Claims (1)
- 【特許請求の範囲】 1. 疾病の治療のための薬物輸送システムを構築する方法であって、薬物あ るいはその他の物質を生体適合性のマトリックス内に固定化することを含み、こ のとき、このマトリックスは、投与した際に該マトリックスの周囲に存在する生 体物質に対する少なくともひとつのレセプターを含有しており、該レセプターは マトリックス内に固定されているが、生物学的活性を保持しており、該生体物質 の量に対応して活性化され、マトリックスの立体構造を変化させ、薬物を可動化 し、周囲にのみ放出するようになっていることを特徴とする方法。 2. 薬物あるいはその他の物質が天然に存在する生物学的物質であることを 特徴とする請求項1に記載の方法。 3. 生物学的物質がホルモンであることを特徴とする請求項2に記載の方法 。 4. ホルモンがインシュリンであることを特徴とする請求項3に記載の方法 。 5. レセプターが結合性高分子であることを特徴とする請求項1〜4のいず れか1項に記載の方法。 6. 結合性高分子がレクチンであることを特徴とする請求項5に記載の方法 。 7. レクチンが、タチナタマメレクチンのコンカナバリンAであることを特 徴とする請求項6に記載の方法。 8. レクチンが、エンドウレクチンであることを特徴とする請求項6に記載 の方法。 9. レセプターが生体物質と可逆的に結合することを特徴とする請求項5〜 8のいずれか1項に記載の方法。 10. 生体物質が炭水化物であることを特徴とする請求項1〜9のいずれか1 項に記載の方法。 11. 炭水化物がクルコース、フルクトースまたはマンノースであることを特 徴とする請求項10に記載の方法。 12. 生体物質が炭水化物重合体であることを特徴とする請求項10に記載の方 法。 13. 炭水化物が分岐鎖デンプン、マンナン、レバン、デキストランまたは合 成炭水化物であることを特徴とする請求項12に記載の方法。 14. デキストランの末端グルコース分子がレセプターに結合することを特徴 とする請求項13に記載の方法。 15. デキストランの末端グルコース分子がコンカナバリンAに結合してゲル を形成すると、薬物がマトリックス内に固定化されることを特徴とする請求項1 〜14のいずれか1項記載の方法。 16. レセプターが共有結合により、マトリックス内部に固定されることを特 徴とする請求項1〜15のいずれか1項に記載の方法。 17. 不溶性の凝集体を形成することなく、レセプターが共有結合により、マ トリックス内部に固定されることを特徴とする請求項16に記載の方法。 18. 不溶性の凝集体を形成することなく、デキストランの重合鎖に共有結合 することにより、コンカナバリンA分子がマトリックス内に固定されることを特 徴とする請求項16に記載の方法。 19. デキストランのビシナルOH基からなるシッフ塩基をヨウ化ナトリウム で酸化し、続いて還元、沈澱により精製することにより;直鎖ポリアクリルアミ ドヒドラジドにアジド結合させることにより;または直鎖ポリアクリルアミドに グルタルアルデヒド結合させることにより、コンカナバリンAをデキストランに 共有結合させることを特徴とする請求項18に記載の方法。 20. レセプターに生体物質が結合することによりマトリックスの立体構造の 変化が起こることを特徴とする請求項1〜19のいずれか1項に記載の方法。 21. 立体構造の変化によりマトリックスが非ゲル化することを特徴とする請 求項20に記載の方法。 22. pH4.0〜8.0の間の変化により、または、Mn++イオンもしくはCa++イオン の濃度の変化により影響を受けないようにシステムを構築することを特徴とする 請求項1〜21のいずれか1項に記載の方法。
