Abstract
Description
Claims (12)
- 1.発現に際して検出可能なポリペプチドを産生するインディケーター遺伝子を 含むべく遺伝子工学的に処理されたリポーター細菌において、該インディケータ ー遺伝子の発現が該細菌のバクテリオファージ感染に際して開始されるリポータ ー細菌。
- 2.インディケーター遺伝子がルシフェラーゼである、請求の範囲第1項に記載 のリポーター細菌。
- 3.流体試料または適切な処理により流体となされた試料中のターゲット細菌を 下記工程により検査する方法: a)存在するターゲット細菌を感染させる条件下で試料にバクテリオファージを 添加し、 b)試料中の細胞外バクテリオファージを破壊、除去、中和または不活化し、c )試料をインキュベートして感染を完了させ、ターゲット細菌にバクテリオファ ージを放出させ、そして d)試料中のターゲット細菌を指示するものとしてバクテリオファージをアッセ イする。
- 4.工程d)が下記により実施される、請求の範囲第3項に記載の方法:di) 得られたバクテリオファージを用いて請求の範囲第1項または第2項に記載のリ ポーター細菌を感染させ、そしてdii)リポーター細菌が発現した検出可能な ポリペプチドを、試料中のターゲット細菌を指示するものとして観察する。
- 5.工程c)とd)の間に下記の追加工程が含まれる、請求の範囲第3項または 第4項に記載の方法: ci)許容宿主細菌を用いて試料中のバクテリオファージを増幅する。
- 6.ターゲット細菌が病原体である、請求の範囲第5項に記載の方法。
- 7.工程b)が試料を不飽和脂肪酸の存在下にフォトン照射することにより実施 される、請求の範囲第3項ないし第6項のいずれかに記載の方法。
- 8.工程b)が酢酸を用いて実施される、請求の範囲第3項ないし第6項のいず れかに記載の方法。
- 9.工程a)が周囲温度より低い温度で実施される、請求の範囲第8項に記載の 方法。
- 10.試料中のターゲット細菌を下記工程により検査する方法:a)請求の範囲 第1項または第2項に記載のリポーター細菌を試料に添加し、b)存在するター ゲット細菌および添加されたリポーター細菌を感染させる競合アッセイ条件下で 試料にバクテリオファージを添加し、c)リポーター細菌が発現した検出可能な ポリペプチドを、試料中のターゲット細菌を指示するものとして観察する。
- 11.試料中のバクテリオファージを下記工程により検査する方法:a)存在す るバクテリオファージがリポーター細菌に感染する条件下で請求の範囲第1項ま たは第2項に記載のリポーター細菌を試料に添加し、b)リポーター細菌が発現 した検出可能なポリペプチドを、試料中のバクテリオファージを指示するものと して観察する。
- 12.殺ウイルス薬を下記工程により試験する方法:a)既知濃度の予め定めら れたバクテリオファージを含有する流体を用意し、b)流体を殺ウイルス薬によ り処理し、c)処理された流体に、生存バクテリオファージが感染する条件下で 、請求の範囲第1項または第2項に記載のリポーター細菌を添加し、d)標識細 菌が発現した検出可能なポリペプチドを、殺ウイルス処理の有効性を指示するも のとして観察する。
