Abstract
Description
Claims (22)
- 1.遺伝子的に変換された微生物において、微生物での遺伝子変換に関連する形 質を修飾することができる哺乳類遺伝子を検出する方法であって、下記工程を含 む:(a)遺伝子的に変換された微生物において、哺乳類cDNAを発現するこ とができる発現ベクターに哺乳類cDNAを備え、(b)遺伝子的に変換された 微生物に発現ベクターを導入し、これにより、発現ベクターを含む遺伝子的に変 換された微生物を生成し、 (c)発現ベクターを含む遺伝子的に変換された微生物を、前記微生物の生育に 適切な条件下に維持し、および(d)微生物の遺伝子変換に換連のある形質変換 が修飾された遺伝子的に変換された微生物を同定する。
- 2.前記発現ベクターが、前記哺乳類cDNAと操作的に関連があるプロモータ ーDNA配列を含み、前記プロモーターDNA配列が、前記微生物に内因性であ る、請求の範囲第1項に記載の方法。
- 3.前記発現ベクターが、pADNS、PADANS、PAAUNおよびpAA UN−ATGからなるグループから選択される、請求の範囲第2項に記載の方法 。
- 4.前記微生物が、酵母および哺乳類細胞からなるグループから選択される、請 求の範囲第1項に記載の方法。
- 5.前記微生物が、S.cerevisiaeおよびS.pombeからなるグ ループから選択される酵母微生物である、請求の範囲第4項に記載の方法。
- 6.前記微生物が,S.cerevisiae株TK161−R2V、S.ce revisiae株10DAB、S.cerevisiae株SKN37および S.pombe株SP65からなるグループから選択される、請求の範囲第5項 に記載の方法。
- 7.前記微生物が、酵母微生物であり、前記形質変換が、熱ショック感受性、窒 素欠乏、標準量グリコーゲン合成不能、酢酸塩での生育不能、および胞子形成不 能からなるグループから選択される、請求の範囲第1項に記載の方法。
- 8.前記微生物での遺伝子変換が、活性化、阻害、あるいは環状ヌクレオチド・ ホスホジエステラーゼが関与する細胞反応の転写減衰の結果に帰するものである 請求の範囲第1項に記載の方法。
- 9.前記遺伝子変換が、RAS−関連タンパク質をコードする遺伝子の変換であ る請求の範囲第1項に記載の方法。
- 10.工程(d)で同定された微生物から前記哺乳類cDNAを単離することを さらに含む、請求の範囲第1項に記載の方法。
- 11.哺乳類RAS−関連タンパク質ポリペプチドをコードし、プラスミドpJ C99(A.T.C.C.68599)、pJC265(A.T.C.C.68 598)、pJC310(A.T.C.C.68597)、PML5(A.T. C.C.68593)、pATC16(A.T.C.C.68599)、および pATG29(A.T.C.C.68591)にある哺乳類cDNA挿入体から なるグループから選択されるDNA配列から本質的に成る、精製され、単離され たDNA配列。
- 12.哺乳類RAS−関連タンパク質ポリペプチドをコードし、請求の範囲第1 1項に記載のDNA配列と正確なハイブリダイゼーション条件下でハイプリダイ ズする、DNA配列から本質的に成る、精製され、単離されたDNA配列。
- 13.変性コドンの手段により、請求の範囲第11項もしくは第12項に記載の DNA配列によりコードされたポリペプチドをコードするDNA配列から本質的 に成る、精製され、単離されたDNA配列。
- 14.請求の範囲第11項、第12項、もしくは第13項に記載のDNA配列の 、原核細胞あるいは真核細胞での発現のポリペプチド生成物。
- 15.哺乳類環状ヌクレオチド・ホスホジエステラーゼをコードし、プラスミド pRATDPD(A.T.C.C.68586)、pJC44c(A.T.C. C.68603)、PTM3(A.T.C.C.68600)、PTM72(A .T.C.C.68602)、pPDE21(A.T.C.C.68595)、 pGB18ARR(A.T.C.C.68596)、pGB25(A.T.C. C.68594)、およびpTM22(A.T.C.C.68601)にある哺 乳類cDNA挿入体からなるグループから選択されるDNA配列から本質的に成 る、精製され、単離されたDNA配列。
- 16.哺乳類環状ヌクレオチド・ホスホジエステラーゼをコードし、請求の範囲 第15項、および配列番号;33,34,35,37および41に記載のDNA 配列から選択されたDNA配列と正確なハイブリダイゼーション条件下でハイプ リダイズするDNA配列から本質的に成る、精製され、単離されたDNA配列。
- 17.変性コドンの手段により、請求の範囲第15項もしくは第16項に記載の DNA配列によりコードされたポリペプチドをコードするDNA配列から本質的 に成る、精製され、単離されたDNA配列。
- 18.請求の範囲第15項、第16項、もしくは第17項に記載のDNA配列の 、原核細胞あるいは真核細胞での発現のポリペプチド生成物。
- 19.遺伝子的に変換された微生物において発現する際に、微生物における遺伝 子変換と関連する形質変換を修飾する、哺乳類遺伝子の発現生成物の活性を変換 する化学薬品の同定の方法であって、下記工程を含む: (a)遺伝子的に変換された微生物において、哺乳類遺伝子を発現し、これによ り、遺伝子変換と関連する形質変換を修飾し、(b)工程(a)の遺伝子的に変 換された微生物と、形質検定に適切な条件下で、検定される化学薬品を接触させ 、および(c)工程(a)で修飾された遺伝子変換と関連する形質変換が逆転を したか否かを決定するものであり、前記形質変換の逆転は、哺乳類遺伝子の生成 物を阻害する化学薬品で表示する。
- 20.前記微生物が、酵母微生物である、請求の範囲第19項に記載の方法。
- 21.環状ヌクレオチド・ホスホジエステラーゼの酵素活性を修節する、化学薬 品の同定の方法であって、下記工程を含む:(a)配列番号;15もしくは16 に記載のDNA配列で、前記DNA配列を発現次第、感受性に変換する形質を有 する、原核宿主細胞あるいは真核宿主細胞を、安定裏に形質転換し、(b)工程 (a)で形成した宿主細胞を、栄養培地で、宿主細胞形質に対応する変換を伴っ て、前記宿主細胞中で前記DNA配列の発現を許容する条件下で生育し、 (c)変換された形質を有する前記宿主細胞を同定し、(d)前記宿主細胞を粉 砕し、 (e)前記粉砕した宿主細胞からサイトソルを単離し、(f)前記サイトソルと 前記化学薬品を接触させ、および(g)前記DNAの発現物質の酵素活性が、変 換されたか否かを決定する。
- 22.前記宿主細胞が、酵母宿主細胞である、請求の範囲第21項に記載の方法 。
