Description
Claims (12)
- 1.タンパク質を、処理されるタンパク質がその天然配座をとることができるp Hで、少なくとも400ミリモル/lの濃度のトリス塩基及び/またはトリスの 塩の溶液とインキュベートする、変性タンパク質の再活性化法。
- 2.インキュベーション溶液のpH値がトリスと異なる緩衝物質により調節され る請求項1に記載の方法。
- 3.トリスがまたpH値を調節するのに使用される請求項1に記載の方法。
- 4.0.4〜2モル/lのトリス溶液が使用される請求項1〜3の一つに記載の 方法。
- 5.0.2〜1.0モル/lのアルギニンがインキュベーション溶液に添加され る請求項1〜4の一つに記載の方法。
- 6.組換えtPAまたはtPA突然変異タンパク質が処理される請求項5に記載 の方法。
- 7.ドメイン組成K2Pを有する非グリコシル化tPA突然変異タンパク質が処 理される請求項6に記載の方法。
- 8.変性タンパク質が復元溶液に連続的またはバッチ式に添加される請求項1〜 7の一つに記載の方法。
- 9.パルス復元法が復元リアクター中で行われる請求項8に記載の方法。
- 10.復元が約1モル/lのトリス濃度のアルギニンを含む緩衝液中で行われ、 そしてタンパク質濃度がパルス当たり約200μg増加される請求項9に記載の 方法。
- 11.組換えG−CSFが処理される請求項1〜5または8〜10の一つに記載 の方法。
- 12.抗体またはそれらのフラグメントが処理される請求項1〜5または8〜1 0の一つに記載の方法。
