Claims (18)
- 1.単子葉植物、特にイネ科植物、特定的にはトウモロコシ、コムギ、イネ、エ ンバク、オオムギ、モロコシ、ライムギ及びアワのような穀類植物、より特定的 にはトウモロコシをDNA、ゲノム特に核ゲノムで形質転換するための方法であ って、a)緻密な胚形成カルスを形成することが可能な前記植物の無傷組織又は 前記植物の前記無傷組織から得られた緻密な胚形成カルス、特にその胚形成セク ターを損傷及び/又は分解させ、i)前記DNAの取り込み、ii)前記植物ゲ ノムにおける前記DNAの組込み的形質転換及びiii)前記細胞からの前記植 物の再生に関して前記無傷組織又はカルスの細胞をコンピテントにする段階と、 b)前記コンピテント細胞を前記DNAで形質転換する段階と、次にc)形質転 換された前記コンピテント細胞から正常表現型稔性成熱植物のような正常表現型 植物として前記植物を再生させる段階とを含む方法。
- 2.最大寸法0.1〜5mm、好ましくは1〜2.5mm、特に1.25〜1. 75mmに切断することにより前記無傷組織を損傷させ、こうして生成された前 記コンピテント細胞を形質転換することを特徴とする請求項1に記載の方法。
- 3.前記無傷組織を切断することにより損傷させた後、酵素で処理することによ り分解し、好ましくは前記無傷組織の細胞壁に穴をあけ、次にこうして生成され た前記コンピテント細胞を形質転換することを特徴とする請求項1又は2に記載 の方法。
- 4.前記植物がトウモロコシであり、無傷未成熟トウモロコシ胚を酵素で処理す ることにより分解し、好ましくは前記胚の細胞壁に穴をあけ、次にこうして生成 された前記コンピテント細胞を形質転換することを特徴とする請求項1に記載の 方法。
- 5.前記カルスを最大長0.5〜2.5mm、特に1〜2mm、特定的には1. 25〜1.75mm、及び好ましくは最小長約0.1mmに切断することにより 損傷させ、次いでこうして生成された前記コンピテント細胞を形質転換すること を特徴とする請求項1に記載の方法。
- 6.前記カルスを切断することにより損傷させた後、酵素で処理することにより 分解し、好ましくは前記カルスの細胞壁に穴をあけ、次いでこうして生成された 前記コンピテント細胞を形質転換することを特徴とする請求項1又は5に記載の 方法。
- 7.前記カルスを酵素で処理することにより分解して前記カルスの細胞壁に穴を あけ、次いでこうして生成された前記コンピテント細胞を形質転換することを特 徴とする請求項1に記載の方法。
- 8.前記コンピテント細胞を直接遺伝子移入、特にエレクトロボレーションによ り形質転換することを特徴とする請求項1から7のいずれか一項に記載の方法。
- 9.植物中で発現可能なDNA、特にキメラ遺伝子で組込み的に形質転換された ゲノム、特に核ゲノムを有するイネ科植物、特定的には穀類、より特定的にはト ウモロコシ又はイネであって、Dattaら(1990) Bio/Techn ology 8:736、Shimamotoら(1989) Nature 338:274、Gordon−Kammら(1990) The PIant Cell 2:603及びFrommら(1990) Bio/Techno logy 8:833により記載されているような従来の培養条件下においては 、1)前記DNAの取り込み、2)前記植物ゲノムヘの前記DNAの組込み的形 質転換及び3)前記DNAで形質転換された正常表現型稔性植物としての前記植 物の再生に関してコンピテントな前記植物の胚形成懸濁液培養物又はプロトプラ ストを得ることが実質的に不可能であることを特徴とする前記植物。
- 10.特定系統の植物であって、特に該系統の未形質転換プロトプラスト100 00当たり約500以下、特に約100以下、特定的には約10以下、より特定 的には約1以下しか正常表現型植物を再生できない場合に、前記培養条件下では 該系統の形質転換胚形成懸濁液培養物又は形質転換プロトプラストから該系統の 正常表現型稔性植物のような正常表現型植物として植物を再生することが実質的 に不可能であることを特徴とする請求項9に記載の植物。
- 11.請求項1から8のいずれか一項に記載の方法により作出される形質転換植 物、特に単子葉植物、特定的にはイネ科植物、より特定的にはトウモロコシ又は イネのような穀類。
- 12.タイプIカルスを形成する能力を有するが、Gordon−Kammら( 1990)The PIantCell 2:603及びFrommら(199 0) Bio/Technology 8:833により記載されている培養条 件下では、10%以上、特に1%以上、特定的には0.1%以上、より特定的に は0.01%以上の頻度でのタイプIIカルスを形成する能力をもたないことを 特徴とする請求項9から11のいずれか一項に記載のトウモロコシ植物。
- 13.前記DNAで組込み的に形質転換された請求項9から12のいずれか一項 に記載の植物の細胞又は複数の該細胞から本質的に構成される培養物。
- 14.請求項9から12のいずれか一項に記載の形質転換植物の種子。
- 15.ハイブリッド植物の作出手段のような授粉植物であることを特徴とする請 求項9から12のいずれか一項に記載の植物。
- 16.請求項15に記載の授粉植物と別の近交系植物とを交配することにより得 られるハイブリッド。
- 17.タンパク質が発現される植物細胞を死滅又は不能にすることが可能な該タ ンパク質をコードし且つタペート特異的PTA29プロモーターの制御下にある コーディング配列で形質転換された雄性不稔性単子葉植物、好ましくはイネ科植 物、特にトウモロコシ植物、特定的には請求項12に記載のトウモロコシ植物、 又は雄性不稔性植物の種子。
- 18.請求項17に記載の形質転換単子葉植物の細胞又は複数の該細胞から本質 的に構成される培養物。
