, worin sowohl die immunologische Aktivität des Immunogens als auch die Fähigkeit des Trägers, spezifisch an das Schleimhautepithel zu binden, im wesentlichen beibehalten wird, und das Trägermolekül ein zur Bindung an die epitheliale Schleimhaut geeignetes Molekül ist, ausgewählt aus dem Ganzen, einem Teil, einem Analogon, einem Homologen, einem Derivat oder einer Kombination hiervon eines bakteriellen Adhesins, eines viralen Hämagglutinins, eines Toxins oder einer Bindungsuntereinheit eines Toxins, oder eines Lectins, zusammen mit einem pharmazeutisch annehmbaren Träger oder einem Streckmittel, ausgenommen Immunogen-Trägerkomplexe, worin das Immunogen an das Trägermolekül natürlicherweise gebunden vorliegt.
35. Arzneimittel nach Anspruch 34, das zur oralen Verabreichung ausgelegt ist.
36. Arzneimittel nach Anspruch 34, das zur nasalen Administration ausgelegt ist.
37. Arzneimittel nach einem der Ansprüche 34 bis 36, worin das Arzneimittel in Kapsel-, Tabletten-, Retard- , Elixier-, Gel-, Pasten- oder Nasensprayform vorliegt.
38. Arzneimittel nach einem der Ansprüche 34 bis 37, das zusätzlich ein Nahrungsmolekül aufweist, um die Größenordnung und/oder die Art der Immunantwort auf das Immunogen des Arzneimittels selektiv zu modulieren.
39. Arzneimittel nach Anspruch 38, wobei das Nahrungsmolekül aus Aminosäuren, Vitaminen, Monosacchariden und Oligosacchariden ausgewählt wird.
40. Arzneimittel nach Anspruch 38 oder Anspruch 39, worin das Nahrungsmolekül ausgewählt wird aus Vitamin A, Vitamin B1, Vitamin B2, Vitamin B6, Vitamin B12, Vitamin C, Vitamin D, Vitamin E, Fructose, Lactose, Mannose, Melibiose, Sorbitol oder Xylose.
