Claims (1)
- 【特許請求の範囲】 1.カチオンポリマーおよびポリアニオンを含む、細胞に遺伝子を輸送するた めの3μmよりも小さい固形のナノスフィアであって、ポリアニオンが核酸を含 み、カチオンポリマーがポリリシンではないナノスフィア。 2.カチオンポリマーが糖類である、請求項1記載のナノスフィア。 3.カチオンポリマーがキトサンである、請求項2記載の固形のナノスフィア。 4.ナノスフィアが、5%(w/w)よりも多量の核酸を含む、請求項1記載のナノ スフィア。 5.ナノスフィアが、20%(w/w)よりも多量の核酸を含む、請求項1記載のナ ノスフィア。 6.核酸が2〜20kbの遺伝子を含む、請求項1記載のナノスフィア。 7.特異的な標的細胞に遺伝子を輸送するために、固形のナノスフィアを形成 する方法であって、核酸とポリリシンではないカチオンポリマーとのコアセルベ ーションによって3μmよりも小さいナノスフィアを形成する段階を含む方法。 8.カチオンポリマーが糖類である、請求項7記載の方法。 9.硫酸ナトリウム存在下でコアセルベーションを行う、請求項8記載の方法。 10.カチオンポリマーがキトサンである、請求項8記載の方法。 11.コアセルベーションの段階において、約0.01〜7%の濃度でカチオンポリ マーが存在する、請求項8記載の方法。 12.コアセルベーションの段階において、核酸が1ng/mlから500μg/mlの濃 度で存在する、請求項8記載の方法。 13.コアセルベーションの段階において、硫酸ナトリウムの濃度が、約5〜100 mMの間である、請求項8記載の方法。 14.下記の段階を含む、遺伝子を細胞へ導入するための方法: (a)トランスフェクトする細胞を(b)カチオンポリマー分子と核酸分子とを含む 3μmよりも小さい固形ナノスフィアと共にインキュベートする段階であって、 それにより該細胞が該核酸分子によりトランスフェクトされ、カチオンポリマー 分子がポリリシンではない段階。 15.カチオンポリマー分子が糖類である、請求項14記載の方法。 16.カチオンポリマー分子がキトサンである、請求項15記載の方法。 17.核酸がDNAである、請求項15記載の方法。 18.核酸がRNAである、請求項15記載の方法。 19.細胞が培養されている、請求項14記載の方法。 20.細胞が動物の体内にある、請求項14記載の方法。
