Claims (37)
- 1.油体を標的化することができ、次式pp1−aa25−aa2■−V−V− T−L−aa21−p−AAAT aa34−G−G−aa3■−L−L−aa3■−L−aa41−M G−I−aa44−L−aa4■−aa47−T−L−I−SLSVV aa■1−L−aa■3−V−A−T−P−L−aa■■−L−VV L−F−S−P−V−L−V−P−A−A−L−aa73−1ILI aa74−aa75−aa76−aa77−aa78−G−F−L−GL S−S−aa■7−G−V−aa■■−aa■0−L−S−TIIT aa93aa94−S−aa96−aa97−aa98−aa99−T aa100−a101−PP2 を有しているポリペプチドであって、ただしこのペプチドが、にんじんからの1 6kdのオレオジンまたはトウモロコシからの16kdまたは18kdのオレオ ジンを自然発生させる全長以外であることを特徴とするポリペプチド。

- 6.(a)油体への榛的化をもたらすのに十分なオレオジンまたはその一部と、 (b)目的のポリペプチドとを暗号化するDNA配列を含有するDNA構成体。
- 7.前記DNA配列と操作可能に会合している少なくとも一種の調節配列を含有 するベクターDNAであって、このDNA配列が、宿主細胞中の前記DNAを複 製させることができるベクターDNA:を更に有する、請求項6記載のDNA構 成体。
- 8.前記調節配列が、更に宿主細胞中で前記DNAを形質発現させることができ る、請求項7記載のDNA構成体。
- 9.前記DNAがcDNAである、請求項6記載のDNA構成体。
- 10.成分として、転写の方向に、種子中で形質発現した遺伝子の領域5′から 翻訳開始部位への相当部分を有しており、種子中でのDNA配列の形質発現をも たらす第一のDNA配列;油体の標的化をもたらすオレオジンまたはその相当部 分を暗号化する第二のDNA配列であって、この第二のDNA配列が、少なくと も一種の天然または合成の認識部位を含んでおり;および翻訳および転写停止領 域を有しており、ここで前記の成分が操作可能に連鎖しており、前記第二のDN A配列の形質発現が、前記第一のDNA配列によって調節されている、形費発現 カセット。
- 11.前記第一のDNA配列が、シリアルまたは穀類の種子細胞中で形質発現し た遺伝子からである、請求項10記載の形質発現カセット。
- 12.「Aradopsisthaliana」のゲノムが、前記第一のDNA 配列および前記第二のDNA配列の少なくとも一種を含有している、請求項10 記載の形費発現カセット。

- 14.油体タンパク質(OBP)遺伝子であって、種子中でこの遺伝子の形質発 現をもらたす、領域5′から翻訳開始部位への相当部分を有しており、かつ前記 OBP遺伝子のちょうど5′からメチオニン開始コドンまでと5′から翻訳停止 信号までとの間に、少なくとも1つの制限部位を有している、油体タンパク質遺 伝子: を含有している、形質発現カセット。
- 15.前記OBP遺伝子によって読み取り枠内の前記制限部位中へと挿入された 目的のポリペプチドを暗号化するDNA配列を更に有する、請求項14記載の形 質発現カセット。
- 16.前記制限部位が、合成の制限部位である、請求項14記載の形費発現カセ ット。
- 17.前記制限部位中へと挿入されたプロテアーゼ認識部位のオリゴヌクレオチ ドアダプター暗号を更に有している、請求項14記載の形質発現カセット。
- 18.前記プロテアーゼがコラゲナーゼである、請求項17記載の形質発現カセ ット。
- 19.油体タンパク質(OBP)遺伝子中への読み取り=鎖内に挿入された目的 のポリペプチドを暗号化し、油体タンパク質遺伝子が種子中でのこの遺伝子の形 質発現をもたらすためにこのOBP遺伝子の調節領域5′から翻訳開始部位への 相当部分を存している、第一のDNA配列を有している形質発現カセットであっ て、この配列が、前記調節領域中で形質発現されるような前記遺伝子中の部位に 挿入されている、形質発現カセット。
- 20.プロテアーゼ認識部位5′から前記第一のDNA配列を暗号化する第二の DNA配列を更に有しており、この第二のDNA配列が、前記第一のDNA配列 および前記OBP遺伝子と共に読み取り枠内に存在している、請求項19記載の 形質発明カセット。
- 21.種子中で目的のポリペプチドを形質発現させる方法であって:ゲノム組み 込み条件下で形質発現カセットによって宿主植物細胞を形質転換させ、ここでこ の形質発現カセットが、成分として、転写の方向に、種子中で形質発現した遺伝 子の領域5′から翻訳開始部位への相当部分を有しており、種子中でのDNA配 列の形質発現をもたらす第一のDNA配列;油体の標的化をもたらすオレオジン またはその相当部分を暗号化する第二のDNA配列であって、この第二のDNA 配列が、少なくとも一種の天然または合成の認識部位を含んでおり、この認識部 位の中へと、目的のポリペプチドを暗号化する第三のDNA配列が読み取り枠内 に挿入されており;および翻訳および転写停止領域を有しており;ここで前記の 成分が操作可能に連鎖しており、前記第二のDNA配列の形質発現が、種子中で の形質発現のために前記第一のDNA配列によって調節されている、種子中で目 的のポリペプチドを形質発現させる方法。
- 22.「Thalianaaridopsis7」のゲノムが、前記第一および 前記第二のDNA配列のうち少なくとも一方を有している、請求項21記載の方 法。
- 23.種子中で目的のポリペプチドを形質発現させる方法であって;ゲノム組み 込み条件下でDNA構成体によって宿主植物細胞を形質転換させ、ここでこのD NA構成体が、油体タンパク質(OBP)遺伝子中へと読み取り枠内で挿入され た目的のポリペプチドを暗号化する第一のDNA配列を有しており、油体タンパ ク質遺伝子が、種子中での前記遺伝子の形質発現をもたらす前記OBP遺伝子の 調節領域5′から翻訳開始部位までの相当部分を有しており、ここで前記配列が 、前記調節領域下で形質発現されるような前記遺伝子中の部位に挿入されており 、これによって前記DNA構成体が前記植物細胞のゲノム中へと組み込まれ始め ;および 前記植物を成長させて種子を生じさせ、これによって目的のポリペプチドを、前 記OBP遺伝子の形質発現産物と共に融合タンパク質として形質発現させる種子 中で目的のポリペプチドを形質発現させる方法。
- 24.前記種子の細胞中で油体から前記融合タンパク質を分離することを更に含 む、請求項23記載の方法。
- 25.前記分離が: 前記種子の細胞を溶菌させて前記油体を放出させ;および前記油体を破壊するこ とによって前記触合ポリペプチドを放出させることを含む、請求項24記載の方 法。
- 26.前記分離が: 目的の前記ポリペプチドのN−末端の前に位置する前記融合ポリペプチド中のプ ロテアーゼ認識部位を認識しうるプロテアーゼに対して前記融合ポリペプチドを 接触させることを更に含んでいる、請求項25記載の方法。
- 27.前記接触に先立って、前記OBP遺伝子の形質発現産物に対して結合しう る抗体を有する固体担体に対して前記融合タンパク質を結合させることを更に含 んでいる、請求項26記載の方法。
- 28.目的の精製ポリペプチドを得る方法であって:ゲノム組み込み条件下でD NA構成体によって宿主植物細胞を形質転換させ、ここでこのDNA構成体が、 油体タンパク質(OBP)遺伝子中へと読み取り枠内で挿入された目的のポリペ プチドを暗号化する列一のDNA配列を有しており、油体タンパク質遺伝子が、 種子中での前記遺伝子の形質発現をもたらす前記OBP遺伝子の調節領域5′か ら翻訳開始部位までの相当部分を有しており、ここで前記DNA配列の形質発現 が前記調節領域によって調節されるように、前記配列が前記遺伝子中の部位に挿 入されており、これによって前記DNA構成体が前記植物細胞のゲノム中へと組 み込まれ始め;前記植物を成長させて種子を生じさせ、これによって目的の前記 ポリペプチドを、前記OBP遺伝子の形質発現産物と共に融合タンパク質として 形質発現させ; 種子の細胞から油体を分離し; この油体を破壊することによって前記融合タンパク質を放出させ;および目的の 前記ポリペプチドを精製する ことを含んでいる方法。
- 29.目的の前記ポリペプチドが、植物ゲノムによって暗号化されたポリペプチ ド以外である、請求項28記載の方法。
- 30.目的の前記ポリペプチドが、油体中に天然に存在しているポリペプチド以 外である、請求項28記載の方法。
- 31.前記分離が: 前記種子からの細胞の溶菌に続いて汕体フラクションを回収することを含んでい る、請求項30記載の方法。
- 32.請求項10、14または19のいずれか一つの請求項に記載の形質発現カ セットを有する植物細胞。
- 33.請求項10、14または19のいずれか一つの請求項に記載の形質発現カ セットを含有する細胞を行ずる植物。
- 34.請求項23の方法によって得られた、種子中の目的のポリペプチドを形質 発現する植物。
- 35.請求項10、14または19のいずれか一つの請求項に記載の形質発現カ セットを含有する種子。
- 36.請求項23の方法によって得られた、目的のポリペプチドを形質発現する 種子。
- 37.油体中の目的のポリペプチドを得る方法であって:種子中の前記ポリペプ チドを、融合タンパク質として、前記油体の標的化をもたらすオレオジンまたは その相当部分によって形質発現させることを含んでいる、油体中の目的のポリペ プチドを得る方法。
