Claims (1)
- 【特許請求の範囲】 1. 生産細胞が、ベタインの異化に関与する遺伝子に関して、少なくとも1 つの改変を発現することを特徴とする、生産条件下で、代謝産物および/または 酵素を生産する細胞を培養することによって該代謝産物および/または酵素を生 産し、次いで、該代謝産物および/または酵素を回収する微生物学的方法。 2. 改変が、1つまたは複数塩基の置換および/または挿入および/または 欠失、および/または破壊であり、そして細胞が、ベタインを徐々に分解するこ とを特徴とする、請求の範囲1記載の方法。 3. 改変が、細胞分裂上安定であり、および/または非可逆的であることを 特徴とする、請求の範囲2記載の方法。 4. 改変が、遺伝子のコーディング部分、および/または該遺伝子の発現お よび/または転写調節に関与する領域に作用することを特徴とする、請求の範囲 2または3記載の方法。 5. ベタインの異化に関与する少なくとも1つの遺伝子が、増幅され、そし て結果として細胞が、ベタインをより急速に分解することを特徴とする、請求の 範囲1記載の方法。 6. 細胞が、シュードモナス(Pseudomonaas)、ストレプトミ セス(Streptomyces)、アクチノミセーテス(Actinomyc etes)、プロピオニバクテリウム(Propionibacterium) 、コリネバクテリウム(Corynebacterium)、バチルス(Bac illus)、エシェリヒア(Escherichia)、サルモネラ(Sal monella)、リゾビウム(Rhizobium)、アグロバクテリウム( Agrobacterium)、ロドシュードモナス(Rhodopseudo monas)、キサントモナス(Xanthomonas)、クロストリシウム (Clostridium)およびメタノバクテリウム(Methanobac terium)属の細胞から選択されることを特徴とする、請求の範囲1〜5記 載の方法。 7. 代謝産物が、アミノ酸、ビタミン、抗生物質、それらの誘導体 もしくはそれらの前駆体から選ばれることを特徴とする、請求の範囲1〜6記載 の方法。 8. 代謝産物が、コバラミン、好ましくは、ビタミンB12であることを特 徴とする、請求の範囲7記載の方法。 9. 細胞が、代謝産物および/または酵素を生産する組み換え細胞であり、 そして細胞が、ベタインの異化に関与する遺伝子に関して、少なくとも1つの改 変を発現することを特徴とする細胞。 10.ベタインの異化に関与する遺伝子に関して少なくとも1つの改変を発現 し、代謝産物および/または酵素を生産する細胞の、代謝産物および/または酵 素の生産のための使用。
