本明細書で述べる場合、スブチリシン変異体は本技術分野で知られているヒュージョンPCRにより合成された(米国特許出願公開番号第2006/0252155参照)。表1-1はフージョンPCRで使用されるプライマーの配列を提供する。
バチルス・スブチリスにおけるスブチリシン変異体の生成
スブチリシン変異体は10mlのTSB培地(トリプトンと大豆ベースの液体培地)で37℃で一晩B・スブチリス(B.subtilis)形質転換体を培養して製造された。250μlの一夜培養液が100mlの振とうフラスコ中の25mlのMOPSベースの調製培地(defined medium)に加えられ、37℃で68時間培養された。この調製培地は、ほぼ本技術分野で知られている(Neidhartら、J.Bacteriol, 119:736-747,1974)ようにして調製されたが、NH4Cl、FeSO4及びCaCl2がこの基礎培地には加えられず、3mMKHPO4が使用され、この基礎培地には60mM尿素、75 g/Lグルコース、及び1%のソイトンが補われた。また、微量栄養素が、1リットル中に400mgのFeSO4・7H2O、100mg MnSO4・H2O、100mgZnSO4・7H2O、50mgCuCl2・2H2O、100mg CoCl2・6H2O、100mgNaMoO4・2H2O、100mgNa2B4O7・10H2O、10mlの1M CaCl2及び10mlの0.5M酒石酸ナトリウムを含有する原液X100として調製された。目的のプロテアーゼ(つまり、プロテアーゼ変異体)が培地から単離された。
スブチリシン変異体の純度を決定する分析方法
本実施例では、B・スブチリス培地から得られる組換えスブチリシンの純度を決定する方法が述べられる。このプロテアーゼは、ゲルクロマトグラフィーと高性能液体クロマトグラフィー(HPLC)のそれぞれによって単一のバンド又はピークが見られたときは純粋であるとみなされる。
スブチリシン濃度の決定
本実施例では、スブチリシン濃度を決定するために使用される方法が述べられる。いくつかの実験では、測定は計算した消光係数(extinction coefficient)(□)を用いて280nmで行われ、下記のように、精製したプロテアーゼ溶液のタンパク質濃度を決定するため活性部位の滴定が使用された。
PB92プロテアーゼについては、モル消光係数は、26,100 M-1cm-1(3Trp+7Tyr残基)であり、280nmで測定したε1%=9.7(Mr=26,729Da)に相当する。トリプトファン及び/又はチロシンの数が変わっている変異体に関しては、それに応じて補正される。
洗浄性能試験
この実施例では、スブチリシン変異体PX3と市販の皿洗浄及び洗濯洗剤に使用されているGCI-P038対照スブチリシンの食器洗い及び布地洗浄性能の評価に適した方法が述べられる。
本実施例では、スブチリシン変異体PX3と市販の食器洗浄洗剤中のGCI-P038の食器洗浄性能を測定するため使用される方法が述べられる。
これらの実験で使用されるステンレス鋼のシート(10x15 cm;片側がブラシで磨かれている)が実験用食器洗い機中で、高アルカリ性の市販の洗剤(例. ECOLAB(登録商標)洗剤;Henkel)で95℃で徹底的に洗浄を受け、清浄で油のないシートが得られた。これらのシートは最初に使用する前には、まくれが除去された。このシートは卵の黄身で汚れが付着される前、加熱庫で80℃で30分間乾燥された。磨かれた表面は汚れが付着する前は何にも触れなかった。また、この表面に水による汚れ又はほこりも付けさせなかった。冷却されたシートは、汚れを付ける前に秤量された。

スブチリシン変異体の安定性と洗浄性能
これらの実施例では、PX3の安定性と洗浄性能を評価する方法が述べられる。
セリンプロテアーゼ変異体の熱安定性はプロテアーゼ変異体を68℃で1時間保温後AAPF分析を用いてプロテアーゼ活性を評価することにより決定された。分析のこの条件下では、対照プロテアーゼの残存活性(例.野生型GG36 = GCI-P036)は、約50%であった。使用した装置は:F-bottom MTPs(Coaster No.9017)、Biomek FX 及び/又はBiomek FXp Robot(Beckman Coulter)、Spectramax Pus 384 MTP Reader (Molecular Devices), iEMS恒温器/振とう器(1mm 振幅)(Thermo/Labsystems)、密封テープ(Nunc No. 236366)、及びアイスバスである。グリシン緩衝液が、960mlの水に3.75gのグリシン(Merck No. 1.04201.1000)を溶解して調製された。1mlの5%TWEEN(登録商標)-80(Sigma No.P-8074)と10mlの1000mM CaCl2(Merck No. 1.02382.1000)原液(200mlに29.4 gが溶解された)がこの溶液に加えられた。pHが4N NaOHで10.5に調整され、容量が1000mlとされた。グリシン、CaCl2及びTWEEN(登録商標)-80の最終濃度は、それぞれ50mM、10mM及び0.005%である。恒温槽は68℃(恒温反応)と25℃(AAPF分析)に設定された。90μlと190μlのグリシン緩衝液が、空の希釈プレートと恒温反応用プレートのそれぞれに加えられた。10μlの上澄み液がこの希釈プレートへ加えられ、続いてこの希釈プレートから10μlが恒温反応用プレートへ加えられた。次いで恒温反応用プレートから10μlの混合物が、suc-AAPF-pNA基質を含む予熱されたプレートへ加えられた。このsuc-AAPF-pNAプレートが410nmでMTP Readerで測定された(t=0 測定)。恒温反応プレートはテープでカバーされ、68℃、400rpmで1時間恒温反応を受けた。この恒温反応の終了後、プレートは恒温器から取り出され、少なくとも5分間、氷上で冷却された。恒温反応用プレートからの10μlの混合物がsuc-AAPF-pNA基質を含むプレートに移され、プレートは410nm(t=60 測定)で測定された。パーセント残存活性は、

焼いた卵黄身の微小布見本分析
スブチリシン変異体の汚れ除去性能は、マイクロタイタープレート(MTP)で市販の洗剤中で決定された。対照スブチリシンのサンプルとスブチリシン変異体のサンプルが、37℃/300rpm/90% 相対湿度で3日間MTPプレート中で培養された液体培地のろ液から得られた。使用した装置は:96孔のポリスチレンプレート(Costar No. 9017 中間的結合性(medium binding)平底)、Biomek FX 及び/又はBiomek FXp(Beckman Coulter)、Spectramax Plus 384(Molecular Devices)、1mm の振幅のiEMS 恒温反応器/振とう器(Thermo Electron Corporation)及び密封テープ(Nunc No. 236366)を含む。使用した試薬は:5mM HEPES、pH8.0または5mM MOPS、pH7緩衝液、中間的な硬度で3:1 Ca:Mg(CaCl2:MgCl2・6H2O)の水(1ガロン当たり15000グレイン(gpg)の原液を6gpgに希釈)、1プレート当たり2枚のBMI(牛乳/血液/インク)微小布見本。CFTで処理されたEMPA-116BMI綿製微小布見本、1孔当たり予洗浄されパンチで打ち抜かれた2枚の微小布見本、及びプロテアーゼ活性がないことが確認されている、熱で不活性化された既製の洗剤TIDE(登録商標)2Xを含む。この分析ではこのプロテアーゼは基質を加水分解し、基質から色素と不溶な粒子を遊離する。
得られた吸光度は、白試験の値(酵素を含まない)に対して補正され、加水分解活性の尺度を与える。各サンプル(変異体)について性能指数(PI)が計算された。この性能指数は変異体の性能(実測値)と対照値(理論値)を同一のタンパク質濃度で比較する。加えて、理論値は、標準酵素のラングミュア式のパラメーターを用いて、計算できる。1より大きい性能指数(PI>1)は標準(例.野生型)と比べてより良い変異体を示し、PI、1(PI=1)は標準と同じ性能を示す変異体を示し、1より低いPI(PI<1)は標準より性能の劣った変異体を示す。従って、PIはある状況において使用に望ましくない変異体と、優れた変異体を示す。
液体洗濯洗剤組成物
この実施例では、種々の処方の液体洗濯洗剤組成物が提供される。本発明の以下の液体洗濯洗剤組成物は以下のようにして調製される。これらの各調合において、本明細書に記載された少なくとも1個のプロテアーゼ変異体は約0.0001から約10重量パーセントの濃度で含まれる。いくつかの他の実施態様では、その必要性に基づき、調合者により決定される、他の濃度が使用される。

Claims (15)
- 配列番号第5で表されたアミノ酸配列を含むスブツリシン変異体。
- 請求項1のスブチリシン変異体を含む組成物。

- 洗濯用洗剤である請求項3の洗浄組成物
- 食器洗浄用洗剤である請求項3の洗浄組成物。
- 自動食器洗浄機用洗剤である請求項5の食器洗浄用洗剤。
- 液体洗剤である請求項3の洗浄組成物。
- ゲル、錠剤、粉末または顆粒洗剤である請求項3の洗浄組成物。
- リン酸塩を含有しない請求項3の洗浄組成物。
- さらに少なくとも1個の漂白剤を含有する請求項3−9のいずれかの請求項の洗浄組成物。
