Claims (1)
- 【特許請求の範囲】 1.実施例欄の後の最初の頁に記載された式[式I上の種々の基(X、Yお よびR1)は次のように定義される:すなわち、 XとYは水素、ハロゲン及びC1−C6アルコキシから独立的に選択される; R1は窒素、酸素及び硫黄から選択される1〜3個のヘテロ原子を含む五員お よび六員ヘテロアリール環、並びにそれらのベンズ誘導体から選択され、当該ヘ テロアリール環、並びにそれらのベンズ誘導体は、C1−C6アルキル、ヒドロキ シ、トリフルオロメチル基を有しうるC1−C6アルコキシ、C1−C6アルコキシ カルボニル、C1−C6ヒドロキシアルキル、ベンジルオキシ、ハロ、C1−C3ア ルキルアミノカルボニルC1−C3アルキル、式:CONRcRd(式中、RcとRd は水素、2−ピロリジニル及びC1−C6アルキルから独立的に選択されるか、又 はRcとRdはそれぞれが結合する窒素原子と共に、前記窒素原子が唯一のヘテロ 原子である五員又は六員複素環を形成する)を有するアミノカルボニル、RcS (O)n、RfNH及びRgS(式中、ReとRfは水素及びC1−C6アルキルから 独立的に選択され、nは0、1又は2であり、RgはC1−C3アルキルカルボニ ルアミノフェニル及びジ(C1−C3)アルキルアミノ(C1−C6)アルキルから 選択される)から選択される1〜2個の置換基を有していてもよく、五員および /または六員ヘテロアリール環上の置換基の少なくとも1つが式:C1−C4アル キルSOを有するキラルスルホキシドである] で示される化合物、およびその薬剤学的に受容される塩。 2.XとYがHから選択され、R1が2−エチルスルフィニル−3−ピリジ ルから選択される請求項1記載の化合物と、その薬剤学的に受容される塩。 3.(−)−1−(9,10−ジヒドロ−9,10−メタノアントラセン− 9−イルメチル)−4−(2−エチルスルフィニル−3−ピリジル)ピペリジン −4−オールから選択され、当該化合物が結晶質であることを特徴とする請求項 1記載の化合物、およびその薬剤学的に受容される塩。 4.当該結晶質化合物が185〜186℃の融点と、[a]−106°(c =1.0,MeOH中)の比旋光度とを有し、非吸湿性であることを特徴とする 請求項3記載の化合物。 5.(−)−1−(9,10−ジヒドロ−9,10−メタノアントラセン− 9−イルメチル)−4−(2−エチルスルフィニル−3−ピリジル)ピペリジン −4−オールから選択され、水性及び非水性媒質中で良好な平衡溶解度を有し、 溶液中で化学的及び立体化学的安定性を有することを特徴とする請求項1記載の 結晶質化合物。 6.請求項1記載の化合物と、薬剤学的に受容される担体または希釈剤とを 含む薬剤組成物。 7.(−)−1−(9,10−ジヒドロ−9,10−メタノアントラセン− 9−イルメチル)−4−(2−エチルスルフィニル−3−ピリジル)ピペリジン −4−オール又はその薬剤学的に受容される塩から選択される結晶質化合物と、 薬剤学的に受容される担体または希釈剤とを含む薬剤組成物。 8.請求項1〜5のいずれかにおいて定義した、式I化合物又はその薬剤学 的に受容される塩の製造方法であって、式I化合物(式中、XとYは請求項1〜 5のいずれかで定義した通りであり、R1は請求項1〜5のいずれかで定義した 通りであるが、但し、式:C1−C4アルキルSOの前記の基の全体すなわちこれ らの基の各々は式:C1−C4アルキルSである)をキラル酸化方法によって酸化 して、式:C1−C4アルキルSを有する前記基を式:C1−C4アルキルSOを有 するキラルスルホキシドに鏡像異性体選択的に酸化することによって、請求項1 〜5のいずれかで定義した通りの式I化合物を製造することを含み; 薬剤学的に受容される塩が必要である場合には、生理的に受容されるアニオン を形成する酸又は生理的に受容されるカチオンを形成する塩基によって式I化合 物を処理することを含む前記方法。 9.請求項1〜5のいずれかにおいて定義した、式I化合物又はその薬剤学 的に受容される塩の製造方法であって、下記工程: (a)式IIを有するピペリドンを非プロトン性溶媒中で式:R1Liの対応 化合物によって処理する工程; (b)式IIaの対応アミドを適当な還元剤によって処理する工程; (c)式III(式中、Gは水素である)のアルデヒドを還元剤の存在下で式 IVの対応ピペリジンによって処理する工程;または (d)X又はYのいずれかの意味がC1−C6アルコキシであることが好ましい 式I化合物に関しては、式I(式中、X又はYがヒドロキシである)の対応化合 物を好ましい塩基及びアルキルハライドによって処理して、アルコキシ化合物を 形成する工程と、 上記工程後に、工程(a)、(b)、(c)又は(d)において得られた化合物 を鏡像異性体選択性の酸化剤によって処理する工程と を含み; 薬剤学的に受容される塩が必要である場合には、生理的に受容されるアニオン を形成する酸又は生理的に受容されるカチオンを形成する塩基によって式I化合 物を処理することを含む前記方法。 10.酸化剤をチタン/タルトレート/過酸化物の混合物とキラルオキサジ リジンとから選択する請求項8又は9に記載の方法。 11.結晶質(−)−1−(9,10−ジヒドロ−9,10−メタノアント ラセン−9−イルメチル)−4−(2−エチルスルフィニル−3−ピリジル)ピ ペリジン−4−オールの製造方法であって、1−(9,10−ジヒドロ−9,1 0−メタノアントラセン−9−イルメチル)−4−(2−エチルチオ−3−ピリ ジル)ピペリジン−4−オールを乾燥されていないトルエンまたは同等な溶媒に 溶解したジエチル D−タルトレートの冷却溶液、チタンテトライソプロポキシ ド及びt−ブチルヒドロペルオキシドによって処理して、鏡像異性体的に富化し たエチルスルフィニル混合物を形成し、これを次に例えばMeCN又は同等物の ような、適当な溶媒中で再結晶して、純粋な(−)鏡像異性体を形成することを 含む前記方法。 12.薬剤に用いるための結晶質化合物(−)−1−(9,10−ジヒドロ −9,10−メタノアントラセン−9−イルメチル)−4−(2−エチルスルフ ィニル−3−ピリジル)ピペリジン−4−オールまたはその薬剤学的に受容され る 塩。 13.治療を必要とするヒトにおける精神医学的障害の治療用薬剤の製造の ための結晶質化合物(−)−1−(9,10−ジヒドロ−9,10−メタノアン トラセン−9−イルメチル)−4−(2−エチルスルフィニル−3−ピリジル) ピペリジン−4−オールまたはその薬剤学的に受容される塩の使用。 14.治療を必要とするヒトにおけるD1/D2ドパミンアンタゴニストと して及び5HT2セロトニンアンタゴニストとしての結晶質化合物(−)−1− (9,10−ジヒドロ−9,10−メタノアントラセン−9−イルメチル)−4 −(2−エチルスルフィニル−3−ピリジル)ピペリジン−4−オールまたはそ の薬剤学的に受容される塩の使用。 15.ヒトにおける精神病の治療におけるD1/D2ドパミンアンタゴニス トとして及び5HT2セロトニンアンタゴニストとしての結晶質化合物(−)− 1−(9,10−ジヒドロ−9,10−メタノアントラセン−9−イルメチル) −4−(2−エチルスルフィニル−3−ピリジル)ピペリジン−4−オールまた はその薬剤学的に受容される塩の使用。
