Description

【発明の詳細な説明】 本発明は式()で示される5,10−エポキシ
化プレビタミンD3に関する。
本発明の式()で示されるプレビタミンD3
−5,10−エポキシドは新規化合物でありビタミ
ンD3と過酸化物との反応、例えばt−ブチルハ
イドロパーオキサイドとモリブデンカルボニル
〔Mo(CO)6〕を用いた反応に付すことにより製造
される。反応混合物から目的化合物()の単離
は常法により例えば反応液を洗浄後、溶媒を留去
し残渣をカラムクロマトグラフイー等の手段に付
すことにより行なわれる。このようにして製造し
た本発明の化合物()は人の骨髄性白血病HL
−60細胞に対し強い顆粒球への分化誘導能を有し
例えば脱癌剤等の医薬として有用である。
実施例 ビタミンD3100mg、モリブデンカルボニル
〔Mo(CO)6〕3mgを乾燥ベンゼン2mlに溶解し室
温で撹拌する。次いで無水t−ブチルハイドロパ
ーオキサイド104μlを加え1.5時間還流後反応液を
亜硫酸ナトリウム50mg/水5ml中に注ぎ十分に振
る。飽和食塩水で洗浄後、硫酸ナトリウムで乾燥
し溶媒を留去する。残渣をシリカゲルカラムクロ
マトグラフイー(シリカゲル7g,溶媒;20%酢
酸エチル−n−ヘキサン)に付しプレビタミン
D3−5,10−エポキシド()25mgを得る。
NMRスペクトル(CDCl3)δ:0.71(3H,
s), 0.88(6H,d,J=7Hz),0.96(3H,d,J
=7Hz),1.31(3H,s),3.71(1H,m),
5.58 (1H,m),5.46,5.86(2H,d,J=12Hz) マススペクトルm/e;400(M+),385,382, 364,357,343,287,269 UVスペクトルλEtOH nax;243nm
