により誘導された形および末端エチルエステルならびに
その生理学的に認容性の塩の形。
Description
リポジぺプチドおよびリポトリペプチド、その製造およ
び薬剤学的作用物質としてのその使用に関する。
よる誘導体、そのD−およびL−エナンチオマー、ジ−
およびトリペプチド、N−保護基により誘導された形お
よび末端エチルエステルならびにその生理学的に認容性
の塩の形は、腫瘍成長に試験管内および生体内で影響を
及ぼしかつそれを押さえ、並びにプロテインキナーゼC
を抑制するための薬剤学的作用物質として適当である。
介入するために、かつ組織炎症過程におけるホスホリパ
ーゼA2−抑制により薬理学的に作用するために適当で
ある。前記の誘導体は、低いマイクロモル(μm)の濃
度でのリポキシゲナーゼおよびシクロオキシゲナーゼを
避けることができ、その際、非特異的ヒスタミン遊離は
観察されない。リポキシゲナーゼ、シクロオキシゲナー
ゼおよびホスホリパーゼA2は、リン脂質からアラキド
ン酸カスケードの分解生成物を生じる;このようなアラ
キドン酸代謝物は炎症的に作用し、かつ例えば気管支喘
息、枯草熱、筋肉リウマチ等の病原論もしくは病理生理
学的過程の維持に決定的に関与している。長い脂肪族基
を有する中性および/または塩基性のアミノ酸からのア
ミノ酸および短鎖ペプチドは、しばしば両親媒性特徴を
有する。両親媒性物質は、親水性および疎水性の分子領
域が同時に存在することにより優れている。これらは、
従って、膜構成成分との構造的な親和性を示す。式:H
−Orn−NHR.2HBr(R=C16H33、C
18H37)を有するリポアミノ酸および式:H−Orn−
Orn−NHC18H37.3HClを有するリポジペプチ

リーフレンダー(M.Lieflaender)により、Z.Naturforsc
h.B.,Chem.Sci.46(8)1098〜1104(1991)中に記載されて
いる。しかしながら、これらの化合物を抗増殖および炎
症抑制特徴を有する薬剤学的作用物質として使用するこ
とは公知でない。
em.Abstr.Vol.73 120904 b(1970))中には、Z−L−L
ys(BOC)−L−Lys(BOC)−L−Pro−
NH−nC16H33の製造が記載されている。しかしなが
ら、相当する薬剤学的使用は示されていない。PCT出
願 国際特許(WO)第8806885号明細書中に
は、とりわけ、H−D−Pro−NH−C16H33の製造
およびホスホリパーゼA2−抑制剤としての使用が記載
されている。これらのまたは使用された化合物の一次腫
瘍の成長に対する抗増殖特徴および抑制作用に関する言
及はなされていない。

る試験において明らかになった。アミノ酸残基Rは、殊
に、Gly、Ala、Val、Leu、Ile、Se
r、Thr、Lys、Arg、Asp、Asn、Gl
u、Gln、Cys、Met、Phe、Tyr、Pr
o、Trp、His、Orn、Thzを表す。例えば、
アミノ酸残基Orn、Lys、Thzは、殊に有利であ
る。個々のアミノ酸残基に関する略語は、アミノ酸の慣
用名に基づくもので、かつ当業者に公知である。アミノ
酸は、主に、L型から由来するが、同様に、D型からの
アミノ酸も有効である。ジ−およびトリペプチド基を生
じるためのアミノ酸の結合は、塩基性アミノ酸、例えば
オルニチンおよびリジンにおいて、α位または末端ω位
を介して行なうことができる。

合物は、非抗源性の部分的に加水分解されたゼラチンを
用いて粘性の液体の形に変えられるか、または薬理学的
に認容性の固体担持物質で吸収されうる。作用物質は、
薬理学的に認容性の液体担持物質中の懸濁液の形でも投
与される。更に、ゲルまたは懸濁液の製造は、水性また
は非水性の薬理学的に認容性の液体担持物質、保存剤ま
たは界面活性剤と共に、非抗源性の保護ヒドロコロイ
ド、例えばナトリウムカルボキシメチルセルロース、ポ
リビニルピロリドン、アルギン酸ナトリウム、ゼラチ
ン、ポリガラクツロン酸または一定のムコ多糖を用いて
も可能である。このような調合物の例は、慣例の薬理学
的ハンドブック、例えばレミトンズ・ファルマツォイテ
ィカル・サイエンシイズ(Remintons's Pharmaceutical
Sciences,Mack Publishing Co.,Easton,Pa.,1975)中
に見られる。遅延作用物質放出を有する長時間作用する
調合物は、薬理学的に認容性の被覆材料、例えばゼラチ
ン、ポリビニルアルコールまたはエチルセルロース中で
のマイクロカプセル化によっても得られる。被覆材料の
ための他の例およびマイクロカプセル化法に関しては、
ヘルビッヒ(J.A.Herbig)、「エンサイクロペディア・
オブ・ケミカル・テクノロジー」(“Encyclopedia of
Chemical Technology”,Bd.13,2.Aufl.,Wiley,ニューヨ

性は、前記した固体調合物、例えば一定の僅かに水溶性
の塩または固体担持物質との塩の懸濁液または吸収体、
例えばエチレングリコールメタクリレートまたは同様に
架橋されたモノマーのポリマーの中性ヒドロゲル中の分
散液を;(米国特許(US−PS)第3551556号
明細書参照)前記作用物質量を遊離するペレットの形に
調製することである。これらのペレットは、皮下または
筋内に移植することができる。
化合物は、水溶液またはエマルション基礎中での懸濁液
として調製することができる。水を基礎とする懸濁液
は、湿潤剤、例えばポリエチレンオキシドおよびアルキ
ルフェノールの縮合生成物、脂肪族アルコールまたは脂
肪酸、および懸濁剤、例えば親水性コロイド、例えばポ
リビニルピロリドンを用いて製造される。エマルション
を基礎とする懸濁液は、湿潤剤および懸濁剤を用いてエ
マルション基礎中でペプチドを懸濁することにより製造
され、その際、前記した乳化剤が使用される。懸濁液
は、付加的に緩衝剤および/または甘味料、芳香剤、染
料、保存剤および酸化防止剤を含有していてもよい。
使用は、滅菌水溶液中で有利である。保存剤のための例
は、p−ヒドロキシ安息香酸メチル−および−プロピル
エステルであり、これらは、他の溶解性成分、例えば、
等張溶液の製造に十分な量の塩化ナトリウムまたはグル

結晶化が起きる。
c)))−NHC18H37 使用量: Boc−Lys(L−Lys(Z−Lys(Boc)))−OSu 4.84g(5.0mmol) オクタデシルアミン 1.35g(5.0mmol) 実施:スクシンイミドエステルをジオキサン50ml中
に溶かし、かつCHCl330ml中のアミンの溶液に
加える。バッチは、11時間後にゲル化する。CHCl
3を添加しかつ40℃まで加温することにより、再び溶
液が得られる。溶剤を除去し、かつ残分をジイソプロピ
ルエーテルを用いて結晶化させる。
37.4HBr 使用量: Boc−Lys(Z−Lys(Z−Lys(Boc)))−NHC18H37 2.24g(2.0mmol) HBr/氷酢酸 15.8ml 化合物に、冷たい33%HBr/氷酢酸を、4℃で注ぎ
入れ、かつ10℃で2〜3時間、湿気を断って撹拌す
る。ヒドロブロミドは、大抵、既に反応の間に、ほとん
ど完全に沈殿する。ジエチルエーテル(DE)の添加後
に、生成物を吸引濾過し、かつDEでよく洗浄する。粗
生成物はまだ強い臭化ベンジルの臭いがし、かつ粗生成
物を直ぐに数回再結晶させる。生成物が酢酸−ジエチル
エーテル−混合物中に溶解性である場合、溶剤を水流真
空で除去し、かつ残分を僅かなエタノール中に取る。D
EまたはEEの添加により、生成物を沈殿させることが
できる。メタノール/EEからの再結晶により精製す
る。
l 当モル量のトリエチルアミン中のBoc−D−Lys−
OHと、DMF中のBoc−D−Lys(Boc−D−
Lys(Boc))−OHとを反応させて、保護された
ジペプチドBoc−D−Lys(Boc−D−Lys
(Boc))−OHを得る。これを、DPPAおよび1
0%過剰のオクタデシルアミンとTHF中で結合させ
て、Boc−D−Lys(Boc−D−Lys(Bo
c))−NH−C18H37にする。シリカゲルでのカラム
クロマトグラフィーにより精製された生成物を、THF
/ジイソプロピルエーテルから晶出させる。Fp.:7
0〜71℃。TFAを用いて保護基を離脱後に、ジペプ
チドアミドのトリヒドロクロリドを、エーテル中のHC
lを用いて、エタノールから沈殿させ、かつ次いでエタ
ノールから再結晶させる。

- 【請求項1】 抗増殖および炎症抑制特徴を有する薬剤
学的作用物質としての、一般式: CH3−(CH2)n−NH−R [式中、n=11〜20およびR=アミノ酸を表す]の
長鎖の非分枝鎖状アルキルアミド、そのD−およびL−
エナンチオマー、ジ−およびトリペプチド、N−保護基
により誘導された形および末端エチルエステルならびに
