Abstract
Description
Claims (29)
- 1.可溶性媒介物質又は感染性粒子の成分のような分子の生細胞による産生を評 価するための方法であって、細胞培養及び場合により刺激が上記産生を誘導する ために行われ、産生された分子の量がインビトロで固相免疫アッセイにより評価 される、即ち: a)細胞培養及び適宜に刺激が、アッセイされる分子に特異的な捕捉成分が結合 されたアッセイの固相を組み込んだ容器において、場合によりトレーサーの存在 下で行われる;及び b)培養期間が経過したとき、固相の洗浄とアッセイに要される様々な他の可能 な試薬、特に固相存在下におけるトレーサー用基質のインキュベート後に、固相 に結合されたトレーサー又はその上記基質により放射されたシグナルが測定され る; ことを特徴とする方法。
- 2.ステップb)においてトレーサー及び固相が培養に用いられた容器中でイン キュベートされる、請求項1に記載の方法。
- 3.放射されたシグナルが培養に用いられた容器中で測定される、請求項1又は 2に記載の方法。
- 4.固相が培養容器の内表面からなり、それにアッセイされる分子に特異的な捕 捉成分が結合されている、請求項1〜3のいずれか一項に記載の方法。
- 5.固相が細胞培養容器と物理的に依存していない1以上の成分からなる、請求 項1〜3のいずれか一項に記載の方法。
- 6.固相が、アッセイされる分子に特異的な捕捉成分で被覆されたビーズ又は把 持できる部分に堅固に付着された成分からなる、請求項5に記載の方法。
- 7.いくつかの分子がアッセイされる、即ち:a)細胞培養がアッセイされる分 子が存在する多数の容器で同時に行われる; b)各容器に組み込まれた固相に、各々が上記分子の1つを特異的に認識するい くつかの抗体群が結合される;及び c)各分子が各対応分子に特異的な異なるトレーサーと共に各容器内でインキュ ベート後にアッセイされる;請求項1〜6のいずれか一項に記載の方法。
- 8.いくつかの分子がアッセイされる、即ち:3)細胞培養が分子が存在する多 数群の容器で同時に行われる; b)各群の容器に組み込まれた固相に、各群の容器において異なる分子を各々認 識する既定特異性の抗体が結合される;及び c)各分子が、対応分子に特異的なトレーサー又は異なる分子に特異的なトレー サーの混合物である共通トレーサーと共に各容器のインキュベート後にアッセイ される;請求項1〜6のいずれか一項に記載の方法。
- 9.免疫アッセイが固相競合タイプ又はサンドイッチタイプである、請求項1〜 8のいずれか一項に記載の方法。
- 10.捕捉成分がモノクローナル又はポリクローナル抗体からなる、請求項1〜 9のいずれか一項に記載の方法。
- 11.捕捉成分がアッセイされる分子に特異的な抗原又は可溶性レセプターのリ ガンドを含めたいずれか他のリガンドである、請求項1〜10のいずれか一項に 記載の方法。
- 12.アッセイが酵素もしくはアイソトープ法、発光、ケイ光又は比濁回折によ り視覚化される、請求項1〜11のいずれか一項に記載の方法。
- 13.アッセイのトレーサー成分が支持体での培養開始時から存在するか又はそ の後に加えられる、請求項1〜12のいずれか一項に記載の方法。
- 14.測定される媒介物質がサイトカイン、ホルモン、神経ペプチド、可溶性レ セプター、脱顆粒マーカー、細胞外ペプチド又はインビトロで細胞により産生さ れる生物学的効果のいずれか他の媒介物質である、請求項1〜13のいずれか一 項に記載の方法。
- 15.感染粒子の成分がウイルス、その他の細胞内微生物又はそれら抗原成分の 1つからなる、請求項1〜13のいずれか一項に記載の方法。
- 16.媒介物質の産生が自発的であるかあるいはレクチン、リポ多糖、抗体、抗 原、アレルゲン、薬剤及び細胞レセプターの天然又は合成リガンドのような特異 的又はそうでない活性化剤で刺激される、請求項1〜15のいずれか一項に記載 の方法。
- 17.アッセイされる分子に関するトレーサー又は捕捉成分としてアッセイに関 与しかつ固相に結合されている抗体が、上記分子の産生及び/又はその生物活性 に影響を与えないエピトープによりアッセイされる分子を認識する、請求項1〜 16のいずれか一項に記載の方法。
- 18.培養中にアッセイされる分子の産生を調節する剤が培地に加えられる、請 求項1〜17のいずれか一項に記載の方法。
- 19.アッセイの減感化により区別され、アッセイされる分子に特異的なトレー サー抗体と同特異性の未標識抗体との混合物により得られ、減感化により既定の 補正率でアッセイの範囲を高い値まで広げることができる、請求項1〜18のい ずれか一項に記載の方法。
- 20.アッセイされる細胞サンプルが、アッセイされる分子に特異的なトレーサ ー抗体を含む第一培地とアッセイされる分子に特異的なトレーサー抗体及び同特 異性の未標識抗体を含む第二培地で同時に培養される、請求項1〜19のいずれ か一項に記載の方法。
- 21.細胞が場合により希釈された全血、精製された細胞群、組織から抽出され た細胞又は細胞系で代表される、請求項1〜20のいずれか一項に記載の方法。
- 22.シグナルを測定する前における固相洗浄用溶液がタンパク質分解酵素、特 にトリプシンの溶液である、請求項1〜21のいずれか一項に記載の方法。
- 23.請求項1〜22のいずれか一項に記載された方法に適し、免疫捕捉用の容 器としても役立っ細胞培養用の支持容器を含み、免疫アッセイの捕捉成分が結合 されたその固相が上記容器に組み込まれるか又は組み込むことができる技術フォ ーマット又はキット。
- 24.固相が培養支持容器の内表面からなる、請求項23に記載のフォーマット 。
- 25.固相支持容器が、滴定プレートの、場合により除去しうるウェルからなる か、又は免疫アッセイに適したチューブもしくはいずれか他の細胞培養容器から なる、請求項24に記載の技術フォーマット。
- 26.細胞培養及び免疫捕捉用の容器がアッセイの固相に物理的に依存していな い、請求項23に記載の技術フォーマット。
- 27.標準サンプル、トレーサー、適宜にその基質と培地、細胞希釈培地、洗浄 培地及び測定されるパラメーターの産生を刺激又は調節する剤を含めたアッセイ の全成分が供給される、請求項23〜26のいずれか一項に記載の技術フォーマ ット。
- 28.マイクロプレートのウェル中に乾燥形で存在するトレーサー及び/又は活 性化剤を含む、請求項23〜27のいずれか一項に記載のフォーマット。
- 29.容器、特にマイクロプレートのウェル中に乾燥形で存在する標準参照サン プルを含む、請求項23〜28のいずれか一項に記載のフォーマット。
