Description

【発明の詳細な説明】 本発明はCSF(コロニー刺激因子)抑制物質に
関する。
本発明者等はヒトの甲状腺癌細胞より樹立した
CSF産生細胞株T3M−5株を培養している際、
CSFの高濃度含有液は濃度に依存したCSF活性を
示さず、低下傾向が見ろれることに気づき、この
知見に基づき検討の結果、CSF抑制物質の分離に
成功し本発明を完成するに至つた。
本発明のCSF抑制物質は次の理化学的性質を有
する。
(1) 分子量:ゲル過法で5〜6万セアデツクス
G−75(フアルマシア社製)を用いたゲル過
法によつた。
カラムサイズ;直径10mm、高さ1000mm ペツト体積;78.5ml 流速;4.0ml/hr 溶出溶液;0.15MNacl含有0.01M−トリス塩酸
緩衝液(pH7.4) 以下に示す標準蛋白の溶出液量から検量線を求
め、CSF抑制物質の溶出液量から分子量を決定し
た。
γ−Glcbulin 160000 Albumin 66000 Ovalbumin 45000 Chymotrypsinogen A 25700 Cytochrome C 12400 (2) 等電点:pH4.7 セフアデツクスIEF(フアルマシア社製)を
用いたフラツトベツト等電点電気泳動法によつ
た。Phfrmalyte(フアルマシア社製)を用いて
8ワツトで45分間前泳動してpH勾配(2.5〜
5.0)を作成したのち、CSF抑制物質(144mg)
を添加し60ワツトで5時間泳動させてCSF活性
抑制物質作用を調べ、等電点を決定した。
(3) pH安定性:pH5〜9で安定。pH3以下およ
び11以上で失活する。
0.01M−トリス塩酸緩衝液中にCSF抑制物質
を加え、氷冷下24時間放置したのち、0.01M−
トリス塩酸緩衝液(pH7)に対して1昼夜透析
後CSF抑制活性を調べた。
(4) 紫外部吸収:280nmに局大吸収を有する。
(5) 酵素反応:DNase(3.175IU)、RNase
(761U)及びトリプシン(0.2%)では失活せ
ず、プロナーゼ(科研化学製)(0.5IU)で完全
に失活する。
(6) 温度安定性:37℃で安定56℃、30分でほぼ失
活。
このような性質は従来よりCSF抑制物質として
知られるPG−E、chalone、ラクトフエリン、血
清中のリポ蛋白、インターフエロン等とはその分
布、分子量、等電点等においてそれぞれ相違し、
新規物質であることを示している。また、培養時
添加するFCSそのものを後記実施例と同様に処理
しても抑制物質は検出できなかつたので、本抑制
物質が血清由来のものでないことは明らかであ
る。
本発明のCSF抑制物質は上記性質の他にCSFに
依存せず分裂増殖を続けれヒトの白血病細胞株K
−562に対して正常細胞には影響を与えない極め
て低濃度でそのコロニー形勢を抑制するという性
質を有し抗白血病剤としても期待される。
実施例 ヒト甲状腺癌より株化されたCSF産生細胞であ
るT3M−5細胞(この細胞は1981年日本血液学
会に於いてCSFの生産性について発表され、東京
大学第三内科、財団法人実験動物中央研究所及び
中外製薬株式会社研究所等に保存されている。)
を1.0〜2.0×107コずつ10%FCS含有F−10培地
500m1に懸濁し、ローラボトル中37℃、で1週
間回転培養した(0.5rpm)。
同時に培養した20本のローラボトル中の培養上
漬を合し(約101)濃縮後、0.01%Tween20及び
0.15MNacl含有0.01Mトリス塩酸緩衝液pH7.4で
膨潤・平均化したセフアデツクスG−75(フアル
マシア社製)によりゲル過し、分子量5〜6万
の画分を分取する。
得られた画分をDEAEセルフアロースCL−6B
に吸着させ、塩濃度を0.04M、0.08M及び0.15M
と段階的に上昇させて溶出する。次いで0.5M−
Nacl含有0.1Mクエン酸緩衝液(pH5)で溶出さ
れる画分をCSF抑制物質画分とした。
このようにして得られたCSF抑制物質画分7ml
中の蛋白量は1000mgであつた。
十分量のCSF存在下でのマウス骨髄細胞を用い
るコロニーアツセイ法で本画分の蛋白量約1200μ
g添加によつて90%阻害が見られたた。尚、ヒト
骨髄細胞に対しては約120μg2¥90%阻害が見
られた。

  1. 【特許請求の範囲】 1 CSF産生の培養液中に蓄積され、次の理化学
    的性質を有するCSF抑制物質。 1 分子量:ゲル過法で5〜6万 2 等電点:pH4.7 3 pH安定性:pH5〜9で安定。pH3以下およ
    び11以上で失活する。 4 紫外部吸収:280nmに極大吸収を有する。 5 各種酵素に対する安定性:DNase
    (3.175IU)、RNase(76IU)及びトリプシン(0.2
    %)では失活せず、プロナーゼ(科研化学製)
    (0.5IU)で完全に失活する。 6 温度安定性:37℃で安定56℃、30分でほぼ失
    活。
