トをフィッシャー344(F344 RT11v1)またはブラウンノルウエー(B

preglomerular vessels)は、損傷なし(0)、軽度(1)そして中程度(2)
の損傷程度を示すように評価され、そして皮質、外部および内部髄質を含む全腎
臓切片において評価された。程度に特定の血管損傷指標は、全腎臓の切片におい
て遭遇した損傷の各程度を有する血管の百分率として規定した。程度1の血管の
数を1倍し、程度2のそれは2の因子を掛けて、程度3のそれは3の因子を掛け
ることにより、全血管損傷のスコアは全血管の合計として血管損傷の全程度を用

リメラーゼで転写することにより、アンチセンスプローブを生じさせ、センスプ
ローブを生じさせるため、該プラスミドをEcoRIで切断してT7 RNAポ
リメラーゼで転写した。脱パラフィン後に腎臓切片を20μg/mlのプロテイ

同時測定を可能にした。20μgの全RNAを各測定に使用した。プロテクトさ

植は、外科手術後7日までに特徴的な単球細胞浸潤物および組織の損傷をもたら
した。組織学的試験は、前糸球体大動脈および尿細管間質の脈間内膜の局所的な
単核細胞の浸潤を示した。この間質の単核浸潤物の主要な成分は、単球/マクロ
ファージ細胞からなった。大動脈、細動脈、尿細管に対する損傷の程度および間
質の単核細胞の浸潤の範囲は、半定量形態計測に基づく前記の手法を使用して、
非存在(0)から、軽度(1)、中度(2)および重度(3)までのスケールで
見積もられた(材料と方法を参照)。

-1β(8.4倍)およびTNFα(4.6倍)において観察された。この時間

物は、未処置動物に比して、IL-1α(25%)、IL-1β(48%)、TN
Fβ(34%)、TNFα(24%)およびIFNγ(24%)の低下した平均
発現を示した。 ブラウンノルウエーラット腎臓のルイスラットへの移植:低投薬量シクロスポリ
ンA(CyA)に関連したMet-RANTESの効果 我々は、次に、Met-RANTESが腎臓移植拒絶において低い投薬量のC
yA処置を補うことが可能か否かを測定するために実験を拡張した。この手法に
関して、我々は、より活発な拒絶の症状の発現を生じる腎臓移植モデル、即ちブ
ラウンノルウエー腎臓のルイスラットへの移植を選択した。両側の腎臓の切除を
移植時に実施した。使用したCyAのレベル、2.5mg/kg体重を1日あた
り皮下投与したがこのモデルにおいては腎臓拒絶を有意にブロックしなかったこ
とが以前に示された(Grone未公開の結果(Stojanovic T.et al.,1996))。
最後に、あらゆる相乗作用をよりよく検出するために、減少させた投薬量のMe
t-RANTES、1日あたりの動物あたり50μgをこれらの実験において使
用した。表3に要約された結果は、低投薬量のCyAでのみ処置された動物に比
しての、Met-RANTES/低投薬量CyAで処置された動物において観察
された血管および尿細管の損傷の統計学上有意な減少を示す。さらに、間質領域
の単核細胞の浸潤の有意な減少が観察された。これらの組織学的な観察は、未処
置の対照に比してMet-RANTES処置動物において血清クレアチンが減少
した機能的測定により確認された(0.98+/−0.12対1.42+0.1
7mg%(n=3))。 活性化された微小血管内皮上の直接のRANTES結合と接着分子発現 血管腔空間への単球の浸潤の減少が両移植モデルにおいてMet-RANTE
S処置の顕著な特徴を表したので、我々は、この効果に関する可能性のある機構
を研究することを計画した。腎臓の移植拒絶におけるRANTESの役割のモデ
ルにおいては、活性化された血小板から放出されるかまたは局所的に炎症してい
る組織から分泌されるRANTES蛋白質が、単球の補充を支持するかもしれな
い炎症した内皮の表面上に蓄積することが予測された(Nelson P.J.et al.,19
98,Pattison J.et al.,1994,Wiedermann C.J.et al.,1993)。微小血管内
皮へのRANTESの直接の結合を研究するため、我々は、組織切片におけるR
ANTESの内皮表面結合を検出するために以前に用いたアッセイの修飾法を使
用して、活性化された皮膚微小血管内皮(DMVEC)がRANTES蛋白質を
封鎖する能力を試験した(Pattison J.et al.,1994,Wiedermann C.J.et al.
,1993)。HRPをコンジュゲートした抗ヒトRANTESモノクローナル抗体
VL1を過剰なRANTESとインキュベートし、そしてその結果の複合体を、
96ウエル平底プレート内で成長させた、休眠しているかまたはIL-1β活性
化された微小血管内皮に加えた。ELISA-様フォーマットを使用して、DM
VECがmAbとは異なり抗原-mAb複合体に結合する能力を測定した。微小
血管内皮は予めの刺激無しにいくらかのRANTES蛋白質を結合できたが、該
結合は前炎症性サイトカインIL-1αによる予めの刺激後に大きく増加した(
図1)。未刺激の内皮または活性化された内皮への未複合mAbのバックグラウ
ンド染色は無視できた。
子、即ちE-セレクチンおよびIgGスーパーファミリーメンバーICAM-1お
よびVCAM-1の発現を、以前に確立されたフローサイトメトリー方法を使用
してDMVECについて測定した(Weber C.et al.,1995)。該分析は、休眠
するDMVECが構成的な表面レベルのICAM-1を発現したが、しかしなが
ら、VCAM-1またはE-セレクチンはほとんど検出されなかったことを明らか
にした(表4)。IL-1による12時間のDMVEC活性化は、ICAM-1の
アップ制御並びにVCAM-1およびE-セレクチンの表面発現の著しい誘導をも
たらした(表4)。

おいて、我々は、RANTES受容体のの妨害が微小血管内皮上での単球の強固
な拘束と血管外遊出を阻害できるか否かを研究した。このために、我々は、成熟
した単球の接着性とインテグリンレパートリーを示し、そしてCCR1を含むい
くつかのケモカインを発現する単球MonoMac 6細胞を使用した(Erl W.
et al.,1995)。DMVECを、刺激しないままに放置したかまたはIL-1β
(5ng/ml)で12時間活性化したペトリ皿上にて芝生状まで成長させた。
微小血管内皮を次に平行壁フローチャンバー中で試験したが、MonoMac
6細胞を該チャンバー中で1.5dyn/cm2の剪断速度にて5分間灌流した
。そのような生理学的フロー条件下で、MonoMac 6細胞は短期間の回転
を受け、そして細胞の一部の接着は剪断耐性拘束に容易に変換できる。蓄積の5
分後、内皮への強固な接着を受けたMonoMac 6細胞の数を測定した(図
2(a))。
胞のRANTES蛋白質への予めの暴露は顕著な効果を示さなかった。IL-1
βによる微小血管内皮の予めの刺激は単球の剪断耐性接着における増加をもたら
した。mAbによる阻害は、このプロセスが、活性化された内皮上に発現される
ICAM-1およびVCAM-1とそれぞれ相互作用する単球α4およびβ2イン
テグリンにより媒介されるとの以前の発見を確証した(Kukerti S.et al.,199
7,Luscinskas F.W.et al.,1994)。直接結合アッセイにおけるRANTES
の固定化と一致して、IL-1β活性化微小血管内皮のRANTES蛋白質への
予めの暴露は、5分以内に単球の強固な拘束と蓄積を顕著に増強した(図2a)
。注目すべきは、さまざまな濃度(0.01-1μg/ml)のMet-RANT
ESと単球の予めのインキュベーションはIL-1β活性化DMVEC上の単球
のRANTES媒介剪断耐性接着を完全にブロックし(図2a,およびデータ示
さず)、平行して、強固な拘束の逆の測定として使用できる活性化微小血管内皮
上で回転する単球の画分はRANTESへの予めの暴露後に減少したが、Met
-RANTESにより回復したことから、活性化された内皮との最初の相互作用
の数は影響されなかった。強固な拘束の後、単球の画分は形態の変化または拡散
を受け、そしていくつかは内皮細胞の間または下で結局移動した。しかしながら
、RANTESまたはMet-RANTESは拡散または移動を変化させなかっ
た(図2b)ことから、他のシグナルの関与が推論される。即ち、これらの結果
は、炎症を起こした微小血管内皮に対する単球の拘束をブロックすることにより
腎臓移植拒絶の間にMet-RANTESが単球の補充を減じるのかもしれない
ことを示す。 表

プレート上に生育させ、そして修飾ELISA法を用いてRANTESを測定し
た。
6細胞の安定した拘束、拡散または移動に対するMet-RANTESの効果。
ペトリ皿上に芝生状に生育させたDMVECをIL-2(5ng/ml)で12
時間刺激するか、または左は無処理にし(対照)、そしてRANTES(10n
g/ml)存在または不在下で30分間プレインキュベートした。MonoMa
c 6細胞はMet-RANTES(1μg/ml)の存在または不在下で30分
間前処理して、1.5dyn/cm2の一定の流速にて灌流した。(a)DMV
ECへのフィルムの接着は5分間ののちに複数のフィールドにおいて安定に接着
した単球の数を計数することにより測定し、そして細胞/mm2として表現され
た。(b)拡散または移動を受けた単球は複数のハイパワーフィールドにおいて
5分後に計数し、そして初期に安定に接着した細胞の百分率で表現された。デー
タは、3つの個別の実験の+/−SDを表す。(注:結果は0.01から1μg
/mlのMet-RANTESの範囲にわたって再現性があった。)

成類似体の間から選択される、請求項1ないし5の何れか1項記載の使用。 - 【請求項7】 シクロスポリンがシクロスポリンAである、請求項1ないし6

いし7のいずれか1項記載の使用。 - 【請求項9】 移植された器官、組織または細胞の拒絶を治療または予防する
ための、ケモカイン受容体アンタゴニストおよびシクロスポリンを含む薬剤組成
物。 - 【請求項10】 移植された器官、組織または細胞の拒絶を治療または予防す
る際に、その活性成分を、同時に、別々にまたは連続して使用するための、一つ
または複数の薬学上受容可能な賦形剤の存在下でケモカイン受容体アンタゴニス
