【発明の詳細な説明】 核酸プローブアッセイ法および組成物 技術分野 本発明は核酸プローブアッセイ法およびそれに用いる組成物に関する。 より詳細には本発明は分析される試料中の標的核酸系被分析体を、固形支持体に 結合した第110−ブにハイブリダイゼーションすることにより溶液から捕獲す る方法に関する。これらの方法にサンドイッチアッセイ法が含まれる。その場合 、標的は第2プローブにより検出され、これは検出のため標識されており、第1 プローブにハイブリダイズしたセグメント以外の標的被分析体セグメントにハイ ブリダイズする。 本発明が目的とするアッセイ法においては、固形支持体に結合したプローブは約 15〜約100塩基のオリゴヌクレオチドである。 発明の背景 試料を特定の核酸系被分析体の存在につき試験するための核酸プローブアッセイ イッチアッセイ法においては捕獲工程を標識付は工程の前に行うか、または捕獲 および標識付けを同時に行いうることが認められている。たとえばラングゾール およびマルコーム、ジーン36.201−210(1985) 、ランキら、ジ ーン21.77−85(1983) ;ダンおよびハッセル、セル12.23− 36(1977) ;ランキおよびソーダーランド1米国特許第4,486,5 39号および第4.563,419号明細書を参照されない。 サンドイッチアッセイ法は分析される試料の核酸が固形支持体に直接に固定化さ れる必要がないという利点を持つ0代わりに捕獲プローブ(capture p robe)が固形支持体に固定化され、これはアッセイを実際に行う当事者より もむしろ供給業者によって行うことができる。サンドイッチアッセイ法は被分析 プローブアッセイ法(サンドイッチアッセイ法を含む)について各種の方法が報 告されている。たとえばラングゾールおよびマルコーム(前掲);ブーネマンお よびウェス1〜ホフ、メソ、エンザイモ、 (Meth、Enzym−)100 .400−407(1983) ;ブーネマン、ヌクレ、アシッズ、リサ、(N ucl。 、Ac1ds Res、)10.7181−7196(1982);ブーネマン ら、ヌクレ、アシッズ、リサ、10,7t63−7180(1982):クレリ シら、ヌクレ、アシッズ、リサ6,247−258(1979)を参照されたい 、しかし、アミン誘導体およびカルボキシル誘導体にした固形支持体への核酸の 付着、特に核酸プローブ調製物の少なくとも約30%が末端デオキシリボーズま たはりポーズ部分の水酸基の酸素原子のみによって共有結合している付着につい ては報告されていない。 本発明はサンドイッチアッセイ法、および約100塩基以下の短鎖オリゴヌクレ オチドをアッセイにおけるプローブとして用いることの双方の利点を実現するこ とを目的とする。 発明の要約 15〜100塩基のオリゴヌクレオチドをハイブリダイゼーション溶液からの核 酸系被分析体の捕獲のために用いる核酸プローブサンドイッチアッセイ法に使用 するのに特に好適な固形支持体が見出された0本発明の固形支持体は、高い割合 の(一般に少なくとも約30%)結合オリゴヌクレオチド型“捕獲プローブが、 支持体におよびオリゴヌクレオチドの5′末端ヌクレオシドの5′−酸素原子ま たは3′末端ヌクレオシドの3′−酸素原子に共有結合したリンカ−のみを介し て支持体に共有結合しており、このためたとえば非共有性会合または反応性塩基 部分を介しての共有結合など、被分析体捕獲に関する利用率が著しく低下するか または失われさえする様式で支持体に会合した場合と異なり、十分に被分析体の 捕獲に利用されることを特色とする。 本発明の固形支持体は下記のものである。(1)アミノ末端基付きリンカ−によ り誘導体にされたマクロ孔質架橋デキトラン、多孔質ゲイ酸塩ガラス、ポリスチ レン、またはラテックス(リンカ一部分の末端アミン基がホスホルアミデー)  (phosphoramidate)基を介してオリゴヌクレオチド型捕獲プロ ーブの末端ヌクレオシドに共有結合しており、ホスホルアミデート基を介してオ リゴヌクレオチド型捕獲プローブの末端ヌクレオシドに共有結合していないアミ 7基は実質的にすべて、オリゴヌクレオチドとの非特異的相互作用がアシル基で ブロックされている);(2)マクロ孔質架橋デキストランを臭化シアンで活性 化したのち、オリボヌクレオチド型捕獲10−ブの末端ヌクレオシドへ誘導体形 成されたアミノアルキルホスホルアミデート基のアミノ基と反応させて、架橋デ キストランをプローブで共有結合により誘導体にしたちの;(3)カルボキシル 末端基付き、または活性エステル末端基付きリンカ−で誘導体にされたマクロ孔 質架橋デキストラン、多孔質ケイ酸塩ガラス、ポリスチレンまたはラテックス( カルボキシル基の部分は結合用ジアミノアルキルまたはジアミノアルキルジスル フィド(たとえばシスタミニル)部分の1個のアミン基に対しアミド結合で結合 しており、その他方のアミノ基はホスホルアミデートに関与しており、このホス ホルアミデートはオリゴヌクレオチド型捕獲プローブの末端ヌクレオシドに直接 に結合している);ならびに(4)スルフしドリル末端基付きリンカ−により誘 導体にされた架橋デキストラン、多孔質ケイ酸塩ガラス、ラテックスまたはポリ スチレン(スルフヒドリル基の部分は、一端に活性ジスルフィド基(たとえば2 −ピリジルまたは4−ピリジルにより活性化)をもちがつ他端にアミノ基をもつ リンカ−に由来するイオウ原子に対しジスルフィド結合で結合しており、上記ア ミン基はホスホルアミデート基の一部をなし、ホスホルアミデート基はプローブ の末端ヌクレオシドに直接に結合しており、上記ジスルフィド結合はデキストラ ン、ケイ酸塩、ラテックスまたはポリスチレンに対し誘導体形成したスルフヒド リル基とプローブに結合したリンカ−の活性ジスルフィドとのジスルフィド交換 によって生じる)。 本発明による固形支持体を製造するための好ましい支持材は下記のものである。 長鎖アルキルアミン誘導体にされた、およびカルボキシル誘導体にされた制御細 孔ガラス:パース・ケミカル社(米国イリノイ州ロックフォード)によりそれぞ れ製品番号24875および23735において、公称細孔直径500オングス トロームおよび粒径約125〜177ミクロンのビーズ状で販売されている;な らびにマクロ孔質架橋デキストラン材料であるセファクリル(Sephacry l)S−500:ファルマシア社(米国ニューシャーシー州ビスカッタウェイ) により、コードNo、17−0475−01において、湿潤直径約40〜約10 5ミクロンおよび分子量約2X1.0’ダルトン以上のデキストランに関する排 除体積(exclusionvo l ume)をもつビーズ状で販売されてお り、このセファクリルが後記のように臭化シアンにより、またはアミノ末端基付 き、カルボキシル末端基付き、または活性エステル末端基付きリンカ−により誘 導体にされるべく改質される。 好ましい支持材と好ましい捕獲プローブ(それらの5′末端に結合した適切な結 合用部分をもつべく改質されたプローブである)との反応については後述する。 本発明の新規な、かつ有利なサンドイッチアッセイ法は、種々の方法で行われる サンドイッチアッセイ法における支持体の性能に関連して本発明による固形支持 体がもつ種々の予期されなかった特性が見出されたことに基づく。 先行技術によれば標的核酸系被分析体が結合している固形支持体くたとえばニト ロセルロースフィルター)と非相同(ここでは時に“不活性”と呼ぶ)核酸(た とえば切断されたサケ精子もしくはニシン精子DNA、または煮沸したtRNA など、検出のための標識付けがなされておらず、標的核酸系被分析体に対し配列 上の相同性をほとんどもたないもの)(これは支持体上の標的核酸に溶液中のプ ローブ核酸をハイブリダイズさせる目的でハイブリダイゼーション条件下に固形 支持体と共にインキュベートされる溶液中に含まれる)とのプレハイブリダイゼ ーションはプローブの非特異的結合を減少させ、これによりバックグラウンド信 号を減少させることが示唆される。たとえばマニアチスら(前掲)を参照された い。先行技術には、オリゴヌクレオチドにより誘導体にされた固形支持体と非相 同核酸とのプレハイブリダイゼーションが相同配列をもつ核酸のハイブリダイゼ ーションに際してオリゴヌクレオチドによる捕獲効率を高めることは示唆されて いない。本発明者らはきわめて意外なことに、本発明による固形支持体と非相同 核酸とのプレハイブリダイゼーションによって、標的を含有する溶液とのハイブ リダイゼーションに際して標的被分析体の捕獲効率がほぼ倍加することを見出し た。 さらに先行技術によれば、短鎖(100ヌクレオチド以下)オリゴヌクレオチド が存在するハイブリダイゼーション溶液中に硫酸デキストランを含有させても、 オリゴヌクレオチドとこのオリゴヌクレオチドの配列に対し相補的な配列をもつ 核酸との間でハイブリダイゼーションが行われる効率には影響がないことが示唆 される。窓外にもこれと逆に、本発明者らは被分析体が長鎖くすなわち約500 塩基以上)である場合、本発明の固形支持体に結合したオリゴヌクレオチドの捕 獲効率はハイブリダイゼーション溶液中に約5〜20%(、/V)、奸才しくけ 約9〜11%(w/v)の硫酸デキストランを用いることによって著しく改良さ れることを見出した。また本発明者らはプレハイブリダイゼーション溶液中にお けるこの濃度の硫酸デキストランが、非相同DNAを除いても捕獲効率と高める ことを見出した。しかし著しく短鎖の被分析体(約30塩基)については硫酸デ キストランは捕獲効率にほとんど効果を与えないことが認められた。さらに本発 明者らは、硫酸デキストランが長鎖被分析体に対する捕獲効率に与える効果は先 行技術において仮定されるような体積排除効果(volume−exclusi on effect)ではないことを見出した。 ポリエチレングリコールが捕獲効率に与える効果は、はるかに小さいからである 0本発明の固形支持体による捕獲効率に対し硫酸デキストランが与えることが見 出された有利な効果についての根拠は明らかではないが、これは電荷に関係があ ると思われる。 また本発明者らは意外にも、被分析体の捕獲、および標識プロ ーブと被分析体とのハイブリダイゼーションが同時に起こるサンドイッチアッセ イ法により得られた結果(すなわち捕獲プローブによってビーズに結合した被分 析体にハイブリダイズした標識プローブからの信号の強度;バックグラウンドに ついて補正されたもの)は、まず被努析体がかなりの程度、溶液中の標識プロー ブとハイブリダイズし、次いで被分析体(全部または一部が標識プローブと複合 体を形成している)の捕獲が行われるサンドイッチアッセイ法の場合と本質的に 等しいことを見出した。捕獲工程が標識プローブとのハイブリダイゼーション工 程の前に行われた場合に得られる結果は、他の2方法の場合より明らかに劣る。 これは、ビーズ結合した被分析体は溶液中の標識プローブにより結合されにくい ためと思われる。 i&後に、下記の表1に挙げた多数のオリゴヌクレオチドがラトナーら、ネイチ ャー、313.277−284(1985)によりめられた配列から、HIV− 1(エイズの原因物質)についての核酸プローブハイブリダイゼーションアッセ イ法(本発明によるアッセイ法を含む)においてプローブとして用いるのに適切 であることが確認された。特にこれらのオリゴヌクレオチドは、サザーンハイプ リダイゼーションにおいて本発明によるアッセイに採用されるものに匹敵するス トリンジエンシー条件下でHIV−1ゲノム(プロウィルスDNAとして感染細 本発明はその一観点においては、それに結合した15〜100塩基の一重鎖オリ ゴヌクレオチドブローブを含み、(A)ケイ素原子において (1)次式の基 (CH2)n(N H)(CO)(CH2)cR1および−(CHz)n(N  H)(P O2) O−(オリゴ)(アミノ基を末端にもつ基により誘導体にさ れたケイ素原子は実質的に無く、式中のCは0〜5であり、nは2〜8であり、 (&)被分析体の第1セグメントの配列に対し相補的な配列をもつ15〜100 塩基の一重鎖オリゴヌクレオチドである第110−ブ(“捕獲プローブ)を含む 固形支持体であって、固形支持体の一部である第110−ブの実質的部分がもっ ばら支持体に末端結合しているもの(すなわち支持体に共有結合により、かつ5 ′末端ヌクレオシドの5′−酸素原子または3′末端ヌクレオシドの3′−酸素 原子に結合したリンカ−のみを介して結合しているもの)、および (b) 被分析体の第2セグメントの配列に対し相補的な配列をもつ一重鎖オリ ゴヌクレオチドでありかつ検出のために標コされている第2プローブであって、 ただし第2プローブの配列は第1プローブの配列に対し非相補的であり、第2セ グメントが第1セグメントと重複しないものを用いて試料中の一重鎖核酸系被分 析体を検出する方法であって、(1)上記固形支持体をプレハイブリダイゼーシ ョン条件下で、(i)非相同核酸もしくは<ii)約5〜20%(、/V)の濃 度の硫酸デキストラン、または(i)および(ii)の両者からなるプレハイブ リダイゼーション液と共にインキュベートし;(2〉 工程(1)に従ってプレ ハイブリダイズされた固形支持体をハイブリダイゼーション条件下で、約5〜2 0%(w/v)の硫酸デキストラン;アッセイされる試料からの核酸(試料中に 存在する場合には被分析体を含む);およびハイブリダイゼーション液中の被分 析体の量に対し過剰モルの第210−ブからなるハイブリダイゼーション液と共 にインキュベートし;(3)工程(2)の後に固形支持体を洗浄して、これから 被分析体に安定にハイブリダイズしていない第2プローブを除去し;そして (4)工程(3)の後に、被分析体にハイブリダイズした第2プローブ上の検出 可能な標識に起因する信号が固形支持体に付随するか否かを測定する ことよりなる方法である。 上記の節に記載した本発明方法における工程く2)の前で、第2プローブを所望 により溶液中でハイブリダイゼーション条件下に被分析試料からの核酸と共にイ ンキュベートしてもよく、これにより試料中に標的被分析体が存在する場合には 標的被分析体の少なくとも一部分はすでにハイブリダイゼーション液中の第2プ ローブにハイブリダイズし、これと固形支持体が工程(2)に従って該工程(2 )の開始時に結合することを理解すべきである。すなわち標識第2プローブと被 分析体の溶液ハイブリダイゼーションは固形支持体の捕獲10−ブによる標的の 捕獲の前に行うこともできる。 本発明による他の方法は、 (a) 被分析体の第1セグメントの配列に対し相補的な配列をもつ15〜10 0塩基の一重鎖オリゴヌクレオチドである第110−ブ(“捕獲プローブ)を含 む固形支持体であって、固形支持体の一部である第110−ブの実質的部分がも っばら支持体に末端結合しているもの(すなわち支持体に共有結合により、かつ 5′末端ヌクレオシドの5′−酸素原子または3′末端ヌクレオシドの3′−酸 素原子に結合したリンカ−のみを介して結合しているもの)、および (b) 被分析体の第2セグメントの配列に対し相補的な配列をもつ一重鎖オリ ゴヌクレオチドでありかつ検出のために標識されている第210−ブであって、 ただし第210−ブの配列は第110−ブの配列に対し非相補的であり、第2セ グメントが第1セグメントと重複しないものを用いて試料中の一重鎖核酸系被分 析体を検出する方法であって、(1) 上記固形支持体をプレハイブリダイゼー ション条件下で、(i>非相同核酸もしくは(ii)約5〜20%(w/v)の 濃度の硫酸デキストラン、または(i)および(ii>の両者からなるプレハイ ブリダイゼーション液と共にインキュベートし;(2)工程(1)に従ってプレ ハイブリダイズされた固形支持体をハイブリダイゼーション条件下で、約5〜2 0%(−/v)の硫酸デキストラン;アッセイされる試料からの核酸(試料中に 存在する場合には被分析体を含む)からなる第1ハイブリダイゼーシヨン液と共 にインキュベートし、この第1ハイブリダイゼーシヨン液は実質的に第2プロー ブを含ます;(3)工程(2)の後に、ハイブリダイゼーション条件下で固形支 持体を第2ハイブリダイゼーシヨン液中の被分析体の量に対し過剰モルの第2プ ローブからなる第2ハイブリダイゼーシヨン液と共にインキュベートしく第1プ ローブへのハイブリダイゼーションにより固形支持体に結合した被分析体を含む );(4)工程く3)の後に固形支持体を洗浄して、これから被分析体に安定に ハイブリダイズしていない第2プローブを除去し;そして (5)工程(4)の後に、被分析体にハイブリダイズした第2プローブ上の検出 可能な標識に起因する信号が固形支持体に付随するか否かを測定する ことよりなる方法である。 本発明はさらに、試料中の一重鎖核酸系被分析体を検出するための本発明方法を 実施するための試験キットを提供する。このキットは本発明による固形支持体、 固形支持体上に捕獲される被分析体に対する標識プローブ、および標識プローブ からの信号を発生させるのに必要な試薬(必要な場合には)(たとえば、標識が ビオチンである場合には酵素結合アビジン;標識がキレート化ユーロピウムイオ ンである場合には蛍光増強液(ムッソら、国際特許出願公開W O877027 08号明細書参照))からなる。 さらに他の観点においては、本発明は前記衣1に挙げたオリゴヌクレオチド配列 をもつDNAもしくはRNA、またはそれらの相補体くすなわち上記オリゴヌク レオチドの配列に対し相補的な配列をもつオリゴヌクレオチド)を提供し、これ らのDNAまたはRNAは所望により検出のために標識されていてもよい。オリ ゴヌクレオチドを検出可能にすべく標識するために当技術分野で知られている多 数の方法をいずれも、本発明のDNAおよびRNAについて採用できる。たとえ ばDNAもしくはRNAを5′−ヌクレオシドの5′炭素原子にあるボスフェー ト基においてT4ポリヌクレオチドキナーゼを用いて32pにより:DNAもし くはRNAの5′末端にあるリンアミデート基にジアミノアルキルリンカ一部分 を通して(チューおよびオルゲル、D N A 4.327−331(1985 )の記載による)、もしくは改質ヌクレオチド塩基にリンカ−を通して(たとえ ばドライヤーおよびダーバン、ブロシ、ナショナル、アカデ、サイ、(Proe 。 Natl、Acad、 Sci、)(U S A)82,968−972(19 85)の記載による)結合したビオチンにより;またはキレート化ユーロピウム イオンによりくムッソら、国際特許出願公開W 087102708号明細書の 記載による)標識することができる。 捕獲プローブとして使用されるオリゴヌクレオチドは下記の方法により固形支持 材に結合させることができる。(])各種の非共有型相互作用くたとえば核酸が ニトロセルロースまたはナイロンフィルターなどの月利に結合するもの)、(2 )支持材」二の、または支持材に共有結合により誘導体形成された基土の反応性 基と、オリゴヌクレオチドのピリミジンまたはプリン塩基部分上の反応性基との 共有結合ならびに(3)支持材上の、または支持材に共有結合により誘導体形成 された基土の反応性基と、オリゴヌクレオチドの一末端ヌクレオシドもしくは他 方に誘導体形成された基(たとえば5′ヌクレオシドの5′−酸素原子または3 ′末端ヌクレオシドの3′−酸素原子を介して)上の反応性基との共有結合、捕 獲プローブは支持体への共有結合の唯一の手段が上記(3)型のものである場合 には、固形支持体に“末端結合”していると言われる。特に捕獲プローブを含む 固形支持体を核酸プローブアッセイ法に用いるためには、高い割合の捕獲プロー ブが末端結合していること、および末端結合プローブと支持材との非共有型相互 作用を最小限に抑えることが有利である。この種の末端結合した捕獲プローブは 標的を捕獲するために最大限に利用されるからである。支持体において捕獲10 −ブの約10%以上が末端結合している場合、固形支持体において“実質的割合 ”の捕獲プローブが末端結合している。少なくとも約30%が末端結合している ことが好ましい。 捕獲プローブが本発明に用いられる固形支持材(たとえばケイ酸塩ガラスピーズ 、架橋デキストラン、ポリスチレン)に、核酸の末端にジスルフィド結合または ホスホルアミデート基を与えるリンカ−によって結合している固形支持体につい ては、末端結合の程度の測定は可能である。ジスルフィド結合は緩和な条件下で 容易に還元されて末端結合しているプローブを放出するが、1個または2個以上 の塩基を介して共有しているプローブは放出しないからであり、またホスホルア ミデート結合は酸不安定性であり、同様に適切な酸性条件下で、末端結合してい るプローブを放出するが、1個または2個以上の塩基を介して共有結合している プローブは放出しないからである。非共有結合は捕獲プローブを支持材と共に、 リンカ−などにより共有結合しない条件下でインキュベートし、支持材により吸 収されたプローブの量を測定するだけで推定できる。これらの共有結合および非 共有結合の測定に際しては、32p標識された捕獲プローブを用いることが好都 合である。この場合、支持体に付随するプローブおよびそれから除かれたプロー ブの量を容易に測定しうるからである。 特に前記のように、捕獲プローブが各種支持材に各種リンカ−によって結合して いる様式を判定することにより、前記のグループ(B)(3)による固形支持体 (特定の水酸基酸素原子において、式 %式% の基により誘導体にされた架橋デキストラン系マクロ孔質材料)の優れた特性が 見出された。これらの支持体のうちきわめて好ましいデキストラン系材料はセフ ァクリルS−500である。ジスルフィドを含まないリンカ−がより好ましい。 好ましいセファクリルS−500以外のマクロ孔質架橋デキストラン系材料も本 発明による固形支持体に使用できることを認識すべきである。たとえばセファク リルS−200およびセファクリルS−1000(ファルマシア社により販売) も、他のマクロ孔質架橋デキストラン系材料、特にセファクリル類のように、ア リルデキストランをN、N′−メチレンビスアクリルアミドで架橋することによ り製造された材料(ファルマシアその他の会社により販売)と同様に使用できる 。 本発明をここにHIV−1の検出について説明するが、当業者には本発明があら ゆる核酸系被分析体、特にアッセイされる試料中に一重鎖の形で存在するもの( すなわち相補的配列をもつ連鎖を大幅に上回る一本の連鎖をもつもの)について 適用でき、HIV−1に限定されないことが明らかであろう。 さらに本発明による固形支持体は、ここでは主としてそれらを核酸プローブアッ セイに用いることにつき記述したが、アフィニティークロマトグラフィー(“オ リゴdTカラム”が長い間使用されているように)に用いて目的核酸を単離する こともできる。 オリゴdTカラムはポリAティル付きRNAの単離に限定されているが、本発明 の支持体と用いると、DNAまたはRNAのいずれであっても、またポリアデニ ル化されていてもいなくても、目的とする特定の核酸を単離することができる。 捕獲プローブがジスルフィド結合をもつリンカ−によって支持材′に結合してい る本発明の支持体は、目的とする核酸をアフィニティークロマトグラフィーによ り得るために有利に用いられる場合がある。これらのジスルフィド型リンカ−を 含む支持体から捕獲プローブ(目的とする核酸にアニールしたもの)を分離する のに必要な条件が緩和であり、実施しゃすいからである。 ここで“活性エステル”とは当技術分野で理解されるように、カルボキシレート 基とN−しドロキシスクシンイミド、5−2−ピリジンなどの化合物により形成 されるエステルを表わす。 本発明のアッセイ法が適用される試料は、通常は粗製検体、たとえば血液その他 の体液、植物または動物から得た組織試料、食品試料、微生物培養物などを、当 技術分野で知られている方法により処理して、これから核酸、または少なくとも 核酸系被分析体(すなわち“標的”)を含むと思われる核酸画分を分離すること によって得られるであろう、単離過程で、アッセイを妨害し、特定の酵素の場合 は核酸系被分析体およびオリゴヌクレオチド型プローブを破壊する可能性のある 他の物質(たとえば蛋白質)から核酸が分離されるであろう。 被分析体が存在する場合に、これがアッセイの検出限界以下の濃度で存在すると 思われる場合には、たとえばサイキら、サイエンス231.1434−1436 (1985)から採用される方法に従って被分析体の量を増幅する必要があろう 。 プレハイブリダイゼーションおよびハイブリダイゼーションの条件、ならびに不 安定な状態でハイブリダイズしたプローブまたはハイブリダイズしていないプロ ーブを洗浄除去するための方法は、核酸プローブアッセイの技術分野の専門家に は周知である。 第2プローブは、存在すると思われる被分析体の最大量が経験から分かっている 大部分の場合、ハイブリダイゼーション液中の被分析体の量に対し過剰モル量で 供給することは容易である。この最大量がもし分かっていない場合には、第2プ ローブ濃度が広範に変化した幾つかのアッセイを異なる試料部分につき実施する ことにより、第2プローブが過剰モルであるアッセイに確実に行うことができる 。 被分析体にハイブリダイズした第2プローブ上の標識からの信号が固形支持体に 付随するものであるか否かの判定は、支持体においてその標識に適した測定を行 って支持体からのこの種の信号を検出することによる。8ましくは、被験試料に 由来する被分析体が結合している可能性のある支持体からの信号を、対照支持体 、すなわち被験試料について採用されたと実質的に同じ方法で処理されているが 、それに被分析体は結合していないことが分かつている支持体からの信号と比較 する。被験試料からの信号が対照支持体からの信号よりも規模が大きい場合、ア ッセイが双方の支持体について操作可能であることが分かつている限り、被験試 料支持体からの信号は被験試料中の被分析体に起因することが確認される。もち ろん当技術分野において理解されるように、アッセイされる核酸を含有すること が知られている陽性対照試料をも被験試料および陰性対照試料と平行使用して、 そのアッセイ系が本発明を以下の実施例に若干詳細に説明する。 実施例I プローブの調製および精製 短鎖オリゴヌクレオチド(溶液からの被分析体の捕獲に関与する成分を含む)を 用いるサンドイッチアッセイにおいては、感度および特異性、従って信頼性のた めに、プローブが高純度でありかつ均一な配列をもつことが重要である。十分な 品質をもつオリゴヌクレオチドを得るために当業者にとっては多数の合成法およ び精製法があるが、本発明者らは下記の方法を受容しうろことを認めた。すなわ ちオリゴヌクレオチドはアプライド・バイオシステム(Applicle B  iosystem)380A型自動合成装置を用いて、シアノエチルホスホルア ミデート化学により合成される0合成は1Mモルの規模で9分間の短い結合サイ クルを用いて行われる。各結合サイクル独自の収率は比色ア・ンセイ法を用いる トリチル検出により測定して98%である。この方法によって30ヌクレオチド のDNAプローブが3時間で合成され、次いで塩基保護基の除去およびトリチル 化オリゴヌクレオチドの精製にさらに18時間を要する。精製プロトコール終了 時に得られるオリゴヌクレオチドの量は合成1回当たりオリゴヌクレオチド40 0〜600μ2である。 特異的標的核酸である被分析体を確実に検出するためには純粋な全長プローブが 必須であるため、本発明者らはオリゴヌクレオチド精製のプロトコールを開発す ることに特別な注意を払った。トリチル化オリゴヌクレオチドの精製はC−18 逆相半調製用クロマトグラフイー(10X 250+*+*のカラム)により行 う、C−18クロマトグラフイーの前に脱トリチル化されたオリゴヌクレオチド を単離する試みは、オリゴヌクレオチドの疎水性がより低いためより効率の低い 精製手段であることが証明された。40分間にわたって0.1M )ジチルアン モニウムアセテート中15〜35%のアセトニトリルの濃度勾配を用いて、トリ チル化された全長オリゴヌクレオチドを溶離した。このオリゴヌクレオチドを次 いで水中の80%酢酸により1時間脱トリチル化した。脱トリチル化ののちG− 50セフアデツクスクロマトグラフイーを行った。単離されたオリゴヌクレオチ ドをさらにRPC−5カラム上でのHPLCにより判定した<4.6X250m mのカラム;溶剤A。 2−Mトリス、pH12、溶剤8.2+mM)リス、200諭M過塩素酸塩、p H12,濃度勾配置0%Bから50%Bまで、40分間)、この最後の工程は少 量のオリゴヌクレオチドについては付加的な精製工程にもなる。 比較的長鎖のオリゴヌクレオチド(〉50ヌクレオチド)については、全長オリ ゴヌクレオチドの単離は12〜20%のゲルを用いる調製用ポリアクリルアミド ゲル電気泳動により行われる(マニアチスら(前掲)に記述されるプロトコール に従う)。 前記の方法(50ヌクレオチド以下のオリゴヌクレオチドに関するもの)を採用 して、表IにDNAとして挙げられるオリゴヌクレオチドを調製した。 実施例■ プローブのリン酸化 オリゴヌクレオチド型プローブを3′−ヌクレオシドの3′−炭素原子(“3′ 末端”)または5′−ヌクレオシドの5′−炭素原子(“5′末端”)において 標準法によってリン酸化する。 3′末端におけるリン酸化についてはバネットおよびコラナ、ジェイ、バイオロ 、ケミ、(J 、B iol、chem、)249.5213−5221を参照 されたい。 オリゴヌクレオチドはそれらの5′末端においてT4ポリヌクレオチドキナーゼ を用いて標準プロトコールに従ってルーチンにリン酸化されたくマニアチスら( 前掲)を参照されたい)。 32p標識プローブを調製するためには、適宜な位1に32p含有ホスフエート を含む基質をリン酸化に用いる。 実施例■ プローブの標識付けおよび検出 検出のためのプローブ標識付けは放射性または非放射性標識により行うことがで きる。 放射性標識付けを採用する場合、実施例Hに記載した32p標識付けが好ましい が、他の同位体、たとえば128工または”Cを用いた既知の方法に従う標識付 けも採用できる。 被分析体(固形支持体に結合した捕獲プローブと複合体を形成)と複合体を形成 した放射性標識プローブを検出するためにはシンチレーション計数法が採用され る。 多数の非放射性標識付けおよび付随する検出のプロ1〜コールが知られている。 標識オリゴヌクレオチド型プローブとこのプローブの配列に相補的な配列をもつ 被分析体セグメントとの間の塩基対合に対する妨害が最小であるこれら標識付は 技術はいずれも本発明に採用できる。たとえばビオチンに結合し、かつホスホル アミデート結合によりオリゴヌクレオチドの5′末端のホスフェートに結合した ジアミン系リンカ−を介するビオチンによる標識付けを採用しうる(チューおよ びオーゲル、DNA、4巻、327−331(1985)に従う)、ビオチンの 代わりにEDTAキレート化蛍光Eu”を用いる同様な標識付けも採用し、うる (ムッソら、国際特許出願公開W O87102708号明細書に記tI’、) 。 ヌクレオシド塩基に結合したビオチン(ドライヤーおよびダーバン(前掲)によ り報告されたもの)による標識付けも採用できる。 実施例■ 多孔質ケイ酸塩ガラスの誘導体形成 (A>“長鎖アルキルアミン”誘導体型の制御細孔ガラスはパース・ケミカル社 (米国イリノイ州ロックフォート)から購入された。製品No、24875.こ の多孔質ゲイ酸塩ガラスは式(CHz)aN H2の基で誘導体にされたケイ素 原子を含む。 (B) カルボキシル誘導体型の制御細孔ガラスはパース・ケミカル社(米国イ リノイ州ロック7オード)から購入された。製品N o 、23735 、この 多孔質ケイ酸塩ガラスは式−(CH2>38H(C○)(CH2)2C○2Hの 基で誘導体にされたケイ素原子を含む。 (C) チオール誘導体型の制御細孔 ガラスもパース・ケミカル社(米国イリノイ州ロックフォード)から購入された 。製品N o、23748.この多孔質ケイ酸塩ガラスは次式の基で誘導体にさ れたケイ素原子を含む。 (D) スクシンイミドエステル誘導体型の多孔質ケイ酸塩ガラスは前記のカル ボキシル誘導体型ガラスを用いて調製される。 カルボキシル誘導体型ガラスをアセトンで3回、次いで乾燥アセトン(4人の分 子篩上に置くことにより乾@)で3回洗浄することにより乾燥させる。これらの ビーズを次いで焼結ガラスろうとにより濾過し、真空中で10時間乾燥させる。 乾燥N、N”−ジメチルホルムアミド(“DMF”) ]、5 z 1中の乾燥 ビーズ500+yにジシクロへキシルカルボジイミド50019およびN−ヒド ロキシスクシンイミド3003+17を添加する。混合物を16時間攪拌し、乾 燥DMF<3回、各20a+1)および塩化メチレン(2回、各20z1)で洗 浄して、過剰の試薬およびジシクロヘキシル尿素を除去する。得られた支持体を 次いで真空中で3時開乾燥させる。 実施例V マクロ多孔質架橋デキストラン系支持体の誘導体形成(A) セファクリルS− 500はファルマシア社(米国ニューシャーシー州ビスカッタウェイ)からコー ドN o 、1.7−0475− OXで購入された。 (B) セファクリルS−500をブーネマンら(前掲)の方法に従って臭化シ アン活性化した。製造業者により供給された湿潤材料100gを大過剰の蒸留水 および焼結ガラスフィルターを用いて洗浄した。オーバーヘッドかい形攪拌機お よびpH[極を備えた10001Nのビーカーに湿潤ケーキを移した。水冷しな 水500R1を添加し、混合物を激しく攪拌し、その間木片を時々添加すること により10℃以下に保った。固体臭化シアン40gを添加し、pHを反復監視し 、5 M−NaOHの添加により約20分間、10.5〜11.5に維持した0 次いでビーズ材料を5容量の氷冷水ののち1容量の氷冷した10mMリン酸カリ ウム(pH8,0)で速やかに洗浄した。 当業者に理解されるように、CNBr活性化には隣接炭素原子上に水酸基を必要 とする。 (C) カルボキシル誘導体型の材料は下記により調製された。 10mMリン酸カリウム(pH8)200zi中の新たに調製した209のCN Br活性化セファクリルS−500の懸濁液に6−アミノカプロン酸16gを4 回に分けて30分毎に添加し、得られた混合物をかい形攪拌機で20分間攪拌し た。(あるいは6−アミノカプロン酸の代わりにヘキシレンジアミンを用いて、 そのアミン誘導体型支持体を得ることができる)。 得られた誘導体型セファクリルを焼結ガラスろうとにより枦通し、10+mMリ ン酸カリウム緩衝液(pH8,0) ; I Mリン酸カリウム緩衝液(pH8 ,0);I M−KCl、0.1M−NaOH5最後に水で徹底的に洗浄した0 次いで湿潤ゲータを0.1M −M E S (pH6,0)に懸濁し、4℃に 保存した。 (D) スクシンイミド誘導体型のマクロ孔質架橋デキストラン材料は下記によ りカルボキシル誘導体型の材料から調製された。0.1M −ME S[衝液( 合物をかい形攪拌機で20時間攪拌する。得られた誘導体型セファクリルをV( C)の場合と同様に濾過し、洗浄する。使用前に誘導体型材料のリンカ−である シスタミンのジスルフィド結合は、この材料を1100eaジチオトレイ1〜− ル(DTT)、50raM −HE P E S (pH7,7>に25℃にお いて1時間懸濁し、次いでこの材料を脱泡した0、2M −HE PE S ( pH7,7)、0.1M −K(Jで十分に洗浄し、最後に0.2M・HE P  E S (pH7,7>で再び洗浄することにより解裂される。 実施例■ ジビニルベンゼン架橋ポリスチレン系支持体の誘導体形成(A) 1%架橋ジビ ニルベンゼン−ポリスチレンクロルメチル樹脂(1,1mMe!/1?、200 −400メツシュ)は、ニー・ニス・バイオケミカル社(米国オハイオ州クリー ブランド)から購入された。 12%ジビニルベンゼン架橋ポリスチレンビーズ(メツシュサイズ200〜40 0)はポリサイエンス(ペンシルベニア州ワリントン)から購入された。 (B)12%架橋ジビニルベンゼン−ポリスチレンビーズを下記によりカルボキ シル誘導体にした。 上記ビーズ10gを1.2−ジクロルエタン150zlに添加し、混合物を還流 した。ニトロベンゼン101R中の塩化アルミニウム20ミリモルおよびグルタ ル酸モノメチルエステルクロリド10ミリモルの濾過混合物を5分間にわたって 添加し、反応混合物をさらに4時間還流した。樹脂を焼結ガラスろうとで濾過し 、ジオキサン:4N −H(J(3:1)、ジオキサン;水(3:1)、ジオキ サン、エタノール、アセトン、最後にメタノールで洗浄し、次いでデシケータ− 内で真空下に乾燥させた。三日フラスコ中のKOH2yにエチレングリコール1 00y/を添加し、混合物を100℃に加熱した。KOHが十分に溶解したのち 、乾燥した誘導体型樹脂5gをヒドラジン水化物3yt(lと共に添加した。混 合物を還流させ、3時間還流したのち還流温度が198℃に達するまで液体の一 部を留去した0次いでフラスコを密栓し、12時間還流を続けた6次いで樹脂を 前記(グルタル酸モノメチルエステルクロリドによる誘導体形成後)と同様に洗 浄した。 (C)12%ジビニルベンゼン架橋ポリスチレンビーズを下記によりアミン誘導 体にした。カルボキシル誘導体型ビーズ5gにジメチルホルムアミド(D M  F >50zl中のジシクロへキシルカルボジイミド1fIおよびN−ヒドロキ シスクシンイミド1.259を添加した。30分間攪拌したのち、1,8−ジア ミノオクタン7fIを添加し、混合物をさらに3時間攪拌した0次いで樹脂を濾 過したのち、順次メタノール、水、メタノール、CH2Cl 2、最後にアセト ンで洗浄した。 (D) 1%ジビニルベンゼン架橋したクロルメチル誘導体型ポリスチレン樹脂 を下記によりアミノ誘導体にした。DMF80111中の1%架橋したクロルメ チル誘導体型樹脂(ニー・ニス・バイオケミカル社により供給) 10.にN− t−ブチルオキシカルボニル−6−アミノカブロンM2.61?およびフッ化カ リウム1.282を添加し、混合物を80℃で16時間攪拌した。次いで樹脂を 濾過し、順次CH2Clx、メタノールおよびアセトンで洗浄したのち、高真空 (0,01)ル)を3時間施した。CH2(J2中の30%トリフルオル酢酸を 用いてアミノ基の脱保護を行い、アミン誘導体型の支持体を得た。アミノ酸のピ クリン酸滴定によって0.17ミリモル/9(樹脂〉の置換水準が示された。 実施例■ オリゴヌクレオチドの5′−アミノアルキル、5′−カルボキシルアルキル、お よび 5′−シスタミニルホスホルアミデート誘導体(A) 5’−アミノアルキルホ スホルアミデート誘導体アミノアルキル誘導体は一般に1〜20μMの濃度で1 〜10ナノモルの規模において調製された。以下のプロトコールはチューら、ヌ クレ、アシッズ、リサ、(Nucl、Ac1ds Res、>11.6513− 6529(1983)のものであり、修飾されたオリゴヌクレオチドはエタノー ル沈澱法により単離された(コレットおよびカワシマ、ヌクレ、アシッズ、リサ 、(Nucl、Aeids Res、H3,1529−1541(1985)に 従う。 すなわちオリゴヌクレオチドのアミノ−へキシル付加物を調製するために、シリ コナイズしたエッペンドルフ試験管内でオリゴヌクレオチド(5′末端において リン酸化されたもの)4ナノモルを、0.3肩βの0 、24 M・1.6−ジ アミツヘキサン、0.1M・N−メチルイミダゾールおよび0.1M・1−エチ ル−3−(3−ジメチルアミノブロビル)カルボジイミド(“CDI”)(pH 6,0)中に入れ、20時間反応させた。得られたDNAを3回エタノール沈殿 させて過剰のジアミンを除去し、次いで0.1M −MOPS (pH7,5) (アミノ基20〜30マイクロモル/g<ガラス)を含む) Logをシラナイズされた工・ンベンドルフ試験管内で0.11+6の0.1M  −N−メチルイミダゾール、0.1M −CD Iおよび5xio−’〜20 X10−’M・5′−リン酸化オリゴヌクレオチドと混和する。攪拌しながら2 4時間反応を続け、次11でビーズをN ao H(pH12)で1分間洗浄し lの0.1M−NaHCO,,0,5M−Na(1!(pH8,3)、および0 .025zfの5′−6−アミンへキシルホスホルアミデート誘導体型オリゴヌ クレオチド(約5xlO−’M)と0.05M・M OP S (+)H7,5 )中で混和する。反応を20時時間待させる。 次いで試験管を遠心分離し、上層液を除去したのち、0.1M・トリス(pHs 、オよL’1mM・EDTA(pH7,7)と共に1時 間インキュベートした0次いで支持体をN ao T4 (pH12)で洗浄し て、離脱したオリゴヌクレオチドを除去した。一般に、シスタミニル誘導体型オ リゴヌクレオチドについての末端結合の測定結果はアミノアルキル誘導体型オリ ゴヌクレオチドについてのものと一致した。 シスグミニル誘導体型オリゴヌクレオチドを脱離するために用いられる条件が緩 和であることは、本発明の固形支持体を核酸の単離に適用するにつき、注目すべ きである。 本発明のきわめて好ましい固形支持体は5’−(6−アミノへキシルホスホルア ミデート)誘導体型オリゴヌクレオチドで誘導体にされたカルボキシル誘導体型 セファクリルS−500である。 この例の方法を採用すると、カルボキシル誘導体型セファクリル5−sooのa g<湿潤重量)当たり約1.2ピコモルのオリゴヌクレオチドが誘導体形成され る。約半量のオリゴヌクレオチドがもっばら末端結合しており(オリゴヌクレオ チドに結合した6−アミノへキシルホスホルアミデートの6−アミン基とセファ クリルに結合したカルボキシルとの間のアミド結合の形成のみによって共有結合 )、わずか約0.5%が支持体に非共有結合により結合しており、約半量はオリ ゴヌクレオチドの塩基(たとえばシトシン)のアミン基を介して共有結合してい ると考えられる(支持体のカルボキシル基と6−アミノへキシルホスホルアミデ ートのアミノ基との間のアミドの形成を伴うと思われる)。 支持材のカルボキシル基がそれらのN−ヒドロキシスクシンイミドエステルとし て活性化されている場合、末端結合の水準はより高く、ヌクレオシド塩基を介し 体、特にヒトにおける後天性免疫不全症候群の原因物質であるHIV−1ウイル スのゲノムの検出に使用する方法につき説明する。 本発明による固形支持体く”ビーズ)はカルボキシル誘導体型セファクリルS− 500および5’−(6−アミノヘキシル)ホスホルアミデート誘導体型オリゴ ヌクレオチド85−241(捕獲プローブ)を用いて調製された。デキストラン の酸素原子とオリゴヌクレオチドの5′−チミジンの酸素原子との間のリンカ− は次式の構造をもつ。 −C(= N H)N H(CHz)s(CO)(N H)(CH2)IN H (P O2)−オリゴヌクレオチド85−233を第2の標識プローブとして用 いるためにT4ポリヌクレオチドキナーゼを用いて32p含有ホスフエートによ り5′−リン酸化した。 以下の方法は85−241および85−233以外の、HIV−1ゲノムの重複 ないし各セグメントの配列に相補的な配列をもつオリゴヌクレオチド対を用いて 行うことができる点を留意すべきである。たとえば表1に挙げたオリゴヌクレオ チド対をいずれも使用できる。さらにビーズの調製に用いられる他の固形支持体 を用いて、また第210−ブ上に検出のための非放射性標識を用いて行うことも できる。
