上記式()の化合物又はその塩は優れた胃酸
分泌抑制作用を示し、抗潰瘍剤の有効成分として
有用である。
ところで胃又は十二指腸に潰瘍が生ずる一つの
大きな要因は胃酸の異常に多量の分泌であり、こ
れに対処するための従来の抗潰瘍剤は、胃酸を中
和する作用をもつものと、抗コリン作用をもつも
のとに大別される。ところが胃酸を中和するタイ
プのものは持続性に乏しく効果も弱く、また、抗
コリン作用をもつタイプのものは副作用が強く望
ましくない。
一方、胃酸の分泌はヒスタミンH2受容体を介
して刺激されることが既に知られており、最近、
このヒスタミンH2受容体拮抗作用を有する新規
なタイプの胃酸分泌抑制剤が開発され、いくつか
提案されている〔例えば、特公昭53−24422号公
報、特公昭56−1309号公報、特開昭53−18557号
公報、特開昭55−85573号公報、特開昭56−40675
号公報、特表昭55−500898号公報等参照〕。
本発明により提案される上記式()の化合物
は、ベンジルアミン誘導体のアミノアルキル基に
対しメター位に結合する脂肪鎖中に不飽和結合
(−CH=CH−)を有する点で特徴的な、従来の
文献に未載の新規な化合物であり、ヒスタミン
H2受容体拮抗作用にもとづく優れた胃酸分泌抑
制作用を有し、新しいタイプの抗潰瘍剤として有
用な化合物である。
本明細書において用いる「低級」なる語は、こ
の語が付された化合物又は基が6個以下、好まし
くは4個以下の炭素原子を有していることを意味
する。
しかして、「低級アルキル基」は直鎖状又は分
岐鎖状のいずれであつてもよく、例えばメチル、
エチル、プロピル、イソプロピル、ブチル、イソ
ブチル、sec−ブチル、tert−ブチル基等が挙げ
られ、「低級アルカノイルオキシ基」は低級アル
キル部分が上記の意味を有する低級アルキルカル
ボニルオキシ基であり、例えばアセチルオキシ、
プロピオニルオキシ基等が包含され、「低級アル
コキシ基」は低級アルキル部分が上記の意味を有
する低級アルキルオキシ基であり、例えばメトキ
シ、エトキシ、プロポキシ基等が挙げられる。
また、「低級アルキレン基」は上記「低級アル
キル」の2価基、例えばメチレン、エチレン基等
を意味している。
本明細書において用いる「置換基の名称を二つ
並べる」表現、例えば「ヒドロキシ低級アルキル
基」なる表現は、ヒドロキシ基が好ましくは1個
置換した低級アルキル基の意味に用い、例えばヒ
ドロキシメチル、ヒドロキシエチル、ヒドロキシ
プロピル基等が包含される。
同様に、「低級アルカノイルオキシ低級アルキ
ル基」、
「ピリジル低級アルキル基」なる表現は、それ
ぞれの基においてその前半に書かれている置換基
1個により置換された低級アルキル基を意味して
いる。
上記式()において、R1及びR2がこれらが
結合している窒素原子と一緒になつて形成しうる
「4〜8員の複素環」は、該窒素原子以外に異種
原子を含まないことが望ましく且つ環は飽和して
いることが好適である。かかる複素環式基の具体

て広範に変えることができるが、一般に、式

モルの範囲内で使用するのが適当である。
かくして、前記式()の化合物が得ら
れ、この化合物又はその塩は次いで式()
の化合物又はその塩と反応せしめることによ
り、初期の化合物に変えることができる。
式()の化合物又はその塩と式()の
化合物又はその塩との反応は、通常、適当な
不活性溶媒、例えば、メタノール、エタノー
ル、イソプロパノールの如きアルコール類;
トルエン、キシレンの如き芳香族炭化水素
類;アセトニトリルの如きニトリル類;テト
ラヒドロフラン;ジオキサンの如きエーテル
類等の中で行なうことができる。反応温度及
び圧力は臨界的ではなく広範に変えうるが、
一般に反応温度は室温乃至反応混合物の還流
温度、好ましくは40℃乃至反応混合物の還流
温度の範囲内であり、また圧力は常圧で充分
であるが、必要により減圧又は加圧を用いる
こともできる。

上記式()の化合物又はその塩に対する
式()の化合物又はその塩の使用割合は臨
界的ではなく、用いる反応条件等に応じて広
範に変えることができるが、一般には、式
()の化合物又はその塩1モル当り式()
の化合物又はその塩を1〜2モル、好ましく
は1〜1.5モルの割合で使用するのが有利で
ある。

に準じて製造することができる。

と式(XI)の化合物との反応は、通常、適当
な不活性溶媒、例えばメタノール、エタノー
ルの如きアルコール類;エチルエーテル、テ
トラヒドロフランの如きエーテル類;アセト

部分の水素原子が互にシス−配位をもつ化合物が
得られる。
他方、上記式(−a)において二重結合部分
の水素原子が互いにトランスに配位した化合物
は、例えば下記の反応式に示す経路によつて合成
することができる。
上記式中、R1、R2、R3、X1及びnは前記の意
味を有し、R16は低級アルキル基を表わし、X2は
ハロゲン原子を表わし、M1は水素原子又はアル
カリ金属を表わす。
上記反応式において、式()の化合物と式
()の化合物の反応はそれ自体公知のウイチ
ツヒ反応に従つて行なうことができ、得られる式
()の化合物をそれ自体公知のエステル化法
に従い、式R16OHのアルコール又はそのエステ
ル形成性反応性誘導体と反応させることにより上
記式()のエステルに変える。次いでこの式
()のエステルを例えばリチウムアルミニウ
ムハイドライド、t−ブトキシリチウムアルミニ
ウムハイドライド等の錯金属水素化物を用い、そ
れ自体公知の方法で還元し、該エステルを式(
)のアルコールに変え、得られる式()の
アルコールを例えばチオニルハライド、スルフリ
ルハライド、五ハロゲン化燐、三ハロゲン化燐等
のハロゲン化剤で処理して上記式()の化合
物を生成せしめる。

ルアミノ基等を表わし、nは前記の意味を有す
る、
の化合物と反応させ、得られる下記式
式中、R1、R2、R3、Q及びnは前記の意味を
有する、
の化合物のアミノ保護基を離脱させることにより
製造することができる。
上記式()の化合物と式()の化
合物との反応はそれ自体公知の方法(例えば特開
昭56−7760号公報に記載の方法)に従つて行なう
ことができ、また得られる式()の化合物
からのアミノ保護基の離脱はそれ自体公知の方
法、例えばヒドラジノリシス又は穏和な加水分解
によつてアミノ保護基を離脱せしめることによ
り、前記式(−b)の目的化合物を得ることが
できる。
なお、前記式(−b)においてR3が水素原
子を表わす場合の化合物は下記の反応式に従つて
合成することもできる。
上記各式中、R1、R2、Q、M2、X3及びnは前
記の意味を有し、X4はハロゲン原子を表わす。
上記反応式において、式(XI)の化合物と
式()の化合物の反応は、それ自体公知の
方法に従つて前記式()の化合物と式(
)の化合物との反応について前記したのと同
様にして行なうことができ、得られる式(
XII)の化合物のハロゲン化は、例えば、チオニル
ハライド、ハロゲン化水素酸等のハロゲン化剤で
処理することにより達成できる。次いで、得られ
る式()の化合物を脱酸剤、例えばトリ
エチルアミン、水酸化ナトリウム等の存在下に式
()のアミンと反応させ、かくして得られ
る式(−a)の化合物から前述の方法でア
ミノ保護基を離脱させることによつても、目的と
する式(−b−1)の化合物が得られる。
以上述べた方法により製造される前記式()
の化合物は、必要に応じて対応する塩に変えるこ
とができる。造塩反応はそれ自体公知の方法に従
い、式()の化合物を前記した如き無機酸又は
有機酸で処理することにより容易に行なうことが
できる。
かくして、本発明の方法に従い製造される前記
式()の化合物又はその塩は、それ自体公知の
手段、例えば再結晶、蒸留、カラムクロマトグラ
フイー、薄層クロルマトグラフイー等の方法によ
り、反応混合物から単離し及び/又は精製するこ
とができる。
以上に説明した本発明の式()で表わされる
置換ベンジルアミン誘導体及びその塩は、優れた
ヒスタミンH2受容体拮抗作用にもとづく胃酸分
泌抑制作用を有し、胃酸に起因する疾病、たとえ
ば胃又は十二指腸潰瘍の治療に極めて有用な化合
物である。
本発明の式()で表わされる化合物が優れた
ヒスタミンH2受容体拮抗作用を有することは以
下の動物実験により立証される。
なお、以下の動物実験に用いた本発明の化合物
は次の記号で代表させる。

(1) モルモツト右心房標本によるヒスタミンH2
受容体拮抗作用の測定
ハートレイ系モルモツト(雄:400〜550g)を
頭部を打撲し放血し、心臓を摘出した。酸素を飽
和したタイロード液内で右心房を剥離し、その両
端に絹糸をつけた。36℃に保つたタイロード液を
含有し、混合ガス(O295%:CO25%)を通気し
ているマグヌス管(25ml)内に、両端につけた絹
糸を用い張力700mgで心房を懸垂した。心房の収
縮運動をフオース・デイスプレイスメント・トラ
ンスジユーサー(Force−displacement−
transducer)により記録し、心搏数を算出した。
ヒスタミン(二燐酸塩の形で用いる、以下同
じ)を、添加量の対数値が1/2の等間隔となる用
量で、心搏数増加の最大反応が得られるまで、1
×10-8M〜1×10-4M濃度で累加的にマグヌス管
内に加え、ヒスタミンの用量反応曲線
(Doseresponse curve)を得た。マグヌス管内を
数回洗浄し、心房を1時間安定させた後再び前述
の操作を繰り返し、ヒスタミンの用量反応曲線を
得た。マグヌス管内を数回洗浄後、組織を50分間
安定させた。次いで、試験化合物(1×10-5M)
をマグヌス管内に加え、20分後に試験化合物存在
下におけるヒスタミンの用量反応曲線を得た。
第2回目のヒスタミンの用量反応曲線と第3回
目の試験化合物存在下のヒスタミンの用量反応曲
線から、J.M.Van Rossumの方法(Arch.int.
Pharmacodyn.,143、299、1963)により、各試
験化合物のPA2値(一定反応をおこすのに要する
マグヌス管内のヒスタミン濃度を2倍にするのに
必要な、試験化合物のモル濃度の対数値の負数
(negative logarithm))を算出した。その結果
を下記表−1に示す。

かくして、本発明の式()で表わされる化合
物は、抗潰瘍剤として、人間その他の温血動物に
対する治療、措置のために、経口又は非経口投与
(例えば筋注、静注、皮下投与、直腸投与、経皮
投与など)、好ましくは経口投与することができ
る。
本発明の化合物は、薬剤として用いる場合、経
口又は非経口投与に適した種々の形態に製剤する
ことができる。例えば、本発明の化合物は、この
種薬剤に通常使用される無毒性の賦形剤、結合
剤、滑沢剤、崩壊剤、防腐剤、等張化剤、安定化
剤、分散剤、酸化防止剤、着色剤、香味剤、緩衝
剤等の添加物を使用して製剤することができる。
かかる薬剤は、その用途に応じて、固体形態
(例えば錠剤、硬カプセル剤、軟カプセル剤、顆
粒剤、散剤、細粒剤、丸剤、トローチ錠など)、
半固体形態(例えば坐剤、軟膏など)及び液体形
態(注射剤、乳剤、懸濁液、シロツプ、スプレー
など)のいずれかの製剤形態に調製することがで
きる。しかして、使用し得る無毒性の上記添加物
としては、例えばでん粉、ゼラチン、ブドウ糖、
乳糖、果糖、マルトース、炭酸マグネシウム、メ
タケイ酸アルミン酸マグネシウム、合成ケイ酸ア
ルミニウム、無水ケイ酸、タルク、ステアリン酸
マグネシウム、メチルセルロース、カルボキシメ
チルセルロースまたはその塩、アラビアゴム、ポ
リエチレングリコール、p−ヒドロキシ安息香酸
アルキルエステル、シロツプ、エタノール、プロ
ピレングリコール、ワセリン、カーボワツクス、
グリセリン、塩化ナトリウム、亜硫酸ソーダ、リ
ン酸ナトリウム、クエン酸等が挙げられる。該薬
剤はまた、治療学的に有用な他の薬剤を含有する
こともできる。
該薬剤中における本発明の化合物の含有量はそ
の剤形に応じて異なるが、一般に固体及び半固体
形態の場合には5〜100重量%の濃度で、そして
液体形態の場合には0.1〜10重量%の濃度で該活
性化合物を含有していることが望ましい。
本発明の化合物の投与量は、対象とする人間を
はじめとする温血動物の種類、投与経路、症状の
軽重、医者の診断等により広範に変えることがで
きるが、一般に1日当り、0.2〜80mg/Kg、好適
には、0.5〜50mg/Kgとすることができる。しか
し、上記の如く患者の症状の軽重、医者の診断に
応じて、上記範囲の下限よりも少ない量又は上限
よりも多い量を投与することももちろん可能であ
る。上記投与量は1日1回又は数回に分けて投与
することができる。
以下実施例により本発明をさらに説明する。
実施例 1
cis−4−〔3−〔1−(1−ピロリジニル)エチ
ル〕フエノキシ〕−2−ブテニルアミン(150mg)
およびN−シアノ−1−メチル−2−(フエニル
メチレン)ヒドラジンカルボキシイミドチオ酸メ
チルエステル(140mg)をクロロホルム(3ml)
中で5時間加熱還流した。反応混合物をTLC〔展
開溶媒;クロロホルム:メタノール(19:1)〕
で分離精製して、N′−シアノ−N−〔4−〔3−
〔1−(1−ピロリジニル)エチル〕フエノキシ〕
−cis−2−ブテニル〕−1−メチル−2−(フエ
ニルメチレン)ヒドラジンカルボキシイミドアミ
ド(210mg)を得た。

N′−シアノ−N−〔4−〔3−〔1−(1−ピロ
リジニル)エチル〕フエノキシ〕−cis−2−ブテ
ニル〕−1−メチル−2−(フエニルメチレン)ヒ
ドラジンカルボキシイミドアミド(150mg)をア
セトン(5ml)中で6N塩酸(3ml)とともに1
時間撹拌した。反応混合物に水を加え、エーテル
で洗浄し、アンモニア水で塩基性にし、酢酸エチ
ルで抽出した。抽出物を無水硫酸マグネシウムで
乾燥後、溶媒を留去し、残留物をTLC〔展開溶
媒;クロロホルム:メタノール(9:1)〕で分
離精製して、1−メチル−N5−〔4−〔3−〔1−
(1−ピロリジニル)エチル〕フエノキシ〕−cis
−2−ブテニル〕−1H−1,2,4−トリアゾー
ル−3,5−ジアミン(115mg)を得た。

trans−4−〔3−(1−ピペリジニルメチル)
フエノキシ〕−2−ブテニルアミン(130mg)およ
び2−ニトロアミノ−5−(3−ピリジルメチル)
−4−ピリミジン(120mg)の混合物をエタノー
ル(1ml)中で16時間加熱還流した。反応混合物
をTLC〔展開溶媒;クロロホルム:メタノール
(9:1)〕で分離精製して、2−〔4−〔3−(1
−ピペリジニルメチル)フエノキシ〕−trans−2
−ブテニルアミノ〕−5−(3−ピリジルメチル)
−4−ピリミドン(50mg)を得た。

trans−4−〔3−(1−ピペリジニルメチル)
フエノキシ〕−2−ブテニルアミン(75mg)のメ
タノール(2ml)溶液を氷冷下に1,1−ジ酸化
3,4−ジメトキシ−1,2,5−チアジアゾー
ル(55mg)のメタノール(10ml)懸濁液中へよく
撹拌しながら滴下した。氷冷下で45分間撹拌後、
反応混合物中へ15Nアンモニア水(1ml)を加
え、さらに室温で18時間撹拌した。反応混合物を
減圧下に濃縮し、TLC〔展開溶媒;メタノール〕
で分離精製して、1,1−ジ酸化3−アミノ−4
−〔4−〔3−(1−ピペリジニルメチル)フエノ
キシ〕−trans−2−ブテニルアミノ〕−1,2,
5−チアジアゾール(70mg)を得た。

を室温で12時間撹拌し、得られる反応混合物を
TLC〔展開溶媒;クロロホルム:メタノール
(19:1)〕で分離精製して、N′−シアノ−N−
〔4−〔3−(1−ピペリジニルメチル)フエノキ
シ〕−trans−2−ブテニル〕カルバミイミドチオ
酸メチルエステル(1.7g)を得た。

上記実施例の原料は以下の製造例に従つて合成
した。
製造例 1
cis−4−〔3−(1−ピペリジニルメチル)フ
エノキシ〕−2−ブテニルアミンの合成
A 法
(i) 60%水素化ナトリウム1.15gを乾燥ジメチル
スルホキシド15mlに懸濁させて、3−(1−ピ
ペリジニルメチル)フエノール5.0gを固体の
ままで少しずつ加えて撹拌する。室温にて20分
間撹拌した後、N−(4−クロロ−cis−2−ブ
テニル)フタルイミド6.75gを加えて、室温に
て2時間撹拌する。反応液に氷水を加えエーテ
ルで抽出する。水洗後10%塩酸にて抽出し、水
層を集めてアンモニアでアルカリ性とした後、
クロロホルムで抽出する。水洗後無水硫酸マグ
ネシウムにて乾燥し、溶媒を留去して、N−
〔4−(3−(1−ピペリジニルメチル)フエノ
キシ〕−cis−2−ブテニル〕フタルイミド8.3
gを油状物として得た。

(ii) N−〔4−〔3−(1−ピペリジニルメチルフ
エノキシ〕−cis−2−ブテニル〕フタルイミド
8.3g、抱水ヒドラジン8.3ml及びエタノール83
mlの混合物を室温にて4時間放置する。析出し
たフタラジンを良くほぐして去し、ベンゼン
を加えて減圧下にて溶媒を留去し、不溶物が析
出したら去する。この操作を数回くり返した
後、残留物を減圧蒸留して、cis−4−〔3−
(1−ピペリジニルメチル)フエノキシ〕−2−
ブテニルアミン4.2gを得た。沸点168〜170
℃/0.55mmHg。

B 法
60%水素化ナトリウム1.15gを乾燥ジメチルス
ルホキシド15mlに懸濁させて、3−(1−ピペリ
ジニルメチル)フエノール5.0gを固体のままで
少しずつ加えて撹拌する。室温にて20分間撹拌し
た後、この液をcis−1,4−ジクロロ−2−ブ
テン9.7gと乾燥ジメチルスルホキシド5mlの混
合物中に滴下する。室温にて1時間反応後、氷水
中にあけて、エーテルで抽出する。水洗後エーテ
ル層を10%塩酸にて抽出し、水層を集めて炭酸カ
リウムでアルカリ性とした後、再び、エーテルで
抽出する。水洗後無水硫酸マグネシウムにて乾燥
し、溶媒を留去して、1−クロロ−4−〔3−(1
−ピペリジニルメチル)フエノキシ〕−cis−2−
ブテン3.3gを油状物として得た。このものはそ
のまますぐに次の反応に用いる。
得られた1−クロロ−4−〔3−(1−ピペリジ
ニルメチル)フエノキシ〕−cis−2−ブテン3.3
gを、60%水素化ナトリウム0.53g、乾燥ジメチ
ルスルホキシド7.3ml及びフタルイミド1.92gよ
り製した液の中へ滴下する。
室温に2時間反応後、氷水を加えてエーテルで
抽出する。水洗後無水硫酸マグネシウムにて乾燥
し、溶媒を留去して、N−〔4−〔3−(1−ピペ
リジニルメチル)フエノキシ〕−cis−2−ブテニ
ル〕フタルイミド2.4gを油状物として得た。こ
の化合物はA法(i)で得た化合物と完全に一致し
た。
製造例 2
製造例1のA法において、N−(4−クロロ−
cis−2−ブテニル)フタルイミドの代りにN−
(4−クロロ−trans−2−ブテニル)フタルイミ
ド6.75gを用いて同様に操作して、trans−4−
〔3−(1−ピペリジニルメチル)フエノキシ〕−
2−ブテニルアミン4.0gを油状物として得た。

ニルメチル)フエノールの代りに3−〔1−(1−
ピロリジニル)エチル〕フエノール1.0gを用い
て同様に操作して、cis−4−〔3−〔1−(1−ピ
ロリジニル)エチル〕フエノキシ〕−2−ブテニ
ルアミン0.9gを油状物として得た。

ミノメチルフエノール1.0gを用いて同様に操作
して、cis−4−(3−ジメチルアミノメチルフエ
ノキシ)−2−ブテニルアミン0.7gを油状物とし
て得た。

ニルメチル)フエノールの代りに3−(1−パー
ヒドロアゼピニルメチル)フエノール1.0gを用
いて同様に操作して、cis−4−〔3−(1−パー
ヒドロアゼピニルメチル)フエノキシ〕−2−ブ
テニルアミン0.8gを油状物として得た。

製造例 9
(i) THF140ml中に3−フタルイミノプロピルト
リフエニルホスホニウムブロミド24gと、60%
水素化ナトリウム1.8gを懸濁させて、氷冷下
15分間撹拌した後、3−(1−ピペリジニルメ
チル)ベンズアルデヒド7.0gを加える。室温
にて1.5時間撹拌した後、減圧下に溶媒を留去
し、氷水を加えてエーテルで抽出する。水洗後
10%塩酸にてエーテル層から抽出し、水層を集
めてアンモニアでアルカリ性とした後、クロロ
ホルムで抽出する。水洗後無水硫酸マグネシウ
ムにて乾燥し、溶媒を留去して、N−〔4−〔3
−(1−ピペリジニルメチル)フエニル〕−cis
−3−ブテニル〕フタルイミド9.2gを油状物
として得た。

−2−ブテニル〕フタルイミドの代りにN−
〔4−〔3−(1−ピペリジニルメチル)フエニ
ル〕−cis−3−ブテニル〕フタルイミド9.2g
を用いて同様に操作して得た油状物を減圧蒸留
して、140〜142℃/0.15mmHgのcis−4−〔3−
(1−ピペリジニルメチル)フエニル〕−3−ブ
テニルアミン4.0gを得た。

製造例 10
trans−4−〔3−(1−ピペリジニルメチル)
フエニル〕−3−ブテニルアミンの合成
A 法
(i) 60%水素化ナトリウム2.1gをテトラヒドロ
フラン15ml及び乾燥ジメチルスルホキシド15ml
に懸濁させて、氷冷撹拌下オキシカルボニルエ
チルトリフエニルホスホニウムクロリド10.1g
を固体のままで少しずつ加える。氷冷下に10分
間撹拌後、3−(1−ピペリジニルメチル)ベ
ンズアルデヒド5.0gのテトラヒドロフラン5
ml溶液を加えて室温にて4時間撹拌する。少量
の水を加えた後、減圧下にテトラヒドロフラン
を留去し、氷水30mlを加え、エーテルで振る。
水層を取り、減圧下にほとんどの水を留去す
る。残留物にエーテルを加えて振り、静置して
エーテル層をデカントで除く。この操作を数回
くり返した後に、メタノール20mlを加え、次い
で20%塩酸エーテル溶液25mlを加える。析出し
た不溶物を去した後加熱してエーテルを留去
し、10分間還流する。減圧下にメタノールを留
去し、氷水を加えて、炭酸カリウムにてアルカ
リ性とした後、エーテルで抽出する。水洗後、
芒硝で乾燥させて溶媒を留去する。残留物を
TLC(展開溶媒;クロロホルム:メタノール
(9:1))にて精製して、3−〔3−(1−ピペ
リジニルメチル)フエニル〕−trans−2−ブテ
ン酸メチル3.0gを油状物として得た。

NMR(CDCl3,δ):1.2〜1.9(6H,多重線),
2.2〜2.6(4H,多重線),3.1〜3.4(2H,多重
線),3.49(2H,一重線),3.68(3H,一重

(ii) 乾燥したテトラヒドロフラン50mlに水素化リ
チウムアルミニウム1.2gを懸濁させておいて、
氷冷撹拌下、3−〔3−(1−ピペリジニルメチ
ル)フエニル〕−trans−2−ブテン酸メチル
3.0gの乾燥テトラヒドロフラン10ml溶液を滴
下する。氷冷下にて1時間反応後氷水を滴下
し、塩化ナトリウム3gを加えて不溶物を去
する。液から減圧下に溶媒を留去し、残留物
をTLC(展開溶媒;クロロホルム:メタノール
(9:1))にて精製して、4−〔3−(1−ピペ
リジニルメチル)フエニル〕−trans−3−ブテ
ニルアルコール2.5gを油状物として得た。
IR(液膜,cm-1):3330。
NMR(CDCl3,δ):1.2〜1.9(6H,多重線),
2.1〜2.7(6H,多重線),3.41(2H,一重線),

(iii) 4−〔3−(1−ピペリジニルメチル)フエニ
ル〕−trans−3−ブテニルアルコール2.5gを
チオニルクロリド中1時間還流する。減圧下に
チオニルクロリドを留去し氷水を加え、炭酸カ
リウムにてアルカリ性とした後、エーテルで抽
出する。水洗後芒硝にて乾燥し、溶媒を留去し
て、1−クロロ−4−〔3−(1−ピペリジニル
メチル)フエニル〕−trans−3−ブテン1.75g
を油状物として得た。
NMR(CDCl3,δ):1.1〜1.9(6H,多重線),
2.1〜2.9(6H,多重線),3.43(2H,一重線),
3.4〜3.8(2H,多重線),5.6〜6.7(2H,多重
線),7.0〜7.5(4H,多重線)。
(iv) 1−クロロ−4−〔3−(1−ピペリジニルメ
チル)フエニル〕−trans−3−ブテン1.7g、
フタルイミドカリウム5.1g及びジメチルスル
ホキシド17mlを蒸気バード上で30分間加熱した
後、冷却し、氷水を加え、エーテルで抽出す
る。水洗後、10%塩酸にて抽出し、水層を集め
てアンモニアでアルカリ性とした後、クロロホ
ルムで抽出する。水洗後、無水硫酸マグネシウ
ムにて乾燥し、溶媒を留去する。残留物を
TLC(展開溶媒;クロロホルム:メタノール
(19:1))にて精製して、N−〔4−〔3−(1
−ピペリジニルメチル)フエニル〕−trans−3
−ブテニル〕フタルイミド0.8gを油状物とし
て得た。

NMR(CDCl3,δ):1.1〜1.9(6H,多重線),
2.1〜2.9(6H,多重線)3.42(2H,一重線),

(v) 製造例1のA法(ii)において、N−〔4−〔3−
(1−ピペリジニルメチル)フエノキシ〕−cis
−2−ブテニル〕フタルイミドの代りにN−
〔4−〔3−(1−ピペリジニルメチル)フエニ
ル〕−trans−3−ブテニル〕フタルイミド2.6
gを用いて同様に操作して、trans−4−〔3−
(1−ピペリジニルメチル)フエニル〕−3−ブ
テニルアミン1.3gを油状物として得た。
NMR(CDCl3,δ):1.1〜1.9(6H,多重線),
2.1〜3.0(8H,多重線),3.43(2H,一重線),
5.6〜6.7(2H,多重線),7.0〜7.5(4H,多重

B 法
(i) 乾燥テトラヒドロフラン−乾燥ジメチルスル
ホキシド(1:1)混液37ml中に60%水素化ナ
トリウムを懸濁させておいて、氷水撹拌下オキ
シカルボニルエチルトリフエニルホスホニウム
クロリド15.8gを一度に加える。氷冷下に20分
間撹拌後、3−(エトキシカルボニル)ベンズ
アルデヒド7.5gの乾燥テトラヒドロフラン
(7.5ml)溶液を一度に加える。氷冷下に2分間
撹拌後、室温で3時間撹拌する。氷水100mlと
エーテル300mlを加えて振り、水層を取つて塩
酸酸性とした後エーテル抽出する。水洗後エー
テルを留去して得られた残留物をメタノール80
mlにとかし、塩酸ガスを吹込んで、30分間還元
する。減圧下にメタノールを留去し、エーテル
を加えて抽出する。5%炭酸カリウム水溶液で
振つた後、水洗し、芒硝にて乾燥し、減圧下に
溶媒を留去する。得られた粗オイルをカラムク
ロマト(wakogel−C−200、クロロホルム流
出)にて精製した後減圧蒸留して、沸点150〜
154℃/0.6mmHgの3−〔3−(エトキシカルボ
ニル)フエニル〕−trans−2−ブテン酸メチル
4.5gを得た。
IR(液膜,cm-1):1720。
NMR(CDCl3,δ):1.37(3H,三重線),3.0〜
3.5(2H,多重線),3.70(3H,一重線),3.39
(2H,一重線),4.45(2H,四重線,J=7
Hz),6.0〜7.0(2H,多重線),7.1〜8.3(4H,
多重線)。
(ii) 水素化リチウムアルミニウム10gを乾燥テト
ラヒドロフラン500mlに懸濁させておいて、3
−〔3−(エトキシカルボニル)フエニル〕−−
trans−2−ブテン酸メチル25gの乾燥テトラ
ヒドロフラン(100ml)溶液を氷冷撹拌下、内
温10℃以下で滴下する。室温にて1時間反応
後、氷冷撹拌下内温10℃以下で水50mlを滴下す
る。
室温にて30分間撹拌した後、塩化ナトリウム
50gを加えて不溶物を去する。減圧下に溶媒
を留去した後残留物を減圧蒸留して、沸点167
〜170℃/1mmHgの4−(3−ヒドロキシメチ
ルフエニル)−trans−3−ブテニルアルコール
14gを得た。
IR(液膜,cm-1):3320。
NMR(CDCl3,δ):2.37(2H,四重線,J=
7Hz),3.57(2H,三重線,J=7Hz),4.50
(2H,一重線),5.7〜6.7(2H,多重線),6.9
〜7.5(4H,多重線)。
(iii) 4−(3−ヒドロキシメチルフエニル)−
trans−3−ブテニルアルコール14gをチオニ
ルクロリド50ml中に室温にて少しずつ加える。
還流下にて1時間反応後、減圧下にチオニルク
ロリドを留去する。残留物をエーテルに溶かし
て5%炭酸カリウム水溶液で2回振り、水洗
後、芒硝にて乾燥させて溶媒を留去する。残留
物をカラムクロマト(wakogel−C−200、ク
ロロホルム流出)にて精製して、1−クロロ−
4−(3−クロロメチルフエニル)−trans−3
−ブテン15.5gを油状物として得た。この化合
物を減圧蒸留すると、沸点123〜126℃/0.6mm
Hgにて蒸留されるが、分解が激しく、収率は
低下する。
NMR(CDCl3,δ):2.56(2H,四重線,J=
7Hz),3.59(2H,三重線,J=7Hz),4.54
(2H,一重線),5.7〜6.7(2H,多重線),7.0
〜7.5(4H,多重線)。
(iv) 1−クロロ−4−(3−クロロメチルフエニ
ル)−trans−3−ブテン4.1g、ピベリジン3.3
g及びテトラヒドロフラン41mlの混合物を2時
間加熱還流する。減圧下に溶媒を留去し、エー
テルを加えて析出した不溶物を去する。液
を10%塩酸にて抽出し、水層を集めて、炭酸カ
リウムにてアルカリ性とした後、クロロホルム
抽出する。水洗後無水硫酸ナトリウムにて乾燥
し、溶媒を留去して、1−クロロ−〔3−(1−
ピペリジニルメチル)フエニル〕−trans−3−
ブテン3.7gを油状物として得た。この化合物
は製造例10のA法(iii)で得た化合物と完全に一致
した。
製造例 11
N−(3−ブロモプロピル)フタルイミド100
g、トリフエニルホスフイン100g及びベンゼン
200mlの混合物を一夜還流する。析出した結晶を
取し乾燥して、融点216〜217℃の3−フタルイ
ミノプロピルトリフエニルホスホニウムブロミド
128gを得た。
IR(KBr,cm-1):3400,1770,1700。
NMR(CDCl3,δ):1.8〜2.5(2H,多重線),3.5
〜4.3(4H,多重線),7.5〜8.1(19H,多重線)。
製造例 12
乾燥ジメチルスルホキシド20ml中に60%水素化
ナトリウム2.8gを懸濁させておいて、フタルイ
ミド10gを固体のまま少しずつ加える。室温にて
20分間撹拌した後、この液を1,4−ジクロロ−
cis−2−ブテン16.8gの乾燥ジメチルスルホキ
シド16ml溶液中に室温にて滴下する。室温にて2
時間放置後、氷水中にあけてn−ヘキサン50mlを
加え、撹拌し、放置する。析出した結晶を取し
水洗した後、n−ヘキサンで洗い、湿つた結晶を
エーテル50mlで6回抽出する。エーテル層を芒硝
にて乾燥した後、溶媒を留去する。粗結晶をn−
ヘキサンより再結晶して、融点66.9〜68.6℃のN
−〔4−クロロ−cis−2−ブテニル)フタルイミ
ド5.0gを得た。

製造例 13
製造例12において、1,4−ジクロロ−cis−
2−ブテンの代りに1,4−ジクロロ−trans−
2−ブテン27.2gを用いて同様に操作して得た粗
結晶を、アセトン−n−ヘキサンより再結晶し
て、融点104.5〜105.7℃のN−(4−クロロ−
trans−2−ブテニル)フタルイミド13.2gを得
た。
